2.Blood glucose and glycosylated hemoglobin levels of type 2 diabetes at different altitudes and different races in Yunnan Province

Objective To investigate the effects of altitude,race,gender and age on blood glucose and glycosylated hemoglobin(HbA1 c) of type 2 diabetes.Methods A total of 410 patients with type 2 diabetes (T2DM) who resided at different altitudes in Jinghong,Kunming and Lijiang city for over 5 years and received antidiabetic treatment without adjustments for over 3 months were enrolled.To compare blood glucose,hemoglobin (Hb) and HbA1 c levels of T2DM among different altitudes,different races,different genders and different ages,The correlation between races and HbA1c was evaluated with Pearson's correlation analysis.Results No significant differences were observed in fasting blood glucose (FBG) (P =0.49,P =0.1,P =0.09,P =0.3),2 h postprandial blood glucose (2 h PBG) (P =0.33,P =0.4,P =0.7,P =0.1),and HbA1 c (P =0.47,P =0.28,P =0.2,P =0.07) levels between patients at different altitudes,different races,different genders and different ages.However,there were significant differences in Hb (P <0.01).There was no significant correlation between races and HbA1c levels(r =0.003,P =0.6).Conclusions There were effects of altitude,race,gender and age on hemoglobin of type 2 diabetes,but no effects on glycosylated hemoglobin.
3.Impact of PPT health education on the anxiety status of patients with PICC

Objective To discuss the effect of PPT health education to release the anxiety of patients with PICC.Methods 60 patients with PICC were randomly divided into two groups,the experimental group (31 cases) and the control group (29 cascs).The control group received conventional PICC procedures and nursing,while the experimental group received PPT health education 60 minutes before PICC insertion.S-AI in ATAI was used to compare two groups' anxiety status before and after the education.Results Before PICC insertion,there was no statistically significant difference between two groups' S-AI scores.After PICC insertion,the S-AI score of the control group was (40.65 ± 5.24),while that of the experimental group was (29.41 ±3.38),and the difference was statistically significant (t =9.938,P < 0.01).Conclusions PPT health education can effectively release the anxiety of patients with PICC.
4.Effect of compound danshen dripping pill on arterial intima-media thickness in patients with newly diagnosed type 2 diabetes mellitus.

OBJECTIVETo investigate the effect of compound Danshen Dripping Pill (CDDP) on peripheral arterial intima-media thickness (IMT) in patients newly diagnosed as type 2 diabetes mellitus.
METHODSOne hundred and eight patients were equally randomized into 3 groups, in addition to the basic hypoglycemic, hypotensive and lipid-regulation treatment, they were administered orally with aspirin (0.1 g, once daily), vitamin E (0.1 g, twice daily) and CDDP (10 pills, thrice daily) for 18 months, respectively. The conventional cardiovascular risk factors, such as hyperglycemia, lipids profile, and homeostasis model assessment of insulin resistance (HOMA-IR) as well as ultrasound measurement of peripheral arterial IMT before and after treatment were compared.
RESULTSIn the group treated with CDDP after treatment, the levels of HbA1c, serum total cholesterol and low density lipoprotein cholesterol (LDL-C) were significantly lower (all P<0.01), HOMA-IR was higher than those in the other two groups; while IMTs of carotid, iliac and femoral arteries were insignificantly different among them (P>0.05); however, the increment of carotid IMT in the CDDP treated group was less than that in the aspirin treated group (P<0.05), and that of femoral IMT was less than both the aspirin and vitamin E treated groups (P<0.05).
CONCLUSIONFor the patients with newly diagnosed type 2 diabetes, additional administration of CDDP to the conventional treatment could exert beneficial effects on blood glucose controling, and lipid profile improvement and delay of arterial intima-media proliferation.

5.The Inhibitory Effect of Baixuan Xiatare Tablet on Model Mouse with Allergic Contact Dermatitis

OBJECTIVE:To investigate the inhibitory effect of Baixuan xiatare tablet on the model mouse with allergic contact dermatitis (ACD). METHODS:60 BALB/c mice were equally randomized into normal control (isometric solvent) group,model (isometric solvent)group,ebastine(positive control,0.003 g/kg)group and the groups of high,middle and low doses of Baixuan xiatare tablet(2.0,1.0 and 0.5 g/kg). The mice were given drugs,ig,once daily for 14 consecutive days. 0.5% 2,4-dinitrofluoro-benzene(DNFB)acetone olive oil solution was applied,for sensitization,on the prepared mouse’s skins one and two days before administration,and 0.2% DNFB acetone olive oil solution on their left ears 16 days thereafter to establish mouse models of ACD. At 48 h after successful establishment of the models,the thickness of the mouse’s left ear margin was measured and the difference value and swelling degree were calculated;flow cytometer was used to determine the levels of T lymphocyte subsets CD4+ and CD8+ in mouse blood and calculate the ratio of CD4+ to CD8+;the levels of interleukin 4(IL-4)and IL-6 in mouse serum were de-termined. RESULTS:Compared with normal control group,those in the model group had higher difference value of ear margin and swelling degree,lower level of CD4+ in blood and ratio of CD4+ to CD8+,and higher content of IL-6 in serum. There was statisti-cally difference (P<0.01). Compared with model group,those in the groups of high,middle and low doses of Baixuan xiatare tablet had lower degree of left ear swelling and higher level of CD4+ in blood;those in the groups of high and middle doses thereof had lower difference value of left ear margin and level of IL-6 in serum;and those in the group of high dose thereof had higher lev-el of CD8+ in blood. There was statistically significance(P<0.01 or P<0.05). CONCLUSIONS:Baixuan xiatare tablet has inhibi-tory effect to some degree on the mouse model with ACD by a mechanism which may be related to the balance of subsets CD4+and CD8+in blood and the reduction of IL-6 in serum.
6.Regulation of proliferation and apoptosis of human vascular endothelial cell by Acheron.

OBJECTIVETo investigate regulatory effect of Acheron (Achn) on proliferation and apoptosis of human vascular endothelial cell.
METHODS(1) Eahy926 cells were cultured in serum-free DMEM medium (96-well plates) and were divided into Achn inhibition group (transfected with plasmid psi-Achn), psi4.1 group (transfected with psi4.1 empty vector), Achn induction group (transfected with pcDNA-Achn), pcDNA3.1 group (transfected with pcDNA3.1 empty vector), cotransfection group [cotransfected with pcDNA-Achn + psi-calcium/calmodulin-dependent serine protein kinase (CASK)], blank control group (treated with PBS) according to the random number table (the same method below). The cell proliferation was determined by MTT assay at post transfection hour (PTH) 1, 24, 48, 72, with expression of absorbance value. (2) Total protein of Eahy926 cells were extracted and quantitated by BCA assay, and then they were divided into Achn antibody precipitation group (100 µg protein), CASK antibody precipitation group (100 µg protein), IgG antibody group (100 µg protein), Western blot group (20 µg protein). Achn and CASK protein levels were determined by immunoprecipitation and Western blot. (3) Synchronously cultured Eahy926 cells were divided into LPS induction group (treated with 5 mol/L LPS), Achn transfection group (transfected with pcDNA-Achn), cotransfection group (cotransfected with psi-CASK and pcDNA-Achn), KCl group (treated with 5 mol/L KCl), and blank control group (treated with 5 mol/L PBS). Cells in transfection groups were stimulated by LPS for 12 hours after PTH 24. Caspase-3 protein level was detected by immunohistochemistry. (4) Synchronously cultured Eahy926 cells were divided into Achn inhibition group (transfected with psi-Achn vector), Achn induction group (transfected with pcDNA-Achn vector), and blank control group (treated with PBS). Apoptosis rate was determined by FITC/PI with flow cytometry. Data were processed with one-way analysis of variance and t test.
RESULTS(1) The cell proliferation in Achn inhibition group was lower than that in psi4.1 group from PTH 24, and the differences were statistically significant at PTH 48, 72 (with t value respectively 10.777, 6.112, P values all below 0.05). The cell proliferation in Achn induction group during PTH 24-72 were higher that in pcDNA3.1 group (with t value respectively 5.367, 6.053, 9.831, P values all below 0.05). The cell proliferation in cotransfection group at PTH 48, 72 were significantly lower than that in Achn induction group (with t value respectively 5.481, 9.517, P values all below 0.05). (2) Achn protein was detected in CASK antibody precipitation group while CASK protein was also detected in Achn antibody precipitation group. (3) Caspase-3 level in Achn transfection group was lower [(15.6 ± 0.5)%] as compared with that in LPS induction group [(32.8 ± 2.6)%, t = 10.083, P < 0.05], and that in cotransfection group showed further inhibition [(7.0 ± 2.0)%, t = 9.827, P < 0.01]. (4) Apoptosis rate in Achn inhibition group [(45.6 ± 10.9)%] was higher than that in blank control group [(13.2 ± 4.3) %, t = 7.043, P < 0.05]; while that in Achn induction group [(5.3 ± 2.9)%] was lower than that in blank control group (t = 6.499, P < 0.05).
CONCLUSIONSAchn can promote human vascular endothelial cell proliferation, and inhibit its apoptosis induced by LPS or burn serum, and the effect is related to CASK.

7.Study on the differences on HA1 regions between epidemic strains and vaccine strains of influenza virus subtype A3 from 1988 to 2005.

OBJECTIVETo study the differences between epidemic strains of influenza virus subtype A3 circulated in China and Occident in past 18 years, in genetic level, and vaccine strains recommended by WHO in corresponding time.
METHODSAmino acid sequences of HA1 regions of the epidemic strains, which circulated in China and Occident from 1988 to 2005, and the vaccine strains of influenza virus subtype A3 were compared by BioEdit and analyzed on the differences of HA1 and it's antigen determinants
RESULTSDifferences between epidemic strains and vaccine strains recommended in corresponding year, both in HA1 and it' s antigen determinant regions, were obviously greater than that between epidemic strains and vaccine strains recommended in next round (P< 0.01). However, differences between epidemic strains in Occident and vaccine strains recommended in corresponding year were slightly greater than that between epidemic strains and vaccine strains recommended in next round and it was not marked (P >0.05). In addition, differences between epidemic strains and vaccine strains which being used for several years, whether in China or in Occident, constantly increased accompanying the used time prolonged.
CONCLUSIONThere was an obvious lag between vaccine strains recommended by WHO, analyzed in genetic level, and epidemic strains of influenza virus subtype A3 circulated in China.

8.Clustering analysis of HPLC chromatographic fingerprints of Paeonia suffruticosa.

OBJECTIVETo explore the utility of chromatographic data for quality control of Paeonia suffruticosa, a Chinese herbal medicine.
METHODChromatographic fingerprints of Paeonia suffruticosa were determined by HPLC, the clustering analysis was used for data processing.
RESULTThe quantitative differences among different growing areas were found and could be used to classify herbals from different growing areas, while there seemed to be no quantitative differences for processing factor.
CONCLUSIONThe method could be used in quality control for monitoring herbal quality control, and the result also suggested that different growing areas may greatly affected the herbal qualities.

Objective Clinical data of 19 Chinese patients with 21 hydroxylase deficiency (21OHD) were analyzed to improve the diagnosis and treatment level. Methods Clinical features and laboratory data were collected from 19 patients with 21OHD before and after treatment. Results In male patients, the average age of early appearance of secondary sexual character was (9.3?2.8)yrs, and excess androgen resulted in phallic enlargement. Primary amenorrhea was the most common complaint in female(87.5%), and the signs included a varying degree of labioscrotal fusion and clitoral enlargement. The average level of 17-hydroxy progesterone(17OHP) was (63.42?35.07) ?g/L, and adrenocorticotrophic hormone(ACTH), dehydroepiandrosterone(sodium) sulfate(DHEAS) and testosterone(T) were obviously elevated. 10.Decreased expression of DICER1 in gastric cancer.

BACKGROUNDThe role of epigenetics in gene expression regulation and development significantly enhances our understanding of carcinogenesis. All the tumor related genes may be the target of epigenetical or genetic regulation. We selected some epigenetically regulated genes for cDNA array analysis and observed variability in the expression of the DICER1 gene in distinct stages of gastric cancer. The aim of this study was to assess the correlation between the expression of DICER1, an epigenetically regulated gene, and gastric cancer.
METHODSTo detect the expression of 506 tumor-associated genes, including DICER1, in the matched cancerous mucosa, pre-malignant lesion (adjacent mucosa), non-cancerous gastric mucosa and distant lymphocyte metastatic lesion in 3 cases of gastric cancers using cDNA array. DICER1 mRNA expression and DICER1 protein expression were further analyzed by Real-time PCR and Western blot in 32 cases of progressive gastric cancer. DICER1 protein expression was also detected in 33 early and 30 progressive gastric cancers by the immunohistochemistry (IHC) method.
RESULTSIn 3 cases of gastric cancer cDNA array showed dramatically decreased expression of DICER1 in pre-malignant lesion, cancerous mucosa and distant lymphocyte metastatic lesions compared with matched noncancerous gastric mucosa, pre-malignant lesion and cancerous mucosa. Real-time PCR results showed that the expression level of DICER1 mRNA in gastric cancer was significantly down-regulated compared to normal gastric tissue (P < 0.05). The IHC assay also showed that the expression of DICER1 was significantly decreased in progressive gastric cancer. Among the 63 cases of gastric cancers, 13/33 early (39.4%) and 19/30 (63.3%) progressive cancers showed negative expression of DICER1 (50.8%). The difference in expression of DICER1 between early and progressive gastric cancers was significant (P < 0.01). The result of Western blotting showed that DICER1 protein was down-regulated significantly in advanced gastric cancer (P < 0.05).
CONCLUSIONSDICER1 expression is decreased during the progression of gastric cancer, especially in progressive gastric cancers, which indicating DICER1 may play an important role in the development of cancer and the epigenetical regulation involved.
