1.Determination of rhien, emodin, and chrysophanol in Huangdanyinchen Granules by HPLC

Objective:To develop a method to determine the contents of emodin, rhien and chrysophanol in Huangdanyinchen Granules. Methods: HPLC was used to determine the contents of emodin, rhien and chrysophanol in Huangdanyinchen Granules. The separation was performed on YWG ODS column with methol perchloric acid (1%) (8∶2) as a mobile phase and the wavelength of UV detector was 280nm.Results: The separating degree and the linearity of this method was well with the average recovery of rhien: 97.43%, RSD= 1.90%; Emodin:98.45%,RSD=1.62%;Chrysophanol: 98.87%, RSD=1.50%, respectively.Conclusions:The method is simple and rapid and gives satisfactory results. It can be suitable for quality controll of Huangdanyinchen Granules.
2.Expression of MACC1 in nasopharyngeal carcinoma and its relationship with prognostic significance

Objective To explore the expression of Metastasis-associated in colon cancer-1 (MACC1) in patients with nasopharyngeal carcinoma (NPC), and its relationship with clinical pathologic characteristics and prognosis Methods The expression of MACC1 was detected in 130 cases of NPC and the relationship among the MACC1 expressions, clinical pathologic characteristics and prognosis of NPC was analyzed. Results Positive expression rate of MACC1was 68.5% in the NPC and MACC1 expression was associated with advanced T stages, lymph node metastasis and advanced clinical stages of NPC (P < 0.05). The results of Kaplan-Meier survival analysis showed that the five year overall survival rate in patients with positive expressions of MACC1 (45.9%) was significantly lower than that of those with negative expressions (73.7%) and the difference was statistically significant (P < 0.05), Cox multi-factor analysis results showed that MACC1 expression was an independent prognostic factor for NPC (P = 0.004). Conclusion MACC1 abnormal expression is closely related to the invasion and metastasis of NPC and it is expected to become a new target for gene therapy of NPC.
3.Therapeutic effects of recombinant hepatitis B virus vaccine combined with interferon α-1 b in patients with chronic hepatitis B

Objective To investigate the therapeutic effects of recombinant yeast hepatitis B virus(HBV)vaccine combined with interferon(IFN)α-1b and determine the rational dosage of HBV vaccine for the further clinical study with larger sample.Methods Two hundreds and sixteen patients with chronic hepatitis B(CHB)were enrolled in this randomized,multi-center,double-blinded and placebo-controlled clinical trial.All the subjects were not treated with antiviral drugs within 6 months and randomly divided into 90μg,60μg and placebo groups with a ratio of 1:1:1.All the patients were intramuscularly administrated with 90μg or 60μg recombinant HBV vaccine or placebo at week 0,2,6,10,14,18,22,respectively.Meanwhile,they were also treated with IFNα-1b 50μg,3 times a wcek for 24 weeks.All patients were followed up for 24 weeks after withdrawal of anti-HBV therapy.Serum HBV DNA level,HBeAg titer and liver function were monitored frequently.Interferon-γ secreting lymphoeytes were detected by Enzyme-linked immunospot(ELISPOT)in part of the patients.Results The serum HBV DNA levels were(6.03±1.79),(5.52±1.82)and(6.29±1.70)log10 copy/mL at week 24 of treatment in high dose,low dose and placebo groups,respectively (P=0.458).And the serum HBV DNA levels were(5.92±1.98),(5.49±1.99)and(6.16±1.76)log10 copy/mL at weck 24 after withdrawal of treatment,respectively(P=0.720).The rates of patients whose HBV DNA<1×105 copy/mL in these three groups were 30.4%,39.4% and 20.8% at week 24 of treatment,respectively and there was significant difference between high dose group and low dose group(P=0.015).The rate of patients whose HBV DNA<1×105 copy/mL at week 24 after withdrawal was highest in low dose group,but no significant differences before and after treatment in these three groups(P=0.257).The HBV DNA negative rates were 17.4%,25.4% and 6.9% in these three groups,respectively,which were significantly different(P=0.012).At week 24 of treatment and week 24 after withdrawal of treatment,the alanine aminotransferase normalization rate,HBeAg seroconversion rate were highest in low dose group,but no significant differences in these three groups.ELISPOT positive rates at week 24 of treatment and week 24 after withdrawal of treatment in high close and low dose groups were higher than that in placebo group(P<0.05).The incidence of adverse events was similar and there was no drug related severe adverse events in each group.Conclusion Recombinant HBV vaccine maybe contribute to anti-HBV therapy and 60μg of dosage seems to be rational.
4.The effects of group experiential education in patients with diabetes based on screening for diabetic complications

Objective To investigate the effects of group experiential education on glycemic control and self-care activities in patients with diabetes based on screening for diabetic complications.Methods Using convenience sampling,totally 134 patients with type 2 diabetes admitted to our department of endocrinology were randomly divided into the intervention group(n=68) and the control group(n=66) from January to August,2016.The control group received regular education on diabetic complications,while the intervention group received group experiential education via diabetic complications-simulated experience tools on the basis of regular education.Differences in glycemic control and self-care activities between two groups before and after intervention were compared.Results After adjusting for age,sex,diabetic duration,blood pressure,hypoglycemic treatments,lipids profiles,educational level,and diabetic complications and indices before interventions,improvements of fasting plasma glucose(FPG),2 h postprandial plasma glucose (PPG),haemoglobin A1c (HbA1c),and self-care activities in the intervention group were much higher than those in the control group after intervention,and the differences were statistically significant(P<O.05).Conclusion The group experiential education based on screening for diabetic complications can significantly increase self-care activities and improve glycemic control in patients with diabetes.

Objective:
To investigate the pathogen spectrum distribution and drug resistance of febrile neutropenic patients with hematological diseases in Shanghai.
Methods:
A retrospective study was conducted on the clinical isolates from the febrile neutropenic patients hospitalized in the departments of hematology in 12 general hospitals in Shanghai from January 2012 to December 2014. The drug susceptibility test was carried out by Kirby-Bauer method. WHONET 5.6 software was used to analyze pathogenic bacteria and drug susceptibility data.
Results:
A total of 1 260 clinical isolates were collected from the febrile neutropenic patients. Gram-positive bacteria accounted for 33.3% and Gram-negative bacteria accounted for 66.7%.
