4.Production of IL-15 and lts Functional Study in Mouse Splenocyte Activation.
Korean Journal of Immunology 1999;21(4):297-302
After the synthesis of IL-15 cDNA from the total RNA of mouse spleen, it was inserted into the prokaryotic expression vector, pRseta, and eukaryotic expression vector, pcDNA3.0, respectively. Subsequently, the insertion of gene and open reading frame were confirmed by sequencing of each plasmid, respectively. Using pRseta- IL-15 plasmid, the recombinant IL-15 protein was induced by IPTG under BL21 (DE 3) host cells and recombinant IL-15 was expressed at 14.5 KDa with time. Then, IL- 15 was separated by His-tag affinity chromatography and analyzed by SDS-PAGE to yield soluble IL-15 at 14.5 KDa as monomer and 29.0 KDa as dimer. In order to inspect the function and contribution of IL-15, the in vitro experiment was established using mononuclear cells separated from the mouse spleen. After 48h exposure of PHA to mouse splenocyte and 24h treatment with recombinant IL-15, the effects of cytokine inductions inspected against IL-2, IL-6, IL-10, IL-12, IFN-r, and GM-CSF. The results showed that comparing with the control, IL-6 increased, IL-2, IL-12 and IFN-r increased and similar, and GM-CSF decreased. In addition, the direct injection of pcDNA3.0-IL-15 plasmid into mice gave the similar results to in vitro studies. Namely, IL-6 and IL-12 increased, and IL-2, IFN-r and GM-CSF were similar or decreased. IL-10 was not induced in in vitro and in vivo experiments. These results suggested that the IL-15 induce the splenocyte activation and can be an important factor in proliferation and fuction recovery of weakened T-cell.

5.Responses of Neck Muscles to Horizontal Semicircular Canal Excitation in Cats .
Jong Seong PARK ; Han Seong JEONG
Korean Journal of Aerospace and Environmental Medicine 1999;9(2):185-191
The vestibular system is a sensory motor system concerned with the sensation of linear or angular acceleration, eliciting several vestibular postural reflexes. This study was designed to figure out the functional relationship in vestibulocollic reflex. The electromyographic responses of the cervical muscles were observed during stimulation of the horizontal semicircular canal nerves. The sternomastoid muscle was inhibited by stimulation of the ipsilateral horizontal canal nerve and excited by stimulation of the contralateral horizontal canal nerve. The splenius capitis muscle was inhibited by stimulation of the ipsilateral horizontal canal nerve and excited by stimulation of the contralateral canal nerve. The semispinalis capitis revealed same pattern responses as splenius capitis or sternomastoid muscles to stimulation of the horizontal canal nerve. These experimental results indicate that the cervical extensor or flexor muscles respond in a uniform fashion to the stimulated horizontal canal nerve. And taken these results and other morphological study results together, the excitatory neural pathways from the vestibular nucleus in brain stem to cervical motoneurons are crossed to the opposite side but the inhibitory pathways from the nucleus to the cervical motoneurons are uncrossed.

7.Psychiatry, Is It Now Okay?-Enlarging the Boundary of Psychiatry in the Neuroscience Era.
Journal of the Korean Society of Biological Psychiatry 2001;8(1):53-61
The authors, in this paper, addressed a variety of problems and difficulties which Korean psychiatrists should cope with. The surprising development of neurosciences, splitting of neuropsychiatry into neurology and psychiatry, easygoing attitude of psychiatrists, changes in the delivery system of health care and ill-balanced education of psychiary were listed as causes of or contributors to them. Social bias to psychiatry and regulations from outside are also considered as contributors. Psychiatric education, including medical school, residency training, continuing medical education and psychiatric textbooks, need to be changed in order to enlarge the boundary of psychiatry. Reestablishment of identity of psychiatry and psychiatrist is unavoidable, considering far-reaching new knowledge of neuroscience and gradually invisible borderzone between neurology and psychiatry. The other ways worth while to consider are : the expansion of psychiatrists' activities, development of medical behavioral science to a clinical specialty, creation of new psychiatric subspecialties, and additional training of psychiatric residencies in the primary medical care.

The Journal of the Korean Orthopaedic Association 1997;32(6):1416-1423
The authors reviewed both the clinical and radiographic results of 36 bipolar hemiarthroplasty using the porous-coated Harris-Galante uncemented femoral stem performed for avascular necrosis in 30 patients from January 1985 to June 1992. The average age was about 45.6 years and the average follow-up was fifty-four months. Clinical results by the Harris evaluation method were excellent in 33 cases, good in 2 cases, and fair in 1 case. The radiographic fixation of the femoral stem at final follow-up was bony ingrowth fixation in 31 cases, fibrous ingrowth fixation in 4 cases, and unstable fixation in 1 case. There was neither medial nor superior migration of the bipolar cup of more than two millimeters.

10.The Role of Ito Cell in Hepatic Fibrosis after Common Bile Duct Ligation: inhibitory role of vitamin A in Ito cell.
Kyung Hee PARK ; Sang Han LEE ; Jong Min CHAE
Korean Journal of Pathology 1995;29(1):1-9
The purpose of this study was to investigate the inhibitory role of vitamin A with respect to activation of Ito cells in fibrosis of the rat liver induced by common bile duct ligation(CBDL). The liver was examined by immunohistochemical staining for a-smooth muscle actin, the known marker of activated Ito cells, and light and electron microscopy after CBDL andCBDL with intraperitoneal injection of retinoic acid (Sigma, USA) 1 mg/Kg in 3 times per week. The results were sumrrlerized as follows: After CBDL, the bile ductules were markedly proliferated in the periportal areas extending toterminal hepatic veins. Interstitial fibrosis and inflammatory cell infiltration appeared, however, cholestasis was minimal. Retinoic acid treatment with CBDL decreased bile ductular proliferationand interstitial fibrosis compared to CBDL only. After CBDL, proliferated and activated Ito ceIs showing positive reaction in smooth muscle actin were present in the periductular andperisinusoidal areas, and areas of increased interstitial fibrosis. Activated ito cells weredecreased in number after CBDL with vitamin A treatment. Electron microscopically, intracytoplasmic fat droplets and the cytoplasmic processes of Ito cells were decreased afterCBDL. Myofibroblasts were frequently appeared in the interstitial fibrosis after CBDL. But,intracytoplasmic fat droplets of Ito cells were well preserved, and myofibroblasts were found lessfrequently after CBDL with vitamin A treatment. The results suggest that vitamin A plays an inbitory role in the activation and fibrogenesis ofIto cells after CBDL.
