Objective To construct adenovirus vector harboring CTLA4Ig-IRES2-I?B? gene for the study of enhancement blockade of T cell costimulatory pathway. Methods The IRES2 fragment was cloned in to the shuttle plasmid pAdtrack-CMV-CTLA4Ig, and then the human I?B? fragment was constructed with the link of the IRES2 to the pAdtrack-CMV-CTLA4Ig. Subsequently, adenovirus vector harboring CTLA4Ig-IRES2-I?B? was constructed by homologous recombination in E. coli BJ5183, and recombinant vector was packaged and propagated in 293 cells. Results The recombinant CTLA4Ig-IRES2-I?B? adenovirus was constructed by homologous recombination and identified by PCR and restrictive enzyme digestion methods. Conclusion The recombinant CTLA4Ig-IRES2-I?B? adenovirus may be used as a novel immunosuppressive agent for gene therapy in organ transplantation.

Objective To observe the effect of herbal medicines with the function of replenish qi and essence for the immune func- tion of patients after mammary cancer operation. Methods The 115 patients of mammary cancer after modified radical operation were randomized into treatment group and control group. The treatment group began to take the decoction of herbal medicines from 7 days before and from the 3rd day after operation. The control group had the same treatment but without taking herbal medicines. The im- mune indices of both groups were tested on the day 7 days before, one day and 14 days after operation respectively. Results In the treatment group,IgA,IgG,IgM were remarkably increased in comparison with the control group 14 days after operation;with the herbal treatment for 2 weeks,CD_4,CD_4/CD_8,IL-2 rised remarkably(P

AIM: To investigate the electrophysiological characteristics of ventricular myocyte both in subendocardium and in subepicardium during ischemia. METHODS: Using modified Ferrier's method of ischemic and reperfusion injury in isolated guinea pig right ventricular wall. RESULTS: The transmembranal electric activities in subendocardium and in subepicardium were all significantly abnormal, and it was more significant in subendocardium. CONCLUSION: The alteration degree of ventricular myocyte in subendocardium and in subepicardium during ischemia and early reperfusion was different, and this might be the electrophysiological basis of the vulnerability of the subendocardium.

China Journal of Chinese Materia Medica 2014;39(16):3128-3130
Cleistocalycis Operculati Cortex is the dry bark of Cleistocalyx operculatus. It is the raw material of Compound Hibiscuse which is external sterilization antipruritic drugs. The quality standard of Cleistocalycis Operculati Cortex in Guangdong Province "standard for the traditional Chinese medicine" (second volumes) only contains TLC identification. It is unable to effectively monitor and control the quality of Cleistocalycis Operculati Cortex. A reversed-phase HPLC method was established for the determination of 3, 3'-O-dimethylellagic acid from Cleistocalycis Operculati Cortex and the content was calculated by external standard method for the first time. Under the selected chromatographic conditions, the target components between peaks to achieve effective separation. 3,3'-O- dimethylellagic acid standard solution at the concentration of 1.00 - 25.0 mg x L(-1) showed a good linear relationship. The standard curve was Y = 77.33X + 7.904, r = 0.999 5. The average recovery was 101.0%, RSD was 1.3%. The HPLC method for the determination of 3,3'-O-dimethylellagic acid in Cleistocalycis Operculati Cortex is accurate and reliable. It can provide a strong technical support for monitoring the quality of Cleistocalycis Operculati Cortex.

Journal of Xinxiang Medical College 2018;35(1):12-15,21
Objective To evaluate the anti-depression effect of astilbin on mice with depressive disorder and to explore its mechanism of action. Methods Seventy-two male mice (C57BL/6J type) were randomly divided into control group, model group, low, middle, high dose of astilbin group and imipramine(IMI) group, with 12 mice in each group. The mice in the control group were adopted in a normal way.The rats in the model group, low, middle, high dose of astilbin group and IMI group were adopted in chronic unexpected mild stress (CUMS) to establish depression model, and the stress was continuously kept for three weeks and daily medicine with physiological saline,10,20,40 mg · kg-1 astilbin and 10 astilbin imipramine were provided in the way of intraperitoneal injection.Examination for behavioural changes of animals was implemented via tail suspension test, forced swimming test, sucrose preference test, open field test.High performance liquid chromatography (HPLC) was adopted to examine the level of dopamine (DA) and 5-hydroxytryptamine(5-HT)in prefrontal cortex. Results There was no significant difference in the number of horizontal movement and vertical movement among the control group, model group,IMI group and low, middle, high dose of astilbin group(P > 0.05).Compared with the control group, the dead time of tail suspension test and forced swimming test in the model group was longer (P < 0.05),and sugar preference,DA and 5-HT in prefrontal cortex decreased dramatically (P < 0.05).Compared with the model group, the dead time of tail suspension test and forced swimming test in the low, middle, high dose of astilbin group was shorter (P < 0.05),and sugar preference and DA level in prefrontal cortex increased dramatically (P < 0.05).The 5-HT level in prefrontal cortex in the middle, high dose of astilbin group and IMI group was higher than that in the model group (P < 0.05),but there was no significant difference in the 5-HT level in prefrontal cortex between low dose of astilbin group and model group (P > 0.05).There was no significant difference in the dead time of tail suspension test and forced swimming test and sugar preference among the control group,IMI group and low, middle, high dose of astilbin group (P > 0.05).Conclusion Astilbin works excellently in anti-depression,and the major mechanism may involve in up-regulating the level of DA and 5-HT in prefrontal cortex.
