1.Measurement of Apoptosis by Staining with 7-Amino-Actinomycin D with Concurrent Dual Color Immunofluorescence by Single Laser Flow Cytometry.

Apoptosis is a crucial mechanism for the selective elimination of mammalian cells and is involved in many physiological and pathological processes. The heightened awareness of the importance of apoptosis has increased the need for rapid and quantitative methods for measurement of apoptotic changes. Recently, 7-amino-actinomycin D (7-AAD) has been introduced as a valuable fluorescent dye for assessing apoptosis by flow cytometry. When the cells are stained with 7-AAD in the concentration of 10 - 20 ug/ml, live cells are not stained (7-AAD ) and early apoptotic cells are weakly stained (7-AAD ) while late apoptotic or dead cells are stained brightly (7-AAD). On scattergram of forward angle light scatter vs. 7-AAD fluorescence, the three populations can be discriminated not only between each other but also from cell debris or clumps. The apoptotic cells, defined as 7- AAD cells, were demonstrated as apoptotic by morphological observation of the sorted cells. The 7-AAD cell fraction was also demonstrated to be parallel with subdiploid fraction of cells stained with PL However, 7-AAD cells, whose definition is based on the alteration of membrane integrity, have never been demonstrated to be subdiploid fraction by simultaneous DNA staining. Here we directly demonstrate that 7-AAD cells, defined on the scattergram of forward angle light scatter vs. 7-AAD fluorescence, are subdiploid fraction by staining with DNA dye whose fluorescence is collected after 530/30 band pass filter (FL-1). We also demonstrate the effects of 7-AAD concentration, fixation of cells, and proliferation of cells, on the fluorescence pattern, for reference during assessment of apoptosis by simple and rapid method for flow cytometric analysis of cells stained with 7- AAD. We also present a flow cytometric analysis of cells stained with 7-AAD Eor sequential change in apoptotic fraction, with concurrent dual-color immunophenotyping.

2.A Study of Health Professionals Awareness, Satisfaction and Desirable Nutrition Labeling of Foods for Special Dietary Uses.

This survey was conducted to evaluate with 457 health professionals from July, 1999 to September, 1999 the awareness of and satisfaction with the nutrition labeling system in Korea and to determine a desirable labeling system. Professors of food and nutrition and nutrition researchers in the food industry had the hightest scores on awareness of the nutrition labeling system(75.6% and 73.4%, respectively), which was significantly different from each other by occupation(p<0.01). Frequently checked labeling information were expiration date, production date, price, and company respectively. Special nutrient food producers(56.9%), dietitians(49.3%), nurses(48.1%), researchers(42.3%) answered moderately on satisfaction with nutrition information for products, which was statistically significant(p<0.01). This suggests that health professionals were unsatisfied with the present nutrition labeling system. In nutrition focusing statements, nutrient content claims, health claims, and working periods were significantly related with nutrition focusing statements and health claims, but not with nutrient content claims : the subject who had been working for more than 10 years and less than 5 years had positive thoughts or those matters while the subjects whose working periods were between 10 and 5 years had the lowest score. The most desirable labeling units were serving size(65.1%). Ninety two percent of the subjects wanted absolute nutrient contents in food and their percentage of the KRDA. Eight nine percent of health professionals agreed that the health claims should be used only with scientific approval by the government or food research institutions. Only 4.4% of subjects were satisfied with current regulations of health claims(p<0.001). More studies regarding labeling units, their range and nutrient reference values are needed. The opinions of health professionals in nutrition labeling system should discussed to establish a desirable nutrition labeling system.

4.Histologic difference of the nasal mucosa of both nasal cavities in patients with nasal septal deviation.

BACKGROUND AND OBJECTIVES: Patients with nasal septal deviation usually manifest compensatory hypertrophy of the mucosa of unilateral nasal cavity. However, the histologic difference of the bilateral septal mucosa of both nasal cavities in patients with nasal septal deviation has not been extensively studied. Thus we tried to investigate histologic difference of both the septal mucosa of both sides in patients with nasal septal deviation. MATERIALS AND MEHTODS: Nasal mucosae taken from 15 patients with nasal septal deviation were studied. About 0.5 x 0.5 cm sized piece of septal mucosae were obtained from the corresponding site of both nasal cavities at 2 cm from the nostril and 2 cm inferior to the nasal dorsum. After staining by hematoxylin-eosin, the histologic differences of the bilateral nasal mucosa were examined under a light microscope. RESULTS: Surface epithelia of both nasal cavities showed squamous metaplasia or epithelial exfoliation. Inflammatory cell infiltration was more severe in the concave side compared with the convex side. The number of nasal gland were significantly higher in the convex side than in the concave side. CONCLUSIONS: The result of this study indicated that the concave and convex nasal mucosae in patients with nasal septal deviation have different histologic features in terms of inflammatory cell infiltration, and number of nasal glands.

5.The effect of irradiation modes on polymerization and microleakage of composite resin.

The aim of this study was to investigate the effect of light irradiation modes on polymerization shrinkage, degree of cure and microleakage of a composite resin. VIP(TM) (Bisco Dental Products, Schaumburg, IL, USA) and Optilux 501(TM) (Demetron/Kerr, Danbury, CT, USA) were used for curing Filtek(TM) Z-250 (3M Dental Products, St. Paul., MN, USA) composite resin using following irradiation modes: VIP(TM) (Bisco) 200mW/cm2 (V2), 400mW/cm2 (V4), 600mW/cm2 (V6), Pulse-delay (200 mW/cm2 3 seconds, 5 minutes wait, 600mW/cm2 30seconds, VPD) and Optilux 501(TM) (Demetron/Kerr) C-mode (OC), R-mode (OR). Linear polymerization shrinkage of the composite specimens were measured using Linometer (R&B, Daejeon, Korea) for 90 seconds for V2, V4, V6, OC, OR groups and for up to 363 seconds for VPD group (n=10, each). Degree of conversion was measured using FTIR spectrometer (IFS 120 HR, Bruker Karlsruhe, Germany) at the bottom surface of 2 mm thick composite specimens. V2, V4, V6, OC groups were measured separately at five irradiation times (5, 10, 20, 40, 60 seconds) and OR, VPD groups were measured in the above mentioned irradiation modes (n=5, each). Microhardness was measured using Digital microhardness tester (FM7, Future-Tech Co., Tokyo, Japan) at the top and bottom surfaces of 2mm thick composite specimens after exposure to the same irradiation modes as the test of degree of conversion(n=3, each). For the microleakage test, class V cavities were prepared on the distal surface of the ninety extracted human third molars. The cavities were restored with one of the following irradiation modes: V2/60 seconds, V4/40 seconds, V6/30 seconds, VPD, OC and OR. Microleakage was assessed by dye penetration along enamel and dentin margins of cavities. Mean polymerization shrinkage, mean degree of conversion and mean microhardness values for all groups at each time were analyzed using one-way ANOVA and Duncan's multiple range test, and using chi-square test for microleakage values. The results were as follows: 1. Polymerization shrinkage was increased with higher light intensity in groups using VIP(TM) (Bisco): the highest with 600mW/cm2, followed by Pulse-delay, 400mW/cm2 and 200mW/cm2 groups. The degree of polymerization shrinkage was higher with Continuous mode than with Ramp mode in groups using Optilux 501(TM) (Demetron/Kerr). 2. Degree of conversion and microhardness values were higher with higher light intensity. The final degree of conversion was in the range of 44.7 to 54.98% and the final microhardness value in the range of 34.10 to 56.30. 3. Microleakage was greater in dentin margin than in enamel margin. Higher light intensity showed more microleakage in dentin margin in groups using VIP(TM) (Bisco). The micoleakage was the lowest with Continuous mode in enamel margin and with Ramp mode in dentin margin when Optilux 501(TM)(Demetron/Kerr) was used.

6.Sclerosing peritonitis mimicking ovarian carcinoma: Associated with ovarian teratoma.

Sclerosing peritonitis is an unusual fibrosing condition predominantly involving the omentum and simulating carcinoma. The presenting signs and symptoms, imaging examination and cancer antigen 125 (CA-125) status in sclerosing peritonitis sometimes resemble those of ovarian cancer. Thus, the possibility of sclerosing peritonitis should be considered in the differential diagnosis of ovarian carcinoma. It may occur idiopathically and secondary to chronic peritoneal dialysis, the use of peritoneovenous shunt, practolol therapy, or in association with ovarian tumors such as ovarian teratoma. We report a case of peritonitis initially suspected as ovarian carcinoma but diagnosed as sclerosing peritonitis associated with teratoma.

7.Long Term Effects of Hospital Information System on Nurses' Job Pattern and Satisfaction, and Attitudes Toward HIS.

OBJECTIVE: The purpose of this study was to examine the long-term effects of Hospital Information System(HIS) on nurses' job pattern and satisfaction, and attitudes toward electronic medical record system. METHODS: Prospective survey using repeated measures design was performed to compare the changes between 2 years and 4 years after introduction of HIS in a tertiary hospital. Participants were 374 nurses working in the target hospital. Questionnaire was developed by researchers based on Walker, Eyman, Krall, Prophet, and Flanagan(1996)'s study and consisted of 21 questions about job pattern, job satisfaction, and attitudes toward computerization and paper record system. RESULTS: Overall time for documentation, shift reporting, communication with other departments were reduced. Otherwise, direct nursing time was not increased after HIS. Nurses showed positive responses on changes of job pattern after HIS while job satisfaction has been decreased. In addition, nurses' attitudes toward hospital information system and paper medical record system showed they were accepting computerized information system. CONCLUSION: This study identified the long-term positive effects of HIS and the need for nursing sensitive hospital information system.

8.Visualization of a Traumatic Pseudoaneurysm at Internal Carotid Artery Bifurcation due to Blunt Head Injury: A Case Report.

Traumatic intracranial pseudoaneurysms occurring after blunt head injuries are rare. We report an unusual case of subarachnoid hemorrhage (SAH) caused by rupturing of the traumatic pseudoaneurysm of the internal carotid artery (ICA) bifurcation that resulted from a non-penetrating injury. In a patient with severe headache and SAH in the right sylvian cistern, which developed within 7 days after a blunt-force head injury, a trans-femoral cerebral angiogram (TFCA) showed aneurysmal sac which was insufficient to confirm the pseudoaneurysm. We obtained a multi-slab image of three dimensional time of flight (TOF) of magnetic resonance angiography (MRA). The source image of the gadolinium-enhanced MRA revealed an intimal flap within the intracranial ICA bifurcation, providing a clue for the diagnosis of a dissecting pseudoaneurysm at the ICA bifurcation due to blunt head trauma. We performed direct aneurysmal neck clipping, without neurological deficit. A follow-up TFCA did not show either aneurysm sac or luminal narrowing. We suggest that in the patient with a history of blunt head injury with SAH following shortly, multi-slab image of 3D TOF MRA can give visualization of the presence of a pseudoaneurysm.

10.COVID-19–Related Functional Impairment in a Community Sample of Korean Adults: Associations With Depression, COVID-19 Infection Fear, and Resilience

Objective:
We aimed to determine the effects of depression, COVID-19 infection fear, and resilience on COVID-19–related functional impairment.
Methods:
We obtained data from 476 community-dwelling adults aged 20–69 years living in Jeju, South Korea, and evaluated the relationships between COVID-19–related functional impairment (work/school, social, and home life) and sociodemographic and healthrelated characteristics, COVID-19–related life changes (financial difficulties since the pandemic, employment change, interpersonal conflict), and clinical characteristics, including depression, COVID-19 infection fear, and resilience.
Results:
Functional impairment in the home life domain was associated with marital status and monthly income. Greater work/school, social, and home life functional impairment was significantly associated with all COVID-19–related life changes. Regression analysis indicated that resilience modulated the positive associations of COVID-19–related functional impairment with symptoms of depression and COVID-19 infection fear when relevant factors were controlled for.
Conclusion
Our results suggest the importance of clinical characteristics, including depression, COVID-19 infection fear, and resilience for understanding functional impairment related to COVID-19. These results have important implications for interventions aimed at reducing depression and COVID-19 infection fear, and enhancing resilience.
