1.Semi-quantitative determination of HLA-DR antigen expression on cell surfaces with dot-ELISA

Using HB55 McAb, we detected HLA-DR antigens on Daudi, H18, PBL and K562 cells by dot-ELISA. Results corresponding to situation of the antigen expressions on various cell lines were obtained: colour reaction in Daudi, H18 cells and PBL was positive in different degree and K562 sample presented negative reaction. Chromatogram peaks were printed by Dual-Wavelength Flying-Spot scanner CS-9000 (SHIMADZU). Areas and high values of peaks, meaning size and colour of spot respectively, were increased with cell concentration. Both cell concentration and volume of cell suspension did not influence the diffusion of sample. An analysis for the antigen expression on cell surface and its dynamic change can be supplied by the technique.The study on quantitative assay of MHC class Ⅱ antigens on cell surface are still going on.
2.Contrast study of quetiapine and risperidone in female first-episode schizophrenia

Objective To investigate the efficacy and adverse reaction of quetiapine compared to risperidone in female first-episode schizophrenia.Methods Eighty cases met the CCMD-3 criteria for schizophrenia were divided into two groups with random numbers table method,treated with quetiapine(300 ~ 750mg/d) and risperidone(2 ~5mg/d) respectively for 8 weeks.The efficacy was measured with the positive and negative symptoms scale(PANSS),and the side effects were observed.Results The difference between two groups in 2-weekend,4-weekend and 8-weekend PANSS had no statistical meaning(all P>0.05).The efficient rate of clinial treatment of quetiapine and risperidone groups were 90% and 87.5% respectively,The difference had no statistical meaning(P>0.05).Risperidone group had higher incidence rate than quetiapine group in adverse reaction such as tremor,muscle rigidity,akathisia,emmenia change and lactation etc.Conclusion Quetiapine haa equivalent clinial effect with risperidone in female first-episode schizophrenia,and had less adverse reaction.
3.TypeⅠ,Ⅱ,Ⅲ,ⅩCollagen Expression in Meniscus and Articular Cartilage and Immunoreaction Study after Xenogenic and Allogenic Meniscus Transpla ntation in Rabbits

Objective To study the type Ⅰ,Ⅱ,Ⅲ and Ⅹcollag e n expression in meniscus and articular cartilage and immunoreactions post xenoge nic and allogenic meniscus transplantation. Methods After men iscectomy,30 adult New Zealand white rabbits were divded into two groups:group A undertook allogenic medial meniscus transplantation and group B undertook xenog enic meniscus transplantation with fitted meniscus tissues harvested from swine. 6, 12 and 24 weeks post transplantation, the histological and immunohistochemical analysis of type Ⅰ ,Ⅱ,Ⅲ and Ⅹ collagens monoclonal antibody were investigated in menisci and a rticular cartilage. Peripheral blood were obtained for immunoreactions study thr o ugh the methods of CDMT (Complement Dependent Microlymphocytotoxicity Test) and RIA (Radioimmunoassay)of IL-2 and IL-6. Results The meniscu s and articular cartilage were good post allogenic meniscus transplantation, but the transplanted xenogenic meniscus appeared to be dissolved and absorbed and c art ilage lesions were observed in 24 week post operation. No significant difference was found between two groups in type Ⅰ,Ⅱ and Ⅲ collagen expression during e xperiment. From 1~12 weeks, no significant diffe rence was found among type Ⅰ,Ⅱ and Ⅲ collagen expression in articular cartil age in two groups. However,24 weeks post operation, type Ⅹ collagen's expressio n in cartilage was abnormally increased. Neither allogenic nor xenogenic meniscu s transplantation caused fatal immunoreaction during the whole experiment. Con clusion The transplantation meniscus began to be dissolved and absorbed after half a year, but allogenic meniscus transplantation achieved good results . Type Ⅰ,Ⅱ and Ⅲ collagen expression in transplanted meniscus and articular cartilage showed no difference between two groups, but the expression of type Ⅹ collagen was abnormally increased in xenogenic group 24 weeks post operation. No fatal immunoreaction was found in both groups.
4.A Prognosis Study on Xenogenic and Allogenic Meniscus Transplantation for Meniscus Deficiency Repair in Rabbit's Knee

Objective To study the prognosis of the implanted xenogenic and allogenic menisci, and the implanted menisci's protection effects on articular cartilage. Methods 30 adult New Zealand white rabbits were divided equally into two groups and the medial meniscus defection models were established by medial meniscectomy. Allogenic medial menisci implantation were done in group A. Small pieces of menisci tissue were taken from pig meniscus and moulded them as the same shape and size as rabbit's medial menisci,then, xenogenic menisci implantation were done in group B with these moulded grafts. 6, 12 and 24 weeks after the operations, rabbits were sacrificed to observe the implanted menisci and the articular cartilage on medial tibial plateau, medial femoral condylus and femoral trochlea with naked eye and histological method. Results After allogenic meniscus transplantation, the state of implanted meniscus was good, and healed well around the capsula. 24 weeks post allogenic meniscus transplantation, the injury of the articular cartilage was not in evidence. In short time post xenogenic meniscus transplantation, the state of meniscus and cartilage was good and after 24 weeks the xenogenic meniscus was dissolved and absorbed, the articular cartilage also showed degeneration and injury. Conclusion With xenogenic meniscus moulded from pig meniscus tissue,the implanted meniscus was resolved and absorbed and the cartilage degeneration also appeared after half a year. Post allogenic meniscus transplantation, the meniscus structure and function were reconstructed well and the articular cartilage was also well protected.

METHODS:The release was studied by dynamic dialysis method.Different equations were selected to fit the release law.RESU_ LTS&CONCLUSION:It is good to fit the release law by1 st order equation and Weibull equation.

In the end, a development prospect of antibacterial metallic materials research in the future is also described.
7.Effect of intrathecal dexmedetomidine on expression of cAMP response element-Wnding protein phosphorylation in spinal dorsal horn in a rat model of bone cancer pain

Objective To investigate the effect of intrathecal (IT) dexmedetomidine on the expression of cAMP response element-binding protein phosphorylation (p-CREB) in spinal dorsal horn in a rat model of bone cancer pain. Methods Sixty-four adult female Wistar rats weighing 200-240 g were randomly divided into 4 groups (n = 16 each): sham operation group (group S); bone cancer pain group (group BP); normal saline group ( group NS) ; dexmedetomidine group (group D) . Bone cancer pain was induced by injecting Walker 2S6 mammary gland carcinoma cell suspension (2 ×106 cells/ml) 10μl into the medullary cavity of the tibia in BP, NS and D groups. Groups S and BP received no IT injection. Croups NS and D received IT injection of NS 10 μl and dexme detomidine 5 μg/kg respectively 7 days after successful establishment of the model. Ten animals were selected from each group at 1 day before IT administration (T0), immediately before IT administration (T1 ) and at 1, 6, 12 and 24 h after IT administration (T2-5 ) and paw withdrawal threshold (PWT) to mechanical stimuli was measured with von Frey filaments. The other 6 rats in each group were sacrificed at T4 and the spinal cord was removed for determination of p-CREB expression in the spinal dorsal horn.Results PWT was significantly decreased at T1-5 and pCREB expression up-regulated at T4 in BP, NS and D groups compared with group S ( P < 0.05) . Compared with group BP, PWT was significantly decreased at T2-5 and p-CREB expression down-regulated at T4 in group D ( P <0.03), while no significant change in PWT and p-CREB expression was found in group NS (P > 0.05) .Conclusion IT dexmedetomidine can reduce the bone cancer pain through inhibiting the phosphorylation of CREB in rat spinal dorsal horn.
8.Minimally invasive clinical pathway in patients after cardiac interventional therapy

Objective To explore the effect of minimally invasive clinical pathway in patients after car-diac interventional therapy. Methods One hundred and twenty patients who received cardiac interventional therapy from June 2005 to October 2006 were divided into the control group and the observation group.The control group received routine nursing while the observation group adopted minimally invasive clinical pathway of nursing.The nursing effect in the two groups was compared. Results The mean hospitalized duration, sat-isfaction degree and health knowledge level in the observation group were superior to those of the control group (P < 0.05). Conclusions The adoption of minimally invasive clinical pathway in patients after cardiac inter-ventional therapy could increase working efficiency nd ensure the nusing quality.
9.Evaluative effects of flash visual evoked potential on the changes of intracranial pressure

BACKGROUND: Flash visual evoked potential(fVEP) can reflect the integrity of visual pathway from retina to pulvinar cortex. The utilization of its characters can accurately evaluate the injury situation of visual conduction pathway.OBJECTIVE: To investigate the feasibility and effects of fVEP on non-traumatic monitoring of intracranial pressure.DESIGN: A repeated measurement design based on patients.PARTICIPANTS: Totally 197 patients with brain injury including 138males and 59 females with an average of(38 ±9) years old were selected from the First Affiliated Hospital of Chongqing Medical University and Xuanwu Hospital Affiliated to Capital University of Medical Sciences. The selected diseases were: epidural hematoma( n=20),subdural hematoma(n = 26),cerebral contusion( n = 4),subdural hematoma companied with cerebral contusion( n = 92),and cerebral contusion companied with intracerebral hematoma( n = 55).METHODS: The level of intracranial pressure of 197 patients with brain trauma was detected with fVEP and epidural manometry. Data of traumatic surveillance were collected after the completion of fVEP. Changes of blood pressure and heart rate in patients were recorded simultaneously.MAIN OUTCOME MEASURES: fVEP results,and levels of intracranial pressure.RESULTS: The average intracranial pressure of 197 patients with brain trauma was(2.75 ± 0.64) kPa,( 1.54 - 4. 02 kPa) . The dispersion of both traumatic and non-traumatic surveillance was relatively big in patients with confirmed visual pathway injury. No typical wave was induced by fVEP in2.79% of patients. Scatter diagram suggested that the consistence between fVEP and epidural manometry was relatively good. Linear regression analysis showed that heart rate was the most sensitive one to the fluctuation of intracranial pressure (β = -0. 369) followed by systolic pressure (β= 0. 316),while diastolic pressure was not so sensitive (β = 0. 147). There was negative correlation between heart rate and intracranial pressure,which could quite sensitively reflect the fluctuation of intracranial pressure,systolic pressure was the next,and diastolic pressure was not easily to be affected by the changes of intracranial pressure.CONCLUSION: Non-traumatic surveillance,fVEP,has favorable consistence with traumatic detection,which can quantitatively evaluate the changes of intracranial pressure non-traumatically.
