2.Expression of estrogen receptor and progesterone receptor and their significance in the endo-metrial tissue of intrauterine adhesion patients

Purpose To investigate the expression of estrogen receptor ( ER) and progesterone receptor ( PR) in endometrial tissue in patients with intrauterine adhesion ( IUA) and their clinical significance. Methods Two experimental methods, immunohistochemical MaxVision two step method and real-time fluorescencequantitative PCR ( qRT-PCR) , were used for detection of ER and PR in endome-trial tissue both in group IUA ( study group) and non IUA group ( control group) . Results MaxVision immunohistochemical method showed that ER protein expression in the study group (3. 52 ± 0. 71) was significantly higher than that (2. 75 ± 1. 00) in the control group (P=0. 01), ER mRNA expression by qRT-PCR was also significantly higher in the study group (1. 59 ± 0. 26) than that (1. 00 ± 0. 19) in the control group (P=0. 00). The immunohistochemical detection of showed that PR protein expression in the study group (3.26 ±0.70) had no significant different from that (3.58 ±0.28) in the control group (P=0.12), qRT-PCR also showed that ER mRNA in the study group (1.15 ±0.21) had no significantly different from that (1.00 ±0.31) in the control group (P=0. 21 ) . Immunohistochemical expression of ER and PR proteins showed a significant difference between the endometrial glands and en-dometrial stroma (χ2 =5. 797, P=0. 016,χ2 =4. 857, P=0. 027). Conclusion The expression of ER in endometrial tissue in the study group is higher than that in the control group, but the expression of PR has no different between the two groups, ER and PR pro-teins expression are higher in endometrial glands than that in endometrial stroma. These provide a theoretical reference for clinical use of estrogen and progesterone to IUA patients.
3.cDNA Cloning, Expression Analysis of Slc24a5 and Its Relationship With Melanin Deposition in Chicken

Melanin is a kind of biopolymer, composed of a complex quinine/indole-quinone derived mixture which is produced in melanocytes from tyrosine. More than 100 genes related to melanin deposition have been found in mouse. Slc24a5 (solute carrier family 24, member 5) is a novel gene first cloned in zebrafish. And it has been confirmed that the Slc24a5 is involved in controlling the melanin deposition in zebrafish. Here the full length of the chicken slc24a5 mRNA sequence, its expression profile and a discussion of its relationship to melanin deposition in chicken were reported. Chicken Slc254a5 has a CDS of 1 269bp, predicting a protein of 423 amino acids which is about 80 amino acids shorter than in human and mouse. It has 9 exons and 8 introns and spans more than 1lkb of genome sequence. The quantitative real-time PCR confirmed that the chicken Slc24a5 was highly expressed in the eye of White Leghorn and Beijing Fatty Chickens, and in the eye, skin and muscle of Silky. The relative expression in Silky eye is more than two times that of White Leghorn. The relative expression in Silky skin is about 70 times that of White Leghorn and about 15 times that of Beijing Fatty Chickens. The relative expression in Silky muscle is about 15 times that of White Leghorn and about 3 times that of Beijing Fatty Chickens. And the expression result is in accordance with the melanocyte and melanin deposition in these tissues.
4.Clinical Significance of Measurment of D-Dimer and High Sensitive C-Reactive Protein in Serum for Acute Cerebral Infarction

Objective To explore the relationship of D-Dimer and high sensitive C-reactive protein in serum in acute cerebral infarction. Methods 64 patients with acute cerebral infarction were measured with the D-Dimer and Hs-CRP and compared with that of 50 healthy subjects as the controls. Their scores of National Institutes of Health Stroke Scale (NIHSS) were recorded and the correlation with the D-Dimer and Hs-CRP level was analyzed. Results Compared with those in controls, the levels of D-Dimer and Hs-CRP in patients with acute cerebral infarction were significantly higher (P<0.05), and those in patients with large infarction area were significantly higher than those in patients with small infarction area (P<0.01). The levels of D-Dimer and Hs-CRP uncorrelated with the scores of NIHSS.Conclusion The D-Dimer and Hs-CRP level is helpful to assess the acute cerebral infarction.

7.Immunological relationship between food allergy induction and occurrence of asthma in mice

Objective To investigate immunological relationship between early induction of food allergy and occurrence of later asthma in mice, and explore the pathological changes in lung tissues. Methods Thirty-seven female BALB/c mice aged 6 weeks were randomly divided into blank control group ( n = 12), food allergy group ( n = 13) and asthma group (n = 12). After being challenged by ovalbumin (OVA), the levels of serum IgE, IL-4 and INF-γ in bronchoalveolar lavage fluid ( BALF) were detected. The numbers of inflammatory cells and eosinophils ( EOS) in BALF were counted. Lung tissues were obtained for pathological sections, and thickness of bronchial wall and EOS infiltration were observed. Results The level of serum IgE and level of IL-4, ratio of IL-4/IFN-γ and number of EOS in BALF in food allergy group and asthma group were significantly higher than those in control group (P <0.05). The level of IL-4 and number of EOS in BALF in asthma group were significantly higher than those in food allergy group (P <0.05) , while there was no significant difference in serum IgE level between these two groups (P > 0.05), and levels of IFN-7 in BALF in both groups were significantly lower than that in blank control group (P<0.05). There were more EOS infiltration in lung tissues and thicker bronchial wall in food allergy group and asthma group than that in blank control group (P < 0.05), and the number of EOS in asthma group was significantly higher than that in food allergy group ( P < 0.05). Conclusion IgE-mediated immune response is involved in both food allergy and asthma mouse models. Lung immune imbalance of Thl/Th2 and inflammatory cell infiltration caused by food allergy may participate in the occurrence of later asthma.
8.Cloning, Expression and Purification of Gene Encoding Human ?-NGF in E.coli

Nerve growth factor is one of the most important factors playing an important role in regulating the growth, development and survival of the neuron. The purified NGF from human placenta has been reported, the tissue from which can be isolated the NGF is very limited. It is important for basic research and clinic application to expression hNGF by genetic engineering. By polymerase chain reaction,gene fragment encoding the mature part of ?-NGF was amplified using the DNA of human placenta as template. The fragment was sequenced and inserted into expression vector pET-15b, and the recombinant expression vector pET15b-NGF was transformed into E.coli BL21(DE3)pLysS. After inducing with IPTG the NGF was higher expressed up to 25% of the total cell proteins. The expression product was purified with metal chelate affinity chromatography on Ni-IDA agarose under denaturing condition. The purity of rNGF was higher than 90% and yield of rNGF was 4.56mg/L expressing bacteria. SDS-PAGE revealed the NGF expression product had a Mr 16kDa. Western-blot displayed the recombinant product had strong immunological activity with rabbit anti-human ?-NGF polyclonic antibodies. The expression products were dealed with solubilizing inclusion bodies and refolding protein. The test of nerve fiber growth of chicken embryo DRG neurons displayed rNGF had biological activity.
9.Assessment of influence factors to curative effect in cool-tip radiofrequency ablation

Objective To investigate the effects of output power,action time and radiofrequency(RF) needle on the cool-tip radiofrequency ablation(RFA) by experimental tools and to determine the value of ultrasonography in size evaluation of RFA zone.Methods The cool-tip RFA to fresh calf liver were monitored by ultrasound.The experiments by single electrode needle were performed with different combination of output power (80 W,120 W) and time (5 min,8 min,10 min).The cluster needle was used for assessment at 5 min with different output power(80 W,120 W).After the end of trial,the longitudinal specimens were cut open.The view and size of the ablation zone were recorded with naked eyes.The pathological changes displayed by optical microscope were recorded as well.Results The measurement of ablation zone with naked eyes showed with the ablation zone expanded with time in 80 W-power cases,but the pace of expansion slowed down,but in 120 W-power cases,expansion of the ablation zone was not obvious; the ablation zone in 120 W-power was bigger than that in 80 W-power at 5 min,their difference decreased with time,and the ablation zones were similar at 10 min.The cluster needle can produce ablation zone with lesser aspect ratio than that of single electrode needle,consequently similar to circle.Ultrasonic measurement of the ablation zone had real discrepancy.Most of longitudinal diameters were greater than the real ones,while in large ablation lesions,vertical diameters were often less than the real ones.Under optical microscope,no change could be found in shape and structure of the cells in ablation zone.Conclusion The output power and performing time have impact on ablation.The high-power output increased heat production as well as reduction of heat conduction.Compared with single electrode needle,the cluster needle produced ablation zone closer to real hepatic tumor,thus has more reliable effect to small hepatocellular carcinoma with diameter around 2 cm.The ultrasond has a great significance in RFA guidance,but it could not accurately define the border of ablation zone.
10.Clinical Characteristics of 100 Children with Cerebral Palsy

ObjectiveTo analysis the clinical characteristics and the high risk factors of children with cerebral palsy.MethodsClinical data of 100 children with cerebral palsy were retrospectively analyzed.Results51% of children were born with asphyxia, 42% were born prematurely, 39% were low birth body weight. 85.7% of children involved had mental retardation, 12% had hearing loss and 7% had visual impairment. 88% of children involved had abnormal cranial CT results and 46.1% had abnormal EEG. 50.0% of the children had abnormal brain stem auditory evoked potentials.ConclusionAsphyxia, prematurely birth and low birth body weight are high risk factors of cerebral palsy. Most of the children with cerebral palsy in this group are mental retarded.
