1.p21WAF1/CIP and Regulation of Breast Cancer Cell Cycle
Journal of Shenyang Medical College 2016;18(3):129-131
Abnormal proliferation is a key point for the occurrence and development of breast cancer. p21WAF1/CIP is essential to maintain a normal growth and development of the organisms, and its dysfunction can lead to organism overgrowth. The function of p21WAF1/CIP occurs mainly in post?transcriptional and transcriptional level. Especially, the transcription regulation of p21WAF1/CIP is very important to cell cycle progression of breast cancer via epigenetic change in histone deacetylation.

Objective:To test the effect of ginseng polysaccharides on anti?tumor activity. Method: The cell inhibitory rates were detected by MTT assay. Results: The starch?like glucan ( WGPN) and the AG type pectin ( WGPA?2?RG) had no effect on tumor cells, the AG+HG type pectin ( WGPA?4?RG) and the HG type pectin ( WGPA?3?HG, WGPA?1?HG) showed stronger effect on tumor cells. Conclusion:Different types of ginseng polysaccharides have different antiproliferation. A neutral fraction WGPN has no inhibition, while ginseng pectin exertes variable inhibition.

Methods:A total of 360 patients with lung cancer confirmed by pathological diagnosis received clinical chemotherapy from Jan 2011 to Dec 2013 were selected as the experimental group who chose chemotherapy drugs for drug?sensitive individualized treat?ment based on molecular diagnostic technology test results. Another 180 patients with lung cancer were selected as the control group who did not carry out individual testing only chose conventional chemotherapy. The efficacy and side effects of drugs were assessed. The progression?free survival and median survival time were followed and recorded, and the survival curve was drawn by the Kaplan?Meier method. Results: There was significant difference in the efficacy between the two groups ( P<0?05) , and there was no signifi?cant difference in side effects of drugs. The progression?free survival time and median survival time were significantly different ( P<0?01) . Conclusions: Molecular diagnostic techniques to guide individual treatment of lung cancer have some clinical significance, it will help to improve the efficacy and worthy of further study.

Methods: A total of 32 male Wistar rats were randomly divided into control group, injured group and med?icine group. The balloon catheter injury was performed on left common carotid artery of rat by imitating the process of angioplasty. In?jured group and control group were injected saline, medicine group were injected IKK inhibitor. After 15 days, the process of neointi?mal and media hyperplasia was observed by HE staining and the expression level of NF?κB, IKK, inhibitor of nuclear factor kappa B ( IκB) was detected by Western blot, the content of IL?6, TNF?αwas detected by radioimmunoassay, and the content of MMP?1 was determined by MMP?1 kit. Results: Arterial neointima hyperplasia hapened in 3 groups. There was significantly difference between control group and medicine group and injured group in intimal area, the expression level of NF?κB, IKK, IκB, and the content of IL?6, TNF?α and MMP?1 ( P<0?05 or 0?01) . There was no significance in these indexes between control group and medicine group ( P>0?05) . Conclusion:IKK inhibitor can reduce the carotid artery restenosis after balloon injury by inhibiting inflammatory reaction induced by IKK?IκB?NF?κB pathway.

Objective: To investigate the differential expression of ST3GalⅤ in human hepatic carcinoma cell lines, HepG?2 and SMMC?7721 and normal hepatic cell line, L?02 by sialyltransferase DNA microarray. Methods: Expression of ST3Gal family was measured with DNA microarray. Western blot was used to verify DNA microarrays data. Result:ST3GalⅤ downregulated in HepG?2 and SMMC?7721 cell lines, compared with control cell line. Conclusion: The downregulation of ST3GalⅤ in HepG?2 and SMMC?7721 cell lines may result in the reduction of GD1a and influence the hepatocyte proliferations.

Journal of Shenyang Medical College 2016;18(3):146-148
Objective:To observe expression level change of miRNA?126 in the myocardial tissue of rats with myocardial infarction and to observe the influence of Shuxuetong injection. Methods:Myocardial infarction model was established in 80 SD rats, 60 surviv?al rats were randomly divided into Shuxuetong group and model group, the other 30 rats were randomly selected as control group. After the success of the modeling, Shuxuetong group received Shuxuetong injection treatment, model group and control group received same amount of normal saline. Real?time quantitative PCR was used to detect miRNA?126 expression levels in rat myocardial tissue at dife?rent time after operation. Results: After 7 days and 14 days, miRNA?126 expression levels in rats myocardial tissue in Shuxuetong group and model group were significantly lower than that in the control group ( P<0?05) , but there was no difference between the two groups. After 14 days, miRNA?126 expression levels in the Shuxuetong group were higher than that after 7 days ( P<0?05) . Conclu?sions:miRNA?126 in 7 days after myocardial infarction significantly reduced. With the passing of time, miRNA?126 expression levels increased gradually and Shuxuetong can increase the expression levels of miRNA?126 .

Objective: To study the therapeutic effect of hyaluronic acid chitosan?based microemulsion ( HAC?ME) and carboplatin in a Wistar rat model bearing cerebral C6 glioma xenografts. Methods:C6 rat glioma cells were cultured normally. A total of 30 Wistar rats bearing orthotopic C6 glioma xenografts were randomly divided into 3 groups, and administrated with physiological saline, carbopl?atin or HAC?ME and carboplatin in each group. There are 10 rats in each group. The rat apoptosis changes and survival time were ob?served after treated with physiological saline, carboplatin or HAC?ME and carboplatin in each group. Results:Glioma cells were nega?tive in saline group with TUNEL staining, the nuclei of individual glioma cells in glioma tissue were stained with brown, indicating ap?optosis occur in carboplatin group, the apoptosis of glioma cells in glioma tissue significantly increased in HAC?ME and carboplatin group with TUNEL staining. The survival time in HAC?ME and carboplatin group was longer than that in saline group and carboplatin group ( P<0?05) . Conclusion:Administration of HAC?ME and carboplatin can suppress the growth of C6 glioma in rats and may pro?vide experimental basis for clinical treatment of glioma.

Objective:To investigate the diagnosis and treatment significance of invasive and noninvasive operation for ventilator?as?sociated pneumonia ( VAP ) . Methods:A total of 80 cases of VAP suspected patients who had received mechanical ventilation at least 48 hours in ICU from Jun 2014 to Mar 2015 were enrolled. Patients were randomly divided into four groups including noninvasive operation group ( F) , invasive operation group ( Q) , mix group 1 ( H1) and mix group 2 ( H2) . VAP diagnosis rate between groups as well as living time, antibiotic use time, survival rate, calcitonin levels and APACHE II score, oxygenation index were analyzed. Results:Specimen from invasive operation had higher specimens to cultivate positive rate than that from noninvasive operation ( P<0?05), but there was no statistic significant difference in VAP diagnosis rate between two methods (P>0?05). Conclusion:Noninva?sive operation collecting samples for VAP diagnosis is also accurate as invasive one. Collecting specimens from sputum suction tube in the clinical treatment on airway suction is a low cost and simple noninvasive operation.
