3.Effects of Microwave Radiation Exposure of Pregnant Mice on Offspring

Objective To study the relationship between microwave radiation exposure during pregnancy and the offspring health. Methods The pregnant Kunming mice were divided into 3 groups and exposed to microwave radiation of 0, 570 and 1 400 ?W/cm2 respectively. The bone marrow cells of the offspring were collected for the determination of chromosome aberration (CA), sister chromatid exchange (SCE) and micronucleus frequency (MNF). Results SCE showed a significant change in high intensity group, as for CA and MNF, no significant change was seen compared with the control. Conclusion Microwave radiation exposure during pregnancy may induce germ plasm damages in the offspring.
4.Changes of Neural Stem Cells in Subventricular in Developing Human Fetal Brain at Different Ages

Objective To observe the changes of neural stem cells(NSCs) in subventricular in developing human fetal brain at(diffe)-rent ages.Methods Thirty cases of embryoes at gestational age 24-28 weeks induced by labor with water bag were collected to determin distribution,shapes and growth modes in subventricular with hybridization in situ under light microscope.Results NSCs expressing Nestin mRNA existed in subventricular from human fetal brain at different ages,NSCs existed in subventricular of different fetal age included astro-NSCs,each had enations from 3 to 6,and all projections crowded together into a reticular plexiform,in which NSCs districuted.Nucli were round in shape,each had nucleoli from 1 to 4.NSCs had rarefaction chromatin,most NSCs existed in a single growth mode,symmetral cleavage growth mode and clony with 3 NSCs would be seen.There were no differences between positive Nestin mRNA NSCs in distribution and cell shapes,but had differences in growth mode.Conclusion NSCs exist in subventricular from different gestational ages and their growth mode are changing with difference of gestational age.
5.Application of Different Experimental Methods for Determining Neural Stem Cells in Hippocampus from Human Fetal Brain

Objective To explore a precise and dependable method for determining neural stem cells(NSCs)in hippocampus from human fetal brain.Methods Ten cases of embryo at gestational age 32 weeks and induction of labor with water bag were collected,affused and sliced including frost slice and paraffin slice for determining alteration of tissue and NSCs in hippocampus from human fetal brain with HE stain and immunohistochemical staining under light microscope.Results Nestin proteins located in NSCs and the number of NSCs were less in paraffin slice of tissue than those in frost slice.Integrality of structure of tissues and cells in paraffin slice excelled in frost slice.Conclusions There is a precise and dependable method including induction of labor with water bag and allusion and frost slice,which is necessary for wide useness in study of NSCs in human fetal brain.

7.Determination of Global DNA Methylation Levels in Medicinal Plants by LC-MS/MS

OBJECTIVE:To develop a detection LC-MS/MS method for global DNA methylation in medicinal plants. METH-ODS:Genomic DNA was isolated using plant DNA extraction kit,and then hydrolyzed by 88% formic acid at 140 ℃. After dried with nitrogen,extracted DNA was dissolved again with mobile phase. LC separation was performed on HILIC column with mobile phase consisted of 7 mmol/L ammonium formate-acetonitrile (gradient elution) at flow rate of 0.3 ml/min. The analysis was con-ducted by tandem MS with positive ion electrospray ionization in multiple reaction monitoring(MRM)mode. The ratio of genomic DNA methylation in 10 commonly used medicinal plants was calculated. RESULTS:The linear ranges of Cyt and 5mC were 1-500 ng/ml(r=0.999 5)and 0.2-100 ng/ml(r=0.999 6). The relative standard deviations(RSDs)of accuracy were 1.12% and 3.68%(n=6). The RSDs of intra-day precision were 2.36% and 4.02% for Cyt and 5mC,respectively (n=5). The RSDs of inter-day precision were 1.04% and 3.54% for Cyt and 5mC,respectively (n=3). The RSDs of repeatability test were 1.53% and 3.27%for Cyt and 5mC,respectively(n=6). The recoveries of Cyt and 5mC were 98.7%-102.1% and 91.2%-103.5%. The percentages of global DNA methylation in 10 medicinal plants were ranged from 17.63% to 25.18%. CONCLUSIONS:LC-MS/MS method is simple,rapid,sensitive and precise,and can be used for the detection of global DNA methylation in medicinal plants.
10.Primary methods and applications of image texture research in computer-aided diagnosis

OBJECTIVE: To analyze recent developments in image texture research both from methodology and from medical image analysis. DATA SOURCE: With the key words of "medical image, texture research, image analysis, application", we retrieved PubMed database (http://www.ncbi.nlm.nih.gov/sites/entrez/), ScienceDirecr database (http://www.sciencedirect.com/) from 1983 to 2009 and CNKI database (http://www.cnki.net/) from 2004 to 2009. DATA SELECTION: Original research thesis, and reviews with clear opinion, sufficient data and reliable conclusion were included. Repetitive studies and studies concerning unrelated to the objective were excluded. MAIN OUTCOME MEASURES: A total 104 literatures were selected, including 10 Chinese literatures and 94 English literatures. These literatures were primarily collected by reading titles and abstracts. A total of 33 literatures with unrelated objective, 18 literatures with repetitive studies were excluded. Finally, 53 Chinese and English literatures were included for further analysis. RESULTS: Primary methods used in texture analysis are structural, statistical, model-based and transform-based-method. When we are interested in identifying texture primitives and their distribution to analyze regular texture, structural approaches are suited. Characteristics of texture like smoothness and coarseness are well analyzed by statistical approaches. Model-based-method is based on the construction of an image model that can be used not only to describe texture, but also to synthesize it. Digital features of texture are got by using some signal processing theories in transformation domain. Texture applications have been widely used in medical imaging domain. CONCLUSION: Because of the specific and complication of medical image and texture, not all texture measure can be used for medical image analysis. One of the development directions of medical image texture research is how to integrate and educe advantages of different methods to fully extract texture features and exactly attribute medical image texture and the relation between its changes and pathological state, resulting in an important component of computer-aided diagnosis.
