Chinese Journal of Immunology 1985;0(02):-
Objective:To clone the outer membrane protein hopX gene of Helicobacter pylori(Hp)and to perform sequencing and analysis of biological information. Methods:Polymerase chain reaction(PCR)was used to amplify the hopX gene from Hp chromosomal DNA.Then the target gene was digested by restricted endonuclease enzyme of BamH I,and inserted into the prokaryotic expression vector pQE30 digested by corresponding restricted endonuclease enzyme. The recombinant vector was used to select and transform for nucleotide sequence analysis. The biological property at the amino acid level was analyzed by Omiga 2.0 and Antheprot v 5.0.The transformant colony was induced with IPTG and the fusion protein was analyzed by SDS-PAGE and Western blot. Results:The recombinant plasmid was constructed.DNA sequence analysis showed the sequence of hopX was 1 284 bp.The homology of the strains in nucleotide acid was 96%~97%.Their homogeneity in the amino acids was 97%~99%.We get a GeneBank accession number EF208122.Omiga 2.0 software predicted its relative molecular mass(Mr.)was 47 kD and possessed good antigencity. The expressed product contained about 37% of total somatic proteins and Western blot method showed good antigenicity of the recombinant protein. Conclusion:A confirmed gene hopX has been obtained, providing a good foundation for recombination, expression and related study. The corresponding peptide of the gene performed the structural characteristics of some typical antigen molecules, which suggest that it might be a novel vaccine candidate.

AIM: To investigate the effect of Auricularia Polytricha Sacc Polysaccharide( APSP) on protein kinase A( PKA) activity in murine peritoneal macrophages METHODS: New ion- pair RP- HPLC method was used to determinate the activity of PKA in murine peritoneal macrophages RESULTS: APSP( 80mg/L) induced activation of PKA in murine peritoneal macrop_ hages; the peak appeared at 8 minutes and the activity of PKA returned to the basic level at 20 minutes CONCLUSION: The immunopoteniating effect of APSP may be associated with its activating effect on PKA in murine peritoneal macrophages

Chinese Journal of Nosocomiology 1994;0(01):-
OBJECTIVE To explore the clinical distribution and drug resistance of extended-spectrum ?-lactamases(ESBLs) producing Enterobacter cloacae.METHODS The clinical distribution and drug resistance of 73 strains of E.cloacae and ESBLs producing E.cloacae identified in our hospital from Jan 2002 to Jan 2004 were analyzed.RESULTS Among them, the highest detectable rate was in respiratory department and from phlegm specimen. No ESBLs producing E.cloacae and non-(ESBLs) were resistant to imipenem;no non-ESBLs were resistant to cefepime, but some ESBLs producing E.cloacae was resistant to it;all ESBLs producing E.cloacae and non-(ESBLs) were resistant to ceftriaxone and amoxicillin/clavulanic acid.CONCLUSIONS E.cloacae is one of the(commonest) pathogens in nosocomial infection, and ESBLs producing E.cloacae has a high detectable rate, its drug resistance has increased;the clinic should choose antimicrobial agents rationally according to drug sensitive tests in vitro.

Journal of Applied Clinical Pediatrics 2004;0(09):-
Objective To study the correlation between single nucleotide polymorphisms of -238,-308 G/A in promoter region of tumor necrosis factor -?(TNF-?) gene and the type of juvenile idiopathic arthritis (JIA).Methods Clinical data and blood preparation of 127 children with JIA and 106 healthy children were evaluated.Subgroups of JIA were defined according to the Edmonton criteria. The -238 G/A and -308 G/A polymorphisms in DNA analysis in this study were extracted from the whole blood. The restricted fragment length polymorphisms were determined in the cases of all JIA children and control group. Results 1.The TNF-?-238 G/A allele frequencies of JIA group and control group:allele frequency of JIA group was 92.9% and 7.1%,and the control group was 95.3% and 4.7%.The distribution of allele frequencies was no significantly different between JIA group and control group(?2=1.149 P=0.284).But there were significant difference between polyarticular JIA (RF negative) and control group(?2=7.621 P=0.006).2.The TNF-?-308 G/A allele frequencies of JIA group and control group:allele frequency of JIA group was 94.1% and 5.9%,the control group was 95.3% and 4.7%.The distributions of allele frequencies was no significantly different between JIA group and control group(?2=0.322 P=0.571).There were significantly difference between polyarticular JIA (RF negative) and control group (?2=7.621 P=0.006).Conclusions The TNF-?-238,-308 polymorphisms of A in the-238 and-308 TNF-? gene are important to the joint destruction of JIA.The study will be beneficial to provide indirect support to the application of anti-TNF drugs to the treatment of JIA.

Chinese Acupuncture & Moxibustion 2014;34(6):615-618
During professor SHAO Jing-ming's academic research and medical practice, his academic opinion of focusing spirit is gradually developed. In terms of nurturing the spirit, attention should be paid on persistence as well as everyday health maintenance and exercise to nurture the spirit of physician. In terms of clinical diagnosis and treatment, patients' psychology, employment and life status should be observed and experienced, which could bring more methods to take essential care of patients' spirit. The treatment should work with psychological counseling, advocating that based on patients' qi and spirit, various forms of treatment methods should be properly used, such as acupuncture or moxibustion or combination of acupuncture and medicine, along with simple acupoint selection and harmony medication. Before clinical treatment of acupuncture, calming the mind is critically emphasized to make a clear diagnosis. During the acupuncture, calming and focusing the mind is necessary as well as emphasizing the details, so acupuncture could be integrated with Chi Gong to create a new warming-sensation technique. In a word, the academic opinion of focusing spirit is shedding an inspiring light upon further study.

8.Study on the law of professor SHAO Jing-ming's clinical point selection.
Chinese Acupuncture & Moxibustion 2006;26(2):126-128
Professor Shao Jing-ming forms an unique academic thought and technique specialty on the basis of carefully study on medical classics, widely extensive learning, and diligent practice. In several ten years' clinical work, he uses TCM theories as basis and channel theories as guidance, following the standards of treatment based on syndrome differentiation, and in clinical acupuncture and moxibustion treatment, he emphasizes "selection of primary and secondary acupoints, and needling in order of priority". In clinical treatment of various diseases, 3 points are commonly used as main points, with accurate and proper selection of acupoints.

Chinese Journal of Infectious Diseases 2010;28(9):519-523
Objective To understand the nucleotide and amino acid differences of glycoprotein gene (G gene) between isolated rabies viruses in Henan Province and rabies vaccine strains used for human and animals. Methods G gene sequences of nine rabies viruses isolated from dogs in Xinyang city of Henan Province in December 2006 were amplified by reverse transcriptase (RT)-heminestedpolymerase chain reaction (PCR), and then were cloned and sequenced. The phylogenetic trees were constructed for analyzing the genetic characteristics of these rabies viruses. Results The homology of G gene among the nine isolates from Henan Province was 97.6% - 98.9% at nucleotide level and 99.2%-99.8% at amino acid level. The similarities between these isolates and CTN vaccine strain were 85.6%-93.0% and 91.9%-92.9% at nucleotide and amino acid level, respectively, which were higher than those between these isolates and other vaccine strains (80.4% - 83.3% and 87.7% - 92.5% at nucleotide and amino acid level, respectively). The nine isolates had several amino acid substitutions when compared to other genotype 1 rabies virus strains. Conclusions The nine rabies viruses strains isolated from Henan Province all belong to genotype 1. CTN may be an effective vaccine for preventing rabies in Henan Province.
