Autophagy is a dynamic process which subcellular membranes undergo morphological changes that lead to the degradation of cellular cytoplasmic organelles and macromolecules.It is regulated by themammalian target of rapamycin ( mTOR ) -dependent or -independent signaling pathways.It has been demonstrated that autophagy is induced or inhibited in various tumor cells,and it is closely related with cell survival and drug resistance.Because of the complex relationships with cell death,the roles of autophagy in cancer developnent,treatment,and drug-resistance are not the same,and thus controlling autophagy properly may become one of new means of cancer therapy.
3.Clinical value of susceptibility weighted imaging in the diagnosis of Parkinson disease

Objective To explore the application value of susceptibility weighted imaging (SWD) in the Parkinson disease (PD) diagnosis by analysis the changes of brain iron content of the basal ganglia and red nucleus.Methods Totally 56 cases of patients with PD and 40 cases of normal control group were diagnosed by SWI with the 3.0 T magnetic resonance imaging system,the corresponding phase image by using SWI processing software was obtained,the signal values of the various regions of the brain basal ganglia and red nucleus were measured and compared.Results In comparison of PD group with normal control group,the signal values showed differences in caudate (0.05±0.01 vs.0.04±0.01,t=1.991),putamen(-0.42±0.13 vs.-0.25±0.07,t=2.544) and globus pallidus(-0.23± 0.07 vs.-0.11±0.02,t=2.893) (all P<0.05).Conclusions Determining the iron deposition signal of the various regions of the brain basal ganglia by SWI is valuable in the clinical diagnosis of Parkinson's disease.
4.Clinical study of continuous subcutaneous Insulin infusion combined with mefformln hydrochloride in the in-teusive treating of type 2 diabetes mellitus

Objective To evaluate the efleet of continuous subcutaneous insulin infusion (CSII)combined with mefformin hydroehloride( Met H)on type 2 diabetic patients. Methods 240 patients with type 2 diabetes melli-tus (T2DM) were divided into 3 groups randomly: CSII combined with Met H ( A group: n=80) ; simple CSII ( B group:n=80) ; multiple deliver insulin (MDI) ( C group: n=80). The levels of blood glucose, HbA1 e, therapeutic time, a-mount of insulin, Fins ,C-P, HOMA-IR and IAI were compared among them before and after 10d of treatment. Results GroupA, B eornpared with group C, the levels of HbA1 e significantly deeroased in other two groups [ ( 8.0±3.2)%vs. (6.9±1.4)% and(7.0±1.2)% ] (aU P<0.05),so did the therapeutic time[(6.0±1.9)d vs. (3.0±1.4) and(3.5±1.5) d] and insulin dose[ (55.0±17.0) U/d vs. (38.0±15.0) U/d and(45.0±16) U/d] (all P<0.05). The insulin dose was much less in group A than in group B. Versus group B and C,the HOMA-IR of group A (0.8±0.3 vs. 1.5±0.4 and 1.6±0.4)decreased(P<0.05) ,and IAI(-2.0±4.3 vs. -3.6±4. 2 and -4.1±4.3) increased (all P<0.05), without the function impairments of heart, liver or kidney. Conclusion CSII in combi-nation with mefformin hydreehloride can significantly enhance the treatment effect of the patients with type 2 diabetes mellitns; reduce blood glucose and HbA1 e, improve insulin resistenee,lessen insulin need.
5.Stress on recognition of dry eye in children with ocular allergic disorders

Most patients with allergic conjunctivitis in children are vernal keratoconjunctivitis.Dry eye accompanied with al- lergic conjunctivitis in children is correlated with the decline of tear film stability.However,tear film stability decline is correlated with eosinophilic activation and concomitant release of inflammatory mediators.Moreover,conjunctival epithelial,goblet cells and tear film are damaged,then dry eye occur.Main symptom of dry eye in children is twinkling frequency increasing,followed by eye itch,eye red and photophobia.When allergic conjunctivitis in children is diagnosed,dry eye should be considered at the same time.Proper manage- ment for those allergic conjunctivitis can decrease ocular surface damages.
6.Molecular epidemiological characteristics of metallo-β-lactamase-producing Pseudomonas aeruginosa from Changsha

Objective To investigate the genotypes and epidemic of metallo-β-lactamase-(MBL )-producing Pseudomonas aeruginosa (P .aeruginosa)in Changsha.Methods P .aeruginosa isolated from seven comprehensive hospitals in Changsha were collected and performed identification and antimicrobial susceptibility testing,pheno-types of MBL were detected with EDTA-disk synergy test and E-test,genotypes were determined by polymerase chain reaction (PCR),homology analysis were conducted by enterobacterial repetitive intergenic consensus PCR (ERIC-PCR).Results Preliminary screening by EDTA-disk synergy test and E-test showed that only 10 of 81 iso-lates were strong positive;PCR result showed that 18 isolates were positive for MBL,11 of which were IMP-9-type MBL,1 was IMP-1-type,and 6 were VIM-2-type.SIM,SPM,GIM,and NDM-1-types were not found.ERIC-PCR showed that 12 strains of IMP-producing P .aeruginosa has multiple types,6 VIM-2-producing strains were of the same type.Conclusion IMP-9 and VIM-2 are main genotypes in P .aeruginosa in Changsha.
7.Effect and mechanism of leptin on bone metabolism in postmenopausal women

Objective To investigate the effects of Leptin on the base of anthropometry,densitometry,and biochemistry and reveal the role of Leptin in bone metabolism in postmenopausal women with osteoporosis (OP).Methods From January 2008 to January 2014,data of 82 postmenopausal females with or without OP were analyzed.They were randomized into two groups:OP (n=40) and conditional control group (CON) (n=42).The expression of Leptin receptors in osteoblasts was observed by immunohistochemistry.1 × 10 ng/ml,1 × 102 ng/ml,1 × 103 ng/ml,1× 104 ng/ml of Leptin were added respectively.Then the proliferation,differentiation and mineralization of osteoblasts were measure at 24 h,48 h and 96 h.The gene expressions of bone marrow mesenchymal stem cells (BMSCs) derived osteoblasts and adipocytes treated with Leptin were examined by quantitative analysis of Real time-PCR.Results The expression of Leptin receptors in osteoblasts was observed on plasma membranes and cytoplasm in the normal group and osteoporosis group.In osteoporosis group,different concentrations of Leptin enhanced cell proliferation,cell differentiation and cell mineralization.The growth rate of MTT and the concentration of ALP in the serum of were significantly increased with the effects of 1×10 ng/ml,1×102 ng/ml and 1×103 ng/ml of Leptin,and the action of 1×102 ng/ml Leptin was the most powerful with time dependence.Within 96 hours,the growth rate of osteoblasts increased gradually,and the concentration of ALP increased gradually in 3 weeks.The expression of RANKL/OPG in bone marrow BMSCs derived osteoblasts treated with Leptin were significantly increased.Conclusion Leptin receptor is present in osteoblasts,and Leptin affects biological behaviors of osteoblasts through receptors.The direct effect of Leptin may relate the expression of RANKL/OPG.Then,the balance between bone resorption and bone formation was broken and finally osteoporosis occurred.Lepin may promote osteogenesis and inhibit bone resorption in the differentiation of BMSCs.

9.Effects of exercise on bone turnover:promoting or inhibiting development and viability of osteoblasts/osteoclasts

BACKGROUND:Bone turnover markers are the products in the blood or urine in the process of bone reconstruction, which can be used to evaluate bone resorption and bone formation rate, thereby indicating potential bone diseases. OBJECTIVE:To analyze the effect of exercise on the levels of bone turnover markers in the blood and urine samples. METHODS:Databases of PubMed and Wangfang were retrieved with key words of“bone formation, bone resorption, alkaline phosphotase, osteocalcin PICP, PINP, hydroxyproline, pyridinoline, tartrate resistant acid phosphatase”by screening titles and abstracts to search papers related to exercise effects on the biological markers of bone formation. Final y, 48 papers were discussed. RESULTS AND CONCLUSION:The effect of exercise on bone turnover refers to the influence on bone formation and bone resorption. Exercise enhances or inhibits the activity of osteoblasts and osteoclasts, and accelerates or delays bone reconstruction by promoting or inhibiting the growth of osteoblasts and osteoclasts. In recent years, exercise exhibits more and more influence on the bone. Bone turnover markers compared with bone mineral density show changes earlier. By measuring the levels of bone turnover markers in blood and urine samples, it help us to understand the metabolism of bone tissue, to evaluate bone metabolic state, osteoporosis diagnosis classification, predicting of fracture risk, to observe the curative effect of drug treatment, and to diagnosis the metabolic bone differential disease.
10.Role of histone deacetylase inhibitors in cell growth arrest and apoptosis

The acetylation status of histones and non-histone proteins regulate chromatin remodeling and gene transcription. Histone deacetylase inhibitors (HDACi), a promising therapeutic approach to cancer, are characteristic of causing accumulation of acetylated histones and other transcriptional regulators. Recent studies demonstrate that HDACi is able to arrest the cell cycle in G1 and/or G2 phase, and to induce apoptosis in a variety form of transformed cells with little toxicity to normal cells. However, the exact antitumor mechanisms of HDACi are still unclear. This review provides an update on the current knowledge of HDACi with a focus on HDACi-regulated cell growth arrest and apoptosis.
