1.MicroRNAs and esophageal squamous cell carcinoma
Journal of International Oncology 2016;43(11):858-860
MicroRNAs (miRNAs)can influence the proliferation, apoptosis, invasion and metastasis of esophageal squamous cell carcinoma (ESCC)cells by signal transduction,epithelial-mesenchymal transi-tion,angiogenesis and other regulating mechanisms.Specific serum miRNAs can serve as novel diagnostic and prognostic biomarkers of ESCC.Recent studies have indicated that miRNAs can improve the radiation sensitivi-ty and even reverse multi-drug resistance of ESCC,holding great clinical value.
2.Cost-effectiveness evaluation of the strategies for prenatal diagnosis of Down's syndrome
Chinese Journal of Hospital Administration 1996;0(07):-
35) and the third strategy of providing diagnosis to those tested positive at screenings in mid-term pregnancy could, respectively, lead to the diagnosis of 0.93 and 1.47 cases of DS per 10 thousand pregnant women, but they could also induce, respectively, 1.58 and 0.28 cases of miscarriage of normal fetuses due to diagnosis procedures. The costs of one DS diagnosis were, respectively, 108890.98 yuan and 443683.11 yuan. Conclusion Taking into consideration both cost-effectiveness and safety factors, priority ought to be given to the third strategy.

4.Purification of Trypsin by Using AOT/Isooctane Reverse Micellar Extraction and Overcoming Denaturation and Precipitation in the Process
China Biotechnology 2006;0(01):-
Trypsin was purified from crude material of pig pancreas by AOT/isooctane reversed micellar system. The influence of the main operating parameters such as ethanol concentration in forward and backward extraction,pH,KCl and AOT concentration, temperature were investigated.Forward and backward extraction recovery of trypsin reached almost 90% and neared 100%,respectively.Finally,about 88% of total yield was obtained, and the specific activity of trypsin was increased to over 1800U/mg with purification factor of 5 folds more.In AOT-isooctane reverse micellar extraction system, denaturation or precipitation of proteins always occured due to strong electrostatic interaction between AOT-proteins molecules. It had been resolved by adding ethanol into reverse micellar system, and no denaturation was observed.Otherwise, the phase separation time was shortened significantly because of ethanol added. It was only 10 minutes or less to reach phases separation after forward and backward extraction. If this method can be applied in industry, efficiency will be greatly improved.

Objective To investigate the peripheral blood Th17 and CD4 + CD25 + regulatory T cell levels and their correlations in patients with primary hepatocellular carcinoma (PHC).Methods Peripheral blood samples were collected from 30 PHC patients and 25 healthy controls in the First Affiliated Hospital of Zhejiang University from June 2008 to May 2009.Mononuclear cells were isolated and the Th17 and CD4 + CD25 + regulatory T cells were detected by flow cytometry and compared between patients and controls by t test.Spearman test was performed to analyze the correlation of Th17 with CD4 + CD25 +regulatory T cell concentrations. Results The levels of Th17 and CD4 + CD25 + regulatory T cells in peripheral blood in healthy controls were (2.10 ± 0.87) % and (7.10 ± 2.32) % ; while those in PHC patients were (3.38±1.68)% and (11.78±5.62)% (t=3.640 and 4.162,P<0.01).The level of Th17 cells was positively associated with that of CD4 + CD25 + regulatory T cells in PHC patients (r =0.821,P <0.01).Conclusion The levels of Th17 and CD4 + CD25 + regulatory T cells in peripheral blood are high in PHC patients and positively correlated with each other, which indicates that CD4 + CD25 + regulatory T cells may contribute to the disease progression and pathogenesis of carcinoma through inducing Th17 cells differentiation.

0.05).There was a positive correlation between the tempo of the engraftment and the progressive increase of the doses of MNC in the range of (3~5.99)?108/kg,but which was not found in the range of 6?108/kg.Conclusion These results indicate that MNC taken as an index for the measurement of the hematopoietic stem cell/progenitor content can reliably predict hematopoietic reconstitution after both HLA-matched and-mismatched sibling donor peripheral blood stem cell transplantation, with the rate and tempo of engraftment comparable with that in CD34+ cell group, and however, the target dose was reached with one apheresis in all 83 donors in MNC group(100%),as compared with 14 donors in CD34+ cell group(37.84%),suggesting that MNC may replace CD34+ cell as an independent index for the assessment of the stem cell/progenitor content in clinical practice.
