1.Clinical efficacy and safety of atorvastatin on atherosclerosis in advanced elderly patients

Objective To investigate the effect and safety of long-term use of atorvastatin on carotid atherosclerosis in advanced elderly patients.Methods Totally 61 elderly patients with carotid atherosclerosis were permitted into study.They were treated with atorvastatin (20 mg/d) and followed-up for five years.The carotid atherosclerosis parameter and the serum levels of TC,TG,LDL-C,HDL-C,fasting blood glucose (FBG),ALT,AST,STB,CB,Bun,Cr and creatine kinase (CK) were measured before and 5 years after treatment.Results During fellow-up,2 patients died of severe pneumonia and 1 patient died of operation complications of intestinal obstruction,with mortality rate of 4.9%.2 patients (3.4%) gave up treatment because of muscle-related side-effect.Data was collected in 56 patients achieving study end.There was significant decrease in LDL-C (P<0.05) and increase in HDL-C (P<0.05),but there was no change in serum levels of TC,TG,FBG,ALT,AST,STB,CB,BUN,Cr and CK (P<0.05) after 5 years.The total area of carotid atherosclerosis plaque was reduced [(0.63±0.41)cm2 vs.(1.07±0.67)cm2,P<0.05] and echo of carotid atherosclerosis plaque was enhanced (2.67±0.52 vs.1.53±0.44,P<0.01),but carotid intima-media thickness(IMT) appeared no change after five years.Conclusions Atorvastatin is safe in the treatment of advanced elderly patients,it can make carotid atherosclerosis plaque smaller and stable after long-term treatment.
2.Protective effect of atorvastatin on blood brain barrier in rats with focal cerebral ischemia-reperfusion injury

AIM:To explore the protective effects of atorvastatin on blood brain barrier (BBB) in cerebral is-chemia-reperfusion ( IR) injury and the potential mechanisms involved .METHODS: SD rats were divided into sham group, IR group and atorvastain group .Intraluminal suture method was used to establish cerebral IR model , and the ische-mic brain was reperfused for 72 h after the occlusion.The rats in atorvastatin group were administered with atorvastatin (20 mg? kg-1? d-1 ) by gavage once a day for 3 consecutive days after operation .At 72 h after reperfusion , neurological func-tion scores, the water content of the brain tissue , Evans blue (EB) content of ischemic hemisphere , the expression of tight junction ( TJ )-associated protein occludin and inflammation factor phosphatidylinositiol 3-kinase-p110 gamma ( PI3K-p110γ) were tested and analyzed .RESULTS: In IR group, the rats showed elevated neurological function scores ( P<0.01), brain tissue water content (P<0.01) and EB content (P<0.01), accompanied with the down-regulation of oc-cludin expression (P<0.01) and up-regulation of PI3K-p110γ(P<0.01) at 72 h after reperfusion.Compared with IR group, decreased brain edema (P<0.01) and EB leakage (P<0.01) were observed in atorvastatin group , accompanied with increased occludin expression (P<0.01) and decreased PI3K-p110γexpression (P<0.01).However, no statistical difference of the neurological function scores between the 2 groups was observed .CONCLUSION:Atorvastain attenuates cerebral IR injury , which may be associated with the inhibition of inflammatory reactions and the up -regulation of TJ-asso-ciated proteins to maintain the stability of BBB .

4.Evaluation of the cytotoxicity of four dental alloys on human gingival fibroblasts

Objective:To identify the cytotoxicity of four dental casting alloys on human gingival fibroblasts.Methods: MIT assay method was used to evaluate the cytotoxicity of four alloys on human periodontal ligament fibroblasts(HPDLFs) isolated from healthy periodontal ligaments of subjects on the 2nd d,4th d and 7th d of culture.Flow Cytometry(FCM) was used to evaluate the cytotoxicity of four alloys to human gingival fibroblasts.Results: It was found that there was no apparent cytotoxicity of all the four alloys.Conclusion:MTT and FCM methods show the same results.These four alloys have good biocompatibilities.
5.Relation between parenting style and self-congruence in Tibet middle school students

Objective:To explore the characteristic of parenting style and its relationship with self congruence in Tibet middle school students.Methods:Totally 1149 Tibet students in 2 middle schools were investigated by the Scale of EMBU (Egma Minnen av Bardndosnauppforstran)and SCCS (Self Consistency and Congruence Scale) from counties resided mainly by Tibet people.Results:The total average scores of Tibet students'self-congruence were lower than the median[(2.8 ± 0.3) < median 3].There was no significant difference between genders [(2.6 ±0.3)vs.(2.7 ±0.4),P >0.05],and the scores of self flexibility had statistical significant difference in gender [(2.9 ±0.5)vs.(2.8 ±0.5),P <0.05].The scores of parenting style in positive elements were negatively correlated with self congruence in Tibet middle school students (r =-0.20,P <0.01 ).Multi-linear regression showed that the element of maternal emotion and understanding had negative relevance with SCCS(β=-0.16,P <0.01),while the element of maternal rejection and denial had positive relevance with SCCS(β=0.16,P <0.01) .Conclusion:It indicates that the level of self-congruence is higher than median,and that parenting style might have a certain relationship with self congruence in Tibet middle school students.
6.Outpatient experiences of caring in the outpatient clinic:a qualitative research

Objective To explore the outpatients′experience of caring and provide references for improving their experience of medical care.Methods The phenomenology research method was adopted in this study,and twenty-one outpatients were subject to an in-depth interview to learn their outpatient experiences.Results Ten themes of experience with caring were extracted as follows:passionate and patient,careful and considerate,respect for others,offers to help,encouraging and comforting,apologies and courteously,humor,truthful considerations,conscientious and responsible,and a caring environment setup.Five themes of experience without caring were extracted as follows:impatient communication, careless seeing of outpatients,delayed service,passive reminders,and facilities without enough details. Conclusions The outpatients may have abundant experiences of caring at the outpatient clinic,also lack of caring sometimes as well.Outpatient managers should keep an eye on outpatients′needs and satisfaction of caring from time to time,to improve the quality of outpatient service.

8.PTEN protein loss is associated with an increased risk of recurrence in Chinese patients after prostatectomy for clinically localized prostate cancer

Background and purpose:Loss of the tumor suppressor phosphatase and tensin homolog deleted on chromosome 10 (PTEN) is one of the most common somatic genetic aberrations in prostate cancer in Western countries and is frequently associated with tumor progression and poor prognosis. This study aimed to investigate the frequency of PTEN protein loss in Chinese prostate cancer patients and to determine its association with the biochemical recurrence of prostate cancer.Methods:The data from 225 diagnosed localized prostate cancer patients with radical prostatectomy from 2006 to 2011 were collected retrospectively, including patient’s age at diagnosis, prostate-speciifc antigen (PSA) level at diagnosis, Gleason score, clinical stage, surgical margin, and time to biochemical recurrence or not. This study performed PTEN protein immunohistochemistry on tissue microarrays, which were made from 225 Chinese prostate cancer patients mentioned above, treated by radical prostatectomy with one case including 2 cancer spots and 2 adjacent normal gland spots. Correlations of PTEN loss with clinicopathological features were analyzed usingχ2 test. Kaplan-Meier survival model and Cox proportional hazards regression model were used to evaluate the predictive role of PTEN protein expression and patient characteristics for biochemical recurrence. Results:PTEN protein loss was observed in 15% of the patients and was associated with increased preoperative PSA levels (P=0.03) and old age (P=0.009). In univariate Kaplan–Meier analysis, the factors associated with the biochemical recurrence of prostate cancer included PSA levels (P=0.000 4), Gleason sum (P=0.019 8), and PTEN status (P=0.013 1). In multivariable Cox regression analysis, PTEN expression (HR=0.536, P=0.044), PSA levels (HR=1.879, P=0.001), and Gleason score (HR=1.361,P=0.03) were signiifcant in predicting biochemical recurrence of prostate cancer.Conclusion:PTEN protein loss is associated with an increased risk of recurrence, independent of known clinicopathological factors.
9.Reversal effect of FG020318 on multidrug resistance in retinoblastoma cell line SO-Rb50

AIM: To investigate the multidrug resistance(MDR),reversal activity of 2-[4-(2-pyridin-2-yl-vinyl) henyl]-4,5-bis-(4-N,N-diethylaminophenyl)-1(H)-imidazole(FG020318) in a retinoblastoma subline SO-Rb50/VCR,which is resistant to vincristine.METHODS: The procedure of stepwise increase in drug concentrations was used to obtain SO-Rb50/VCR,which was resistant to 200 ?g/L vincristine. The chemosensitivity of this drug resistant cell line with and without FG020318 or cyclosporine A(CSA) were detected by MTT assay and the function of p-glycoprotein(P-gp) was examined by rhodamine 123 accumulation detected with flow cytometry(FCM).RESULTS: FG020318(2.5 ?mol/L) significantly reduced IC50 and increased the rhodamine accumulation in a concentration-dependent manner. It was much stronger than the positive control CSA in reversal of MDR.CONCLUSION: A new tumor MDR modulator FG020318 partly reverses MDR in SO-Rb50/VCR.It may be a promising new drug to tackling MDR.
10.Study on Antioxidant Activities of Ethanol Extract of Paeonia lactiflora and Different Polar Parts in vitro

OBJECTIVE:To study antioxidant activities of ethanol extract of Paeonia lactiflora (CREt) and different polar parts in vitro. METHODS:CREt was prepared with 95% ethanol. CREt (extracted by 50% ethanal) was extracted with aether petrolei,acetic ether and n-butyl alcohol to obtain aether petrolei part (CRP part),acetic ether part (CRE part),n-butyl alcohol part(CRB part)and water part(CRW part). The ability of CREt and different polar parts eliminating DPPH·,O2-· and ·OH were investigated (mass concentrations of CREt and different polar parts were respectively 0.75-12,0.5-6,1.25-15 mg/ml in above 3 tests) and compared with ascorbic acid (VC,0.2 mg/ml) group. RESULTS:The maximum elimination rate of CREt to DPPH·, ·OH and O2-· were(97.55±0.25)%,(81.45±0.20)% and(75.28±0.41)%,IC50 were 1.629,1.789 and 5.268 mg/ml;those of CRE part to those radicals were (82.54 ± 0.36)%,(77.74 ± 0.42)% and (72.16 ± 0.73)%,IC50 were 2.481,1.918 and 6.005 mg/ml;that of CRB part to ·OH reached to(62.53±0.83)%,IC50 was 7.232 mg/ml,but to DPPH·、O2-· were less than 45%;those of CRP part and CRW part to those radicals were all lower than 40%. Each part could eliminate 3 radicals in dose-dependent manner,but were all poorer than VC group,with statistical significance (P<0.01). CONCLUSIONS:The CREt and CRE part show strongest antioxidant activities in vitro,and other parts have weak antioxidant effect.
