Gap junctions are membrane structures made of intercellular channels that allow directly exchange of material and information between adjacent cells. It is the structural basis of gap junctional intercellular communication. Gap junction is aberrant frequently in variety of cancers, which relatives with tumor genesis and metastasis. New anti-cancer drugs targeting gap junctions will be a new direction for tumor treatment.
2.Expression and significance of interleukin-34 level in patients with Henoch-Schoenlein purpura

Objective:To investigate the serum level of IL-34 in patients with acute phase Henoch-Schoenlein purpura ( HSP) and its potential significance .Methods:38 HSP patients were recruited ,together with 21 healthy volunteers as control .The serum level of IL-34,IL-6 and TNF-αwere determined using enzyme-linked immunosorbent assay (ELISA) or chemiluminescence.The correlation analysis was performed to explore the correlation of IL-34 with clinical parameters and the serum level of IL-6,TNF-α.Results:The se-rum levels of IL-34,IL-6 and TNF-αin HSP patients were significantly higher as compared to the control group (P<0.001).The serum level of IL-34 was positively correlated with that of high sensitive C-reactive protein ( hs-CRP) and IL-6 ( r=0.453 ,P=0.004 and r=0.469,P=0.003,respectively).Conclusion:IL-34 might be involved in the pathogenesis of HSP ,and the potential mechanisms of ac-tion is associated with the aberrant expression of pro-inflammatory factors in HSP ,which promote the development of inflammation in the vascular system.
3.Analysis of VOCs in the headspace of SHI-1 acute mononuclear leukemia cell

Objective To find out the impact of SHI-1 cells on VOCs in the headspace, and to assess the feasibili-ty of VOCs analysis in leukemia patients or with other tumors in the differential diagnosis by determing VOCs in the headspace of acute mononuclear leukemia cells(SHI-1). Methods The air samples from the headspace of SHI-1, Human macrophages cells and medium control were collected by syringes, and then determined by means of solid phase microextraction-gas chromatography/mass spectrometry ( SPME-GC/MS ) in order to have a better under-standing of the concentration distributions and changes of VOCs in SHI-1 cells' headspace. Results Using Mann-Whitney U test, we found that eight kinds of VOCs were different among them. They were 2,4-dimethyl heptanes, 4-methyl octane, chloroform, benzene, 3,7-dimethyl dodecanese, hexanol, cyclohexanol, cetane. And alkane, benzene were significantly higher than the control group ( P<0.05 ) , alcohol decreased significantly ( P<0.05 ) . Conclusion SHI-1 cells have an impact on the concentration distributions of VOCs in their headspace. The most outstanding VOCs are alkanes, alcohol and benzene compared with the blank control group. They may be potential markers characteristic of leukemia cells.
4.SUBACUTE ORAL TOXICITY STUDIES OF BCDMH EFFERVESCENT TABLETS FOR HOS-PITAL SEWAGE

Objective To study subacute oral toxicity of a disinfectant for hospital sewage which contained 1-bromo -3 -chloro -5, 5 -dimethylhydantoin (BCDMH) in order to know about its toxicological safety .Methods Animal test and biochemical examination methods were adopted to evaluate its subacute toxicity .Results During the subacute toxicity test, none of the rats died.At the end of the experiment , urine routine and hematological exami -nation were in the normal ranges .There were no difference compared with the control (p >0.05).But serum alanine aminotransferase in both sexes as well as creatinine in male SD rats of the high dose group were increased obviously compared with the control (p <0.05).Mild steatosis and spotty necrosis of liver cells were found in the high dose group during the pathological examination .Conclusion The BCDMH effervescent tablets'no -observed -adverse -effect -level (NOAEL) is 84 mg? kg-1 for male SD rats, and 98 mg? kg-1 for female SD rats.

7.Application of automated high-performance liquid chromatography in the diagnosis of thalassemia

Objective To evaluate the application of high-performance liquid chromatography (HPLC) in diagnosis and screening of thalassemia. Methods Automated HPLC was used to measure HbF and HbA2 in 100 genetically diagnosed thalas-semic patients and 35 normal children. The results were compared with those from traditional tests including alkali denaturation test and cellulose acetate electrophoresis. The diagnose accordance rates, sensitivity and specificity were compared. Results Seventy-fourβthalassemia, 64 were heterozygous with single mutations and 10 were compound heterozygous with double muta-tions. Twenty-sixαthalassemia, 25 were compound mutations and one was heterozygous with single mutation. The HbF percent-age from HPLC was higher than that from alkali denaturation tests in either thalassemia or normal children (P<0.01). HbF level from HPLC inα-thalassemia was signiifcantly different from that in the normal children (P=0.011). The percentage of HbA2 from HPLC was higher than that from cellulose acetate electrophoresis (P=0.010). HbA2 in the single heterozygousβ-thalassemia were twice higher than that in the double heterozygous mutatedβ-thalassemia (P<0.01). The combination of HbF-HbA2 (≥4.0%) from HPLC with MCV (<80 lf) and MCH (<27 pg) had high accordance rates (99.3%), sensitivity (99.0%) and speciifcity (100.0%) in diagnosis of thalassemia. Conclusions When the results of HPLC are combined with MCV and MCH, it can be applied to the diagnosis of thalassemia with high speciifcity, high sensitivity and has high diagnostic accordance rate with genetic results. HPLC can be an ideal approach to screenβthalassemia.
8.Attenuated store-operated Ca2+ entry in mesenteric artery endothelial cells of aged rat

Objective To observe attenuated store-operated Ca2+ entry in superior mesenteric artery endothelial cells of aged rat.Methods The pressure myograph was applied to measure the vasodilation of superior mesenteric artery induced by bradykinin (BK) in aged versus non-elder rats.Primarily cultured MAECs were treated by thapsigargin (TG) and BK to deplete intracellular Ca2+ stores for comparing SOCE.SOCE of MAECs was detected by calcium imaging technology and was compared between aged and non-elder rats.The expression levels of Orai 1 and stromal interaction factor 1 (STIM 1) were observed by immunofluorescence method.Results Compared with the nonelder rats (70.0 ± 4.1 %),BK-induced vasodilation in aged rats (27.0 ± 4.9)% was declined by (60.7±4.3) %.SOCE induced by BK and TG in primarily cultured MAECs was attenuated by 37.6% and 39.2% in aged rats (1.71±0.18 and 1.06±0.03) respectively as compared with non-elder rats (2.72±0.39 and 1.75±0.06).The expressions of SOCE's components Orai 1 and STIM 1 in MAECs were decreased obviously in aged rats than in non elder rats.Conclusions SOCE of MAECs and SOCE-induced vasodilation are significantly attenuated in aged rats.The decreased expression level of Orai 1 and STIM 1 may contribute to this alteration.
9.Bibliometric analysis of the literature on nursing professional basic experiment research in China ten years

Objective To describe the research status, analyze the existing problems and summarize development trend of basic experiment research in China by analyzing the papers publication year, publication journals, geographical distribution, fund status, cooperation situation and the main experimental methods or technologies.It was aimed at providing guidance for further research and development of nursing scientific research.Methods All literatures related to nursing professional basic experiment research were extracted by searching through the China National Knowledge Infrastructure, VIP Database for Chinese Technical Periodicals and Wan-fang database from January 2005 to November 2014.Bibliometric analysis methods were used to analyze the search results.Results 472 references were retrieved to meet the requirements.about 76.3% (360/472) papers were published in the statistical source journals, 10.0% (47/472) of the literatures were published in the nursing professional journals;68.9% (325/472) of the literatures had funding source, 21.0% (99/472) for national fund support, 35.4% (167/472) were supported by provincial fund;literature cooperation degree was 4.38(2 065/472), the co-authored rate was 97.2% (459/472).The level of research focused on tissue and cells, a few involved in molecular level.Conclusions All the literatures on the nursing professional basic experiment research show a increa~d trend year by year.Nursing professional journals' publication volume is not high for such research.Scientific research fund contributions remain to be improved.The interdisciplinary cooperation needs to be expanded.Nursing experimental research will provide a possible for discipline development.
10.Application value of TEE on occlusion of secondum atrial septal defect in adult patients

Objective:To explore the application value of transesophageal echocardiography (TEE)in secondum atri-al septal defect (ASD)intervention and minimally invasive occlusion surgery in adult patients.Methods:A total of 134 cases adult patients (>15 years)with pure secondum ASD (including 91 cases undergoing interventional occlu-sion and 43 cases undergoing minimally invasive occlusion surgery),who were screened via transthoracic echocardio-graphy (TTE)and TEE from Jan 2012 to Dec 2012,were selected.Intervention group and minimally invasive sur-gery group received TTE and TEE monitoring respectively during operation.Preoperative TTE and TEE parameters and operation results were compared and analyzed.Results:The parameters of ASD diameter measured by TEE were significantly higher than those measured by TTE in two groups (P <0.01 all).Maximum defect diameter of ASD measured by TEE was significantly higher than that of measured by TTE [(19.8±5.2)mm vs.(18.7±4.9)mm], P <0.01. The correlation between maximum defect diameter measured by TTE and occluder size (intervention group r =0.926,minimally invasive surgery group r =0.215)was all lower than that of TEE (intervention group r=0.965,minimally invasive surgery group r =0.627),P <0.01 both.Conclusion:TEE is better than TTE in as-sessment of ASD size.It should be regarded as a routine preoperative screening method for adult ASD patients.It has important value in real-time monitoring,guidance and immediate evaluation of therapeutic effects during mini-mally invasive ASD occlusion surgery.
