1.Enhancement of cytotoxic effect of epirubicin on gastric carcinoma cell line SGC-7901 by survivin antisense RNA and signaling pathway

Objective:To investigate whether interfering survivin has the potential to sensitize stomach cancer cells to Epirubicin.Methods:By the method of liposome transfection,survivin antisense oligodeoxynucleotide was transfected into human stomach cancer SGC-7901 cell line.Treat cells with different concentrations of survivin ASODN and Epirubicin respectively to select the best combination trough assessing the growth of cells by MTT assay.Cells were randomly divided into control group,ASODN group,Epirubicin and combination group.Western blot analysis was used to check the survivin protein expression;inhibitive rate were detected by MTT assay;Flow cytometric analysis was used to detect the cycle status of SGC-7901 cells.The apoptosis was screened according to the result of Annexin V-FITC.Reverse transcription polymerase chain reaction(RT-PCR)were applied to detect the expression of bcl-2、SMac mRNA.Results:MTT assay showed the proliferation of SGC-7901 cells was retarded obviously contrasting to control group.The level of survivin protein expression in ASODN group are lower than control group according to Western blot analysis.Annexin V-FITC show that the apoptosis rate of ASODN group cells increases.The level of SMac mRNA expression in ASODN group are higher than control group according to RT-PCR analysis.Conclusion:Survivin siRNA can sensitize stomach cancer cells to Epirubicin.Up-regulating the SMac expression might be one of mechanisms.
2.Formulation and preparation of glycyrrhizic acid solid lipid nanoparticles

Objective To prepare the glycyrrhizic acid solid lipid nanoparticles(GL-SLN).Methods GL-SLN was prepared by conventional rotary-evaporated film-ultrasonication method.Based on the single-factor experiment,GL-SLN was prepared using orthogonal design for optimization of formulation and technology.The obtained GL-SLN was also evaluated.Results GL-SLN had even and regular appearance and the average diameter was 75.8 nm.Zeta potential was-19.7 mV.The drug loading was 8.27% and the entrapment ratio was 91.76%.Conclusion The entrapment ratio of GL-SLN and the stability of GL-SLN prepared by conventional rotary-evaporated film-ultrasonication method are high and good.
3.Effect of human factor Ⅸ intron 1 inserted in forward orientation into retroviral vectors on expression of factor Ⅸ in muscle cells

Objective:To investigate the effects of human factor Ⅸ (FⅨ) intron 1 inserted in forward orientation into retroviral vector on the expression of FⅨ in muscle cells. Methods: Two FⅨ mini-genes, FⅨm1 and FⅨm2, were inserted in forward orientation into retroviral vector backbone, LⅨSN, constructing 2 vectors named LⅨm1SN and LⅨm2SN, containing shortened intron of 1.4 and 0.3 kb, respectively. Retrovirus was made by transfecting PA317 packaging cells. Transient and stable expression of FⅨ was tested in SCID mouse muscle cells (SCID-MB). Results: The vector titers and expression levels of LⅨm1SN and LⅨm2SN in SCID-MB were 1 to 2 folds higher than that of LⅨSN. Some SCID-MB colonies transfected with LⅨm2SN contained intact FⅨm2 inton elements, suggesting that splicing of virus mRNA may not be complete in packaging cells. Conclusion: FⅨ intron 1 inserted in forward orientation in retroviral vectors can increase retroviral titer and the expression level of FⅨ in muscle cells.
5.Progress of studies on molecular immunology of human platelets.

In autoimmune thrombocytopenic purpura (AITP) specific autoantibodies bind platelet GP via their Fab fragments. Both splenic CD5+ B and CD5- B cells produce platelet glycoprotein-specific antibodies. There is limited number of antigenic determinants, and the GP-specific autoantibodies are derived from a restricted number of B-cell clones in chronic AITP. Blocking co-stimulatory signals could induce platelet-specific T cell anergy. MMF could be used as a second line agent for the treatment of steroid-resistant AITP. Detection of plasma thrombopoietin levels play an important role in the differentiation of thrombocytopenic states caused by platelet destruction or due to bone marrow hypoplasia. Endogenous TPO level is also important on the differential diagnosis of ET and RT. Quinine- or heparin-dependent antibodies could induce thrombocytopenia. PCR-SSP is useful for the genotyping of the platelet-specific alloantigen HPA. Biotinylated platelets have an impaired response to agonists as evidenced by in vitro platelet aggregation tests.

6.Optimization of the Preparation Technology of Berberine Hydrochloride Solid Lipid Nanoparticles by Orthogonal Experiment

OBJECTIVE: To prepare Berberine hydrochloride solid lipid nanoparticles(BH-SLN). METHODS: BH-SL-N was prepared by conventional rotary-evaporated film-ultrasonication method. Based on the single-factor experiment, BH-SLN was prepared using orthogonal design for optimization of formulation and technology. The quality of the obtained BH-SLN was evaluated. RESULTS:BH-SLN had even and regular appearance with a mean diameter of 60.5 nm,Zeta potential of —29.7 mV,drug loading of 8.69%, and entrapment ratio of 97.58%. CONCLUSION: The entrapment ratio of BH-SLN is high and the stability is good for BH-SLN prepared by conventional rotary-evaporated film-ultrasonication method, and the method is reliable.
7.Application of Functional Magnetic System in osseous deformity

BACKGROUND:Osseous deformity is one of the most common Jaw deformities in clinic, which give patients serious impact on functionality and aesthetics, so patients want to cure it strongly. Recently, Functional Magnetic System has played an increasingly important role in correcting osseous deformity because of its advantages, so it is gradual y known by people.
OBJECTIVE:This article wil give a summary of Functional Magnetic System in the progress and deficiencies, in order to have a greater development in the field of correct osseous deformity.
METHODS:We searched the PubMed database, from January 1990 to June 2013 with the key words of“magnetic, orthodontics”in English. An online search of CNKI database, Wanfang database, Chongqing VIP database from January 1990 to June 2013 was also conducted with the key words of“magnetic, orthodontics”in Chinese.
RESULTS AND CONCLUSION:Functional Magnetic System is the combination of magnetic force and the traditional function correcting device, with the aid of attraction and repulsion, we could make up for the inadequacy in the treatment of Jaw osseous deformity, including the problems of length, width and height. With the development of magnetic materials and the research of magnetic materials, Functional Magnetic System wil have a bigger development.
8.Preliminary study on the influence of enhancer Ⅰ upon the immune response of HBV DNA vaccine

Objective To study the influence of ENH Ⅰ of HBV on immune response of HBV DNA vaccine. Methods DNA fragments of HBsAg and HBsAg-ENH Ⅰ region of HBV, after being amplified by PCR using the complete genome DNA of HBV adr subtype, were inserted into VR1012 vectors. The recombinant plasmids were transfected into COS-7 cells and HepG2 cells, and injected into Balb/C mice. The expression of COS-7 cells, HepG2 cells and the cellular and humoral immune response of mice were determined with Western blot, ELISA and ELSPOT. Results HBsAg was expressed by transfected HepG2 cells and COS-7 cells. The quantity of expression by HepG2 cells increased apparently when transfected by that combined with ENH Ⅰ. In transfected COS-7 cells, there was no significant difference in the expression of HBsAg between those transfected by two recombinant plasmids. The HBsAb and the HBsAg specific CTL were found in mice in the second week after immunization, and there was no marked difference between those immunized by two recombinant plasmids. Conclusions When inserted into HBV DNA vaccine, ENH Ⅰ can promote the expression of HBsAg in transfected HepG2 cells, but can not affect the expression of transfected COS-7 cells and the immune response of immunized Balb/C mice.

Chelation therapy could remove transfusional iron burden. Three chelators are currently available, including deferoxamine, deferiprone, and deferasirox, which can be used as monotherapy or in combination.Several factors must be considered in the design of optimal and individualized chelation regimens,which include chelator availability and its properties,degree of organ-specific iron loading,ongoing transfusional iron burden, and patient preference. Comparative effectiveness trials may help to determine the ideal strategy. This article reviews latest research presented at the 53rd annual meeting of the American Society of Hematology (ASH) on the use of iron chelators.

Outcomes for patients with hemophilia have improved dramatically over the past 50 years.With the increased availability of safe clotting factor concentrates,the primary focus in clinical management is now the prevention of long-term complications,most notably the debilitating hemophilic arthropathy that is associated with severe disease.Definitive evidence of improved clinical results from primary prophylaxis started in young patients-with severe hemophilia A and a minimal bleeding history is presented.Furthermore,recent studies showing benefits for initiating prophylaxis in older adolescents and adults with established joint disease are examined.lnhibitors to factor Ⅷ are the most problematic complication of factor replacement therapy.Patient-specific and treatment-related factors that contribute to the risk of inhibitor formation are discussed and controversies and clinical evidence related to approaches for tolerance induction are reviewed.Immune tolerance induction is the proven method for eradication of inhibitors.This article reviews latest research presented at the 55th ASH annual meeting on the hemophilias and their clinical management.
