Objective To investigate the distribution characteristics and antimicrobial resistance of carbapenem-resistantAcinetobacter baumannii (CRAB),and to guide effective clinical prevention and rational antimicrobial use. Methods Data about clinically isolated CRAB between January 2009 and December 2013 were analyzed retrospec-tively,distribution and antimicrobial resistance were analyzed by WHONET 5.5 software.Results A total of 888 Acinetobacter baumannii strains were isolated,421 of which were CRAB,the isolation rate was 47.4%,the isola-tion rates in 2011 ,2012 and 2013 were all about 50.0%;CRAB strains were mainly isolated from sputum (73.4%) and mostly distributed in intensive care unit (ICU)(61 .3%),followed by neurosurgery department (12.4% ). CRAB presented highly antimicrobial resistance.Except cefotaxime and cefatriaxone,resistant rates of CRAB to the other detected antimicrobial agents(ceftazidime,cefepime,cefoperazone/sulbactam,aztreonam,imipenem,amika-cin,gentamycin,minocycline,chloramphenicol,levofloxacin,ciprofloxacin,and compound sulfamethoxazole)were all higher than non-CRAB isolates(all P ≤0.01),Compared with non-CRAB isolates,The resistant rate of CRAB to cefoperazone/sulbactam was the lowest(<15%),followed by minocycline,resistant rates to other antimicrobial agents were all >80.0%.Conclusion Surveillance of CRAB should be further strengthened.It is necessary to fo-cus on the control and prevention of healthcare-associated infection in ICU patients and respiratory system.

Objective HCV NS3 protein plays an important role in disease caused by HCV. We investigate the gene expression of HCV NS3 in yeast for future study of the function of the protein. Methods PCR was performed to amplify the gene of HCV NS3 from the plasmid pBRTM/HCV containing the whole fragment of HCV and the gene was cloned into pGEM T vector. Thereafter, HCV NS3 gene was cut from pGEM T vector and cloned into yeast expression plasmid pGBKT7, and recombinant pGBKT7∶NS3 was transformed into yeast AH109. The yeast protein was isolated and analyzed with sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE) and Western blotting. Results HCV NS3 gene was successfully cloned into pGBKT7. The results of SDS PAGE and Western blotting assay showed that the molecular weight of the expressed product was about 22000 Da and HCV NS3 protein was existed within yeast cells.Conclusions HCV NS3 was successfully expressed in yeast expression system.

The nonstructural protein 5A (NS5A) of the hepatitis C virus (HCV) has been shown to interact with a variety of cellular proteins and implicated in the regulation of cell growth, interferon resistance, and other cellular signaling pathways. Using the yeast-two hybrid method, we have isolated a clone that encodes a novel NS5A--associated binding protein: NS5ABP37. Reverse transcription polymerase chain reaction (RT-PCR) method was employed to amplify the full fragment,and the plasmid pGADT7-NS5ABP37 with the Saccharomyces cerevisiae vector pGADT7 was constructed. To prove the interaction, yeast cell Y187 transformed with pGADT7-NS5ABP37 was mated with yeast cell AH109 containing pGBKT7-NS5A to verify the interaction between the novel protein coded by the new gene NS5ABP37 and NS5A.

Objective To investigate the effect of bFGF on the proliferation of olfactory epithelium ensheathing cells. Methods With BrdU incorporation method,the effect of bFGF on the proliferating rate of olfactory epithelium ensheathing cells was observed in the research. Results The olfactory epithelium ensheathing cells can proliferate without any proliferating factor.The bFGF(10?g/L)can enhance the proliferating rate in a moderate way.In this experiment BPE(bovine pituitary extract) can not enhance the stimulating effect of bFGF.Conclusion The bFGF(10?g/L)can stimulate the proliferating rate of the olfactory epithelium ensheathing cells in vitro.

Objective Studying the expression of the Nogo(N\|18) in olfactory ensheathing cells(OECs) to investigate the functional relationship between Nogo and the OECs in promoting the regeneration of neurite axon. Methods The expression of the Nogo(N\|18) in the primary cultured ensheathing cells from olfactory bulb of the adult rats was studied with the method of immunocytochemical staining and double\|immunofluorescence staining examed under the confocal laser scanning microscope. Results The Nogo\|A protein was located in primary cultured ensheathing cells from the olfactory bulb.The protein was mainly distributed in the cytoplasm.The member and processes were less stained.Conclusion\ The ensheathing cells in vitro contain Nogo protein.It suggests that Nogo protein is not a depressive factor in the axon regeneration of olfactory system.\;[

Objective To investigate the clinical features of patients with multi-cranial nerves impairments as the onset of Guillain-Barré syndrome (GBS). Methods 10 patients of GBS with complaint of multiple cranial neuropathy were analyzed retrospectively. Results The cranial nerves Ⅶ, Ⅸ and Ⅹ were involved at the onset of GBS, tending to affect men rather than women (4∶1), aged of 18～55 years old (8/10), and with less the antecedent of infection. The knee and ankle jerk reflexes were minimal or absent in all the patients, and the meningeal irritation signs were observed in 4 patients. Assisted ventilation was required in 4 patients during the course of their illness. The cerebrospinal fluid (CSF) characterized with increased protein concentration but a normal cell count in 2 patients in the first week, and all the patients in the following 3 weeks. The incidence of motor conduction velocity (MCV) and F waves abnormalities of electrophysiological evaluation were 81.25% and 94.44% respectively. The Hughes scales were (4.00±0.82) before treatment, and were (2.25±0.96, P=0.012) and (0.50±1.00, P=0.000) 14 d and 28 d after intravenous immunoglobulin (IVIG). Conclusion The probability of GBS should be considered in patients with multiple cranial neuropathy, especially the cranial nerves VII, IX and X impairments without precise causes. The early electrophysiological studies and CSF examinations may be useful for diagnosis. IVIG can be preferred as an effective treatment.

The effects of subclinical epileptiform discharges (SEDs) on children with cerebral palsy cannot be ignored. Data from neurodevelopmental clinic studies showed that the overall incidence of SEDs in cerebral palsy was 18%～40%, with the highest in spastic hemiplegia and diplegia. The major pattern of SEDs was focal and multifocal, and was usually found in centro-temporal and parietal regions. The cortex impairment and other complications were risk factors related to SEDs in cerebral palsy. Paroxysmal or frequent long-time SEDs, with the Results of transient or chronic cognitive impairment, have been found to lead to subsequent death of cortical neurons of cerebral palsy patients thereby worsening their prognosis. Valproic acid (VPA), benzodiazepines (BZs) and lamotrigine (LTG) have a role in inhibiting SEDs, while adrenocorticotropic hormone (ACTH) and glucocorticoids have a great role in it.

Objective To investigate the characteristics of pathogens and risk factors of the catheterassociated infection (CAI) in emergency ICU (EICU) in order to design an appropriately therapeutic strategy for the future.Methods From January 2008 to December 2010,a total of 1363 patients were enrolled for this retrospective study.Blood sample taken from the vein with indwelling catheterization and the tips of catheters cut in 5 cm after withdrawn from the veins in 1363 patients were collected for bacterial culture.Results Of 1363 catheters,pathogens were found in 147 (10.79％) after venous catheterization.The daily occurrences of CAI were 3.05 ones per 1000 catheters.Of 147 cases of infection,46.94％ pathogens were gram-negative bacilli,40.14％ gram-positive cocci,and 12.92％ fungi.Unconditional Logistic regression analysis suggested that repeated catheterization,femoral vein catheterization,the application of multi-lumen catheter and long-term indwelling catheterization were the independent risk factors responsible for CAI.Conclusions The risk factors responsible for catheter related infections should be controlled to prevent the occurrence of nosocomial infection.

Objective To evaluate the clinical effect of IPSe. max press ceramic endocrowns being used in preserving endodontically treated molars. Methods Forty-five endodontically treated molars were selected from 45 patients in this study. All the molars had been endodontically treated and performed with ultrasonic scaling. After 1-2 weeks, the crowns and pulp chambers were prepared. The impressions and master casts were gained and IPSe. max press ceramic endocrowns were fabricated in the lab. The endocrowns were cemented with resin cement. The further consultations were offered to these molars after 6 months and 1 year. This study investigated the expression of hemeoxygenase-1 (HO-1) in rats with acute lung rejection and its implication. A valid rat orthotopic left lung transplantation model (SD rat-->Wistar rat) was established by using an improved three-cuff anastomosis technique. The rats were divided into control group, CoPP (HO-1 inducer)-treated group and ZnPP (HO-1 inhibitor)- treated group. The severity of acute rejection was graded on the basis of the morphologic changes of the lung samples stained with HE. The expression of HO-1 protein in lung tissue was detected by using immunohistochemistry and Western blot, and HO-1 mRNA activity was assayed by RT-PCR. The results showed that the expression of HO-1 protein was significantly increased with the acute rejection grading in rats (P<0.01). As compared with control and ZnPP-treated groups, the severity of acute rejection was not alleviated and the grade not reduced significantly in CoPP-treated group (P>0.05). It was concluded that HO-1 protein might be involved in the pathological process of post-graft acute rejection. The expression of HO-1 protein was increased gradually with aggravation of acute rejection, and HO-1 protein might be used as an index to monitor acute rejection after lung transplantation.
