3.Imbalance of Th1/Th2 in patients with systemic lupus erythematosus combined with coronary heart disease

Objective To investigate the imbalance of Th1/Th2 cell response in patients with systemic lupus erythematosus (SLE) combined with coronary heart disease .Methods SLE patients ,SLE patients with coronary heart disease and healthy con-trols were enrolled and blood samples were collected .T-bet/GATA-3 ,the transcription factors of Th1/Th2 cells ,were detected by real-time PCR ;the intracellular cytokines IFN-γ and IL-4 in CD4 + T cells were stained by fluorescent antibodies and detected by flow cytometry ;the level of serum IFN-γ and IL-4 were detected by ELISA .Results Comparing with healthy control group ,the ex-pression level of Th1 transcription factor T-bet ,the introcellular secretion of IFN-γ in CD4 + T cells and the serum IFN-γ were all decreased in non-coronary heart disease patients with SLE( P < 0 .05) .Comparing with non-coronary heart disease patients with SLE or healthy control group ,the expression level of Th1 transcription factor T-bet ,the introcellular secretion of IFN-γ in CD4 + T cells and the serum IFN-γ were all increased in patients with SLE combined coronary heart disease(P< 0 .05) ;while the expression level of Th2 transcription factor GATA-3 ,the introcellular secretion of IL-4 in CD4 + T cells and the serum IL-4 were all decreased in patients with SLE combined coronary heart disease(P< 0 .05) .Conclusion There were imbalance towards Th1 cell response in patients with SLE combined coronary heart disease ,which may related to the occurrence and development of disease .
4.Analysis on the influencing factors of academic publications written by young doctors in affiliated hospital of a university

Objective By analyzing academic publications written by young doctors in a University Affiliated Hospital during 2013-2015,todescribe the characteristics and rules,and analyze the influencing factors,and to build a basis for providing better service and making effective incentive policies.Methods Materials were collected from questionnaire survey and administrative departments,SPSS software was used to analyze the relationship between factors and the numbers of academic papers.Results 214 young doctors published 486 papers in 2013-2015.Results show that people with senior titles and supervise the graduate students,as the PIs of research projects,as a research team PI,fixed time for research,belonging to mature science research team,interdisciplinary cooperation,cooperation with other units,would be able to have more publications.Conclusions Management staffs should know that research fund is important to young doctors to start and follow-up their research work;they Should help the research team to develop and mature,should help young doctors to find and integrate into the research team;ensure young doctors have fixed research time by making policy,should help young doctors to form the good habit of research;Strengthen cross disciplinary construction,help young doctors to carry out inter unit and inter professional collaboration.
5.Biomarkers of preterm infant low birth weight brain damage

Along with contemporary development of life support technology in perinatology and neo-natal intensive care,mortality of preterm infants has been reduced.However,the problems caused by the brain damage have been attracting more and more attention.The long-term outcome of prematures depends on their nervous system sequelae among the total complications.Preterm infants'brain damage mainly includes intravent-ricular hemorrhage(IVH)and periventricular leukomalacia(PVL).IVH and its complications are the main causes of early death,mental and physical developmental disorder of neonatals.Thus,the early diagnosis of brain dam-age,especially PVL,has become a serious problem that we are going to face.The scholars in or aboard hao payed close attention to these markers,such as MBP、S100B、ACT A、NF-L、NGB、MMP、IL-6、IL-10、IL-1 1 、NSE.The paper reviews these biological markers.
6.Expression of p63 and p73 Protein in Capillary Hemangioma

Objective To investigate the relationship between the expression of p63, p73 proteins and the development of hemangioma. Methods The immunohistochemical technique and quantitative image analysis were used to detect the expression of p63 and p73 proteins in 40 cases of capillary hemangioma and 20 specimens of normal skin. Results The absorbance value (mean ? SD) of p63 and p73 expression in normal skin tissue, proliferative phase of hemangioma and involuting phase of hemangioma were 0.923 ? 0.191 and 0.953 ? 0.120, 8.271 ? 1.953 and 6.408 ? 2.151, 0.920 ? 0.187 and 1.073 ? 0.516, respectively. The expression of p63 and p73 in proliferative phase of hemangioma was significantly increased as compared with those in involuting phase of hemangioma and normal skin tissue (P 0.05). Conclusions It is suggested that p63 gene is not a tumor suppressor gene but an oncogene in hemangioma and may contribute to the proliferation of endothelial cell and be associated with angiogenesis, and p73 may play an important role in the proliferation of hemangioma.
7.Application of comprehensive nursing intervention in patients with gastroesophageal reflux

Objective To investigate the effect of comprehensive nursing intervention on patients with gastroesophageal reflux disease(GERD).Methods 68 patients with GERD from January 201 2 to January 201 5 in the 1 st Affiliated Hospital of Bengbu Medical College were randomly divided into observation group (n =33)and control group(n =35).The symptom scores and life quality scores of the patients in the two groups were compared. Results 4 weeks after discharging,the symptom scores of reflux,heartburn,chest pain,belch in the observation group were (2.5 ±1 .0)points,(2.8 ±1 .2)points,(2.8 ±1 .3)points,(2.1 ±1 .2)points respectively,which in the control group were (4.8 ±1 .6)points,(4.4 ±2.1 )points,(3.8 ±1 .2)points,(3.4 ±1 .4)points respectively,the differ-ences were statistically significant(t =7.34,4.87,4.32,4.71 ,all P <0.05).1 2 weeks after discharging,the symptom scores of reflux,heartburn,chest pain,belch in the observation group were (1 .8 ±0.8)points,(2.2 ±1 .0)points, (1 .5 ±0.4)points,(1 .5 ±0.6)points respectively,which in the control group were (4.7 ±1 .4)points,(4.5 ± 1 .5)points,(3.0 ±0.6)points,(3.2 ±1 .6)points respectively,the differences were statistically significant(t =1 0.98,7.43,6.1 2,6.02,all P <0.05).Meanwhile,the symptom scores of the observation group 1 2 weeks after discharging were lower than those of the observation group 4 weeks after discharging (t =2.96,1 .86,7.55,4.62,all P <0.05).The life quality scores of the observation group were significantly higher than the control group,the differ-ences were statistically significant(all P <0.05).Conclusion Comprehensive nursing intervention can significantly improve symptoms and quality of life in patients with GERD.With the comprehensive intervention of patients under-taking,patients can benefit more.
8.Application of continuous infusion of remifentanil during induction of general anesthesia in elderly patients

Objective To study the application of continuous infusion of remifentanil combined with propofol with different velocity du-ring induction of general anesthesia in elderly patients. Methods Sixty elderly patients were divided into 4 groups randomly(n=15) and given remifentanil with continuous infusion rate of 0.1,0.15,0.2 and 0.3 ?g?kg-1?min-1,respectively.After given midazolam and propofol,remifentanil infusion started with different velocity.Three minutes later,vecuronium was given and intubation performed 2 min later.After that,propofol infusion rate was adjusted according to the changes of blood pressure and kept at 4 mg?kg-1?h-1 5 min before incision.Blood pressure(BP),heart rate(HR),intubation score(following Grant's method) and all side effects and adjuvant drugs used were recorded. Nitrites play multiple characteristic functions in invasion and metastasis of hepatic cancer cells, but the exact mechanism is not yet known. Cancer cells can maintain the malignant characteristics via clearance of excess mitochondria by mitophagy. The purpose of this article was to determine the roles of nitrite, reactive oxygen species (ROS) and hypoxia inducing factor 1 alpha (HIF-1 α) in mitophagy of hepatic cancer cells. After exposure of human hepatocellular carcinoma SMMC-7721 cells to a serial concentrations of sodium nitrite for 24 h under normal oxygen, the maximal cell vitality was increased by 16 mg x (-1) sodium nitrite. In addition, the potentials of migration and invasion for SMMC-7721 cells were increased significantly at the same time. Furthermore, sodium nitrite exposure displayed an increase of stress fibers, lamellipodum and perinuclear mitochondrial distribution by cell staining with Actin-Tracker Green and Mito-Tracker Red, which was reversed by N-acetylcysteine (NAC, a reactive oxygen scavenger). DCFH-DA staining with fluorescent microscopy showed that the intracellular level of ROS concentration was increased by the sodium nitrite treatment. LC3 immunostaining and Western blot results showed that sodium nitrite enhanced cell autophagy flux. Under the transmission electron microscopy (TEM), more autolysosomes formed after sodium nitrite treatment and NAC could prevent autophagosome degradation. RT-PCR results indicated that the expression levels of COX I and COXIV mRNA were decreased significantly after sodium nitrite treatment. Meanwhile, laser scanning confocal microscopy showed that sodium nitrite significantly reduced mitochondrial mass detected by Mito-Tracker Green staining. The expression levels of HIF-1α, Beclin-1 and Bnip3 (mitophagy marker molecular) increased remarkably after sodium nitrite treatment, which were reversed by NAC. Our results demonstrated that sodium nitrite (16 mg x L(-1)) increased the potentials of invasion and migration of hepatic cancer SMMC-7721 cells through induction of ROS and HIF-1α mediated mitophagy.

10.Preparation and Identification of Recombinant Vasoactive Intestinal Peptide

In order to prepare the recombinant vasoactive intestinal peptide (VIP) using intein mediated rapid purification system,the cDNA encoding the recombinant VIP was designed and synthesized according to the preference of E.coli,and then was cloned into the expression vector PTWIN. The recombinant plasmid PTWIN-VIP was transformed into expression host E.coli strain ER2566.The fusion protein consisting of the recombinant VIP,intein and chitin binding domain was expressed and purified by chitin affinity chromatography. The target peptide was released from the fusion protein by changing the temperature and the pH of the cleavage buffer. The molecular weight of the recombinant VIP was determined by the mass spectrometry and the results was conformity with the theoretical value. The preliminary bioactivity assay indicated that the recombinant VIP decreased the serum resistin levels significantly in LPS-induced acute inflammation. The preparation and the characterization of anti-inflammatory effects of the recombinant VIP layed the foundation for its further application.
