1.Anti-proliferation role and effect on PI3K/Akt and Erk1/2 signal pathways of a chimeric anti-erbB2 antibody(chA21)on SKBR3 cells

Aim To study the inhibitory effect of anti-ErbB2 chimeric antibody chA21 on proliferation of human malignant breast cancer cell lines SKBR3 and explore its possible mechanisms through studying its effects on ErbB2 distribution and PI3K/Akt,Erk1/2 signal transduction.Methods The inhibitory effect of chA21 on SKBR3 was assessed by MTT assay.The effect of chA21 on distribution of ErbB2 on cell plasma membrane was determined by FACS(Fluorescence-activated cell sorting)analysis;p-Akt and p-Erk1/2 were detected by Western blot.Results The inhibitory rate of chA21 on SKBR3 cells in vitro increased in a dose and time dependent manner.Furthermore,it could down-regulate the distribution of ErbB2 on plasma membrane and decrease the activation of p-Akt and p-Erk1/2.Conclusions ChA21 can inhibit the proliferation of SKBR3 cell in vitro.Down-regulation ErbB2 on cell plasma membrane resulting in decreasing the activation of p-Akt and p-Erk1/2 may be one of the mechanisms of the inhibitory effect.

3.Evaluation of left ventricular systolic function with tissue Doppler imaging in patients with type 2 diabetes

Objective To evaluate the clinical application of the tissue Doppler imaging (TDI) in investigating the cardiac longitudinal systolic function in the patients with type 2 diabetes mellitus. Methods Forty-two patients with type 2 diabetes mellitus and 25 volunteers underwent TDI. The long axis myocardial peak systolic velocity (Sm), displacement (Dm) and strain rates (SRs) of 18 segments in different left ventricular walls were measured.Results The Dm and SRs of left ventricular wall in diabetic group dramatically decreased (P<0.01). Meanwhile, the diabetic combined hypertension group had much lower Sm of ventricular middle segments than the control group (P<0.05).Conclusion The stain rate and tissue tracking are sensitive and reliable to evaluate the early local cardiac systolic function in the patients with diabetes mellitus. TDI can be used as an noninvasive method to detect early systolic dysfunction.
4.Neurotoxicity induced by multiple exposures to sevoflurane anesthesia in neonatal rats

Objective To evaluate the neurotoxicity induced by multiple exposures to sevoflurane anesthesia in the neonatal rats.Methods Thirty-two healthy SPF Sprague-Dawley rats of both sexes,aged 7 days,weighing 15-20 g,were randomly divided into 2 groups (n=16 each) using a random number table:control group (group C) and multiple exposures to sevoflurane anesthesia group (group Sev).On postnatal day 7,14 and 21,2.6% sevoflurane was inhaled for 2 h in group Sev,while the mixed gas of oxygen and air was inhaled instead of sevoflurane in group C.Morris water maze test was carried out on postnatal day 32-36 to assess the cognitive function.On postnatal day 21 and 36,8 rats in each group were selected and anesthetized,and the cerebrospinal fluid was collected for determination of the concentrations of amyloid β-protein by enzyme-linked immunosorbent assay.Results Compared with group C,no significant change was found in the escape latency,movement time spent in the effective region of the platform,movement distance,the number of entries into the effective region,percentage of residence time,percentage of movement distance,and percentage of the number of entries (P>0.05),and the concentrations of amyloid β-protein in the cerebrospinal fluid were significantly increased on postnatal day 36 in group Sev (P<0.05).Conclusion Multiple exposures to sevoflurane anesthesia can induce central neurotoxicity,but do not induce changes in the cognitive function in the neonatal rats.
5.Comparison of two different gastric tube insertion methods on neonatal pain response

Objective To compare two different gastric tube insertion methods on neonatal pain responae.Methods From April to July 2010,47 neonatal patients underwent gastric tube insertion in the neonatal department of our hospital were randomly classified into two groups:the nasal gastric tube insertion group(28 cases) and the oral gastric tube insertion group( 19 cases).Neonatal pain score and incidence of complications during gastric tube insertion between both groups were compared.Results The mean rank of pain scores were significantly higher in the nasal gastric tube insertion group than in the oral gastric tube insertion group.The incidence of pain and complications were higher in the nasal gastric tube insertion group than in the oral gastric tube insertion group.Conclusions The impact of oral gastric tube insertion on neonatal pain response is not significant,and few complications occur during oral gastric tube insertion,so it is worthy of clinical application.
6.The Effect of Dezocine Analgesia on Cognitive Function of Patients after Gynecological Laparoscopic Surgery

Objective To discuss the application of Mini Mental State Scale (the mini-mental state examination MMSE) test in assess the effect of dezocine analgesia therapy on the cognitive function of patients with agitation in recovery period of general anesthesia after gynecological laparoscopic surgery.Methods We observed 210 cases of elective gynecologic laparoscopic surgery in our hospital from January 2015 to December 2015,ASA grade Ⅰ or Ⅱ.All patients were transrered to PACU (ICU) after extubation,and were divided into three groups:A,B,C group,each with 70 cases in each group.Patients with severe agitation in A group were timely given 10mg of dezocine,patients with mild agitation in B group were given 10mg of dezocine after a delay of 5 ~ 10 minutes,and patients without agitation in C group were not given dezocine.Then the MMSE test was performed to evaluate the cognition scores of patients in the three groups.The MMSE scores immediately,15 minutes,30 minutes,60 minutes after given dezocine and 24 hours after operation were recorded.Results The statistical results showed that there were significant differences in the MMSE scores of patients immediately and at 30 minutes after given dezocine between three groups (P<0.05);and there was no significant difference in the MMSE scores of patients at 60 minutes after given dezocine and 24 hours after operation between three groups (P >0.05).Conclusion Dezocine analgesia therapy has no effect on the cognitive function of patients with agitation in recovery period of general anesthesia after gynecological laparoscopic surgery.Dezocine can be safely used for analgesia and sedation treatment for patients with agitation in recovery period of general anesthesia after gynecological laparoscopic surgery analgesia,sedation agitation.
7.Effects of neonatal repeated inha lation of sevoflurane on ability of learning and memory and hippocampal volume in infantile rats

Obj cetive A large number of recent studies show that sevoflurane anesthesia may cause learning and memory dysfunction.The aim of this study was to explore changes of learning and memory ability and hippocampal volume in infantile rats after neonatal interrupted and repeated inhalation of 2.6% sevoflurane through detecting the learning and memory ability by Morris water maze and the hippocampus volume by MRI.Method s Thirty two neonatal SD rats were randomly devided into two groups (n=16):experimental group and control group.Rats inhalated 2.6%sevoflurane in the experimental group and 1 L/min O2 +1 L/min Air in the control group at the postnatal days of 7, 14 and 21 (P7, P14, P21). The learning and memory ability was determined by the Morris water maze test from P31 to P37;The brains of rats were scanned by mag-netic resonance imaging ( MRI) machine under anesthesia with 1%sodium pentobarbital at P37, and the brain and bilateral hippocampal volumes were measured. Results ①In the place navigation test, the escape latency had no significant difference between the two groups (P>0.05).In the spatial probe test, the dwelling time, movement distance and number of entering times in platform quadrant decreased slightly in experimental group compared with those in the control group, while there was no significant difference (P>0.05).②The brain volume [(1.53 ±0.18) cm3 vs (1.60 ±0.13) cm3] and right hippocampal volume [(16.15 ±1.76)mm3 vs(16.46 ±1.71)mm3] had no significant difference between the two groups (P>0.05).The left hippocampal volume [(16.46 ±1.71)mm3] was decreased in the experimental group compared with the control group [(18.10 ±2.53)mm3](P<0.05). Conclusion The learning and memory ability has no significant changes in in-fantile rats after neonatal interrupted and repeated sevoflurane inhalation and MRI examination of hippocampal volume is not sufficient for the diagnosis of cognitive dysfunction.
9.Adverse Drug Reactions of 6 Kinds of Quinolones

OBJECTIVE To analyze the adverse drug reactions of 6 kinds of quinolones in order to provide information for clinically rational use of drugs.METHODS The adverse drug reactions of quinolones such as norfloxacin,ofloxacin,levofloxacin,ciprofloxacin,fleroxacin and rufloxacin used in outpatients during Feb to May 2007 were collected and retrospectively analyzed.RESULTS The main adverse drug reactions were seen in gastrointestinal system,nervous system,immune system,respiratory system,urinary system etc.From them the symptoms of gastrointestinal system and nervous system were prominent.CONCLUSIONS In use of quinolones it should think highly to their adverse drug reactions.
10.Effect on production of avermectins of spore pigment biosynthesis in Streptomyces avermitilis NRRL8165.

The flanking fragments of the whiE(a) gene cluster was PCR amplified, cloned and used to construct the gene replacement plasmid pHL643. pHL643 was conjugated into Streptomyces avermitilis NRRL8165 followed by screening for double crossover event, yielding three apramycin resistance and thiostrepton sensitive isolates named ZJ1, ZJ2 and ZJ3, which were deficient in biosynthesis of the grey spore pigment. The whiE(a) gene replacement of these isolates was confirmed by Southern hybridization. Fermentation of the mutant strains in shaking flasks and HPLC analyses showed that the production of avermectins increased by 47% compared with that of the wild type, indicating that the spore pigment biosynthesis competes with the avermectins biosynthetic pathway.
