1.miR-200 family and its mechanism

MiR-200 family is a cluster of miRNAs closely linked to EMT. The miR-200 family contains miR-200a, miR-200b, miR-200c, miR-141 and miR-429. Many studies have shown that miR-200 family are key regulators of EMT and determine the epithelial phenotype. They are important to both EMT development and tumor progression. This paper reviews the advancement in the research of miR-200 family and its mechanism.
2. Expression of SHH protein and its downstream transcriptional factor GLI1 in hedgehog signal pathway in peutz-jeghers syndrome

Objective To detect the protein expression of SHH and its downstream transcriptional factor GLI1 in Peutz-Jeghers syndrome (PJS) and to investigate the roles of SHH and GLI1 in the development and carcinogenesis of PJS polyps. Methods Immunohistochemical (IHC) staining was employed to detect the expression and localization of SHH and GLI1 protein in 20 PJS polyp samples, 25 adenomatous polyp tissues, 25 colon adenocarcinoma samples, and 20 normal intestinal mucosal tissues. Results IHC revealed that SHH protein and GLI1 protein were mainly localized on cell membrane and in the cytoplasm, and the nuclear staining of GLI1 protein in the colon adenocarcinoma was increased. The expression of the two proteins gradually increased in order in the normal intestinal mucosa, PJS polyp, adenomatous polyp, and colon adenocarcinoma (P<0. 001). Spearman rank correlation showed that SHH expression was significantly correlated with GLI1 expression in PJS polyp(rp =0. 503, P = 0. 024). Conclusion SHH-GLI1 is present in the tissues of normal intestinal mucosa, PJS polyp, adenomatous polyp, and colon adenocarcinoma, and the expression has an increasing tendency in the above tissues, indicating SHH-GLI1 signal pathway may be associated with the development and carcinogenesis of PJS polyp.

The metabolic syndrome leading to cerebrovascular disease attracts more and more attention by the medical circle,and its exact nosogenesis is not clear.There is no effective therapeutic method for this disease either.The author believes phlegm and blood stasis being the key factors for metaboric syndrome.Clinically,the author got good effects by treating the disease according to the different nosogenesis and stages of disease.
4.Autologous peripheral blood stem cell transplantation for progressive multiple sclerosis

Objective To explore the clinical effect of treatment for progressive multiple sclerosis(PMS)by autologous peripheral blood stem cell transplantation(APBSCT).Methods Between September 2001 and July 2004,thirteen patients with PMS were treated with APBSCT in Xuanwu Hospital.Peripheral blood stem cells (PBSCs)were mobilized with G-CSF alone.The stem cell transplants were pured of lymphocytes in 7 patients.The high dose regimen was BEAM conditioning regimen (Carmustine,Etoposide,Cytosine Arabinoside,and Melphalan).The median follow-up time was 22 (3~36)months.Results The result showed that the mean EDSS rating score of patients in 18 months after APBSCT was significantly decreased[(4.05?0.66) vs(6.00?0.30),P
5.Quality standard for Compound Yiqi Granules

AIM: To establish the quality standard for Compound Yiqi Granules (Radix Astragali, Radix Achyranthis Bidentatae, etc.). METHODS: Radix Achyranthis Bidentatae; Herba Epimedii were identified by TLC. The content of astragaloside IV in Compound Yiqi Granules was determined by TLCS. RESULTS: The methodological study showed a good linear correlation at the range of 1.12-5.60 ?g of astragaloside IV. The average recovery of astragaloside IV was 97.69% and RSD was 2.10%, respectively. CONCLUSION: The method is simple, accurate and with good repeatability. The method can be used for quality control of Compound Yiqi Granules.

Gonorrhea is a sexually transmitted disease caused by Neisseria gonorrhoeae,and substantially harms human health and socioeconomic development.Due to inappropriate treatment and the presence of drug resistance genes in patients,antibiotic resistance has emerged in Neisseria gonorrhoeae,such as resistance to penicillin,tetracycline,ciprofloxacin,or other antibiotics.Currently,extended-spectrum cephalosporins (ESCs) are the first-line treatment of gonococcal infection.With the wide use of ESCs,the sensitivity of Neisseria gonorrhoeae to ESCs has been decreasing gradually,and there have been reports on cases of treatment failure in clinical practice.In order to control gonorrhea and deal with drug resistance in Neisseria gonorrhoeae,combined therapy,alternative therapy and new drugs have been developed in clinic.
7.Role of long non - coding RNA in the pathogenesis of diabetic retinopathy

Long non-coding RNA ( LncRNA) is a class of transcript (>200 nucleotides) that do not encode proteins. It plays an important role in epigenetic regulation and gene expression at transcriptional or post transcriptional level. The abnormal expression of LncRNA may lead to various pathological processes. Diabetic retinopathy ( DR ) is a multifactorial disease. Recent studies have shown that many specific expressions of LncRNAs are closely related to the genesis of DR. In this review, we summarized the recent advances in the function of LncRNA, the regulatory mechanisms of LncRNA involved in the development of DR, and the related therapies.
8.The value of flow cytometry in diagnosis and subclassification of non-Hodgldn lymphoma

Objective To evaluate the utility of flow cytometry (FCM) in diagnosis and subclassification of non-Hodgkin lymphoma (NHL). Methods The samples of lymph nodes biopsy from 59 cases clinically suspected of NHL were detected by flow cytometry; and clonal lymphocytes and their immunophenotypes were identified analyzed. The concordance between the results of flow cytometry and histopathology was analyzed. Results Among the 59 cases, flow cytometry was able to identify aberrant clonal lymphocytes in 24 of 28 NHL cases identified by histopathology, the neoplastic lymphocytes ranged from 4.28 % to 89.10 %; 23 cases were diagnosed as B-NHL and 1 case was diagnosed as T-NHL. Compared with histopathology, the accuracy of FCM was 85.71% in diagnosis of NHL. The specificity and sensitivity of FCM was 100 % and 92% in diagnosis of B-NHL. The accuracy of flow cytometry immunophenotyping in classification of 24 cases of NHL was consistent with that of histopathology. Conclusion Flow cytometry could be an ancillary technique in diagnosis of NHL by identifying aberrant clonal lymphocytes, and enable identification of B-NHL subtype.
9.Effect of natural killer cells expressing KIR2DS1 receptor on efficiently killing dendritic cells in vitro

Objective To study the killing effects of the killer cell immunoglobulin-like receptors (KIR)2DS1-mediated natural killer (NK) cells against dendritic cells (DCs) and analysis the expression of chemokine receptor CCR7 after KIR2DS1-positive NK cells interact with the C2 epitope DCs in vitro,and explore the mechanism of alloreactive NK cell to limit graft-versus-host disease (GVHD).Methods NK cells were separated by Rosettesep NK sorting kit from healthy donor peripheral blood and taken as effector cells.The mononuclear cells from peripheral blood of newly diagnosed acute myeloid leukemia (AML) patients,and DCs were induced as target cells.Polymerase chain reaction and sequence specific primers (PCR-SSP) genotyping techniques and direct sequencing SBT were used to detect KIR gene,HLA-Cw of the healthy donors and patients.The cytotoxic activity of KIR2DS1-positive NK cells or KIR2DS1-negative NK cells against DCs were detected by CCK-8 assay.The expression of CCR7 after NK cells interacted with DCs was detected by flow cytometry.Results The purity of NK cells analyzed by flow cytometry was (94.20±1.23) %.Under same target ratio (10∶1),the killing effect of KIR2DS1+ NK cell against DCs was higher than that in KIR2DS1-NK cells (t =3.70,P < 0.05).Under same target ratio (10∶1),the cell lysis in C1/C1 group,KIR2DS1+ NK cells against C2/C2 group DCs (15.06±1.81) % was significantly higher than that against C1/C1 group (10.07±1.03) % and C1/C2 group (8.65±0.93) % DCs (F =56.368,P =0.001).KIR2DS1 + NK cells efficiently acquired CCR7 upon coculture with C2 epitope DCs.Conclusions NK cell expressing the KIR2DS1-activating receptor efficiently kill DCs,prevent GVHD,while enable NK cell acquire CCR7,obtaining migratory properties in vitro.
10.Diagnostic value of multi-slice CT in diagnosis of focal pulmonary ground-glass opacity nodules of 1 cm or less

Objective To investigate the clinical value of multi-slice spiral CT (MSCT)in the diagnosis of focal pulmonary ground-glass opacity nodules(fGGO)of 1 cm or less.Methods The MSCT examination data of 95 patients with subcentimeter fGGO was analyzed and compared with pathology results.The different pathological types of fGGP lesion size,internal solid component size,mixed type ground glass nodules (mGGO)proportion and pleural sag,lesion shape,grave leaves,burr,cavitation and the boundary conditions were compared. According to the relationship of the lesions and the surrounding blood vessels,3 types were classified,and the relationship of fGGO and blood vessel were analyzed.Results The minimal lesion was 0.41 cm of 95 cases and the maximum was 0.99 cm.There was statistically signifi-cant difference in the lesion size,solid component sizes,mGGO and lobulation occupied the percentage of the group before infiltrating and the adenocarcinoma group (P <0.05),and there was no statistically significant difference in pleural indentation,lesion shape,burr,vacuoles and boundary conditions (P >0.05).The difference was statistically significant in the leaflet occupied the percentage of the benign group com-pared with the adenocarcinoma group,and the differences of the remaining features were not statistically significant compared with the adeno-carcinoma group(P >0.05).According to the relstionship between fGGO and the vessels,patients in the benign group were type Ⅰ or typeⅡ,type Ⅲ was found in 10 cases of the group before infiltrating (25.6%),and type Ⅲ in 17 cases of the adenocarcinoma group (39.5%), the proportion of type Ⅲ of the group before infiltrating and the adenocarcinoma group was higher than that of the benign group (P <0.05). Conclusion The qualitative diagnosis of pulmonary fGGO is difficult,and the relationship between the morphology of the lesions under the MSCT and the surrounding vessels have some value for the diagnosis of the lesions.
