Objective To investigate the drinking water quality in the countryside in different river systems areas in Hefei, China and to provide scientific data for the improvement of drinking water quality and protection of human health. Methods In Hefei, according to hydrological geology, the rural areas were classified into the Lake-Chaohu river system, the Huaihe-River river system, the Yangtze-River-Huaihe-River watershed area and the general area, the multi-stages sampling method were employed in the investigation of water quality. Results 82.15% of local people drank the shallow-layer well water. 82.73% of water wells exposed to a pollution source which was no more than 30 meters from the well. Dissolved total solid and total coliforms were higher in the rainy season compared with that in the dry season, but the content of the other contaminants were higher in the dry season. Among the river systems areas, the content of some contaminants showed a significant difference. When the distance of pollutant source was fixed, increasing the well depth could reduce the bacterium count in water. Conclusion In the countryside of Hefei, the total bacteria count and the total coliforms in drinking water greatly exceed the allowed limits, water supply improvement should be made according to the local hydrological and geological condition.

The Laboratory medicine is a practice subject.Department of Medical Laboratory in the first clinical medicine college(Nanfang Hospital),Southern Medical University strengthens communication and cooperation with enterprises,such as third parted laboratory medicine institutions and independent laboratories.The enterprises participate in teaching work including reforming curriculum system.cultivating students'practice and innovation abilities,constructing exercitation bases,sdvancing students'occupation competition,and so on.Along with emphasizing practice and innovation abilities cultivation,the hospital-enterprise cultivation pattern for Laboratory medicine was established and made primary progress.

OBJECTIVE:To establish the HPLC fingerprint for Anemone raddeana. METHODS:HPLC was performed on the column of Phemomenex Gemini C18 with mobile phase of 0.1%phosphoric acid-acetonitrile(gradient elution)at a flow rate of 1 ml/min,the detection wavelength was 206 nm,the column temperature was 30℃,and the injection volume was 20μl. With the refer-ence of raddeanin A,13 batches of A. raddeana were analyzed,chromatographic fingerprint similarity evaluation system software was conducted for similarity analysis,and SPSS 13.0 was conducted for cluster analysis. RESULTS:There were 11 common peaks in the 13 batches of A. raddeana with similarity of higher than 0.90. According to the verification,the fingerprint and control fin-gerprint shows good consistency. The drugs in Huadian,Jiaohe, Tiangang,Shulan,Tonghua and Fusong of Jilin and Shangzhi of Heilongjiang were regarded as category 1,and in Harbin,Yabuli town and Yimianpo of Heilongjiang,Qingyuan of Liaoning,Ji-nan of Shandong were category 2. CONCLUSIONS:The established fingerprint can provide reference for the identification and quality evaluation of A. raddeana.

Objective To evaluate the effect of trimetazidine pretreatment on myocardial injury induced by propofol in rats.Methods Forty healthy Sprague-Dawley rats,weighing 240-250 g,aged 2.5 months,were randomly divided into 4 groups (n =10 each):fat emulsion group (group L),propofol group (group P),and different doses of trimetazidine groups (TL and TH groups).In group L,10％ fat emulsion was infused intravenously for 12 h at a rate of 30 mg·kg-1 ·h-1.In group P,1％ propofol was infused intravenously for 12 h at a rate of 30 mg·kg-1 ·h-1.In TL and TH groups,trimetazidine 2 and 5 mg/kg were injected intravenously,respectively,and 1.5 h later propofol was infused intravenously for 12 h at a rate of 30 mg·kg-1 ·h-1.At 6 and 12 h after beginning of administration,blood samples from the internal jugular vein were taken for determination of the levels of serum creatine kinase (CK),creatine kinase isoenzyme-MB (CK-MB),lactate dehydrogenase (LDH),α-hydroxybutyric acid dehydrogenase (α-HBDH),cardiac troponin Ⅰ (cTnⅠ),triglycerides (TG),low-density lipoprotein (LDL) and high-density lipoprotein (HDL).After blood sampling at 12 h after beginning of administration,myocardial specimens were obtained for microscopic examination of the pathological changes with light and electron microscope.Results Compared with group L,the serum CK,CK-MB,LDH,α-HBDH,cTnⅠ and TG levels were significantly increased,HDL was decreased,and no significant changes were found in LDL concentrations in P and TL groups,and the serum TG,HDL and LDL were increased,and no significant changes were found in the other parameters in group TH.Compared with group P,the serum CK,CK-MB,LDH,α-HBDH,and cTnⅠ levels were significantly decreased in TL and TH groups,the serum TG level was increased in TL group,and the serum TG,HDL and LDL levels were increased in TH group.Compared with group TL,the serum CK,CK-MB,LDH,α-HBDH and cTnⅠ levels were significantly decreased,and the serum TG,HDL and LDL levels were increased in group TH.Conclusion Trimetazidine pretreatment can attenuate myocardial injury induced by propofol,but it induces increase in blood lipids.

AIM To explore the mercury accumulation in KM mice after being given Zuotai at different doses and time.METHODS KM mice were randomly divided into blank group,Zuotai low-,middle-and high-dose (6.07,60.70 and 606.97 mg/kg,42 d;606.97 mg/kg,14 d) groups.The mercury contents in brain (olfactory bulb,cortex,hippocampus,hypothalamus,brain stem,cerebellum),heart,lung,kidney,liver,spleen,serum,muscle of mice were measured after administration.RESULTS Compared with the blank group,Zuotai at low-dose significantly increased the mercury contents in hippocampus,cerebellum,lung,kidney,liver and serum of mice after 42-day treatment;Zuotai at middle-dose markedly increased the mercury contents in olfactory bulb,cortex,hippocampus,brain stem,cerebellum,heart,lung,kidney,liver,spleen and serum of mice after 42-day treatment;the mice treated with high-dose of Zuotai for 42,14 days significantly increased the mercury contents in olfactory bulb,cortex,hippocampus,hypothalamus,brain stem,cerebellum,heart,lung,kidney,liver,spleen,muscle and serum.CONCLUSION Mercury can be accumulated in different tissues of mice after intragastric administration of Zuotai in a dose-and time-dependent manner,which suggests that Zuotai and its compound preparations should not be used in high-dose and long-term.

Objective To investigate the expressions of aquaporin 4 (AQP4) in the bacterial meningitis in rats and to explore the molecular mechanism for brain edema caused by bacterial meningitis.Methods Totally 40 of 3-week-old-Sprague-Dawley healthy rats,body weight 60-80 g,male or female,were divided into a normal control group(n =10),and infection groups:24 hours after injection(n =10),48 hours after injection(n =10),and 5 days after injection(n =10).The expressions of AQP4 in the brain were detected by immunohistochemistry and Western blot methods respectively after 24 hours,48 hours,5 days of inoculation.Results Mortality rate:no rats in the control group and the infection group after 24 hours were dead.Two rats in the infection group after 48 hours and 4 rats in the infection group after 5 days were dead because of serious sickness,with the mortality rates 20％ and 40％,respectively.AQP4 expression was slightly positive under light microscope,and the positive cells mainly surrounded glial cells and blood vessels,while neurons were not dyed.Immunohistochemical staining showed that AQP4 expression in the model group increased with the severity of edema;compared with the control group,the AQP4 expression in the brain tissues increased in different periods after rats were infected,and the differences between groups were statistically significant (F--91.84,P ＜ 0.01).Western blot analysis showed that after the brain received streptococcus pneumoniae injection,expression of AQP4 began to increase in 24 hours after streptococcal injection,and reached to the peak in 48 hours,but decreased in 5 days,but the expression still remained higher than that of the normal control group.Each group had statistically significant difference(F =14.23,P ＜ 0.01).Conclusions Expression of AQP4 in the models with bacterial meningitis may increase initially and decrease later.It suggests that AQP4 plays a protective role during the development of infectious brain edema.

Objective:To observe the effect of TGF-β1 on collagen degradation of rabbit keratocytes.Methods:The activity of collagen degradation of keratocytes with three-dime nsional culture in collagen gel was determined with spectrophotometer.Results:TGF-β inhibited collagen degradation of keratocytes in dose-dependent manner.Conclusion:The present result suggests that TGF-β1 has the inhibitory effect on collagen degradation of keratocytes and may help in the treatment of corneal ulceration.

Objective To observe the effects and mechanism of lung inflation with carbon monoxide (CO) during the cold ischemia phase on lung ischemia-reperfusion injury (IRI) after rat lung transplantation.Method Twenty-four pairs of SD rats were selected to establish the model of lung transplantation,and random number method was used to divide 24 donors into 3 groups with 8 rats in each group.(1) CO inflation group (CO group):During the cold ischemia phase,500 ppm CO +volume fraction 40％ O2 + N2 was used for lung inflation,and the volume was 5 mL/kg;(2) O2 inflation group (O2 group):During the cold ischemia phase,volume fraction 40％ O2 + volume fraction 60％ N2 was used for lung inflation;(3) Control group:The lung was deflated during the cold ischemia phase.The gas was replaced every 30 min in the CO and O2 groups,and the lung transplantations were performed after 180 min of cold ischemia.The arterial blood gas analysis was performed at baseline,3 min after reperfusion,and 60,120,and 180 min after reperfusion.The recipient serum levels of relative inflammatory factors,lung tissue cell apoptosis and nuclear factor kappa B (NF-κB) protein expression were detected after 180 min of reperfusion.Result As compared with the control group (238 ± 61 mm Hg),the oxygenation index in the O2 group (293 ± 78 mm Hg) and CO group (361 ± 48 mm Hg) was increased (P＜0.05),and as compared with the O2 group,that in the CO group was increased (P＜0.05).Furthermore,as compared with the control group,the interleukin (IL)-8,tumor necrosis factor (TNF)-α,and cell apoptosis in the O2 group and CO group were decreased significantly,and as compared with the O2 group,those in the CO group and NF-κB protein expression were significantly decreased (P＜0.05).Conclusion Lung inflation with CO during the cold ischemia phase ameliorated the rat lung IRI via reducing the inflammatory response and cell apoptosis mediated by the NF-κB pathway.

Medical virtual simulation teaching environment is an advanced and complex combination of virtual simulation technology and medical technology training .As an advanced information technology , it has been attracting the attention of medical education researchers .Based on the characteristics of medical laboratory technology education , the virtual simulation experimental teaching center was established and a framework of the virtual simulation training environment was designed in the Department of Laboratory Medicine, Southern Medical University .Virtual simulation softwares and tool softwares were put into use widely in our experimental teaching .As it turns out, this simulated training environment has many merits such as risk-free safety , great efficiency , genuine learning experience and wide application potential , which partly overcomes the restriction of the teaching process brought by the condition of the practical operation and improves the teaching effect significantly .

Aim To investigate the effect of PLK1 on epithelial-mesenchymal transition (EMT)of human e-sophageal squamous cell carcinoma (ESCC)cells TE-1 5 and its relevant molecular mechanisms.Methods PLK1 overexpressed ESCC cells and control vector were used as the experimental cells.The expression of EMT-related protein markers E-cadherin and vimentin were measured by Western blot.vimentin mRNA was measured by Real-time PCR.Total cellular protein and nuclear protein were respectively extracted,and then they were used to detect the expression of β-catenin by Western blot.β-catenin siRNA and non-specific siR-NA were transiently transfected into the cell clones overexpressed PLK1 ,and then vimentin was detected by Western blot.β-catenin protein degradation com-plex was detected by immunoprecipitation and Western blot.Results The mesenchymal marker vimentin was distinctively upregulated and the epithelial marker E-cadherin was distinctively downregulated in the cell clones overexpressed PLK1 ,compared with those in the vector clones.This indicated that EMT occurred in ESCC cells.vimentin mRNA was also markedly in-creased.In the cell clones overexpressed PLK1 ,β-catenin were both elevated from the total cells and the nucleus.The expression of vimentin was reduced whenβ-catenin was knocked down.APC and GSK-3βwere both reduced from Axin immunoprecipitate in the cell clones overexpressed PLK1 .Conclusion PLK1 up-regulates vimentin and promotes EMT in ESCC cells probably by inhibiting the formation of protein degrada-tion complex and stabilizing β-catenin.
