1.Invasion Suppressor Role of E-Cadherin in Epithelial Cancer Cell Lines.

BACKGROUND: The generation of the invasiveness in transfromed cells represents an essential step of tumor progression. The primary cause of the scattering of the cells in invasive carcinoma is a loss of the integrity of the intercellular adherens junction often involving loss of a functional cell-cell adhesion molecule E-cadherin. Therefore, the perturbation of E-cadherin function causes diaggregation of tumor cells and may promote the invasion and metastases. OBJECTIVE: The reduction in E-cadherin activity seems to correlate with the infiltrative ability of tumor cells. The purpose of this study was to compare the E-cadherin expression among different cell lines which were normal to undifferentiated and to check the virtual relationaship between E-cadherin and invasiveness. METHODS: We used 5 cell lines, HaCaT, A431, C3, SiHa and HeLa cell. To check the expression patterns and amounts of E-cadherin in each cell line, immunofluorescence staining, Western blot anlysis and Northern blot analysis were done. An in vitro invasion assay using the collagen gel and MRC-5 fibroblast under the influence of HECD-1 antibody which block the E-cadherin function was done to measure the invasiveness of tumor cells. Collagenase activity in culture supernatants of each cell were analyzed by zymography. RESULTS: Immunofluorescence staining revealed a homogenously well preserved pattern in HaCat, A431, C3 cells. SiHa cells showed patch distribution but HeLa cells did not express the E-cadherin. Western blot analysis and Northern blot results largely corresponded with the immunofluorescence results. The in vitro invasion assay revealed invasion into the collagen matrix of the HeLa cells. When HECD-1 antibody was added to the medium, other cells showed partially disrupted stratification. The collagenolytic activity at 72 kDa sixe was detected in the HeLa cell line only. CONCLUSION: There is an inverse relationship between E-cadherin expression and tumor invasion. Therefore, through their regulation of cell adhesion and motility, cadherin plays a crucial role in the suppression of tumor invasion and metastasis.

2.An Epidemiologic Investigation of Typhoid Fever Outbreak in 0hchun Middle and High Schools Located in Young-il Gun Kyungpook Province.

An epidemiologic investigation was conducted to identify the source of infection and the mode of transmission of typhoid fever outbreak occurred in Ohchun middle and high schools located in Young il Gun, Kyungpook Province from May 21 to Jun 4, 1994. Four out of 13 hospitalized students in three general hospitals in Pohang City were culture positive typhoid fever (Salmonella typhi Group D). Review of the in and outpatient logbooks of the three general hospitals in Pohang City, county health center and local clinics in the vicinity of the Ohchun middle and high schools suggested that the outbreak was confined to the schools. Drinking water source of the schools was two underground water pumps that had no treatment facilities; one in northern end and the other in southern end of the main school building. However, the southern water pump was closed on April 28, 1994 because a recent test for the water revealed E-coli suggesting a possible fecal contamination. Two hundred twenty students stated that they had experienced symptoms related to typhoid fever since April l, 1994 in a self administered questionaire survey of all of the 2, 226 students. Personal interviews with these 220 students were performed to confirm the clinical history together with a rectal swab culture for typhoid fever and Widal test. A total of 39 patients were detected; 15 confirmed cases (culture-positive) and 24 suspected cases (symptom positive with equal to or greater than 1:160 titer of Salmonella O-antibody or 1:320 titer of H-antibody titer in Widal test). The epidemic curve showed a unimodal curve with a peak on the second week(17 May, 1994) and tailed down to the 5th week (22 28 May, 1994). The overall incidence rate of typhoid fever was 1.8 per 100 students; 1.5 (1.6 for male, 1.5 for female) in the middle school and 2. 4 (6. 2 for male, 0. 4 for female) in the high school. When contrasted with the students who did not drink water at all in the school, the relative risks of incidence rate of typhoid fever for the students who drank the underground water of the school, both underground water and water brought from home, or water brought from home only were 55.5, 19.0, and 3.0 respectively. Three classes which had especially high incidence rates of typhoid fever were located in the southern part of the main school building, where the contaminated underground water pump is located. These findings suggested that the source of infection was the contaminated underground water of the school. Both of the two underground water pumps were closed permanently and the public running water was supplied to the schools.

3.Albumin as a Predictor of Survival in Ambulatory Community Dwelling Korean Elderly.

PURPOSE: This study performed to identify the association between various nutritional indices including biochemical, anthropometric variables and mortality in Korean elderly. METHODS: Among a total of 151,000 subjects over 40 years of living in Jung-up district, 1051 was selected using a random cluster sample method. Between Dec 1996 and Feb 1997, they underwent the blood tests and anthropometric measurements. Three years later, on Dec 2000, we investigated the mortality of the 340 subjects that had been over 65 years at the enroll time. RESULTS: Of the 1051 subjects(M:F=410:641), 340 were over 65 years at the enroll time, 34(10%) had died by 3 years follow up. After adjustment of age and sex, decreased Hb, Hct, serum protein, albumin, increased serum creatinine were significantly associated with an increased risk of mortality(p<0.05) among the variables. Especially serum albumin remained strong predictors of mortality after cont rolling for other significant variables(RR=0.15, 95% CI=0.03-0.63, p=0.01). CONCLUSION: We confirmed the strong independent association with serum albumin and mortality in the rural community dwelling ambulatory elderly Koreans.

4.Studies on the B Cell Proliferation and Differentiation Factors in Human B Cell System.

We have studied the function of lymphokines on human tonsillar B cell prolifertion and differentiation. B cells were stimulated with Staphylococcus aureus Cowanl (SAC) or anti- bead. The followings showed the results of this study. 1) In B cell activation, SAC induced B cell DNA synthesis but anti-mubead did not. SAC could activate and proliferate B cells. Minimal number of B cells were required to proliferate effectively. 2) In B cell proliferation, SAC could proliferate B cell in the abscence of lymphokines. Exogenous IL-2 or IL-4 enhanced B cell proliferation. The roles of IL-2 were very important in B cell proliferation. The effect of IL-4 on the IL-2 induced B cell proliferation was inhibitory in SAC-B cells. IL-4 could enhance the proliferation of anti-mu bead activated B cells. 3) In B cell differentiation, IL-2 was a major factor to differentiate SAC activated B cells, but IL-4 did not. IL-6 had a synergistic effect on the differentiation. The results of this study showed that the different signal transduction mechanisms were involved in B cell proliferation and differentiation. The B cell resposes to lymphokine were different, and it is depend upon antigens or mitogens.

5.Characteristics of Obstructive Sleep Apnea Syndrome Patients Proven with Nocturnal Polysomnography as Correlates of Age and Gender.

OBJECTIVES: The purpose of this study was to assess the clinical and polysomnographic characteristics of Korean patients with obstructive sleep apnea syndrome (OSAS), especially in relation to differences due to age and gender. METHODS: All subjects were consecutive patients who were proven to have OSAS with nocturnal polysomnography. They were interviewed with a structured interview format including sociodemographic information, past medical history, medication, and sleep-related history. Simultaneously, they were also given Pittsburgh Sleep Quality Index (PSQI) and Epworth Sleepiness Scale (ESS) to answer in order to check subjective sleep quality and subjective sleepiness. RESULTS: Mean age of the 308 subjects was 49.5+/-13.3 years, with 77.6% of the subjects being males and 22.4% of the subjects being females. The aging effects on the sleep architecture in Korean OSAS corresponded with normal aging, but with the effect of OSAS itself superimposed, the extent of aging effects was more marked than that of normal aging. The severity of Korean patients of OSAS was not correlated with age. When divided into age subgroups, significant correlation was found between RDI and BMI in patients of each subgroup of those in the 4th to 7th decades. The oldest subgroup (>70 years) described their subjective sleep quality as poorer than any other age subgroups, despite of less subjective drowsiness. The severity of OSAS and the change of sleep architecture of male subjects turned out to be severer than those of female ones. The female/male ratio of the subjects tended to increase with aging. CONCLUSIONS: The aging effect on the sleep architecture in Korean OSAS seems to be a mixture of the changes by normal aging and sleep disorder per se. The severity of OSAS was not correlated with age, but highly correlated with BMI. The severity of OSAS and the change of sleep architecture of male patients were severer than those of female ones.

8.Effects of Nitric Oxide on the Maturation of Mouse Oocyte in vitro.

OBJECTIVE: Nitric oxide (NO) produced in ovary may contribute to follicle maturation, ovulation, oocyte maturation and luteinization. In this study, the effect of nitric oxide on the spontaneous maturation of mouse oocyte was observed. Method: The index of oocyte maturation was checked by the germinal vesicle breakdown (GVBD) and appearance of polar body (PB) under microscope in the denuded oocytes and oocyte-cumulus complexes (OCCs) from mouse ovarian follicles after 24 hours pregnant-mare serum gonadotropin treatment. RESULTS: The GVBD appeared 50 %, 1 hour and 80 %, 2 hrs after changes of oocytes from dibutyryl cAMP (dbcAMP, 0.5 mM) contained media into dbcAMP-free media. dbcAMP (0.5 mM) completely blocked the GVBD until 24 hrs but dbcGMP (5 mM) delayed the GVBD by 1 hr. Sodium nitroprusside, the NO generator, inhibited the GVBD dose-dependently at 2 hr incubation in denuded and OCCs. The appearance of GVBD was not different between control and dbcGMP or SNP in denuded oocytes and OCCs at 24 hrs incubation. The guanylate cyclase activity in denuded oocyte cytosol was not detected whereas the guanylate cyclase activity in OCCs cytosol was 1.3 nmole/min/mg protein which was increased about 3 times by SNP (100 micrometer). CONCLUSION: These results suggest that the NO in ovary may delay the spontaneous oocyte maturation in early stage by acting on the maturation signaling protein as well as guanylate cyclase.

9.A STUDY ON ACCUARACY AND DIMENSIONAL STABILITY ACCORDING TO IMPRESSION METHODS.

Accurate reproduction of intraoral tissue is essential in the fabrication of fixed prosthesis. This selection of impression material and method is very important. In this study, vinyl polysiloxane type impression material Panasil was used. Three impression methods ; one-step technique, individual tray technique, and individual tooth tray technique using the vinyl polysiloxane type bite registration material Futar Occlusion, were used to take horizontal and vertical impression. Improved stone models were fabricated. The amount of occlusal surface discrepaneies, interabutment distance discrepancies, amount of marginal defect were evaluated with the X-PLAN360d and photoscanning. The results were as follows. 1. There was no significant difference in occlusal surface according to the directions and techniques of impression taking. 2. There was no significant difference in interabutment distance discrepancies according to the directions and techniques of impression taking 3. There was no significant difference in marginal discrepancies according to the direction of impression taking but there was significant difference between one-step technique and individual tray technique, individual tooth tray technique (P<0.05). Taken together, these results suggest that individual tray technique and individual tooth tray technique are more accurate for impression taking than one-step technique.
