Journal of Zhejiang Chinese Medical University 2015;(5):372-377
Objective]To establish a new method for grafting ratio(GR) analysis of deoxycholic acid grafted chitosan(CS-DCA). [Methods]In this study, linear potentiometric titration(LPT) and 2,4,6-trinitrobenzenesulfonicacid(TNBS) were introduced to measure the GR of CS-DCA, using the 1H-nuclear magnetic resonance(1H-NMR) as reference, the results obtained by the two methods were analyzed to find a method to substitute H-NMR. [Results]There was no significant difference of the data determined by 1H-NMR and LPT(P>0.05), however, it didn't apply to 1H-NMR and TNBS(P<0.05), the results indicated that LPT had equivalent accuracy of 1H-NMR. Using 1H-NMR method as the standard reference, the relative error was no more than 3.8% for the method of LPT, and the maximum relative error for the method of TNBS was up to 12.8%, it meant that LPT had higher precision. [Conclusion]LPT is an economical, rapid and accurate method for the GR analysis of CS-DCA.
7.Effects of QingReYangYin Recipe on the Ovaryhormone and Local Insulin Resistance in Rat Model of Polycystic Ovary Syndrome
Journal of Zhejiang Chinese Medical University 2015;(5):361-365
Objective]To observe and understand the effect and mechanism of QingReYangYin Recipe(QRYYR) on the ovary hormone level and mRNA expressions of PI3K and PKB in adipose tissue around womb and ovary in rat model of polycystic ovary syndrome(PCOS).[Methods]Rat PCOS model was established by Poretsky method and divided randomly into four groups: model group, western medicine group, low dose QRYYR group and high dose QRYYR group. Normal rats were used as normal group. All rats were orally administrated for 14 days and were weighed every day. After 14 days treatment, serum sample of each rat was obtained for analysis of estradiol(E2),progesterone(P), testosterone(T) contents. Each ovary was collected and weighed for measurement of the ovarian index. The fat pad around womb and ovary was obtained for analyzing the mRNA expressions of PI3K and PKB by PCR. [Results]Compared with the model group, the serum E2 and P levels in QRYYR group were significantly higher but T level was significantly lower(P<0.05) than the model group. The levels of E2, P and T in high dose QRYYR group had no difference from that in the western medicine group and the normal group(P>0.05).The ovarian index in the QRYYR groups was significantly decreased as compared with the model group(P<0.05),and that in the high dose QRYYR groups had no difference with the normal group(P>0.05).The mRNA expressions of PI3K and PKB were significantly increased in the QRYYR groups as compared with the model group(P<0.05),and the expressions in high dose QRYYR group, western medicine group and the normal group had no significant difference from each other(P>0.05). [Conclusion] QRYYR could significantly increase the content of E2 and P in the rat serum, significantly decrease the content of T and the ovarian index. Meanwhile, it could significantly increase the mRNA expressions of PI3K and PKB in adipose tissue. It could improve the ovarian function and decrease the local insulin resistance. The effect of the high dosage group on PCOS was superior or similar to Metformin.
