2.Clinical Observation of Medicinal Thread Moxibustion plus Chinese Medicinal Washing for Acute Eczema

ObjectiveTo observe the efficacy and safety of medicinal thread moxibustion plus Chinese medicinal washing in treating acute eczema.MethodEighty patients with acute eczema were randomized into a treatment group and a control group, 40 in each group. The control group was externally treated with 3% boric acid solution, while the treatment group was by medicinal thread moxibustion at acupoints from Zhuang Medicine, including Changzi point, Kuihua point, and Jumei point, plus Quchi (LI11), Shousanli (LI10), and Xuehai (SP10), in association with Chinese medicinal washing. The clinical efficacies were then analyzed. ResultAfter 2 treatment courses, the total effective rate was 100.0% in the treatment group versus 77.5% in the control group, and the difference was statistically significant (P<0.05), indicating that the total effective rate of the treatment group washigher than that of the control group; besides, there was no adverse events happened in the treatment group during the whole treatment. ConclusionMedicinal thread moxibustion plus Chinese medicinal washing is efficient and safe in treating eczema.
3.Therapeutic Observation of Comprehensive Muscle-region Therapy in Chuang Medicine for Cervical Spondylosis of Neck Type

Objective To observe the clinical efficacy of comprehensive muscle-region therapy based on Chuang Medicine in treating cervical spondylosis of neck type.Method A hundred patients with cervical spondylosis of neck type were randomized into a treatment group and a control group, 50 cases in each group. The treatment group was intervened by the comprehensive muscle-region therapy based on Chuang medicine (muscle-region tuina, fire-needle acupuncture of Chuang medicine, and cupping), while the control group was by regular acupuncture treatment. The Visual Analogue Scale (VAS) score and symptoms and signs scores were observed before and after intervention, and the clinical efficacies were compared.Result The VAS scores respectively after 1 and 2 treatment courses were significantly different from that before intervention in the treatment group (P<0.05,P<0.01). The VAS score after 2 treatment courses was significantly different from that before intervention in the control group (P<0.05). After 2 treatment courses, the VAS score in the treatment group was significantly different from that in the control group (P<0.05). After treatment, the symptoms and signs scores (pain intensity, pain-affected area, number of tender points, limited motion, and general score) were significantly different from that before intervention in the treatment group (P<0.01, P<0.05). In the control group, the pain intensity, limited motion and general score after treatment were significantly different from that before intervention (P<0.05,P<0.01). After treatment, the symptoms and signs scores in the treatment group were significantly different from that in the control group (P<0.05,P<0.01). The total effective rate was 100.0% in the treatment group versus 84.0% in the control group, and the difference was statistically significant (P<0.05).Conclusion Comprehensive muscle- region therapy in Chuang medicine is an effective approach in treating cervical spondylosis of neck type.
4.Efficacy of splenectomy plus selective pericardial devascularization under en-doscope in treatment of advanced schistosomiasis patients with portal hyper-tension and hypersplenism

Objective To evaluate the efficacy of splenectomy plus selective pericardial devascularization under endoscope in the treatment of advanced schistosomiasis patients with portal hypertension and hypersplenism so as to explore the minimally in-vasive and safer surgical treatment. Methods A secure splenectomy was performed with laparoscope and its supporting devices, and at the same time,the ligation of the left gastric vein and the ligation of esophageal vein perforating vertically into the esopha-gus were also performed in 14 advanced schistosomiasis patients with portal hypertension and hypersplenism. Results Among the 14 patients,the splenic artery was separated and clipped before the treatment of splenic pedicle. One patient was of conversion to open laparotomy for the splenic vein rupture bleeding in the separation. There was no death. Conclusion The operation of sple-nectomy plus selective pericardial devascularization under endoscope is effective,truly minimally invasive,and safe in the treat-ment of advanced schistosomiasis patients with portal hypertension and hypersplenism.
5.Association Study Between the rs4373814 Polymorphisms of Calcium Channel Gene and the Susceptibility of Essential Hypertension in Han Population at Northeastern Area of China

Objective: To investigate the relationship between rs4373814 polymorphisms of calcium channel (CACNB2) gene and the susceptibility of essential hypertension (EH) occurrence in Han population at northeastern area of China. Methods: A case-control study in Han population at northeastern of China was carried out and the research included 1998 participants as 2 groups: EH group,n=1006 patients treated in our hospital, and Control group,n=992 subjects with normal blood pressure from physical examination. The rs4373814 polymorphisms were examined by Snapshot technique and the statistical analysis was performed by SPSS 19.0 software. Results: The rs4373814 risk allele C frequency of CACNB2 gene was signiifcantly different between EH group and Control group,P=0.039, OR=1.068, 95% CI (1.004-1.137). Logistic regression analysis showed that with adjusted gender, age, BMI, smoking, drinking, in dominant model, the genotype rs4373814 carring CC + GC/GG obviously increased the risk of EH occurrence,P=0.037, OR=1.260, 95% CI (1.013-1.567). Conclusion: The rs4373814 polymorphisms of CACNB2 gene are related to the susceptibility of EH occurrence in Han population at northeastern area of China.
6.Effect of human tissue kallikrein gene therapy on insulin resistance and nephropathy in type 2 diabetic rats

Objective To evaluate the therapeutic effect of recombinant adeno-associated viral vector (rAAV) expressing human tissue kallikrein gene (rAAV-HK) on insulin resistance and renal complications in tyDe2 diabetic rats. Methods Male Wistar rats were injected low dose streptozotocin and fed with high fat and sucrose diets to form type 2 diabetic model. rAAV mediated HK gene (HK group) or LacZ gene (LacZ group) were introduced to the diabetic rats, and their systolic blood pressure, fasting blood glucose and insulin, serum creatinine, urine creatinine, urine osmolarity and urine microalbumin were measured. The homeostasis model assessment of insulin resistance (HOMA-IR), urinary albumin excretion rate (UAER) and creatinine clearance rate (Ccr) were calculated. The expression of PI3-kinase p11o catalytic subunit (p110) and Akt phosphorylation on Thr-308 were detected by Western blot. The morphology of kidney wag observed. Results Delivery of rAAVHK resulted in a reduction in blood pressure at 2 weeks and the hypotensive effect lasted for the duration of the study. The HOMA-IR was significantly lower in HK group than LacZ group (4.76±0.33 vs 8.36±0.48, P<0.01) at the end of the study, fasting insulin level was reduced [(8.19±2.45 vs 13.85±3.76)mIU/L. P<0.01], but there was no significant change in fasting blood glucose [(13.09±3.01 vs 13.58±2.88)mmol/L].The phosphorylation of p11o and Akt Thr-308 were significantly decreased in skeletal muscle and liver in LacZ group and were almost corrected by HK gene therapy. The UAER and Ccr were significantly lower and urinary osmolarity were higher in HK-treated rats compared with LaeZ rats. Histological assessment indicated that the renal complication was relieved by HK gene delivery. Conclusion The rAAV-mediated HK gene delivery efficiently attenuated insulin resistance partly through PI3K/Akt pathway and diabetic nephropathy in type 2diabetic rats.
7.Investigation about nurses'congnition of reporting adverse events and errors voluntarily in Guangxi Province

Objective In order to know nurses'congnition of reporting adverse events and errors voluntarily by questionnaire,and then analyze the related factors. Methods Investigated 275 nurses from 25 hospitals in Guangxi Province by self-desinged questionnaire to know their cognition about reporting adverse events and errors voluntarily,analyzed the datum of invetigation. Results There was different ideas about reporting adverse events and errors voluntarily in nurses with different professional title and duty.Nurses'attitude was different under the different system of reporting adverse events and errors. Conclusions The safety awarness of nurses should be strengthened.The existing reporting system must be imported. It is necessary to establish a comprehensive reporting system of care mistakes and adverse events.
8.Interference of human tissue kailikrein on renal interstitial fibrosis in rats with 5/6 nephrectomy

Objective To investigate the interference and associated mechanism of hnman tissue kallikrein (HK) gene on renal interstitial fibrosis in rats with 5/6 nephrectomy. Methods Human kallikrein cDNA was packed in a recombinant adeno-associated virus(rAAV)-based plasmid vector. The rAAV-HK was produced by transfection in 293 cells. Twenty-four male Wistsr rats were divided into sham operation and operation groups. The rats with 5/6 nephrectomy were randomly divided into simple operation, control and experiment groups. The rats in experiment group received single dose rAAV-HK via the tail vein with 1×1011 pfu. Before nephrectomy and every month after surgery until the rats were sacrificed, the caudal arterial pressure was measured using tail cuff blood pressure determinator. Three months after HK gene delivery, the rats were sacrificed. The expression of HK in rats was assessed by RT-PCR, Western blot and enzyme-linked immunosorbent assay (ELISA). The pathological changes of renal interstitium were evaluated by Masson stainning, and the distribution of bradykinin B2 receptor (BKB2R) and angiotensin Ⅱ typel receptor (ATIR) was examined by immunohistochemistry. The expressions of BKB2R, AT1R, p-MAPK protein in renal tissue were detected by Western blot. Results Three months after HK gene delivery, the systolic blood pressure of experiment group was significantly decreased compared with the control group [(163±13) nun Hg vs (217±16) mm Hg, P<0.01](1 mm Hg=0.133 kPa). Compared with sham rats, the rats in simple operation group and control group had much more renal interstitial collagen deposition and more serious fibrosis performance, but renal interstitial collagen deposition and fibrosis were significantly ameliorated in the rats of experiment group. In addition, the tubulointerstitial injury index of HK transgenic rats was significantly lower than that of the rats in control group (1.33±0.73 vs 3.01±0.62, P<0.01). Up-regnlating expression of bradykinn B2 receptor protein and down-regulating expression of AT1 receptor and p-MAPK protein were found in renal tissues of experimental group after three months (P<0.05). Conclusion HK gene delivery significantly alleviates renal interstitial fibrosis in rats with 5/6 nephrectomy through regulating the expression of bradykinin B2 receptor, AT1 receptor and p-MAPK in renal tissue.
9.The effects of venom peptide liquor on adjunctive arthritis in mice

Objective To investigate effect of the venom peptide liquor on adjunctive arthritis (AA) in mice .Methods The male Kunming mice were randomly divided into 6 groups ,named as the control group ,model group ,positive control group(1 mg/kg dex-amethason) ,wine group(10 mL/kg) ,low dosage of venom peptide liquor group(8 .3 mL/kg) and high dosage of venom peptide liq-uor group(33 .2 mL/kg);the right toes ,ankle diameter and whole body weight were measured at 7-day intervals ;the spleen index , serum level of circulating immune complexes(CIC) were determined after the thirty-fourth day when the mice was put to death .Re-sults The level of CIC in AA mice decreased significantly (P<0 .05) ,the decrease of spleen index and the reduction of toe and an-kle swelling in the low dose group were significant(P<0 .01) .Conclusion The venom peptide liquor exhibited apparent inhibitory effect on AA in mice .
10.Establishment and evaluation of a mouse model of circulating hepatoma cells

Objective To establish a mouse model of circulating tumor cells (CTCs) by applying mouse hepatoma Hapa 1-6 cells.Methods 108 healthy male C57BL/6 mice were randomly divided into 3 groups according to their body weights.Hepa 1-6 cell suspension was intravenously injected to each mouse in the three groups at a concentration of 1×106,5×106 and 1×107/mL,0.2 mL per mouse,respectively.Blood samples were collected from the mice on the 1st,5th,9th,13th,17th and 21st days after tumor cell injection.The number,ratio and relative inhibition rate of CTCs were calculated in 20,000 nucleated cells.The mortality of mice was recorded.②80 male C57BL/6 mice were averaged into 2 groups according to their body weight: control and sorafenib tosylate groups.0.2 mL of Hepa 1-6 single cell suspension was injected to each mouse through the caudal vein at a concentration of 5×106/mL.The mice were gavaged with sorafenib tosylate (50 mg/kg) for 21 days and blood samples were collected at the 3rd,8th,15th,and 21st days for CTC assessment.Results For the 1×106/mL group,the CTC inhibition rate was 25.1%,18.1%,8.9%,4.4%,2.9% and 0.3% on the 1st,5th,9th,13th,17th and 21st days,respectively,and all the mice were alive.For the 5×106/mL group,the CTC inhibition rate was 40.4%,35.4%,15.4%,9.0%,6.6% and 4.1% on the 1st,5th,9th,13th,17th and 21st days,respectively,and all the mice were alive.For the 1×107/mL group,the CTC inhibition rate was 39.1% and 33.5% on the 1st and 5th days,respectively.Some mice died immediately after intravenous injection and all mice died within 7 days.②The relative clearance of CTCs was-7.5%,4.6%,55.3% and-94.5% on the 3rd,8th,15th and 21st days of sorafenib tosylate administration.Compared with the control group,there were significant differences among the three groups (P<0.05 or P<0.01).Conclusions A mouse model of circulating hepatoma cells has been established by intravenous injection of 0.2 mL of 5×106/mL mouse Hepa 1-6 cell suspension.This mouse model can be used for screening and evaluation of drugs for circulating tumor cell inhibition.
