BACKGROUND:Bone marrow mesenchymal stem cels play an osteogenic role under the assistance of scaffold materials. The scaffold cannot only deliver the cels to the bone defect area, but also act as a new bone growth framework. Colagen-chitosan composite is one of ideal scaffold materials in bone tissue engineering, which has osteoinductive ability and better osteogenic ability than conventional scaffolds. Bone transport technology has been widely used in the clinical repair of long bone defects, but it has some deficiencies, such as slow osteogenesis, long time for external fixation and nonunion. How to further accelerate bone formation and reduce complications has become the current problem to be solved. Here, it is hypothesized that bone marrow mesenchymal stem cels/ colagen/chitosan composite scaffold can increase the therapeutic effect of bone transport in the repair of tibial bone defects.
METHODS/DESIGN:This study is a randomized controled animal experiment, includingin vitro andin vivo tests. In vitro test: Bone marrow mesenchymal stem cels are isolated from the bone marrow of New Zealand rabbits aged 1-2 months, and passaged to the third generation. Then, cel suspension is added onto the colagen-chitosan scaffold to construct the bone marrow mesenchymal stem cels/colagen/chitosan composite scaffold. In vivo test: Twenty-four New Zealand rabbits at 3-4 months are selected and randomly assigned to receive bone transport, scaffold implantation, bone transport+scaffold implantation, respectively. The primary outcome measures are the growth of implant materials and bone defect interface, X-ray detection of bone defect repair, hematoxylin-eosin staining and scanning electron microscope observation of bone formation in the bone defect region, immunohistochemical detection of type I colage expression in the osteogenic region, scanning electron microscope observation of interface bonding between implant materials and host bone, ultrastructure and bone formation.
DISCUSSION:The results from this animal experiment wil help to determine the feasibility of bone marrow mesenchymal stem cels/colagen/chitosan composite scaffold to accelerate bone repair during bone defect repair using bone transport technology.

Objective To evaluate the clinical outcome and the acute toxicity in nasopharyngeal carcinoma (NPC) treated with tomotherapy followed by the anti-EGFR monoclonal antibody. Methods Between March 2008 and November 2009,34 newly diagnosed NPC patients were treated with helical tomotherapy combined with nimotuzumab or cetuximab. All the patients underwent tomotherapy at the dose of 70 Gy/33F for the gross tumor volume (pGTVns) and positive lymphnodes (GTVnd) ,and 60 Gy/33F for the high risk clinical target volume (PTV1),and 56 Gy/33 F for the low risk clinical target volume (PTV2),respectively.17 patients in group N were given weekly injection of 200 mg for 6-7 times and 17 patients in group C were given initial dosage 400 mg/m2 followed by subsequent weekly dosage of 250 mg/m2 for 6-7 times.Acute lesions were evaluated with the RTOG/EORTC criteria.Result The median follow-up time was 22 months. The effective rates (CR + PR) in 3,6 and 12 months were 14/17,12/17,12/17 in group N and 15/17,14/17,14/17 in group C.The 1 year survival rate was 15/17 in group Nand 17/17 in group C.Nimotuzumab had less acute mucositis reaction (u = 2.25,P < 0.05),weight loss(t=2.56,P=0.02) and rash (u=4.36,P<0.01) compared with cetuximab. Conclusions Helical tomotherapy combined with nimotuzumab or cetuximab was effective and made no difference in the shortterm efficacy and 1 year survival rate for the patients with NPC.Nimotuzumab has less acute reaction than cetuximab.More studies should be done to prove long-term effects.
8.District differences and regional division of heights among Chinese Han children and adolescents
Chinese Journal of Epidemiology 2012;33(2):197-200
Objective To analyze the district differences of height among Chinese Han population aged 7 to 18 years, and to divide the levels of heights into several regions accordingly,to provide basis for evaluation the development of height in the above said population. Methods Standard Deviation was used to compare the relative lavels of height in different provinces and to divide them into several regions. The results were then showed in map through the Geographic Information System (GIS).Results All the 30 provinces in China were divided into three regions:high, medium and short regions according to the stature.Region with high stature was mainly located in northern, while medium stature was in central and northwest parts and short stature was in southwest parts of China. The differences of height appeared to be significant among regions in every age group. The differences of height were 1-3 cm between adjacent regions, and were even lager between the regions with high or short stature which appeared to be 4-5 cm.Conclusion Overall, heights among the Chinese Han children and adolescents were taller in the northern and shorter in the southern areas. The highest and lowest levels of heights were in the Northern and the Southwest areas respectively. It was observed that the levels of height in the coastal areas were higher than that of the inland. There were no absolute boundaries between different regions because of the gradual transition in the levels of heights in various districts.
9.Study on the references regarding height and prevalence of stunting among children and adolescents aged 7 to 18 years of Chinese Han ethnicity
Chinese Journal of Epidemiology 2012;33(6):576-579
Objective To develop the reference on height among children and adolescents aged 7 to 18 years of Chinese Han ethnicity in different administrative districts of China and to use this height reference for screening stunting. Results from the national screening program were compared with the WHO references. Methods Data from the Chinese National Survey on Students Constitution and Health in 2005 was used to develop the height references through LMS method. Results Differences among the references on height for children and adolescents in different districts ( Ⅰ - Ⅳ ) were remarkable. The highest was seen in the Second Districts while the lowest was seen in the First Districts among those at the age of 18,with differences as 3.18 cm for boys and 2.92 cm for girls. The heights from the inland were shorter than that of the WHO references, with the differences ranging from 0.31 cm to 5.07 cm for boys, and from 0.98 cm to 4.22 cm for girls. The prevalence rates of stunting were 2.22%,2.29%,5.25%,respectively according to local references, national references and the WHO references. Conclusion Universal height references from the whole nation could not discriminate the variations caused by geographic conditions and hereditory factors related to socio-economie situations.Height references should be developed locally,to evaluate height levels of the areas.

To investigate the impact of phenotypic knockout of CXCR4 on Molt-4 cells via intrakine technology, the C-terminal alpha-helix gene SDF-1alpha/54/KDEL of human stromal cell-derived Faceor-1 deletion is fused to a retention signal 4-peptide -KDEL that retains the newly synthesized receptor within the Molt-4 cells endoplasimc reticulum. Subsequently, PCR is used to amplify the target gene SDF-1alpha/54/ KDEL from the constructed plasmid SDF-WT-Gly x 4-Dec/PET-30a(+) at its C-terminal and subclone it into eukaryotic expression vectors pEGFP-C3 for generating recombinant vector cells by lipEGFP-C3/SDF-1alpha/54/KDEL, and then have it sequenced. After the transfection of recombinant plasmids into COS-7 posome, SDF-1alpha/54/KDEL protein is confirmed with Western blot. The recombinant plasmids pEGFP-C3/SDF-1alpha/54/KDEL are isolated and transiently transfected in Molt-4 cells by electroporation. Flow cytometric analysis shows a dramatic reduction of CXCR4 expression on Molt-4 cells. The conclusion is that SDF-1alpha/54/KDEL could assume a role in the phenotypic knockout of CXCR4, and the findings suggest that the inhibiting effect of SDF-1alpha/54 against CXCR4 is not influenced by the deletion of SDF-1alpha helix at the C terminal.
