1. Identification of Velvet Antler by mitochondrial DNA fingerprint

OBJECTIVE: To compare specific primer method and r andom amplified polymorphic DNA (RAPD) method for identification of Velvet Antler, thus to choose a simpler method. METHODS: DNA samples were extracted from sika deer antler, wilddeer antler, reideer pilose antler and commercially available Velvet Antler, and then purified and amplified with specific primer amplification and RAPD amplification. RESULTS: The sika deer antler, wild deer antler, reideer pilose antler and some of the commercially available Velvet Antler all showed 313bp segment in the agarose gel with different brightness while the other commercial products did not show this segment. RAPD not only effectively showed positive and negative DNA amplification results, but also showed the difference in PCR products of sika deer antlerwild deer antler with different strip numbers and strip brightness. CONCLUSION: RAPD method is more accurate and rapid and it has wide application prospect.
2.Effects of pulmonary static inflation with different pressures on postoperative lung function in patients undergoing cardiac valve replacement with cardiopulmonary bypass

Objective To evaluate the effects of pulmonary static inflation with different pressures on postoperative lung function in patients undergoing cardiac valve replacement with cardiopulmonary bypass (CPB).Methods Sixty ASA Ⅱ or Ⅲ patients,aged 26-70 yr,weighing 47-78 kg,undergoing elective cardiac valve replacement with CPB,were randomly divided into 2 groups (n =30 each):pulmonary static inflation with 5 cm H2O group (group L) and with 10 cm H2O group (group H).In L and H groups,pulmonary static inflation was performed with the pressure maintained at 5 and 10 cm H2O,respectively,after stopping mechanical ventilation during CPB.Arterial blood samples were taken before skin incision and at 1,3 and 6 h after termination of CPB for blood gas analysis.The alveolar-arterial oxygen pressure difference (D(A-a)O2),respiratory index (RI) and oxy.genation index (OI) were calculated.The indwelling time of endotracheal tube after operation and duration of ICU stay were recorded.Results Compared with group L,D(A-a)O2 and RI were significantly decreased and OI was increased at 1,3 and 6 h after termination of CPB,the incidence of OI less than 300 mm Hg was decreased (P < 0.05),and no significant change was found in the indwelling time of endotracheal tube after operation and duration of ICU stay in H group (P > 0.05).Collusion Pulmonary static inflation with 10 cm H2O can better improve postoperative pulmonary diffusion function than with 5 cm H2O in patients undergoing cardiac valve replacement with CPB.

Intelligent prosthesis is going to be a research focus recently with the increment of the transfemoral amputees and the devel-opment of microelectronic technology, control technology and so on. As the fundamental part of the lower extremity prosthetics, the knee prostheses with high performance is still hard to design. This paper reviewed the development of the intelligent knee prostheses, classified the current available prostheses in the term of construction design, and discussed the drawback of their design and the trend of the further de-velopment.
4.Effects of Shuxuetong on Hemodynamics and Serum sICAM-1, IL-6, TNF-α in Patients with Cerebral Infarction

Objective To investigate the effects of Shuxuetong on hemodynamics and serum sICAM-1,IL-6,TNF-α in patients with cerebral infarction.Methods A hundred patients with cerebral infarction were randomly divided into observation group and control group from Jan 2011 to Jan 2015.Fifty patients in control group were treated with conventional therapy alone and 50 patients in observation group were treated with conventional therapy combined with intravenous injection with Shuxuetong.The changes of hemodynamics were observed before and after treatment and the levels of serum sICAM-1,IL-6,TNF-α were detected by enzyme linked immunosorbent assay (ELISA).Results No statistical significance was found in hemodynamics of control group before and after treatment (P >0.05).The indexes of hemodynamics in observation group were significantly improved compared with those in observation group before treatment and those in control group after treatment (P< 0.05).The levels of serum sICAM-1,IL-6,TNF-α were significantly reduced in two groups after treatment (P <0.01) and the levels of serum sICAM-1,IL-6,TNF-α after treatment were significantly lower in observation group than those in control group (P<0.01).Conclusion Shuxuetong can effectively improve the indexes ofhemodynamics and reduce the levels of serum sICAM-1,IL-6 and TNF-α.It has a positive effect on the treatment of cerevral infarction.

Osteoclastic bone resorption and osteoblastic bone formation are coordinated as a coupled mechanism to effect the development of bone and to maintain bone homeostasis. Recently reported Eph/ephrin bidirectional signaling between osteoclasts and osteoblasts plays a pivol role in bone homeostasis and casts new light on coupling of bone resorption and bone formation, which is gaining more and more attention in researches of bone biology and bone diseases. The present article aims to address the researches on the Eph/ephrin bidirectional signaling between osteoblasts and osteoclasts with molecular constitution, mechanism of the signal transduction, biological significance and so on.
6.Effects of different doses of intrathecal magnesium on bone cancer pain in mice

Objective To investigate the effects of different doses of intrathecal magnesium on bone cancer pain (BCP) in mice.Methods Two hundred and eighty-eight male C3H/HeJ mice,aged 8-10 weeks,weighing 18-22 g,were randomly divided into 6 groups (n =48 each):control group (group C) ; sham operation group (group S) ; BCP + artificial cerebro-spinal fluid (aCSF) 5 μl group (group BCP) ; BCP + MgSO4 14.4 μg group (group M1 ) ; BCP + MgSO4 43.2 μg group (group M2 ) and BCP + MgSO4 86.4 μg group (group M3 ).BCP was produced by injecting fibrosarcoma cells of bone into the medullary cavity of right femur.Intrathecal catheter was placed in the 4 BCP groups.The aCSF 5 μl or MgSO4 14.4μg/5 μl,43.2 μg/5 μl,or 86.4μg/5 μl was injected intrathecally on 14th day after inoculation of tumor cells.The paw withdrawal threshold to mechanical stimuli (PWMT) and paw withdrawal lateney to thermal stimuli (PWTL) were measured at 0.5 h before administration (T0 ) and at 0.5,2,4 and 8 h after administration (T1-4).Eight animals chosen from each group at T0-4 were sacrificed,and L4-5 segment of the spinal cord was removed for determination of NR2B expression (by immuno-flurorescence) in the spinal cord.Results PWMT and PWTL were significantly decreased at T0-4,and NR2B expression was significantly up-regulated at T0-4 in groups BCP,M1,M2,M3 compared with groups C and S ( P <0.05).Compared with group BCP,PWMT and PWTL were significandy increased at T1-3,and NR2B expression was significantly down-regulated at T1-3 in groups M2 and M3 ( P < 0.05).Compared with group M2,PWMT and PWTL were significantly increased at T1-3,and NR2B expression was significantly down-regulated at T1-3 in group M3 ( P < 0.05).Conclusion Intrathecal magnesium can reduce BCP in a dose-dependent manner in mice.
7.A retrospective study of risk factors and prevention for shivering after intrathecal anesthesia

Objective To investigate the risk factors and prevention for shivering after intrathecal anesthesia.Methods Atotal of 1008 patients which received intrathecal anesthesia were visited after operation.Patient gender,age,ASA status,type of surgery,anesthesia method,preventive medications,local anesthetics and postanesthetic shivering were recorded.Results Age was negatively correlated with shivering (B =-0.019,P < 0.05 ).Type of surgery,anesthesia method was correlated with shivering,too( P < 0.05).The likelihood of shivering after combined spinal epidural anesthesia was 1.851 times than spinal anesthesia.Preventive medications were correlated with shivering(P < 0.05 ).The likelihood of shivering after prophylactic use of ondansetron plus dexamethasone was 0.196 times than that without prophylactic medication.Selection of local anesthetics were correlated with shivering (B =0.443,P < 0.05 ).The likelihood of shivering after use of bupivacaine was 1.558 times than that use of ropivacaine.Conclusions Risk factors for shivering after intrathecal anesthesia are patient age,type of surgery,anesthesia method,the selection of local anesthetics.Prophylactic use of dexamethasone plus ondansetron can be an effective prevention of shivering after intrathecal anesthesia.
8.Mitochondrial damage in proetin-energy wasting of skeletal muscle in rats with diabetic kidney disease and the effect of low-protein diet combined with α-keto acids

Objective To observe the mitochondrial damage associated with protein-energy wasting of skeletal muscle in diabetic kidney disease (DKD) model of Goto-Kakizaki(GK) rats and evaluate the effects of low-protein diet supplemented with α-keto acids on muscle wasting.Methods Forty-five male 24-week-age GK rats were randomly divided into three groups,normal protein diet group (NPD),low-protein diet group (LPD) and LPD +or-keto group (Keto).Fifteen gender and age matched Wistar rats were served as control group (CTL).The living condition of GK rats was observed and the weight was measured once a week.Urine albumin,serum glucose,creatinine and urea nitrogen were measured at 24,32,40,48 week age.Soleus muscle was observed to calculate the muscle size and the percentage of Ⅰ and Ⅱ type muscle fiber with software after SDH and NADH staining at 48-week-age.Tissue ultrastructure was observed under the transmission electron microscopy.The activity of citrate synthase was detected by spectrophotometer.Expression of mitochondrial DNA was examined by Q-PCR.Results Compared with the CTL group,NPD,LPD and Keto groups had lower body weight,higher urine albumin,higher serum creatinine and urea nitrogen (P < 0.05).The crosssectional area of muscle fibers was larger in CTL group.Compared with CTL group,the muscle fiber was partly broken,the mitochondrial morphology was obviously changed,the percentage of type Ⅱmuscle fiber was increased significantly (P < 0.05),and the activity of citrate synthase and the number of mitochondrial DNA were decreased significantly in NPD,LPD and Keto groups (P < 0.05).In Keto group,muscle wasting was improved compared with NPD and LPD group (P < 0.05),the crosssectional area of soleus muscle increased and the percentage of type Ⅱ muscle fiber decreased,levels of urine albumin,semm creatinine and urea nitrogen decreased (P < 0.05).Under transmission electron microscopy,the muscle fiber of keto group was intact and mitochondiral morphology was close to that of CTL group.The activity of citrate synthase and number of mitochondiral DNA were higher as compared to CTL group (P < 0.05).There were no significant differences between NPD and LPD group.Conclusions In DKD condition,protein degradation in the skeletal muscle is accelerated,mitochondrion is swelling,the number of mitochondrial DNA is decreased and mitochondrial function is impaired.Low-protein diet supplemented with α-keto acids can improve mitochondrial damage and muscle wasting induced by DKD.
9.Subjective well-being simplify scale in college students:development, reliability and validity

Objective To develop subjective well-being simplify scale (SWBSS),and examine its reliability and validity.Methods A self-report SWBSS containing 13 items was developed on college student and research literature.2374 college students completed SWBSS,Index of Well-Being(IWB),World Health of Organization Quality of Life Brief Scale (WHOQOL-BREF),Beck Depression Rating Scale (BDI) and Self-rating Anxiety Scale(SAS).Results ①Exploratory factor analysis results showed that SWBSS had 1 factor,and accounted for 41.768% of variance.②Confirmatory factor analysis results indicated that x2/df =2.208,RMR =0.016,RMSEA =0.032 ; GFI =0.986,AGFI =0.974,NFI =0.978,RFI =0.966,IFI =0.988,TLI =0.981,CFI =0.988 ; PGFI =0.552,PNFI =0.639,PCFI =0.646 ; construct reliability =0.901.③The Cronbach'α coefficient,split-half reliability,stability coefficient of SWBSS was 0.876,0.817 and 0.740 (P < 0.01).The SWBSS scores was significantly correlated with the scores of IWB,WHOQOL-BREF,BDI and SAS (r =0.706 ~ 0.892,r =-0.650 ~-0.580,P<0.01).Conclusion The stability,internal consistency,and validity of the SWBSS are good and meet with psychometric standard.
10.Arsenic trioxide inhibits KM3 cell growth and induces apoptosis and potentiated by bortezomib

AIM: To observe if there is a synergistical effect on induction of apoptosis when arsenic trioxide alone or combination with bortezomib in KM3 cells. METHODS: KM3 cells were treated with arsenic trioxide alone or combined with bortezomib, the numbers of viable cells were determined by trypan blue exclusion. Cell growth inhibition was examined by MTT method. The cells were simultaneously stained with annexin V-FITC and PI and apoptosis was determined by bivariate flow cytometry using a FACScan. Reverse trascriptional-PCR (RT-PCR) method was used to examine the change of p65 mRNA and Western blotting to measure the expression of protein p65, p-p65, caspase-3, -8, -9, and poly ADP-ribose polymerase (PARP). RESULTS: Arsenic trioxide inhibited the cell growth and induced apoptosis. The mechanism was responsible for the activation of caspase-mediated induction of apoptosis. A synergistic effect of combination with bortezomib on apoptosis was observed. CONCLUSION: Arsenic trioxide inhibits KM3 cell growth and induces apoptosis with a synergistical effect when cotreated with bortezomib.
