1.Expressions of human fibrinogen-like protein 2 in peripheral and cerebrospinal fluid white blood cells in patients with cryptococcal meningitis

Objective The purpose of this study was to explore the expression of human fibrinogen-like protein 2 (hFgl2) in patients with cryptococcal meningitis (CM).Methods The expression of hFgl2 in the peripheral blood leukocytes from 20 CM cases and 20 healthy individuals,and the expression of hFgl2 in cerebrospinal fluid (CSF) leukocytes in 11 pairs of CM patients and matched controls was detected using immunohistochemical methods.Medical image analysis system (MIAS) was used to analyze the expression of hFgl2 gray values,which were negatively correlated to the expression of hFgl2 protein in both peripheral blood and CSF.Results The average gray values for hFgl2 protein in peripheral monocytes of CM patients and healthy controls were 114.49± 11.82 and 137.56± 10.64,respectively,and those in peripheral neutrophils were 111.71 ± 8.62 and 143.56±12.57,respectively.Compared with the healthy control group,expressions of hFgl2 in peripheral blood monocytes and neutrophils were both significantly increased in CM patients (t =21.21,t=27.19; P<0.01).The average gray values for hFgl2 protein in CSF monocytes from CM patients and negative controls were 115.44±8.67 and 149.25±8.57,respectively,and those in CSFneutrophils were 111.71±8.62 and 143.56 ± 12.57,respectively.The expressions of hFgl2 in CSFmonocytes and neutrophils were significantly increased in CM patients (t =29.95,t=28.87; P<0.01),while the expression intensity of hFgl2 was similar when peripheral blood monocytes and neutrophils compared with their CSF counterparts (t=0.924,t=0.607; both P>0.05).Conclusions hFgl2 can be expressed in both peripheral and CSF monocytes and neutrophils in CM patients.
2.CT and MRI Diagnosis of Trigeminal Nerve Tumors and Relative Anatomic Analysis

Objective To investigate the CT and MRI findings of trigeminal nerve tumors and relative anatomic characteristics.Methods CT and MRI findings in 17 cases of trigeminal neuroma proved surgically and pathologically were analysed.Results Of 17 cases,the tumors located in middle and posterior carnial fossa in 8 cases and 5 cases respectively,these tumors appeared as oval or lobulated structure.The tumors localized in transcranial fossa in 4 cases with dumbbell or irregular shape.CT and MRI findings were as following:(1)Higher density of tumors in 3,low density in 4,the same density with adjacent graymatter in 2 and cystic low density in 4;(2)Homogeneous and inhomogeneous low signal intensity on T_1WI in 5 and 7 respectively,homogeneous and inhomogeneous high signal intensity on T_2WI in 4 and 8 respectively;(3)Inhomogeneous and homogeneous enhancement in 10 and 7 cases respectively on CT and MRI contrast-enhanced scan;(4)The tumors grew along trigeminal nerve and across the cranial fossa with dumbbel appearance in 4,the root of trigeminal nerve were thickened and connected with the tumors body in 3,enlargement of Meckel's cave in 6;(5) The erosion of petrous bone in 3 cases clear boundary of tumors and without calcificaton or adjacent edema in 15 cases.Conclusion CT and MRI are reliable methods in diagnosing trigeminal neuroma.
3.Antiviral effect of chinonin against herpes simplex virus.

In order to investigate the antiviral effect of chinonin against Herpes simplex virus (HSV), the encephalitis model in mice and skin infection model in guinea pigs were established by HSV- I and HSV-II infection respectively. Acyclovir was used as the positive reference drug to evaluate the antiviral capacity of chinonin. Chinonin showed an obvious therapeutic effect on encephalitis in mice at doses of 25 and 50 mg/kg. At both dosages, chinonin demonstrated stronger protection than acyclovir (1 and 5 mg/kg) to the infected mice from death. It was also found that chinonin could treat the skin infection in guinea pigs effectively. The therapeutic effect of chinonin was similar to that of acyclovir (5 mg/kg) at 25 mg/kg but obviously better than that at 50 and 75 mg/ kg. In conclusion, chinonin is a potential candidate for the treatment against HSV.
4.The Choice of Therapeutic Methods and MRI Diagnosis of Acoustic Neuroma

Objective To evaluate the choice of therapeutic methods and MRI diagnosis of acoustic neuroma.Methods The MRIimaging and clinical materials of 60 patients with acoustic neuroma were analyzed,48 cases were underwent an operation,12 cases weretreated conservatively or gamma knife treatment,follow-up ranged from 1 to 4 years.Results There were 62 tumors round the internalauditory canal.There were 58 cases with single acoustic neuroma and two cases with couples acoustic neuroma.28 tumors demonstratedhypointense and 30 tumors demonstrated hypo-and isointense signals on T_1 weighted image.38 tumors demonstrated hyperintense and 24tumors demonstrated hyper-and isointense signals on T_2 weighted image.The Ⅶ,Ⅷ nerves affected side were thickened than that ofopposite side in 32 patients.After Gd-DTPA administration 24 tumors were homogeneously enhanced,26 tumors were inhomogeneously or circularly enhanced in 50 acoustic neuromas of 48 cases.Operation was still the main election for acoustic neuroma.Conclusion MRI is an effective method in the diagnosis of acoustic neuroma,and providing advice for clinics in making therapeutic programs.
5.Effect of heroin on prolactin gene expression in female rats

Objective To investigate effects of heroin on prolactin(PRL) mRNA in female rats.Methods Fifty female Wistar rats were randomly divided into five groups(10 rats each group): control,heroin-treated-3 d,heroin-treated-9 d,heroin-withdrawal-3 d and heroin-withdrawal-9 d groups.PRL mRNA of pituitary was assessed by(RT-PCR) and PRL concentration in rat plasma was determined by radioimmunoassay(RIA).Results The expression levels of PRL mRNA of pituitary were declined significantly in heroin-treated-3 d,9 d and heroin-withdrawal-3 d groups compared with control group(P0.05)).Conclusion The PRL mRNA in the pituitary and the PRL concentration in plasma in female rats are decreased by heroin and the effect can not be reversed in a short term after heroin withdrawal.
6.Clinical features, levels of protein carbonyl in serum of obstrucitive sleep apnea syndrome with type 2 diabetes mellitus.

OBJECTIVE:
To explore the prevalence, clinical feature and levels of protein carbonyl (PCO) in serum of type 2 diabetes mellitus combining obstrucitive sleep apnea syndrome (OSAHS).
METHOD:
Two hundred and three patients with type 2 diabetes were taken multi lead sleep detection and their AHI, age, height, body mass index (BMI),waistline, duration of diabetes, fast blood glucose, HbA1c level and level of PCO in serum were recorded.
RESULT:
The prevalence of OSAHS was 79.2% in 203 patients, serious apnea 30.4%, moderate apnea 45.4%, mild apnea 24.2%. BMI, waistline, fast blood glucose, HbA1c level and level of PCO in serum were higher in patients complicated with OSAHS (P < 0.01). HbA1c was independently and positively correlated with patients with OSAHS risk (P < 0.05, OR 6.11). The independent correlation factors of AHI included HbA1c level,BMI, waistline, duration of diabetes and level of PCO in serum,with HbA1c as the predominant factor (P < 0.05).
CONCLUSION
The prevalence of OSAHS was higher in patients of type 2 diabetes mellitus combining (OSAHS). Moreover, in these patients poor glucose control and aggravated protein oxidative injury were observed.

7.Analysis of enzymological characterization of N-V proteinase with fibrinolytic activity

Objective To detect the optimum reaction temperature,pH,Km of N-V proteinase and determine its enzymological classification.Methods The fibrin plate method was used to determine the optimum reaction temperature and pH of N-V proteinase.The Km of N-V proteinase was detected by using Leu-pNA as substrate.E64,PMSF,zymofren and 1,10-phenanthroline were used as inhibitors to determine proteinase classification of N-V proteinase.Results The optimum reaction temperature was 50℃,pH was 8-9,Km was 1.97?10-3mol?L-1.PSMF inhibited the fibrinolytic activity of N-V proteinase(P0.05).Conclusion N-V proteinase belongs to serine proteinase.
8.Application of nursing intervention in prevention of urinary retention after total knee arthroplasty

Objective To investigate the effect of preoperative nursing intervention in the prevention of postoperative retention of urine of patients after total knee arthroplasty. Methods 128patients underwent total knee arthroplasty were divided into the intervention group and the control group (64 cases in each group) by random number table.The patients in the control group received routine nursing care and the patients in the intervention group accepted preoperative nursing intervention 3 to 5 days before surgery. The incidence of post-operation urinary retention, catheterization, satisfaction degree and compliance of the patients were compared between the two groups. Results The incidence rate of post-operation urinary retention and catheterization of the patients was respectively 15.6%(10/64) and 7.8%(5/64) in the intervention group,lower than 34.4%(22/64) and 23.4%(15/64) in the control group. The difference was statistically significant (χ2=6.00, 5.93, respectively, P<0.05). The satisfaction degree and compliance of patients were respectively 95.3%(61/64) and 75.0%(48/64) in the intervention group, higher than 85.2%(52/64) and 31.3%(20/64) in the control group,the difference was statistically significant (χ2=6.12, 24.60,respectively, P < 0.05). Conclusions Preoperative nursing intervention have good effect in the prevention of postoperative retention of urine and improving satisfaction degree of patients underwent total knee arthroplasty.
9.Role of CD4~+ CD25~+ regulatory T lymphocytes in the pathogenesis of murine pulmonary tuberculosis

Objective To investigate the role of CD4 ~+ CD25~+ regulatory T lymphocytes (Treg)in modulating the cellular immune response and pathogenesis of murine pulmonary tuberculosis.Methods Inactivation of Treg was achieved by intraperitoneal injection anti-CD25 (clone PC61,50 μ/mouse) in PC61 group, and rat-IgG (50 μ/mouse) was injected intraperitoneally in control group. All the mice were inoculated intravenously with H37Rv 0. 1 mL (1 × 10~6 CFU) 3 days after Treg inactivation. The effects of Treg inactivation in different tissues were analyzed by flow cytometry. The cellular immune response, pulmonary histopathology and bacterial load were determined in vitro at different time points. The data were compared using homogeneity of variance F test and non-paired t test. Results In spleen, the percentages of Treg/CD4 T lymphocytes in PC61 group and control group were (21. 13± 3. 58)% and (30. 42± 4. 20)%, respectively at day 10 of inoculation (t = 2. 38, P < 0. 05), and those were (16. 12 ± 1. 26)% and ( 17. 34± 1. 62)%,respectively at day 30 of inoculation (t = 0. 84,P>0. 05). The percentages of Foxp3~+/CD4~+ T lymphocytes in PC61 group and control group were (32. 07 ± 3. 95)% and (60. 55 ± 5. 48)%,respectively at day 10 of inoculation (t = 5. 96, P<0. 05). Similar results were achieved in the peripheral blood. The lung bacterial load at day 10 of inoculation in PC61 group was lower than that in control group (t = 4. 63, P < 0. 01), but the differences were not significant thereafter. The changes of lung histopathology at late stage of infection (day 120) in PC61 group were less severe than those in control group. Conclusions Murine Tregs increase dramatically after Mycobacterium tuberculosis infection. Treg could inhibit the specific cellular immunity against Mycobacterium tuberculosis, and therefore, may facilitate the persistent infection of Mycobacterium tuberculosis and development of tuberculosis.
10.Observations on the application of LMA in the airway management in total laparoscopic hysterectomy in obese patients

Objective To observe the curative effects of applying laryngeal masks in the airway management in total laparoscopic hysterectomy in obese patients.Methods 60 cases of obese patients who underwent total laparoscopic hysterectomy were collected and were randomly divided into,the Endotracheal intubation (ET) group(n =30) and the laryngeal mask (LMA) group (n =30).The pre-operative co-existent diseases,applications of analgesia and intra-operative vital signs,the peak airway waves (Paw) 60min after pneumoperitoneum,blood gas;revival time;the incidences of complications were observed.Results 60min after pneumoperitoneum,MAP,HR,Paw and SpO2 values were:ET group:[(114.6 ± 22.4) mmHg,(108.2 ± 19.6) times/min,(25.4 ± 3.1) mmHg,(96.1 ± 1.2%)] LMA group:[(97.5 ±20.8)mmHg,(86.5 ±20.7) times/min,(22.6 ±3.7)mmHg,(98.9 ±0.8%)].The difference was statistically significant (P < O.05);The blood gas analysis,PH,PaO2,PaCO2 and BE values are:ET group:[(7.34 ± 0.05),(177.5 ± 44.5) mmHg,(42.6 ± 6.1) mmHg,(-4.9 ± 0.8) mmol/L],LMA group:[(7.38 ± 0.05),(216.7 ± 46.6) mmHg,(38.4 ± 5.5) mmHg,(-3.1 ± 0.6) mmol/L].The difference was statistically significant (P < 0.05);the status of complications:23 cases in ET group,accounting for 77%,10 cases in LMA group,accounting 30%,presenting significant differences (P < 0.05).Conclusion The application of laryngeal masks in the airway management in total laparoscopic hysterectomy in obese patients facilitated the maintaining the stability of hemodynamics and blood gas,resulted in smooth post-anesthesia recovery and fewer complications.
