1.THE ROLE OF ELASTIN IN EXPERIMENTAL ANIMAL MODEL OF ABDOMINAL AORTIC ANEURYSM

To investigate the relationship between the diameter of abdominal aortic aneurysm(AAA) and the concentration of elastin, forty five healthy male Wistar rats were selected,5 rats of them were used as normal control, the other 40 rats were randomly divided into 2 groups. A 1 cm segment of the abdominal aorta of the rat was isolated and perfused with pancreatic elastase in the experimental group, and the control group was given normal saline solution. Laparotomy was performed immediately after operation and on the 2, 7, 14 day, the aortas were measured and harvested. Histological studies were carried out to analyze the changes in elastin. The aortic diameter progressed to aneurysmal dimension in the experimental group. Histological study revealed loss of elastin in tunica media of the aorta in the experimental group. Neither macroscopic nor microscopic lesions were found in control group. It is suggested that there is a significant correlation between the aortic diameter and the decreased elastin content.
2.Early therapeutic effect of coronary artery bypass grafting with and without cardiopulmonary bypass

Objective:To evaluate whether off pump coronary artery bypass grafting (OPCABG) has advantages over conventional coronary artery bypass grafting(CCABG). Methods:A total of 362 patients undergoing coronary artery bypass grafting were divided into 2 groups: OPCABG group:152 cases undergoing OPCABG and beating heart via sternotomy; CCABG group,210 cases undergoing CCABG with extracorporeal circulation. Preoperative and postoperative variables of 2 groups were compared. Results:There were no significant differences in sex, age, weight, acute or remote myocardium infarction, hypertension, diabetes and type of bypass grafts between 2 groups.The numbers of bridge vessels were not significantly different between the 2 groups; the incidences of low cardiac output syndrome, atrial fibrillation and atelectasis in OPCABG group were significantly less than those of CCABG group( P

Objective:To evaluate the therapeutic effectiveness of peritoneal dialysis(PD) for acute renal failure(ARF) following cardiac surgery with cardiopulmonary bypass and its effect on hemodynamics. Methods:Retrospective data from 88 patients with ARF following cardiac surgery undergoing PD were analyzed. 5.Comparison of training in department of anesthesiology for flexible fibreoptic intubation with application of virtual reality simulator and high-fidelity manikin

Objective To compare the efficacy and efficiency of simulation-based training of flexible fibreoptic intubation in novices with virtual reality simulator.Methods A total of 46 anaesthesia residents in their first stage of training in anaesthesiology with no experience in flexible fibreoptic intubation at Peking University People's Hospital were enrolled in the study,and were divided into 2 groups randomly,which were virtual reality simulator group (group S,n=23) and manikin group (group M,n=23).The group S was then trained for 25 times on simulator,while the group M did the same processes on manikin.After training,participants in both groups had their performance assessed with the fibrescope evaluated through the oral route using a simulation manikin,who were instructed to attempt to advance the fibrescope 5 consecutive times to view the carina in the shortest amount of time.The time required to view the carina of each practice during training in both groups were recorded as pooled data to construct group learning curves with the application of SPSS 20.0.By using repeated measures analysis of variance and Ttest,the procedure time and global rating scale (GRS) of fibreoptic bronchoscope manipulation ability were compared between groups,so did the participant's confidence between before and after the training both within-subjects and between-subjects.Results The plateaus in the learning curves were achieved after 19 (15,26) practice sessions in group S and 24 (19,31) in group M,respectively.There was no significant difference in the procedure time [(13.7 ± 6.6) s and (11.9 ±4.1) s] and GRS [(3.9 ±0.4) vs.(3.7 ±0.3)]between groups.There were significant increases in participant's confidences in both groups after training [group S:(1.8 ± 0.5) vs.(3.9 ± 0.6),t=10.928,P=0.000;group M:(2.0 ± 0.7) vs.(3.9 ± 0.5),t=15.306,P=0.000],but there was no significant difference between groups.Conclusion The simulation-based training of flexible fibreoptic intubation in novices with virtual reality simulator is more efficient than the one with manikin,but the similar effects can be achieved in both modalities,after adequate trainings.In the related training a balance between time cost and economic cost should be considered and the appropriate teaching methods and forms should be taken.
6.Effects of interleukin-36ot on psoriasiform skin lesions and C-C motif chemokine ligand 20 expression in mice

Objective To evaluate effects of interleukin-36α (IL-36α) on psoriasiform skin lesions and C-C motif chemokine ligand 20 (CCL20) expression in mice.Methods Totally,30 BALB/c female mice were randomly and equally divided into 3 groups:control group treated with topical vaseline cream on the shaved back and intracutaneous injection with phosphate buffer saline (PBS),model group treated with topical imiquimod cream on the shaved back and intracutaneous injection with PBS,experimental group treated with topical imiquimod cream on the shaved back and intracutaneous injection with IL-36α solution.Psoriasis area severity index (PASI) was used to evaluate changes of psoriasiform skin lesions in mice,and light microscopy to observe morphological changes of skin lesions and to measure the thickness of the epidermis.Real-time fluorescence-based quantitative PCR (qRT-PCR) and Western blot analysis were performed to determine the expression of IL-36α in skin lesions in the control group and model group,and qRT-PCR,Western blot analysis and immunohistochemical study to evaluate changes of CCL20 levels in skin lesions.Results The model group showed significantly increased mRNA (△ Ct value:0.0195 ± 0.0059) and protein expression (3.922 ± 0.248) of IL-36α compared with the control group (mRNA:0.0012 ± 0.0004,P < 0.05;protein:0.690 ± 0.025,P < 0.05).The mRNA and protein expression of CCL20 were significantly higher in the experimental group than those in the model group (mRNA:2.152 ± 0.793 vs.0.999 ± 0.178;protein:0.397 ± 0.033 vs.0.145 ± 0.030;both P < 0.05),and higher in the model group than those in the control group (mRNA:0.378 ± 0.075;protein:0.025 ± 0.009;both P < 0.05).Immunohistochemical study showed that the expression intensity of CCL20 in skin lesions significantly increased in the experimental group compared with that in the model group (Z =2.294,P < 0.05).Conclusion IL-36α may aggravate psoriasiform skin inflammation in mice by promoting CCL20 expression.

Nine compounds were isolated from an ethanol extract of the roots of K. roxburghii by using a combination of various chromatographic techniques including column chromatography over silica gel, MCI gel, Sephadex LH-20, and reversed-phase HPLC. On the basis of physical-chemical properties and spectroscopic data analysis, their structures were identified as munjistin (1), 1-methoxy-3,6-dihydroxy-2-hydroxymethyl-9,10-anthraquinone (2), 1,2,3-trihydroxy-9,10-anthraquinone (3), arjunolic acid (4), hyptatic acid-A (5), hyptatic acid-B (6), 2α,3β,24-trihydroxyurs-12-en-28-oic acid (7), 2α,3β,23-trihydroxyurs-12-en-28-oic acid (8), and daucosterol (9). Compounds 1-9 were obtained from this genus for the first time.

8.Characterization of microstructure of ibuprofen-hydroxypropyl-beta-cyclodextrin and ibuprofen-beta-cyclodextrin by atomic force microscope.

The microstructures of ibuprofen-hydroxypropyl-bets-cyclodextrin (IBU-HP-beta-CyD) and ibuprofen-beta-cyclodextrin (IBU-beta-CyD) were observed by atomic force microscope (AFM). The high resolving capability of AFM has the tungsten filament probe with the spring constant of 0.06 N x m(-1). Samples were observed in a small scale scanning area of 10.5 nm x 10.5 nm and 800 x 800 pixels. The original scanning images were gained by tapping mode at room temperature. Their three-dimensional reconstruction of microstructure was performed by G3DR software. The outer diameters of HP-beta-CyD and beta-CyD are 1.53 nm. The benzene diameter of IBU is 0.62 nm, fitting to the inner diameters of HP-beta-CyD and beta-CyD. The benzene and hydrophobic chain of IBU enter into the hole of cyclodextrin at 1:1 ratio. The results were evidenced by IR, X-ray diffraction and the phase solubility.

9.Experimental study of mandibular periosteal distraction in rabbits.

OBJECTIVETo investigate a novel technique for new bone formation--periosteal distraction osteogenesis.
METHODSA custom made periosteal distraction device was fixed to bilateral surface of the mandible in three rabbits. Periosteal distraction was performed on the left side of the mandible, the right side of the mandible served as the control. The animals were sacrificed at the end of distraction process. All the specimens were X-rayed and histologically examinated.
RESULTSAll three animals survived with no obvious complications. Both in mass specimens and X-rays, there showed new bone formation on the distracted side of the mandible. In histological examinations, there was osteoblast-like cell infiltration and bone tissue formation in the distracted area.
CONCLUSIONPeriosteal distraction osteogenesis can provide a novel technique for the repair of bone defects.

10.Study on stability of oridonin solution.

OBJECTIVETo investigate the stability of oridonin (ORI) solution for research and development of novel ORI prepartions.
METHODThe effect of pH on the degradation rate of ORI was evaluated, the pH values of the oridonin solutions were adjusted to the setting pH, with 1 mol x L(-1) HCl or NaOH, respectively, and stored at room temperature for 60 h. The constant temperature method was applied to evaluate the stability of ORI solution at room temperature and at 4 degrees C.
RESULTThe pH-rate profile of ORI was V-shaped, and the pHm was 5. The t90 of ORI solution at room temperature was 53.2 h and 91.5 h at 4 degrees C CONCLUSION: The ORI solution is not stable. The pH-dependent degradation of ORI solution confirms to specific acid-base catalysis reaction.
