Objective To evaluate the difference in efficacy and safety between ~(131)Ⅰand antithyroid drugs (ATD)in the treatment of Graves'disease in children and adolescents.Methods MEDLINE(1966-2005), Cochrane Central Register of Controlled Trials(Cochrane Library Issue 2,2006),EMBASE(1984-2004), CBMDISK(1978-2005)and CNKI(1994-2006)were searched by computer.Isotopes(1989-2004),Radiologia Pratica(1986-2005),Chinese Journal of Endocrinology and Metabolism(1985-2004),and Endocrinology and Metabolism Clinics of North America(1988-2001)were manually searched.Trials comparatively analyzed ~(131)Ⅰand antithyroid drugs on the treatment of Graves'disease in children and adolescents were included.The quality of the study methodologies such as randomization,blinding and allocation concealment was evaluated and meta-analysis was performed by Revman 4.2 software.Results Five non-randomized controlled trials involving 538 patients were included.Among these trials one was prospective and the rest were all retrospective.~(131)Ⅰwas more effective in increasing the complete remission and decreasing the rate of recurrence as compared with ATD,but the rate of hypothyroidism was significantly increased after ~(131)Ⅰtreatment.Conclusion Based on the five studies,the evidence suggests that ~(131)Ⅰtherapy is effective and safe for children and adolescents,and the total curative effects in Graves'disease are superior to ATD.However great shortage of randomized controlled trial(RCT),and problems concerning randomization,blind method,follow-up and statistic analysis still exist in clinical controlled trials, hence more RCT with high quality should be conducted.

Objective To investigate the adverse event reporting of nurses and analyze the influenc-ing factors. Methods A sample of 801 nurses were assessed by completing AHRQ Hospital Survey On Patient Safety Culture (Chinese version). Results 82.65% of respondents within hospital didn't reported a single event in the past 12 months. The positive responsive rates for "frequency of event reported"and "nonpunitive response to error"was 29.92% and 29.38% respectively, where higher scores were found in the respondents possessing bachelor degrees and the average score of the respondents from out-patients unit was lower than those from other units. Conclusions The findings illustrate that it is necessary to improve the performance of event reporting of nurses and the influence factors lie in the level of the nurses' educa-tion degree and whether the event reporting system is punitive or not.

Cervical carcinoma is the most common malignant tumor in gynecology department.The reason for its appearance,development,the prognosis has very close relationship with psychological factors and social factors besides the biology factors.From researching the physiological and psychological change of cervical carcinoma patient after surgery and the relevance between psychology and physiology,this article attempts to comprehensively and systematically discuss the influence of psychological factors and social factors to cervical carcinoma prognosis and providing theoretical basis and practical methods for cervical carcinoma comprehensive treatment under the new medical pattern.

Objective To investigate the characteristics of attention in children with comorbid attention deficit hyperactivity disorder (ADHD) and tic disorder (TD) (CAT). Methods 60 children with comorbid ADHD and TD (CAT), 79 children with ADHD, 60 children with TD and 71 normal control children (NG) were collected. All of those cases were evaluated with the integrated visual and auditory continuous performance test (IVA-CPT). Results It was slightly higher for visual response control quotient (VRCQ) in CAT group(85.15 ±21.10) than those in ADHD group(78.52 ± 21.85) (P ＜ 0.05), there was no significant different in the other five scores between CAT group and ADHD group (P＞0.05). It was significantly lower for the full response control quotient (FRCQ), auditory response control quotient (ARCQ) ,visual response control quotient (VRCQ), full attention quotient (FAQ), auditory attention quotient (AAQ) and visual attention Quotient (VAQ) in CAT group (83.43 ± 20.98,86.12 ± 19.88,85.15 ± 21. 10,64.88 ± 16.88,67.92 ± 17. 11 ,67.97 ± 20.04) than those in TD group(96.47 ± 17.83,97.50 ± 17.62,95.72 ± 18.87,85.37 ± 18.54,86.47 ± 17.32,87.43 ± 19.89) and those in normal control group(98.42 ± 12.67,101.50 ± 15.39,95.92 ± 11.79,99.33 ± 12.98,98.50 ± 12.09,100.00 ± 16.12) (P＜0.01). Conclusion There have serious reduced control ability and attention deficit in children with CAT. The attention deficit is attributed to the existence of ADHD.

Objective To study the expression and pathologic significance of CD147,cyclophilin A (CyPA)and cyclophilin B(CyPB)in psoriatic lesions.Methods Immunohistochemical method was ap- plied to detect the expression of CD147,CyPA and CyPB in skin specimens of 15 patients with psoriasis pustulosa(PP),20 patients with progressive psoriasis vulgaris(PPV),and 20 patients with inactive psori- asis vulgaris(IPV).Immunoreactivity intensity distribution index(IRIDI)was calculated to assess the expres- sion intensity of CD147,CyPA and CyPB.Results CD147,CyPA and CyPB were detected in all speci- mens.The IRIDI scores of CD147 and CyPA were significantly higher in keratinocytes of psoriatic lesions than in those of the control specimens(all P0.05).The IRIDI score of CyPB in T lymphocytes of PP lesions was significantly elevated than that in PPV lesions,which was in turn higher than that in IPV lesions(all P0.05).Conclusion CD147,CyPA and CyPB may play a role in the occurrence and development of psoriasis.

OBJECTIVEThis study further explores the stromal cell-derived factor-1 (SDF-1)/chemokine receptor 4 (CXCR4) signaling axis mechanism in temporomandibular joint osteoarthritis (OA) by detecting the changes in CXCR4, interleukin (IL)-6, and collagen X expression in the ATDC5 cell line stimulated by the cyclic tensile strain and SDF-1.

METHODSInsulin-transferrin-selenium (ITS) was used to induce ATDC5 cells to differentiate into chondrocyte-like cells. After three weeks, the cells were divided into two groups: those with and without cyclic tensile strain. These groups were further divided into the negative control and SDF-1 groups. Strain force of 20% was applied. After 12 h, the total proteins were extracted from cells of the four groups, and Western blot analysis was used to detect the changes in CXCR4, IL-6, and collagen X expression.

RESULTSSDF-1 could enhance CXCR4, IL-6, and collagen X expressions in the chondrocytes, and 20% tensile strain force could further upregulate the three factors.

CONCLUSIONUnder abnormal tensile force, SDF-1 can upregulate its specific receptor CXCR4, thus increasing its-binding efficiency and resulting in the activation of the SDF-1/CXCR4 axis. This condition enhances the expressions of IL-6 and other inflammatory factors and directly damages to cartilage tissue. Such damage directly promotes chondrocyte hypertrophy, which enhances collagen X expression.

Objective To investigate the effects of small interfering RNA(siRNA)on the expression of CD147 on peripheral blood T-lymphocytes from patients with psoriasis vulgaris,and its effect on the proliferation and activation of these cells.Methods Peripheral blood monouclear cells(PBMC)were obtained from 10 patients with psoriasis vulgaris,and T lymphocytes were isolated.CD147 siRNA was chemically synthesized,then electroporated into the peripheral T-lymphocytes.Untransfected cells,blank-transfected cells and unspecifically transfected cells served as the control.After 24-,48-,72-and 96-hour incubation,RT-PCR was used to detect the mRNA expression of CD147 in these cells.MTT assay and flow cytometry were utilized to assess the proliferation of these cellas,and the expression Of CD25 at 24,48,and 72 hours after the transfection.Results Compared with untransfected cells,the mRNA expression of CD147 declined significantly in CD147 siRNA-transfected cells at 24 hours(P＜0.05),reached to the minimum at 48 hours (P＜0.01):there was no significant difierence in the expression of CD147 between the two groups of cells at 96 hours after the transfection(P＞0.05).There was a decrease of cell proliferation level by(44.5±3.13)%,(50.7±3.5)%and(53.98±4.15)%in CD147 siRNA-transfected cells 24,48 and 72 hours following the transfection,respectively;the corresponding decrease in blank-transfected cells was (37.28±3.56)%,(33.73±3.29)%,and(28.80±1.49)%,respectively,and that in unspecifically transfected cells,(31.29±2.46)%,(36.1±2.62)%and(32.08±2.78)%,respectively.A significant decrease was observed in the proliferation of CD147 siRNA-transfected cells compared with that of blank-transfected cells and unspecifically transfected cells at these three time points(P＜0.05,0.01,0.01 respectively).The expression rate of CD25 at 24,48 and 72 hours was(47.23±3.65)%,(31.50±4.22)%and(23.05±4.15)%,respectively,on CD147 siRNA-transfected cells,and,(80.2±4.8)%,(81.6±3.35)%and(83.5±4.1)%,respectively,on untransfeeted cells;the differences between the two groups at the three time points were statistically significant(all P＜0.01).Conclusion CD147 is correlated with cell proliferation and activation of peripheral T lymphoeytes from patients with psoriasis vulgaris,and may serve as a new treatment target for psoriasis.

Objective To explore the correlation between family environment and self-concept of children with attention deficit hyperactivitydisorder (ADHD). Methods Family Environment Scales Chinese Version (FES-CV) and Piers-Harris Children's Self-concept Scale(PHCSS) were used to evaluate 79 children with ADHD and 71 normal children. Results The scores of behavior, intelligence and school,physical appearance, sociability, happiness and satisfaction, and total scores of PHCSS were lower in ADHD group than in normal group (P<0.05). Compared with the normal group, the ADHD group presented lower scores in cohesion, independence, intellectual-cultural orientation,moral religious emphasis, organization (P<0.05) and obviously higher score in conflicts (P<0.01) in FES-CV. The stepwise regressionanalysis showed that moral religion emphasis of FES-CV was a major risk factor to total scores of PHCSS in ADHD children (P<0.05). ConclusionChildren with ADHD show more family dysfunction, which may correlated with their poor self-concept.

Overexpression of procollagen gene can cause the extraordinary increase of collagen's synthesis and therefore lead to the keloid and hypertrophic scar. To utilize ribozyme to suppress the expression of procollagen genes, a eukaryotic expression recombinant plasmid containing a dual-ribozyme gene against alpha 1 (I) and alpha 1 (III) procollagen genes was constructed. The ribozyme from in vitro transcription was incubated with target transcripts from recombinant plasmids which separately contained the fragments of the second exons of pro alpha 1 (I) and pro alpha 1 (III) collagen genes under various experimental conditions. The results showed that the dual-ribozyme could efficiently catalyze the specific cleavage of the target RNAs at 37 degrees C, 42 degrees C, 50 degrees C and Mg2+ concentration from 10 mmol/L to 20 mmol/L. This work provided a basis for further study on the ribozyme to suppress the expression of procollagen genes and control the cicatrization.

OBJECTIVE: To investigate the effect of pSUPER/CD147siRNA on the formation, proliferation, and angiogenesis of malignant melanoma in nude mice. METHODS: The nude mouse subcutaneous xenotransplantation models of malignant melanoma were constructed and observed using morphological analysis. The protein expression of PCNA and CD31 in tumors was detected using immunohistochemical technique. Tumoral proliferative activity in the nude mice was quantitatively evaluated by proliferation cell nuclear antigen (PCNA). Microvessel density( MVD) was counted based on the endothelial cells positively stained with anti-CD31 antibody. RESULTS: The subcutaneous tumors appeared in all the nude mice 5 days later after the transplantation, but the volume of malignant melanoma in the experiment group significantly decreased compared with the control group (P<0.01). PCNA expression and MVD were significantly lower in the experiment group than those in the control group ( P<0.01). CONCLUSION CD147 siRNA inhibits the growth and angiogenesis of malignant melanoma in vivo, suggesting that CD147 might be a new gene therapy target molecule for malignant melanoma.
