1.The relationship between LPS-induced iNOS mRNA expression and macrophages cytotoxic function

Purpose:To study LPS induced iNOS mRNA expression and NO production by murine peritoneal macrophages, and their relationship with cytotoxic function of activated macrophages.Methods:LPS induced iNOS mRNA expression, production of NO and effects of activated macrophages on anti tumor were investigated separately by RT PCR, nitrate reductase method and 3HTdR incorporation.Results:LPS can upregulates iNOS mRNA expression in a dose dependent manner. L NIL(specific inhibitors of iNOS) pretreatment significantly reduced cytotoxic effect on the proliferation of HL 60 cells, but had little effect on the K562 tumor target cells on condition that NO production is sufficiently inhibited. Conclusions:LPS can effectively induce iNOS mRNA expression and NO production in murine peritoneal macrophages. NO is one of the important effector molecules in unspecific immunity of activated macrophages, but it plays different roles in cytotoxic effects of macrophages on different target tumor cells.

Sorafenib is a molecular targeted drug for the treatment of advanced primary liver cancer.However,along with the occurrence of drug resistance,the therapeutic effect was effected.At present,there is clear evidence that the emergence of drug resistance of live cancer is closely related to the epithelial-mesenchymal transition,liver cancer stem cells and the heterogeneity of liver cancer,and the PI3K/AKt signaling pathway as the vital common signaling channel was involved in the above mentioned process.From this we can conclude that complementary inhibtion of PI3K/AKt signaling pathway at the same time is the method that can strengthen the effect of sorafenib on the treatment of liver cancer so far.
4.RAPID DETERMINATION OF GLUTAMATE DECARBOXYLASE ACTIVITY FROM LACTIC ACID BACTERIA BY SPECTROMETRIC METHOD AND ITS APPLICATIONS

In this paper, a colorimetric method with high sensitivity, reproducibility and low cost for determining glutamate decarboxylase (GAD) activity was developed based on Berthelot reaction. The optimum substrate system is 0.2 mol/L MacIlvaine buffer pH4.7, containing 0.1 mmol/L pyridoxal phosphate (PLP) and 10 mmol/L L-monosodium glutamate (L-MSG). The reaction mixture consisted of 200 ?L substrate solution and 1~100 ?L enzyme preparation from lactic acid bacteria and was incubated at 30℃. The enzymatic reaction was terminated by immersion in ice-water and addition of 200 ?L 0.2 mol/L sodium borate buffer, pH9.0, then 1 mL 6% phenol solution and 400 ?L sodium hypochloride were added. Color development was carried out in boiling water for 10 min then immediately put in ice-water bath for 20 min. The optical density is read at 630 nm. ?-aminobutyric acid (GABA) produced was calculated using the standard curve. Applications of the method in GAD studies were discussed.
6.Application of PBL teaching combined with flipped classroom in standardized residency training of orthopedics

Objective:To explore the application of self-teaching model based on PBL teaching combined with flipped classroom in standardized residency training of orthopedics.Methods:There were 102 cases of residents who received standardized residency training from March 2017 to February 2018 in orthopedics department of Yueyang Hospital of Traditional Chinese and Western Medicine, among whom 55 were randomized into the observation group and 47 were divided into the control group. Traditional teaching was applied in the control group and self-teaching model was applied additionally in the observation group. At the end of the month, the teaching effect was evaluated by the department graduation examinations and questionnaire survey. SPSS 19.0 was used for t test. Results:The ability of history inquiry, diagnosis and physical examination of the residents in the observation group were higher than those in the control group. The self-study and data access ability, confidence in presenting, and satisfaction with teaching in the observation group were better than those in the control group.Conclusion:The self-teaching mode can arouse the learning initiative of residents, and increase residents' literature retrieval ability and their learning satisfaction.

Vertebral artery dominance is generally considered to be a congenital vascular variation or meaningless clinical finding.Many studies have shown that the bilateral vertebral artery blood flow asymmetry caused by vertebral artery dominance is easy to result in basilar artery curvature.Some studies have shown that there are certain correlations between both the vertebral artery dominance and the basilar artery curvature and the posterior circulation stroke.
8.Role of CTLA4-Ig and anti-CD40L monoclonal antibody in the immune rejection of pancreas transplantation in rats

Objective To investigate the role of CTLA4-Ig and anti-CD40L monoclonal antibody in the acute rejection of pancreaticoduodenal transplantation model of rats.Methods Pancreaticoduo- denal transplantation model was established from the donor F344 rats to the Lewis recipients(diabetes models).The models were divided into 4 groups:groups A,B,C and D with 12 rats in each group. Two days after transplantation,recipients were injected intraperitoneally with saline,CTLA4-Ig(200?g),anti-CD40L monoclonal antibody(200?g),CTLA4-Ig(200?g)combined with anti-CD40L monoclonal antibody(200?g)respectively.On the day 1,4,7,10 after transplantation,the grafts were harvested for histopathological examination and RT-PCR to determine the levels of interleukin (IL)-2,interferon(IFN)-?,IL-4 and IL-10;The blood CD3~+,CD4~+and CD8~+ T cells were detected by flow cytometry.On the day 4 after transplantation,the CD4~+ CD25~+ T cells in the grafts were de- tected by flow cytometry.Results As compared with group A,the severity of the rejection of grafts in groups B,C and D were depressed;Down-regulation of IL-2 was observed in the groups B,C and D, and the levels in group D were lowest.Down-regulation of IFN-7 was detected in the groups B,C and D,but there was no significantly difference between groups D and B or groups D and C.Up-regulation of IL-4 was observed in the groups B and C,and the levels in group D were lower than in groups A,B and C.Up-regulation of IL-10 was observed in groups B and C,and there was significant difference between groups D and B or groups D and C.The CD3~+,CD4~+ and CD8~+ T cells in groups B,C and D were less,but more CID4~+ CD25~+ T ceils in transplanted pancreas were observed,more notably in group D than in group C.Conclusions Combined use of CTLA4-Ig and anti-CD40L monoclonal anti- body can remarkably diminish the severity of the rejection,which might be mediated by altering the balance in Th1/Th2 and increasing the number of CD4~+ CD25~+ regulatory T cells.Co-stimulation blockade with CTLA4-Ig and anti-CD40L monoclonal antibody induction seems to be an attractive strategy to control allograft rejection.
9.Measurements of normal inner ear on multi-slice computed tomography

Objective To evaluate the anatomical and developmental characteristics of inner ears and establish a normalized inner ear multi-slice computed tomography (MSCT)measurement standard.Methods The raw data of 81 normal head MSCT cases were amplified and reconstructed in inner ear areas and multi-plannar reformation (MPR)was used to measure diameters of the inner ears.-test was used to inspect the difference between sides and genders,one-way analysis of variance was adopted for different age groups. Significance level α=0.05,P <0.05 was considered statistically significant.Results There was no statistical significance in measured values of inner ears in different age groups,genders and sides.Not all the shapes of bony semicircular canals were completely circu-lar or elliptical.In lumen,posterior semicircular canal was maximal and superior semicircular canal was minimal.In height,posterior semicircular canal was maximal and lateral semicircular canal was minimal.In width,superior semicircular canal was maximal and lateral semicircular canal was minimal.Conclusion Measurement specifications of inner ear on MSCT are initial established,main structures of inner ear can be clearly displayed by post-processing of head MSCT raw data.
10.Anti-infective Effect of Levofloxacin During Perioperation of Appendectomy due to Acute Appendicitis
