1.Effect of Aqueous Extract of Ginger on Vascular Dementia in Rats

Objective To observe the therapeutic effects of aqueous extract of ginger on vascular dementia(VD) in rats.Methods Male SD rats were randomly divided into sham-operation group, model contral group, huperzine A group and three ginger aqueous extract groups for high, middle and low dosage. The VD rat model was established by bilateral corotid ligation and repeated cerebral ischemia reperfusion. Spatial learning and memory abilities in both place navigation test and spatial prob test were evaluated with Morris water maze. The activitie of acetylcholinesterase (AChE) in cortex and striatum tissue and SOD and MDA content in brain tissue was detected by spectrophotometry.Results Compared with sham-operation group, the rats in model control group exhibited serious learning and memory deficits in both place navigation test and spatial prob test and decreased significantly AChE activity in striatum tissue. Aqueous extract of ginger could not only improve the learning and memory abilities in both place navigation test and spatial prob test, but also increase AChE activity and the ratio of SOD/MDA and decrease MDA content in brain tissue. Conclusion Aqueous extract of ginger has obvious effects on improving learning and momery abilities of rats with VD. Increasing the function of cholinergic neuron and inhibiting AChE activity are likely to be the partial mechanism.
2.Effects of aspirin-niacinamide-zinc complex (WUY) on platelet aggregation and experimental thrombosis

AIM: To study the effects of aspirin-niacinamide-zinc complex (WUY) on platelet aggregation and experimental thrombosis. METHODS: With adenosine diphosphatethe (ADP), arachidonic acid (AA) and collagen, the effects of aspirin-niacinamide-zinc complex (WUY) on platelet aggregation in vitro or in vivo were investigated by Born's method. The mouse mortality caused by intravenous injection of AA and experimental thrombus formation in rats were observed. Radioimmunoassay was used for measuring thromboxane B_2 (TXB_2) and 6-keto-PGF_(1?) in plasma of rabbits. RESULTS: In high, middle and low dose groups, drugs, in vitro, inhibited ADP-, AA-and collagen-induced platelet aggregation and the effect of WUY was stronger than that of ASP in high dose groups. In vivo, WUY showed more potent inhibitory effects on AA-induced aggregation in 1 h and 3 h. WUY had a powerful inhibitory effect on mouse death as a result of pulmonary thrombi induced by AA injection into the tail vein and ED_(50) was lower than that of ASP. In addition, WUY exhibited strong inhibitory effect on thrombus formation in rat arteri-venous shunt and significantly reduced plasma level of TXB_2 while it markedly increasing 6-keto-PGF_(1?) in high dose groups and ASP significantly reduced plasma level of both TXB_2 and 6-keto-PGF_(1?). CONCLUSION: The effect of WUY on platelet aggregation and experimental thrombosis is stronger than that of ASP and can increase plasma level of 6-keto-PGF_(1?).
3.Synergistic inhibitory effects of pirarubicin combined with hyperthermia on proliferation and invasion of gastric cancer cells

Objective To investigate the synergistic effects of pirarubicin (THP) combined with hyperthermia on proliferation and invasive potential of gastrointestinal cancer cells in vitro and to discuss the underlying mechanisms. Methods Gastric cancer cell line MGC-803 was established, and they were divided into 4 groups: control group (37℃), high temperature group (43℃), drug group (37℃+THP), and drug and high temperature group (43℃+THP). After THP and hyperthermia treatment, cell proliferative ability was determined by soft agar clonogenic assay, cell nuclear antigen PCNA was detected by flow cytometry, cyclin E mRNA level was assessed by reverse transcription PCR (RT-PCR). 4.Protective effect of polypeptides from Chlamys farreri on murine thymocytes damaged by 60Co γ-rays

Objective To study the effect and mechanism of polypeptide from Chlamys farreri(PCF) on the thymocytes damaged by 60Co γ-rays. Methods The ceils were randomly divided into six groups: control group, 60Co groups (2 Gy, model),60Co + 0.5% PCF, 60Co+ 0.25% PCF, 60Co + 0.125% PCF, 60Co +0.1% VitC. The concentration of GSH-Px, ROS, A-SAC, T-AOC and the cells' viability were determined. The mitochondria membrane potential were tested. The expressions of P53, Bax and Bel-2 proteins were examined.Results The activities f GSH-Px, A-SAC, T-AOC in cells were enhanced, and the amounts of ROS were decreased by PCF. The expression of Bcl-2 gene was up-regulated, and down-regulated for the expression of P53 and Bax. All observed indexes of the PCF groups were significantly different compared with model group (P <0.05). Conclusions PCF has the protective effects on damages of thymocytes caused by 60Co irradiation. The mechanisms might be related to inhibiting of lymphocyte apoptosis and enhancing the activities of lymphocytes.
5.Comparison of routine shunting and selective shunting in patients with moderate and severe carotid artery stenosis undergoing carotid endarterectomy

Objective To compare the therapeutic effect between routine shunting and selective shunting in patients with moderate and severe carotid artery stenosis undergoing carotid endarterectomy (CEA). Methods One hundred and ninety-two patients with moderate and severe carotid artery stenosis undergoing CEA were selected, and the patients were divided into control group (routine shunting) and observation group (selective shunting) according to the random digits table method with 96 cases each. The intraoperative carotid artery occlusion time and incidences of stroke event 30 d after operation were recorded. Results In the observation group, the rate of carotid artery shunting was 35.4% (34/96), among which the rate of carotid artery shunting in patients with contralateral severe carotid artery stenosis or occlusion was 8/13, the rate of carotid artery shunting in patients with unilateral carotid stenosis was 31.3% (26/83), and there was statistical difference (χ2 = 13.006, P<0.01). There was no statistical difference in intraoperative carotid artery occlusion time between control group and observation group ( t=2.091, P>0.05). In the observation group, the intraoperative carotid artery occlusion time in patients with carotid artery shunting was significantly shorter than that in patients without carotid artery shunting:(4.36 ± 0.48) min vs. (10.15 ± 0.91) min, and there was statistical difference (t=7.884, P<0.05). There was no statistical difference in the incidence of stroke event 30 d after operation between control group and observation group (χ2 = 1.189, P>0.05). Conclusions The selective shunting during CEA can reduce the incidence of postoperative stroke event in patients with carotid artery stenosis, and especially it can give a good clinical effect in the patients with contralateral severe carotid artery stenosis or occlusion.
6.STUDIES ON THE EXPRESSION CONDITION OF SEGMENT OF GABA_A RECEPTOR a1 SUBUNTT IN E. COLI

In order to improve the expression level of segment of GABAA receptor a1 subunit in E. coli, growth conditions of the recombatant, which influence the final yield of protein expression, including growth medium, inoculation ratio, temperature, pH, rotation speed, inducing time and concentration of IPTG and so on, were studied in shaking flasks. The results indicated that, with 3% inoculation ratio, cultured 3.5 hours at 37℃, and then induced 5 hours by IPTG at 32℃, the yield of GABAA receptor protein was 95mg/L and the biomass was 3.25 g/ L. In contrast, using a 16 L stirred fermentor instead of shaking flasks, the highest level of the protein expression, 136mg/L with 4.95g/L of biomass, was achieved after fermenting 5.5 hours.
7.Treating chronic persistent bronchial asthma children with abnormal myocardial enzyme spectrum by Yupingfeng powder: an efficacy observation.

OBJECTIVETo observe the clinical efficacy of treating chronic persistent bronchial asthma (CPBA) children with abnormal myocardial enzyme spectrum (AMES) by Yupingfeng Powder (YP) combined routine therapy.
METHODSFrom January 2010 to December 2012, 156 CPBA children patients with AMES were randomly assigned to the treatment group (80 cases) and the control group (76 cases). All patients received routine treatment (inhaled corticosteroids and/or leukotriene regulator). Besides, those in the treatment group took YP. The treatment duration was 3 months. The scores of children asthma control test (C-ACT), pulmonary function (FEV,% and PEF%), myocardial enzyme spectrum were observed before and after treatment, and 3 months before and after treatment. The myocardial enzyme spectrum of 40 healthy children at the baby clinics during the same period were recruited as the control.
RESULTSCompared with the control group, creatine kinase isoenzyme (CK-MB), creatine kinase(CK), and lactate dehydrogenase (LDH) increased in the two treatment groups (P <0.01), but there was no statistical difference in AST (P >0.05). Compared with before treatment in the same group, CK-MB, CK, LDH, and AST decreased in the treatment group after treatment and 3 months after treatment (P <0.01). CK-MB, CK, LDH, and AST decreased in the control group 3 months after treatment (P <0.01, P <0.05).Compared with after treatment, CK decreased in the control group 3 months after treatment (P <0.01). C-ACT score, FEV(1),%, and PEF% all increased in the two groups after treatment and 3 months after treatment (P <0.01, P <0.05). Compared with after treatment in the same group, CK decreased in the control group 3 months after treatment (P <0. 01). Compared with the control group in the same period, post-treatment CK-MB and CK decreased (P <0. 01, P <0. 05), while post-treatment C-ACT score, FEV, %, and PEF% increased (P <0.05) in the treatment group (P <0.05).
CONCLUSIONYP could strengthen specific and non-specific immunity of the organism, and improve clinical symptoms and the level of myocardial enzyme spectrum.

Therefore, based on the traditional method, we have developed a new type of laparoscopic teaching system for the standardized training residents and increased and integrated the LAP GAME R operations training system and the real-time multimedia teaching platform. The preliminary practice effect is good.
9.Correlations of plasma levels of brain natriuretic peptide and Meprin-α with the severity of coronary-artery stenosis

Objective To explore the relationship of the severity of coronary-artery stenosis with plasma levels of brain natriuretic peptide (BNP) and Meprin-α.Methods Totally 237 patients in our hospital were divided into control group (CON group), stable angina group (SA group) and acute coronary syndrome group (ACS group), according to coronary artery angiography.Patients with acute coronary syndrome were divided into 3 subgroups: unstable angina (UA) group, non-ST segment elevation myocardial infarction (NSTEMI) group and ST segment elevation myocardial infarction (STEMI) group.Patients with coronary artery disease (CAD) were divided into 3 subgroups: low-score, medium-score and high-score groups, according to coronary angiography and Syntax score.BNP and Meprin-α levels were determined in patients with coronary artery disease, and the degree of coronary artery stenosis was evaluated.The differences in above indexes were analyzed and compared among the three groups.Results Plasma levels of BNP and Meprin-α were higher in ACS group than in CON group [(233.16± 78.22)ng/L vs.(33.48 ± 13.71)ng/L, (26.89 ± 6.45) nmol/L vs.(12.83±0.66)nmol/L, both P<0.05].Compared with UA group, plasma levels of BNP and Meprin-α were increased in NSTEMI and STEMI groups (all P<0.05).Compared with the control group, plasma levels of BNP and Meprin-α in the Syntax scores-divided subgroups were increased (all P< 0.05).The plasma levels of BNP and Meprin-α in CAD patients were significantly increased along with the increase of Syntax Score.Spearman correlation analysis showed that low density lipoprotein cholesterol, glucose, BNP and Meprin-α levels had positive correlations with the occurrence of coronary heart disease, while high density lipoprotein level was negatively correlated with the occurrence of coronary heart disease (all P<0.05).Conclusions BNP and Meprin-α levels in peripheral blood are significantly elevated in patients with coronary heart disease, and they are correlated with Syntax score.The risk of ACS is increased along with the increased BNP and Meprinα levels.
10.Influence of Mild Hypothermia on Nervous Cell Apoptosis and Activity of Caspase - 3 after Hypoxia - Ischemia Brain Damage in Neonatal Rats

Objective To study the effects of mild hypothermia on nervous cell apoptosis and activity of Caspase - 3 after hypoxia -ischemia brain damage (HIBD) in neonatal rats. Methods Seven - day SD rats were divided into sham operation group,normothermia HIBD group (HIBD group) and mild hypothermia HIBD group(mild hypothermia group). In HIBD group,the left carotid of rats was ligated and animals were exposed to 8% oxygen for 2 h.and in mild hypothermia group,hypothermia of 31 - 32 1C lasting 8 hours was used just after HIBD. L sing methods of TUNEL and AC - DEVI) - AMC, the apoptotic cell rate and Caspase - 3 activity level were determined in sham operated group, HIBD group and mild hypothermia group separatively. Results The apoptotic cell rate and Caspase - 3 activity level were significantly lower in sham - operated group and mild hypothermia group than those in HIBD group. Conclusion One of the mechanisms of brain protection by mild hypothermia may be contributed to the decrease of neural cell apoptosis and the activity level of Caspase 3 HIBD.
