1.The Expression of Metallothionein-1 in Mouse Kidney

Objective To investigate the expression and cellular localization of metallothionein-1 (MT-1) in mouse kidney. Methods The adult male CD-1 mice were selected as experimental animals. The protein of MT-1 in mouse renal cortex and medulla was detected by Western blot assay. Immunohistochemistry and immunofluorescence methods were ap-plied to observe the location of MT-1 in renal tubules and collecting ducts of mouse kidney. Double-immunofluorescence and confocal laser scanning microscopy were used to observe the subcellular localization of MT-1 in the epithelial cells of re-nal tubules and collecting ducts of mouse kidney. Results The expression of MT-1 was detected in cortex and medulla of mouse kidney. The expression of MT-1was mainly found in proximal convoluted tubules and distal convoluted tubules of cor-tical labyrinth, and proximal straight tubules, distal straight tubules and collecting ducts of ray medullary and medulla of kid-ney. The strongest expression of MT-1 was found in proximal tubules, secondary in distal tubules and last in collecting ducts. The intracellular location of MT-1 was in brush border of proximal tubules and intracytoplasm, in the distal tubule epi-thelial cell cytoplasm and in the basolateral membranes of collecting duct epithelial cells. The subcellular localization was found in the lysosome. Conclusion The different locations of MT-1 expressed in proximal tubules, distal tubules and col-lecting ducts may determine different roles of them in kidney.
2.Effects of interleukin-1 ,interleukin-6 and tumor necrosis factor-α on migration of human lens epithelial cells

Background Lens epithelial cells (LECs)play an important role in maintaining the lens transparency.Inflammatory cytokines have been clarified to participate in the formation of traumatic and after cataracts.However,the influence of inflammatory cytokines on the migration of LECs is still studying.Objective This study was to investigate the effects of interleukin-1 alpha (IL-1α),interleukin-1 beta (IL-lβ),IL-6,tumor necrosis factor alpha(TNF-α) and their mixture on the migration of human LECs in vitro.Methods HLE-B3 cells were cultured in DMEM containing 0.5% fetal bovine serum.10 μg/L of IL-1α,IL-1β,IL-6,TNF-α or the mixture was added in the medium for 24 hours,respectively.Wound-scratch assay and transwell monolayer permeability assay were used to evaluate the effects of different cytokines on LECs migration.The migrating cell numbers at the scratch zone and the transmembrane cell numbers at 12:00,3:00,6:00,9:00 and the center areas on the transwell chamber were calculated under the inverted microscope.In the control group,LECs were cultured in DMEM with 0.5% fetal bovine serum only.Results Wound-scratch assay revealed that 24 hours after cytokine incubation,the LECs numbers in the scratch zone were significantly less in the control group compared with the IL-1β group and TNF-αgroup (P =0.000,0.000),and those in the mixture group were higher than the IL-1β group and TNF-α group (P=O.000,0.000).However,no significant change was found in migrating LECs number between IL-1o group or IL-6 group and the control group (P =0.600,0.098).Transwell assay displayed that after 24-hour culture with cytokines,the largest density of transmembrane LECs was obtained in the mixture group,with a significant difference in comparison with the IL-1β group or TNF-α group (P=0.000,0.000).However,the transmembrane LECs were much more in the IL-1 β group or TNF-α group compared with the control group (P =0.000,0.000).Only a few transmembrane LECs were seen in the IL-1α group and IL-6 group in comparison with the control group(P =0.056,0.327).Conclusions IL-lα and IL-6 can not stimulate the migration of human LECs,but TNF-α and IL-1β can enhance their migration markedly.Mixture of these cytokines may play a much powerful influence on LECs migration.

6.Intestinal Absorption of Nicorandil in Rats

Objective To explore the absorption properties and kinetics of nicorandil from small intestinal segments. Methods The absorption kinetics and permeability of nicorandil under different conditions were investigated by HPLC via the in situ perfusing method in rats. Results There was no significant difference in Ka of nicorandil under different concentrations and different pH.The absorption rate in rats descended in order of jejunum,duodenum,ileum and colon, as (4.69±0.05)×10-2 ,(3.35 ±0.04) × 10-2 ,(2.66± 0.05) × 10-2 and (0.89± 0.08) × 10-2 .h-1 . Conclusion The absorption of nicorandil in intestine complies with the passive transport mechanism and first order kinetics.Nicorandil is well absorbed in the whole intestine and can be prepared as delayed release and controlled release forms.

Objective To analysis the genetic context of armA gene identified in different species .Methods BLASTN searches were used to analysis sequences harboring armA submitted to the GenBank database .Results armA gene was mostly found down-stream of ISCR1 element and was most commonly detected in Enterobacter spp ..Its flanking sequences were highly conserved in Enterobacter spp ..Conclusion The genetic context of armA gene reported in different species scattered in different geometrical lo-cations share some common characteristics .
8.Mammographic appearance of accessory breast tissue in the axilla

Objective To observe the mammographic appearance of accessory breast tissue in the axilla. Methods In the past 3 years, 7 562 women were underwent bilateral screen-film mammography. All of the mediolateral oblique(MLO) films were reviewed retrospectively to look for whether there was accessory breast tissue and what the mammographic features were like. Radiographically the accessory tissue resembled the main normal breast glandular tissue but was separated from it. Results Of the 7562 cases, accessory breast tissue in the axilla was detected in 161 cases. The prevalence was 2%. The age ranged from 17 to 70 years (mean, 39 years). 38% of them were found in the bilateral axilla, 42% only in the right, and 20% only in the left. The dimensions on the right ranged from 0.7 to 8.0 cm (mean, 3.5 cm), and that on the left ranged from 1.0 to 7.0 cm (mean, 3.3 cm). There were four types among the accessory breast tissue: patchy type was the most (35%), then the branched type(26%),mixed type(20%), and mass-like type(19%). 3 cases were proved by pathology. Conclusion It is important that the radiologist be familiar with the mammographic appearance of accessory breast tissue in the axilla in order that they could be distinguished from other pathological changes.
9.Clinical Value of Percutaneous Dilatational Tracheotomy in Cardiosurgery

Objective To discuss the value of percutaneous dilatational tracheotomy(PDT) for cardiosurgery in intensive care unit by comparing its outcomes with that of open tracheotomy(OT).MethodsA prospective study was conducted on 40 severe patients undergoing cardiosurgery.The patients were divided into PDT(treated from January 2002 to July 2003) and OT groups(treated from August 2003 to February 2008) with 20 cases in each.The operation time,length and heal time of the incision,blood loss,vital signs,and postoperative complications were compared between the two groups.ResultsThe operation time,length and heal time of the incision in the PDT group were significantly different from that in the OT group [(8.8?2.2) min vs(21.5?5.3) min,t=-9.897;(1.3?0.2) cm vs(3.4?0.5) cm,t=-17.440;and(3.9?0.9) d vs(5.9?1.2) d,t=-5.963;P=0.000].One patient in the PDT group(5%) had Ⅱ degree blood loss during the operation that was significantly less than those in the OT group(13 patients,65%,?2=15.824,P=0.000).The level of the vital signs change in the PDT group was significantly milder than those in the OT group [MAP: 1-5 mm Hg(median 2) vs 1-20 mm Hg(median 3.5),Z=-2.959,P=0.003;heart rate: 3-12 beats/min(median 5.5) vs 7-70 beats/min(median 10),Z=-3.956,P=0.000;SpO2: 0-4%(median 2%) vs 0-31%(median 3.5%),Z=-3.548,P=0.000].Furthermore,the rate of incisional leakage of sputum and overall rate of complications in the PDT group were significantly lower than those in the OT group.[1(5%) vs 8(40%),?2=5.161,P=0.023;and 1(5%) vs 12(60%),?2=13.789,P=0.000].ConclusionsPDT is a safe and minimally invasive technique with a high rate of success and low rate of complications.The method is easy to operate,and thus could be a good choice for severe patients after cardiovascular surgeries.
10.Preparation of nano-liposome enveloping Flos Magnoliae volatile oil

OBJECTIVE: To discuss the influences of the concentrations of sodium cholate, cholesterol and drug, preparation temperature, ultrasonic time and power on the particle size and encapsulation efficiency of Flos Magnoliae volatile oil nano-liposome through film-ultrasonic wave technique together with membrane filter method. METHODS: The nano-liposomes were prepared by film-ultrasonic wave technique together with membrane filter method. The drug content in liposomes was determined by gas chromatography and the particle size was determined by laser light scattering granularity equipment. RESULTS: By film-ultrasonic wave technique together with membrane filter method, the liposomes in the range of 30-40 nm were prepared. The prepared nano-liposomes demonstrated good decentralization and homogeneity, and the encapsulation efficiency was (91.7+/-3)%. CONCLUSION: Film-ultrasonic wave together with membrane filter method is a simple method to fabricate nano-magnoliae liposome.
