The secondary structure of the protein of DM0 and DMT in Oreochromis aureus were predicted by the methods of Garnier-Robson, Chou-Fasman and Karplus-Schulz based on the amimo acid sequences of DM0 and DMT. To investigate the induction of apoptosis of mouse colonic adenocarcinoma CT26 cells by recombinant Clostridium difficile toxin B (rTcdB), CT26 cells were exposed to different concentrations of rTcd B. Inhibition of cell proliferation was measured by MTT assay. The activation of Caspase 3 was measured by colorimetric method. Cell morphological analysis and flow cytometry were performed to confirm cell apoptosis. rTcd B inhibited the proliferation of CT26 cells in a timeand dose-dependent manner. Caspase 3 activity in CT26 cells was elevated remarkably after rTcd B exposure for 6 h, 12 h, 18 h or 24 h, as compared with the control group. Morphological changes were observed by fluorescence microscopy. The exposure of rTcd B to CT26 cells induced a timeand dose-dependent apoptotic cell death as determined by flow cytometry analysis. The results showed that recombinant Clostridium difficile toxin B induced apoptosis of CT26 cells.

Objective To explore psychological stress factors of patients in the ophthalmology department and psychological nursing interventions as well as effect, to promote the rehabilitation of patients. Methods A total of 289 patients from our ophthalmology department was investigated with the self-designed questionnaire. Results Major psychological stress factors of Ophthalmology inpatient were knowing nothing a variety of points for attention(92%), fearing and worrying the disease and surgery(85%), worrying about the level of health care technology of medical staff (84%). Psychological stress of patients released through psychological nursing intervention measures such as cognitive intervention and environmental intervention. 243 patients of fearing and worrying the disease and surgery reduced to 35 before and after intervene, The difference was statistically significant (X2 = 299.84, P < 0. 01). Conclusions We should properly guide and inspire positive mental patients based on patients' mental state, give mental nursing intervention, promote the rehabilitation of eye disease.
7.Research progress on translesion synthesis DNA polymerase ζ-Rev7 in tumor development
Basic & Clinical Medicine 2018;38(5):727-730
Translesion DNA synthesis is a kind of post-replication repair process, which mainly involves DNA polymerase kappa, eta and zeta.Translesion synthesis is a tolerance mechanism of DNA damage inside the cell, in which DNA polymerase zeta plays an important role.DNA polymerase zeta is mainly composed of subunits Rev7 and Rev3, and play a main role of participate in the damage repair in cell metabolism.Rev7(also known as MAD2L2)is a multifunctional protein that can be combined with a variety of proteins to participate in DNA dam-age repair, cell cycle regulation, gene expression and carcinogenesis, which is also involved in the prognosis of various adverse tumors.

AIM:To investigate the expression of extracellular matrix metalloproteinase inducer(EMMPRIN)and hepatocyte growth factor(HGF)in non-small-cell lung carcinoma(NSCLC)and their relationship with lymphoid metastasis and prognosis.METHODS:Expression of EMMPRIN and HGF in 77 cases of patients with NSCLC was detected immunohistochemically. The relationship of expression of EMMPRIN and HGF with tumor size, smoking, histological type, differentiation, lymphoid metastasis, clinical stage, and prognosis was analyzed.RESULTS:The expressive rates of EMMPRIN and HGF were 68% and 44%,respectively.The expressions of EMMPRIN and HGF were associated positively with lymphoid metastasis(r=0.371 and 0.339,P0.05).The expression of EMMPRIN was associated with the expression of HGF in NSCLC.CONCLUSION:The expression of EMMPRIN and HGF is associated with lymphoid metastasis and prognosis in NSCLC.Overexpression of EMMPRIN and HGF implies infavourable prognosis in NSCLC.

OBJECTIVETo study the influence of alcohol on the liver sinusoids endothelial cell (LSEC) fenestrae of rats.
METHODSSetting up the rat model of alcoholic liver disease by orogastric administration of alcohol, then kill the experimental and control groups of rats at the end of 4 weeks, 8 weeks and 12 weeks after alcohol feeding, and also at the end of another 12 weeks after balance foods feeding succeeding with alcohol feeding for 12 weeks. Staining the liver tissue by means of HE method and observing the successive change of LSEC fenestrae by transmission electron microscope.
RESULTSThe normal LSEC was flat with nucleus and organelle arranged regularly. The distal cytoplasm displayed as lamina with many fenestrae, not accompanied by basement membrane (BM) formation under the endothelial cell. At the end of 4 weeks of alcohol feeding, fenestrae decreased at the partial distal LSEC cytoplasm, but no BM developed. At the end of 8 weeks, fenestrae decreased significantly, even disappeared, with the BM developed incompletely under the endothelial cell. Concomitantly, fibroblast with active function developed. At the end of 12 weeks, the changes became more obvious; the complete BM could even be seen. However, this kind of changes was mostly limited in the single or adjoining sinusoids, as well as with little widespread formation of fibrosis. At the end of 12 weeks of stopping alcohol feeding, defenestrae and development of BM attenuated obviously.
CONCLUSIONThe defenestrae and BM of LSEC develop gradually with the chronic alcohol stimulation. Sinusoid capillarization and liver fibrosis even form when significant changes happen. The early change of the limited defenestrae and capillarization may be the basis of alcohol periportal fibrosis formation. This kind of liver fibrosis can be reversible after stopping alcohol feeding.

Chinese Journal of General Practitioners 2018;17(1):56-59
One hundred ten patients with Parkinson′s disease(PD)and 110 age-matched healthy controls were enrolled in the study. The non-motor symptoms(NMS)were assessed with Non-motor symptoms of Parkinson′s disease questionnaire(PDNMSQ30).The PD patients were classified as gait abnormality posture type(PIGD, n=73)and tremor type(TD, n=37).The non-motor symptoms were compared between two groups. Results showed that the NMS in PD patients was higher than that in healthy controls[(35.2 ±3.0)vs.(6.0 ±2.0)symptoms, t=6.43, P=0.01].The NMS score of PIGD group was higher than that of TD group[(28.9 ±1.1)vs.(6.2 ±0.7)symptoms, t=2.73, P<0.01]; Mini-mental state examination(MMSE)and Montreal cognitive assessment scale(MoCA)scores of TD group were significantly higher than those of PIGD group(t=11.52,P<0.01; t=8.79,P<0.01).The results indicate that for PD patients with different motor types, their NMS are different, and PD patients with PIGD have more axial involvement, and more NMS.
