This paper described the results of the optimal method for preparation and extraction of lectin from Phaeoporus obliquus(Pers ex Fr).(J.Schroet).We determined the optimal buffer solution using the ag-glutination test with 2% hare blood cells.The optimal extraction process included the following parameters that were conformed by optimization analysis: material-water ratio,extraction time,concentration of sodium chloride and the pH value.The results are that the agglutination indexes of POL using TBS and PBS extrac-tion buffer were 64 and 16,respectively.The optimal extraction process is that,the material-water ratio was 1:50,the extraction time was 20 h,while the concentration of sodium chloride was 0 mol/L and pH was 8.0.The agglutinability of POL was 256 examined by this method.The optimized extraction process is stable and practicable,which may supply some basis for the application of the lectin from Phaeoporus obliquus in immunoregulation.
2.Emodin ameliorates high-glucose induced mesangial p38 over-activation and hypocontractility via activation of PPARgamma.

Early stage diabetic nephropathy is characterized by elevated glomerular filtration. Recent studies have identified high-glucose induced p38 MAPK (p38) over-activation in mesangial cells. Mesangial hypocontractility is the major underlying mechanism, however, no ameliorating agents are currently available. We investigated the protective effects of emodin on high-glucose induced mesangial cell hypocontractility. Mesangial cells were cultured under normal (5.6 mM) and high glucose (30 mM) conditions. Emodin was administrated at doses of 50 mg/l and 100 mg/l. Angiotension II stimulated cell surface reductions were measured to evaluate cell contractility. p38 activity was detected using Western blotting. To further explore the possible mechanism of emodin, expression of the peroxisome proliferator-activated receptor gamma (PPARgamma) was measured and its specific inhibitor, gw9662, was administrated. Our results showed: (1) high-glucose resulted in a 280% increase in p38 activity associated with significant impairment of mesangial contractility; (2) emodin treatment dose-dependently inhibited high-glucose induced p38 over-activation (a 40% decrease for 50 mg/l emodin and a 73% decrease for 100 mg/l emodin), and mesangial hypocontractility was ameriolated by emodin; (3) both the PPARgamma mRNA and protein levels were elevated after emodin treatment; (4) inhibition of PPARgamma using gw9662 effectively blocked the ameliorating effects of emodin on high-glucose induced p38 over-activation and mesangial hypocontractility. Emodin effectively ameliorated p38 over-activation and hypocontractility in high-glucose induced mesangial cells, possibly via activation of PPARgamma.

3.Effects of arsenic trioxide, ginseng saponin and beta-elemene on telomere-telomerase system in K562 cell line.

The aim was to explore the modulating and inhibiting effects of arsenic trioxide, ginseng saponin and beta-elemene on telomere length and telomerase activity in K562 cell line, and to study their anti-tumor mechanism and seek new method of therapy for acute leukemia. Human erythroleukemia cell line K562 was co-cultured with arsenic trioxide, ginseng saponin, beta-elemene separately, cells were collected after 24, 48 and 72 hours for further detecting. Telomere length and telomerase activity were detected by the methods of Southern-blot and PCR-ELISA respectively. The effects of these drugs on telomere length and telomerase activity were observed at different concentrations and length of time. The results showed that (1) telomerase activity of K562 cells decreased after co-cultured with arsenic trioxide, ginseng saponin and beta-elemene. The inhibiting effects depended on drug concentrations and length of time. When co-cultured at proper concentration and period of time, telomerase activity could be inhibited; (2) viability of K562 cells decreased after co-cultured with arsenic trioxide, ginseng saponin and beta-elemene, the inhibiting effect depends on drug concentrations and length of time; (3) after co-cultured with arsenic trioxide, ginseng saponin, and beta-elemene for 72 hours, telomere length of K562 cell line prolonged a little. It is concluded that (1) arsenic trioxide, ginseng saponin and beta-elemene can inhibit telomerase activity in K562 cell line, the suppression of telomerase activity may be one of the mechanisms of anti-tumor effect; (2) arsenic trioxide, ginseng saponin and beta-elemene can inhibit the growth of K562 cell line, the inhibiting effect depends on concentration and time; (3) when telomerase activity was suppressed, the telomere length prolonged a little, indicating that in K562 cell line may exist another mechanism to regulate telomere length, except telomerase activation.

4.Expression of cell adhesion molecules in acute leukemia cell.

OBJECTIVETo investigate the role of cell adhesion molecule in the development and extramedullary infiltration (EI) of acute leukemia.
METHODSThe expressions of neural cell adhesion molecule (NCAM) gene, intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule (VCAM-1) genes in 25 acute leukemia patients bone marrow cells were detected by microarray and reverse transcriptase-polymerase chain reaction (RT-PCR).
RESULTSThe expressions of NCAM, ICAM-1 and VCAM-1 gene were significantly higher in acute leukemia cells and leukemia cells with EI than in normal tissues and leukemia cells without EI, respectively, both by cDNA microarray and by RT-PCR.
CONCLUSIONThe cDNA microarray is a powerful technique in analysis of acute leukemia cells associated genes. High expressions of cell adhesion molecule genes might be correlated with leukemia pathogenesis and infiltration of acute leukemia cell.

Objective To explore the interaction between HT036(hypothetical protein HT036)and P311 by co-immunoprecipitation.Methods HA-tagged fusion protein(HA-HT036)expression vector was constructed,identified and transfected into human embryo kidney 293(HEK293)cells alone or with Myc-tagged fusion protein(Myc-P311)expression vector pCMV-Myc-p311.The interaction between P311 and HT036 was detected by co-immunoprecipitation.Results Double restriction enzyme digestion showed that pCMV-HA-HT036 was constructed correctly.When Myc-P311 was immunoprecipitated by anti-Myc antibody,HA-HT036 was identified by Western blotting with anti-HA antibody from immunoprecipitated complex.Conclusion The recombinant vector pCMV-HA-HT036 was constructed successfully.The interaction between HT036 and P311 could be identified by co-immunoprecipitation after co-expression of pCMV-HA-HT036 and pCMV-Myc-p311.The result provides an important basis for further study of the intracellular signal transduction of P311.
6.Accuracy of respiratory variations of internal jugular vein in monitoring fluid responsiveness in patients undergoing radical gastrectomy for gastric cancer

Objective To evaluate the accuracy of respiratory variations of internal jugular vein (IJV) in monitoring fluid responsiveness in patients undergoing radical gastrectomy for gastric cancer.Methods Fifty American Society of Anesthesiologists physical status Ⅰ or Ⅱ patients of both sexes,aged 40-64 yr,scheduled for elective radical gastrectomy for gastric cancer,were enrolled in this study.Before induction of anesthesia,the hemodynamic parameters such as heart rate,central venous pressure,cardiac index,stroke volume index (SVI),stroke volume variation and respiratory variation of IJV were recorded after haemodynamics was stable and were recorded again at 10 min after endotracheal intubation,and a loading dose of 6% 130/0.4 hydroxyethyl starch 7 ml/kg was infused over 15 min.The parameters mentioned above were recorded within 5 min after loading dose.Patients were divided into 2 groups according to the percentage of increase in SVI (△SVI) after volume expansion:△SVI≥ 15% was considered to be a positive response (responder group) and △SVI<15% was considered to be a negative response after volume expansion (non-responder group).Results The area under the receiver operating characteristic curve of respiratory variations of IJV in monitoring fluid responsiveness and 95% confidence interval were 0.852 (0.744-0.961).Respiratory variation of IJV 24.6% was considered as the cut-off value and used to monitor fluid responsiveness,and the sensitivity and specificity were 67.6% and 92.3%,respectively.Conclusion Respiratory variation of IJV can be considered as an effective index in monitoring fluid responsiveness in the patients undergoing radical gastrectomy for gastric cancer.
7.Trophoblast cells invaing the placenta bed and change of spiral arteries and microvessels in pre-eclampsia.

OBJECTIVE:
To investigate the invason of trophoblasts in the placenta bed and the change of spiral arteries and microvessels in pre-eclampsia and normal pregnancy.
METHODS:
Twenty cases of normal pregnancies, mild pre-eclampsia and severe pre-eclampsia were chosen as Group A, Group B, and Group C. HE staining and immunohistochemistry staining (SP method) were used to observe the depth and the density of trophoblasts invading the placenta bed and the change of spiral arteries and microvessels.
RESULTS:
The significant difference in the degree of invasion was in the superficial myometrial segment. Group C was the most superficial in the 3 groups (P<0.01). The density of trophoblasts which invaded the placenta bed in the lower half of the basal decidual segment and the myometrial segment showed us Group C was the lowest (P<0.01). There was statistical difference among the 3 groups (P<0.01). The average lumen area of the spiral arteries in the decidual segment and the superficial myometrial segment of the placenta bed was the smallest in Group C among the 3 groups(P<0.01) and there was statistical difference among the 3 groups (P<0.01). The spiral arteries were the thickest in Group C with statistical difference among the 3 groups (P<0.01). The physiological and pathological change of the spiral arteries was mainly in the superficial myometrial segment. The incidence rate of physiological changes in the spiral arteries was the lowest in Group C with statistical difference among the 3 groups (P<0.01). The incidence rate of pathological changes was the highest in Group C (P<0.01) and the normal group was the highest. There was significant difference among the 3 groups(P<0.01). There was positive correlation between the physiological change of the spiral arteries and the invaing degree of the trophoblasts (P<0.05), there was negative correlation between the pathological change of the spiral arteries and the invasion depth as well as the invasion density of the trophoblasts(P<0.05). There was negative correlation between the physiological change and the pathogenetic condition of pre-eclampsia(P<0.05)while there was positive correlation between the pathological change and the pathogenetic condition degree of pre-eclampsia(P<0.05). There was negative correlation between the invasion depth as well as density in uteruso superficial myometrial segment by trophoblast and the pathogenetic condition degree of pre-eclampsia(P<0.05). There was invasion trophoblast in 62.50% lumen wall of spiral arteries in uterus superficial myometrial segment of the placental bed in normal pregnancy while 27.5% was seen in severe pre-eclampsia. Microvascular density in the decidual segment and the superficial myometrial segment of the placenta bed in Group C was the lowest among the 3 groups with statistical difference (P<0.01).
CONCLUSION
The invasion depth of the trophoblasts in pre-eclampsia was more superficial than normal pregnancy.The changes of the invasion of the trophoblasts and the pathological changes of the spiral arteries in the placenta bed mainly existed in the superficial myometrial segment which was closely related to the severity of the illness. That microvascular density in the placental bed of pre-eclampsia started to decrease from the basal decidual segment shows that the microvessel development in the placenta bed is impaired in pre-eclampsia.

8.Psychosocial factors of female patients with recurrent depression

Objective To investigate psychosocial factors in the recurrence of female depression,and provide reliable basis for effective nursing.Methods There were 210 female patients with recurrent depression (observational group) and 217 female patients with a history of depression (control group) who were adopted into the study.Self-made questionnaire was used to record their general conditions,Hamilton Rating Scale for Depression (HAMA-17) was used to evaluate their clinical symptoms,Social Support Rating Scale (SSRS),Life Events Scale (LES),and Eysenck Personality Questionnaire (EPQ) were used to determine their psychosocial factors,and relative analysis as well as logistic regression analysis was used during the process.Results ( 1) Compared with control group,observational group was older,but younger on first onset,had more evident family history,and more obvious anxiety symptoms (P <0.05).In SSRS,both social subjective and objective support factor scores of observational group were significantly lower than those of control group [ (21.69 ± 5.24) vs (23.18 ± 4.80),(9.00 ± 2.77) vs (9.70 ± 3.14) ],and the differences were statistically significant (t =3.07,2.45,respectively; P < 0.05 ).In LES,the quantity of total life events and the quantity of negative life events in observational group were higher than those of control group [ (46.25 ± 18.38) vs (32.39 ± 19.34),(37.01± 16.04 ) vs ( 26.86 ± 12.07 ) ],and the differences were statistically significant ( t =2.04,2.02,respectively ; P < 0.05 ).In EPQ,the N factor score of observational group was ( 16.37 ±4.93),higher than that of control group (9.21±4.52),and the E factor score of observational group was (6.75 ± 4.56),lower than that of control group ( 13.44 ± 4.37 ),and both the differences were statistically significant (t =9.47,9.51,respectively; P < 0.05 ).(2)The logistic regression analysis showed that marital status,family heredity,negative life events,neurotic personality and objective support were major hazard factors for the recurrence of female depression.Conclusions Recurrent attacks of female depression are related to patients' general conditions as well as psychosocial factors,and the results of this study provide a theoretical foundation for the implementation of mental nursing intervention.
9.Effect of rhynchophylline on behaviors of methamphetamine-dependent zebrafish and the mechanism.

OBJECTIVETo observe the effect of rhynchophylline on methamphetamine-dependent zebrafish and explore the possible mechanism.
METHODSZebrafish were divided into control group, amphetamine group, low- (50 mg/kg) and high (100 mg/kg)-dose rhynchophylline groups, and ketamine (150 mg/kg) group. Conditioned place preference (CPP) was induced in zebrafish with methamphetamine, and the staying time in the drug box and the tracking map of the zebrafish were observed with Noldus Ethovision XT system. The protein expressions of TH, NR2B and GLUR2 in the brain of zebrafish with CPP were detected with Western blotting.
RESULTSCompared with the control group, zebrafish in methamphetamine group showed significant variations in the staying time and swimming distance in the drug box after conditioning (P<0.05) with obvious alterations of NR2B, TH and GLUR2 expressions in the brain (P<0.05). Treatment of methamphetamine-dependent zebrafish with high-dose rhynchophylline significantly reduced the variations in the staying time and swimming distance in the drug box (P<0.05) and in the expressions of NR2B, TH and GLUR2 in the brain (P<0.05).
CONCLUSIONRhynchophylline can inhibit methamphetamine dependence in zebrafish, the mechanism of which may involve the expressions of TH, NR2B and GLUR2 proteins in the brain.
10.Responsiveness evaluation of mental intervention services system in Wenchuan earthquake area.

OBJECTIVETo understand the mental intervention service system responsiveness.
METHODSA stratified, multi-stage random cluster sampling method was used, and a total of 211 residents in the central earthquake area were face to face interviewed by using the evaluating questionnaire of mental intervention service system responsiveness (including confidentiality, autonomy, prompt attention and so on, in sum of eight indicators). Analytic hierarchy process method was used to determine the weight of each index, carrying out a single index evaluation and fuzzy comprehensive evaluation on the mental intervention service system responsiveness, and using Spearman rank correlation and Binary logistic regression model to analyze the relationship of total satisfaction of mental intervention with each index.
RESULTSThe dignity and confidentiality indicators were higher weight and rating. Prompt attention and the autonomy indicators were higher weight but lower rating, while surroundings and choice of providers indicators were lower weight and rating. It was also found that communication and social support network indicators were lower weight but higher rating. The overall assessment of mental intervention service system responsiveness ranged between "good" and "very good". All rank correlation coefficients of the indicators and the total satisfaction of mental interventions were significant (r(s) = 0.186 - 0.362, P < 0.05), except for confidentiality. The logistic regression analysis showed that the main factors of the individual variables influencing the total satisfaction were dignity (adjusted OR = 3.047, P < 0.001), surroundings (adjusted OR = 1.619, P = 0.019), and social support network (adjusted OR = 1.527, P = 0.005).
CONCLUSIONThe overall assessment of mental intervention service system responsiveness was high. Mental interventions should be taken positive and effective measures to improve the prompt attention, autonomy, the choice of providers and service environment.
