1.Cheiloplasty by incising along the outline for correction of the defection caused by cheiloschisis operation

Objective To find a satisfied repairing method which can complete the functional cheiloplasty while avoiding the visible scar after the cheiloschisis II operation for bilateral cleft lip. Methods The functional operation was carried out in subcutaneous layer by incising along the outline of the nose and the mouth. Results A satisfied effect was achieved after 6 months to 2 years' follow up in 15 cases of the patients (4 cases of bilateral cleft, 11 cases of cheiloschisis). Conclusions The new operation method has improved the traditional method of cheiloplasty. It can effectively repair the secondary defection in nose and mouth resulted from the cheilochisis operation. It can rebuild the orbicularis oris muscle and repair the diseased alar catilage as well. Furthermore, it not only brings a natural looking appearance, but also a natural function and an invisible incision scar. [
2.Treatment of recurrent cavernous hemangiomas by ethanol thrombosis and pinyangmycin sclerothery

Objective To investigate a safe, effective and mini-invasive method in the treatment of recurrent cavernous hemangiomas. Methods Under local or ketamine anesthesia, ethanol was injected into hemangioma to embolize the draining vessels, and 8~16 mg pinyangmycin was then injected into the lesion slowly. After 7~10 days, second injection of pinyangmycin and total 3~5 times were needed as a course of treatment. Results In the 6 cases reported, shrinkage and degradation were observed and finally tumor was disappeared. There was no skin necrosis, no recurrence and other complications in the 1~5 years follow-up period. Conclusion Ethanol embolization and pinyangmycin injection is a safe, simple, mini-invasive, and effective method in treating the recurrent cavernous hemangiomas.
3.Comparison of the efficacy and safety of different synchronization chemotherapy for middle -advanced cervical cancer

Objective To explore the efficacy and safety of different synchronization chemotherapy for mid-dle -advanced cervical cancer.Methods 96 patients with middle -advanced cervical cancer were selected as the research objects.According to the digital table,they were divided into the observation group and control group, 48 cases in each group.The observation group was used radiotherapy combined with nedaplatin chemotherapy,The control group was used radiotherapy cisplatin and 5 -fluorouracil chemotherapy,The efficacy,transfer rate,pelvic recurrence rate,4 year survival rate and side effect of the two groups were compared.Results The effective rate in the observation group was 91.8%,87.5% in the control group,there was no significant difference between the two groups (χ2 =0.446 5,P >0.05);The rate of distant metastasis,pelvic recurrence and 4 year survival rates between the two groups had no significant difference(χ2 =1.200 0,1.338 0,1.523 8,all P >0.05);The incidence rate of Ⅲneutropenia in the observation group was 4.2%,which was lower than 22.9% in the control group(χ2 =7.206 7,P <0.05);The incidence rate Ⅲ gastrointestinal reaction of the observation group was 20.8%,which was lower than 47.9% of the control group(χ2 =7.803 8,P <0.05).Conclusion The two schemes of radiotherapy clinical cura-tive effect of chemotherapy in the treatment of middle -advanced cervical cancer has a same effect,and the patients of nedaplatin chemoradiotherapy has better tolerability,safety,which is worth of clinical application.
4.Contraction of rat TRESK gene recombinant adenovirus vector

Objective To construct rat TRESK gene recombinant adenovirus expression vector.Methods TRESK full-length cDNA was cloned from rat dorsal root ganglion cells and confirmed by RT-PCR and sequencing. pAd/CMV/V5-DEST-TRESK was then constructed under the control of CMV-promotor. DH5α colibacillus was translated and the positive recombinants were subsequently identified by PCR and DNA sequencing. 293T cells were cotransfected and packed to produce adenovirus. Results The titer of virus was tested using Hole-by-dilution titer method. The full length of TRESK from rat dorsal root ganglion cells is 781 bp. It was demonstrated that the DNA sequencing was completely consistent with TRESK sequencing of rat recorded in GeneBank. The PCR amplification of the pAd/CMV/V5-DEST-TRESK gDNA was matched with pAD-GFP blank vector as anticipated. The titer of the concentrated virus was 1.31×109 TU/ml. Conclusion Rat TRESK gene recombinant adenovirus vector is constructed successfully.
5.Changes in TRESK mRNA expression in dorsal root ganglion in a rat model of neuropathic pain

Objective To evaluate the changes in the expression of diplopore potassium ion channel TRESK mRNA in dorsal root ganlion (DRG) in rats with neuropathic pain (NP) .Methods Thirty-two male SD rats weighing 220-250 g were randomly divided into 2 groups ( n = 16 each) : group sham operation (group S) and group NP. NP was induced by ligation and severance of left tibial and common fibular nerves according to the technique described by Decosterd. Eight rats in each group were sacrificed 1 day before and 14 day after operation and their L4,5 DRGs in the operated side were isolated for determination of TRESK mRNA expression by RT-PCR. In the remaining 8 rats in each group paw withdrawal threshold to mechanical stimuli ( MWT) and paw withdrawal latency to a thermal nociceptive stimulus (TWL) were measured at 1 day before (baseline) and 1, 3, 5, 7, 14 day after operation. Results MWT was significantly lower in group NP than in group S. The TRESK mRNA expression in L4,5 DRGs in the operated side was significantly decreased after operation as compared with the baseline before operation in group NP and was significantly lower in group NP than in group S. Conclusion The development and maintenance of NP may be closely related with down-regulation of TRESK mRNA.
6.Effect of sevoflurane preconditioning on TREK-1 channel expression in hippocampus after focal cerebral ischemia-reperfusion in rats

Objective To investigate the effect of sevoflurane preconditioning on TREK-1 channel expression in hippocampus after focal cerebral ischemia-reperfusion (I/R)in rats and the mechanism.Methods Thirty-six male SD rats weighing 240-280 g were randomly divided into 3 groups(n=12 each):group Ⅰ sham operation (group S),group Ⅱ I/R and group Ⅲ sevoflurane preconditioning (group Sevo).Focal cerebral ischemia was produced by inserting a 4-0 nylon thread with rounded tip into right internal jugular vein.The nylon thread was the nylon thread Wag about 18-20 Innl.The right middle cerebral artery(MCA)Wag occluded for 2 h and then released for 24 h reperfusion.The Sevo group inhaled 2.4% sevoflurane for 30 min at l h before ischemia.Neurological deftcits were assessed and scored at the end of 24 h reperfusion (the higher was the score,the severer was the deficit).The cerebral infarct size was determined by TTC staining and the TREK-1 mRNA in hippocampus by RT-PCR.Results The cerebral infarct size was significantly smaller and the neurological deficit scores were significantly lower in Sevo group than in I/R group.The TREK-1 mBNA expression was significantly up-regulated in Sevo group as compared with I/R group.Conclusion Sevoflurane preconditioning Can protect the brain against I/R injury by activating TREK-1 in hippocampus.

Objective To investigate the morbidity, diagnostic profile and perinatal outcome of pregestational diabetes mellitus (PGDM) in 15 hospitals in Guangdong province. Methods A total of 41338 women delivered in the 15 hospitals during the 6 months,195 women with PGDM(PGDM group) and 195 women with normal glucose test result(control group)were recruited from these tertiary hospitals in Guangdong province from January 2016 to June 2016. The morbidity and diagnostic profile of PGDM were analyzed. The complications during pregnancy and perinatal outcomes were compared between the two groups. In the PGDM group, pregnancy outcomes were analyzed in women who used insulin treatment (n=91) and women who did not (n=104). Results (1)The incidence of PGDM was 0.472%(195/41338). Diabetes mellitus were diagnosed in 59 women (30.3%, 59/195) before pregnancy, and 136 women (69.7%,136/195) were diagnosed as PGDM after conceptions. Forty-six women (33.8%) were diagnosed by fasting glucose and glycohemoglobin (HbA1c) screening. (2) The maternal age, pre-pregnancy body mass index (BMI), prenatal BMI, percentage of family history of diabetes, incidence of macrosomia, concentration of low density lipoprotein were significantly higher in PGDM group than those in control group (all P<0.05). Women in PGDM group had significantly higher HbA1c concentration((6.3±1.3)% vs (5.2±0.4)%), fasting glucose [(6.3±2.3) vs (4.8±1.1) mmol/L], oral glucose tolerance test(OGTT)-1 h glucose((12.6±2.9) vs (7.1± 1.3) mmol/L)and OGTT-2 h glucose [(12.0±3.0) vs (6.4±1.0) mmol/L] than those in control group (P<0.01). (3)The morbidity of preterm births was significantly higher (11.3% vs 1.0%, P<0.01), and the gestational age at delivery in PGDM group was significantly smaller [(37.6±2.3) vs (39.2±1.2) weeks, P<0.01]. Cesarean delivery rate in the PGDM group (70.8% vs 29.7%) was significantly higher than the control group (P<0.01). There was significantly difference between PGDM group and control in the neonatal male/female ratio (98/97 vs 111/84, P=0.033). The neonatal birth weight in PGDM group was significantly higher((3159±700) vs (3451±423) g, P<0.01). And the incidence of neonatal hypoglycemia in the PGDM group was higher than the control group (7.7% vs 2.6%, P=0.036).(4)In the PGDM group, women who were treated with insulin had a smaller gestational age at delivery [(36.9±2.9) vs (37.9±2.5) weeks, P<0.01], and the neonates had a higher neonatal ICU(NICU)admission rate (24.2% vs 9.6% , P<0.01). Conclusions The morbidity of PGDM in the 15 hospitals in Guangdong province is 0.472%. The majority of PGDM was diagnosed during pregnancy; HbA1c and fasting glucose are reliable parameters for PGDM screening. Women with PGDM have obvious family history of diabetes and repeated pregnancy may accelerate the process of diabetes mellitus. Women with PGDM have higher risk for preterm delivery and neonatal hypoglycemia. Unsatisfied glucose control followed by insulin treatment may increase the need for NICU admission.
9. Evaluation of the role of platelet derived growth factor in papillary thyroid carcinoma by targeted ultrasound contrast agent

Objective:
To explore the role of targeted ultrasound contrast agent in the evaluation of angiogenesis in papillary thyroid carcinoma (PTC) in nude mice by constructing platelet derived growth factor receptor α (PDGFRα)-targeted ultrasound contrast agents, and to explore the effect of platelet derived growth factor (PDGF) on angiogenesis in PTC.
Methods:
PDGFRα-targeted ultrasound contrast agents were constructed through biotin-avidin linkage, and blank micro-bubbles were served as control group. After the siRNA-PDGF BCPAP cell line was established, well prepared BCPAP cells or siRNA-PDGF-BCPAP cells were injected subcutaneously into the back of male BABL/C nude mice. The growth of the tumors was observed closely. All tumors of the normal group and the siRNA-PDGF group were examined by PDGFRα-targeted ultrasound contrast agents or blank micro-bubbles, and the dynamic images were analyzed quantitatively by QontraXt software.All mice were sacrificed after ultrasonography. Microvessels density (MVD) in the tumors was counted by CD31 staining and the expressions of vascular endothelial growth factor(VEGF) and PDGF were detected by Western Blot.
Results:
PTCs in nude mice were enhanced well by PDGFRα-targeted ultrasound contrast agent or blank micro-bubbles. The peak of tumors in siRNA-PDGF group was significantly lower than that in the normal group [(43.085±13.244)% vs (57.428±10.952)%,
10.Expression and significance of NT-proBNP,GFAP and HIF-1α in inflammatory injury in patients with cerebral hemorrhage

Objective To investigate N terminal pro B type natriuretic peptide (NT-proBNP),glial fibrillary acidic protein (GFAP)and hypoxia inducible factor-1 (hypoxia inducible factor-1α,HIF-1α) expression and there significance in inflammatory injury in patients with cerebral hemorrhage.Methods We selected 80 patients with acute intracerebral hemorrhage treated in our hospital from January 2018 to December 2019 as the observation group and 80 healthy people who underwent physical examination in our hospital in the same period as the control group.After routine treatment,NT-proBNP,GFAP and HIF-1α in patients serum and cerebrospinal fluid were detectedα、Tumor necrosis factor-α(TNF-α) interleukin-6 (IL-6),and correlation analysis was carried out.Results NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid of patients in the observation group、TNF-α and IL-6 levels in the control group were higher than those in the control group (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid of the observation group after treatment,as well as TNF-α and IL-6 levels were lower than those before treatment (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid increased with the aggravation of the disease、TNF-α and IL-6 levels were positively correlated with the degree of intracerebral hemorrhage (P<0.05).NT-proBNP,GFAP and HIF-1α in serum and cerebrospinal fluid were associated with inflammatory injury in intracerebral hemorrhage.There was a significant positive correlation with IL-6 level (P<0.05).NT-proBNP,GFAP and HIF-1α.It was closely related to inflammatory injury in patients with intracerebral hemorrhage and was an independent risk factor for intracerebral hemorrhage complicated with inflammatory injury (P<0.05).Conclusion The levels of NT-proBNP,GFAP and HIF-1α are closely related to inflammatory injury and prognosis in patients with cerebral hemorrhage.The detection of NT-proBNP,GFAP and HIF-1α levels has certain clinical value in judging the disease and prognosis.
