4.Assessment of Life Quality of Children with Complex Congenital Heart Defects after Surgical Treatment

Objective To assess the life quality of children with complex congenital heart defects after surgical treatment.Methods Ten children with complex heart defects after surgical treatment were given comprehensive assessment in physical development(using weight and height standard deviation score,SDS);the symptoms and signs that easily occured;startting time for enrolment to a kindergarten or primary school enrolment.Results The weight SDS of 9 cases were below average value of reference crowd,only 1 case was above average value of refere nce crowd;all the 10 cases of the height SDS were below average value of reference crowd;the symptoms prone to appear:2 cases had cyanosis,9 cases had breathlessness,and all 10 cases of heart auscultation had heart murmur;8 cases stayed at home,having reached the school or kindergarten age.Conclusions The life quality of children with complex congenital heart defects after surgical treatment is inferior.The congenital heart disease prevention show focus on pre-birth intervention.
5.Construction of Recombinant Adenoviral Vector Carrying Mus-?NGF Gene and Its Expression in the MSCs

Objective To construct the adenoviral vector carrying Mus-?NGF gene and to study the expression of the gene in the marrow mesenchymal stem-cells(MSCs).Methods Mus-?NGF cDNA was cloned from mice glandulae maxillaries whole mRNA by reverse-transcription polymerase-chain-reactions.The Mus-?NGF cDNA was positively cloned into the adenoviral shuttle vector pAdtrack carrying cytomegalovirus promoter(CMV)and tag-protein(GFP),and then homologous recombined into competent BJ5 183 cells with the adenoviral backbone plasmid pAdeasy-1.The recombinant adenoviral plasmid was identified by restriction enzymes and transfected into HEK293 cells to package and amplify recombinant adenoviral particles which was capable of infecting and would express ?NGF and GFP proteins.After the adenovirus infected the MSCs,the expression was observed via fluorescent microscrope and detected using immunocytochemical method.The secretion of NGF was detected by WB.Results The recombinant adenoviral vector carrying Mus-?NGF gene was constructed and confirmed by restriction endonuclease enzyme analysis.The relucent green fluor-light would be observed in the MSCs.IHC showed that Mus-?NGF gene was exactly transcripted and expressed in the MSCs,and the secretion of NGF was confirmed.Conclusion The exogenous gene,such as ?NGF,an be imported into the MSCs efficiently by recombinant adenoviral vector,and can be expressed and secreted successfully in the cells.It improves foundation for the celluar transplant and gene intervention of MSCs expressing NGF in inner ear.

7.Distribution and significance of NGF in cochleae of neonate rats

Objective To investigate the distribution of NGF in the cochleae of neonate rats and to explore the mechanism of NGF in the auditory path. Methods Twenty healthy neonate SD rats aged 5-7 d were selected and their cochlear paraffin sections were labeled with a rabbit anti-NGF polyclonal antibody by SP method. Results The membranous labyrinth of neonate rats appeared similar to triangle and NGF expression was observed in immature corti’s organ, in spiral ganglion cells and in nerve fibers throughout the cochlea. The distribution located in both cytoplasm and nucleus. Conclusion The distribution of NGF was closely related to survival and maturation of spiral ganglion cells and hair cells in rats aged 5-7 d.

9.How Aware Elderly Subjects are of Medical Device Clinical Trials and Their Adverse Events - A Survery.

BACKGROUND: Along with the recent greater number of medical devices for the elderly in the market, there has been a parallel increase in the number of clinical trials for these devices. It is uncertain, though, whether the target population has the awareness and understanding of these clinical trials and the possible adverse events of the devices. METHODS: A total of 147 elderly subjects (mean age, 65.8+/-8.3 years; 39 males) responded to the questionnaire. A pilot study during the development of the questionnaire was conducted in 2009 with the actual survey taking place from June to November 2011. The questionnaire included 22 items-10 items on awareness and understanding of the clinical trial; 4 items on the understanding of adverse events; and 8 items on demographical characteristics. RESULTS: Of the responders, 62.5% had participated in at least one clinical trial and had heard about the clinical trials through a doctor, newspaper, or television. And 63.3% of the responders understood that the purpose of the trial was to show efficacy and safety for the medical device. There were significant differences on the awareness of the trial by gender and life level. However, there was no statistically significant difference in adverse events awareness. CONCLUSION: Although elderly subjects participate in clinical trials, it seems they do not receive enough information when involved in clinical trials of medical devices. In the future, before these trials are carried out, medical device companies need to ensure that they provide their elderly subjects with better education and information.

10.Treatment of Asthenozoospermia Men with Shen-essence Deficiency Syndrome by Yishen Zhongzi Pill: a Clinical Study.

OBJECTIVETo observe the effect of Yishen Zhongzi Pill (YZP) on the sperm motility of Shen-essence deficiency syndrome (SEDS) patients.
METHODSTotally 144 patients were randomly assigned to the treatment group (71 cases) and the control group (73 cases). Ten cases dropped out during the therapeutic course. A total of 134 patients completed this clinical study, 67 in each group. Patients in the treatment group took YZP (6 g each time, twice daily), while those in the control group took Wuzi Yanzong Pill (6 g each time, twice daily). The therapeutic course for all was one month. Changes of the sperm motility, clinical efficacy, and Chinese medical symptoms scores were observed between the two groups before and after treatment.
RESULTSCompared with before treatment in the same group, percentages of class a spermatozoa and class a + b spermatozoa were enhanced in the two groups after treatment (P <0. 01). Besides, they were higher in treatment group than in the control group (P <0. 05). The total effective rate of the treatment group was 77. 61 % (52/67 cases), while it was 58. 21 % (39/67 cases) in the control group. It was better in the treatment group than in the control group (Z= -2. 914, P =0. 004). Compared with before treatment in the same group, scores for Chinese medical symptoms decreased in the two groups (P <0. 01). The difference of pre-post integrals was higher in the treatment group than in the control group (P <0. 01).
CONCLUSIONYZP showed favorable effect in treating asthenospermia patients with SEDS and could effectively elevate the motility of sperm.
