1.Distribution of the larvae of Toxocara canis and Ascaris lumbricoides in the tissues of mice.

Mice were infected by feeding the embryonated eggs of Toxocara canis and Ascaris lumbricoides. Each mouse was killed daily for a week and then at several days interval after infection and the distribution of larvae in the various tissues of mice was investigated after the macerating the tissues and digesting with artificial gastric juice. It was confirmed that the migratory behaviour of larvae of T. canis and A. lumbricoides is referred to as the somatic and tracheal type of migration in the mice respectively. Toxocara larvae were found in the carcass on the third day after infection and in the brain after the sixth day of infection. From the thirty-fifth day to the seventy-sixth day after infection, Toxocara larvae were not found in the tissues of mice except in the carcass and brain and they did not develop further than the second-stage larvae. The size of Ascaris larvae, from the embryonated eggs was 0.228-0.271 mm length by 0.010-0.013 mm width and in the third day of infection the size of larvae was 0.271-0.343 mm length by 0.017-0.020 mm width. Between the fifth and tenth day after infection, lavrae molted twice in the lungs and grew to the fouth-stage larvae; 1.357-2.0 mm by 0.034-0.071 mm. These larvae migrated to the intestinal canal after the tenth day of infection and disappeared from the mouse after the twenty-fifth day of infection. No larvae were found in the carcass and brain. The inflammatory reactions in the tissues of infected mice were also observed.

8.Clinical Observation of the Chronic Prostatitis.

There were observed on the 154 cases of the chronic prostatitis confirmed by subjective complaints, rectal finding of the prostate and microscopic examination of the prostatic fluid of the patients, who had been visited to Department of Urology. Seoul Red Cross Hospital from April 1 to Sep. 30, l973. The results obtained were summarized as follows, 1) The distribution of age was between 19 years and 67 years, but ':the most common age was. from 24 to 30 years as 50 percent of the total cases. 2) The complications were urethritis on 82 cases (53.3%), epididymitis on 18 cases(11.7%), cystitis on 13 cases(8.5%), tuberculosis of urinary tract on 9 cases (5.9%). 3) Major subjective symptoms were burning on urination, perineal discomfort morning drop and generalized weakness. 4) On rectal palpation of the prostate of all the patients 83 cases(53.9%) were firm, 24 cases (15.6%) indurated, 6 cases(3.9%) nodular but 39 cases(25.9%) were normal consistency. 5) Microscopic examination of prostatic fluid was revealed of normal limit in 28 cases(18.2%) of all the patients.6) 58 percent of cages treated acculately for 2 to 16 weeks have been improved.

10.Production of IL-15 and lts Functional Study in Mouse Splenocyte Activation.

After the synthesis of IL-15 cDNA from the total RNA of mouse spleen, it was inserted into the prokaryotic expression vector, pRseta, and eukaryotic expression vector, pcDNA3.0, respectively. Subsequently, the insertion of gene and open reading frame were confirmed by sequencing of each plasmid, respectively. Using pRseta- IL-15 plasmid, the recombinant IL-15 protein was induced by IPTG under BL21 (DE 3) host cells and recombinant IL-15 was expressed at 14.5 KDa with time. Then, IL- 15 was separated by His-tag affinity chromatography and analyzed by SDS-PAGE to yield soluble IL-15 at 14.5 KDa as monomer and 29.0 KDa as dimer. In order to inspect the function and contribution of IL-15, the in vitro experiment was established using mononuclear cells separated from the mouse spleen. After 48h exposure of PHA to mouse splenocyte and 24h treatment with recombinant IL-15, the effects of cytokine inductions inspected against IL-2, IL-6, IL-10, IL-12, IFN-r, and GM-CSF. The results showed that comparing with the control, IL-6 increased, IL-2, IL-12 and IFN-r increased and similar, and GM-CSF decreased. In addition, the direct injection of pcDNA3.0-IL-15 plasmid into mice gave the similar results to in vitro studies. Namely, IL-6 and IL-12 increased, and IL-2, IFN-r and GM-CSF were similar or decreased. IL-10 was not induced in in vitro and in vivo experiments. These results suggested that the IL-15 induce the splenocyte activation and can be an important factor in proliferation and fuction recovery of weakened T-cell.
