1.Effects of the recombinant plasmid of rat interstitial collagenase on experimental liver fibrosis

Objective To observe the effects of recombinant plasmid of rat's interstitial collagenase on experimental liver fibrosis. Methods We constructed the recombinant plasmid of rat's interstitial collagenase fused Flag peptide which can be expressed in eucaryotic cells. After bound to galactose terminal glyco poly L lysine(G PLL), we transferred the recombinant plasmid to the rat's fibrotic liver in vivo by intravenous injection, then observed the effects of the plasmid on the fibrotic liver by RT PCR, immunohistochemistry and the observation of pathology. Results The recombinant plasmid of interstitial collagenase could be expressed in rat liver, the expression of the plasmid could increase the degradation of type Ⅰ and type Ⅲ collagen in the rat's fibrotic liver compared with fibrotic modal group ( P 0.05). Conclusion The recombinant plasmid of interstitial collagenase have some effects on liver fibrosis, but this effects is limited.
2.Effect of various rewarming regimes on hemodynamic parameters of dogs after open abdominal injury and seawater immersion

Objective To evaluate the effect of various rewarming regimes on rectal temperature and hemodynamic parameters of dogs after open abdominal injury and seawater immersion. Method 30 healthy dogs were subjected to open abdominal injury and 2-hour seawater immersion. Then dogs were divided into three groups: group A (n=10) received no special warming treatment, in group B(n=10) electric blanket was used to warm up to 42℃, in group C (n=10) animals were warmed by 1/3 saline solution in 42℃ for 20 minutes and continuous perfusion of the abdominal cavity with 1/2 saline solution for 2 hours. The changes in body temperature, heart rate, MAP, CVP and CO were observed before and after rescue. Results The results of group A were not satisfactory. The rectal temperature, MAP, HR and CO were not recovered to normal after 6 hours of treatment. and the rectal temperature was 34.8℃ after 6 hours. The rectal temperature rose to 36.9℃ after being rewarmed with an electric blanket in group B. The result was better than that of group A. But there was tachycardia. The effects of rewarming from hypothermia in group C was satisfied. All hemodynamic parameters and rectal temperature recovered after 4 hours treatment. Conclusion Cold seawater immersion after open abdominal injury would result to hypothermia, and intra-corporeal rewarming should be the optional treatment.
3.Hemodynamic and pathological changes after seawater immersion in dogs with perforating injury of the intestine

Objective To evaluate the effect of seawater immersion on hemodynamics and pathological changes in dogs with perforating injury of the intestine. Methods Thirty healthy adult dogs were subjected to perforating injury of the intestine. Ten injured dogs were immersed in artificial seawater after the injury. An equal number of injured dogs were immersed in normal saline solution. Control group dogs (n=10) were not subjected to salt water immersion. Hemodynamical and pathological changes were observed at different time intervals after injury. Results Obvious hemodynamic disorder and serious pathological changes were found after seawater immersion. Normal saline solution group and control group dogs did not show obvious disorders in hemodynamics and pathology. Conclusion Seawater immersion is one of the main factors leading to the disturbance in hemodynamic and pathological changes after perforating injury of the intestine.
4.Relationship between plasma carbon monoxide and hyperdynamic circulation of cirrhotic rats

Objective To explore the relationship between the plasma level of carbon monoxide and the hyperdynamic circulation of cirrhotic rats. Methods S.D. rats were divided into cirrhotic ( n =10) and control ( n =10) groups. Liver cirrhosis was induced by percutaneous administration of carbon tetrachloride. Mean arterial pressure (MAP, kPa),heart rate (HR,beat/min) and portal pressure (PP,cm H 2O) were measured by indwelling catheters. Plasma CO was measured by Chalmers method, and plasma NO 2/NO 3 was determined by the reduction of cadmium column. Heme oxygenase (HO) activity in splanchnic organs from cirrhotic rats and controls was also determined. Results Typical features of cirrhosis were histologically observed in carbon tetrachloride treated animals. 5.Effects of antisense tissue inhibitor metalloproteinase-1 on hepatic stellate cells

Objective To study the expressing status of antisense tissue inhibitor of metalloproteina se-1(TIMP-1) in hepatic stellate cells (HSC) constructed in vitro, and to eval u ate the effects on the production of type Ⅰ and Ⅲ collagens secreted by activated rat HSC. Methods HSC were extracted from normal rat liver by pronase and co llagenase digestion and purified by centrifugal elutriation, and were cultured pla stic until they were activated to a myofibroblastic phenotype after 7-10 days. RT-nest-PCR and gene recombinant techniques were used to construct the rat ant isense TIMP-1 expression plasmid which can express in eukaryotic cells, and seg uenced after being counstructed. The expressing plasmid and the pcDNA3 empty pla smid were transfected into HSC by Effectene reagent separately. The cells were sel ected after growing in DMEM containing 400 ?g/ml G418 for 3 to 4 weeks. Exp ression of TIMP-1 in HSC was d etermined by Northern blot and Western blot. We tested the interstitial collagen ase activity in culture media with FITC-labled type Ⅰ collagen as substrate. U ltimately, we quantified the typeⅠ and type Ⅲ collagen in HSC by Wester n blot. Results The exogenous antisense TIMP-1 recombinant plasmid could block the expression of TIMP-1 greatly, while there were not the same outcome i n pcDNA3 empty plasmid g roup and non-transfecting control group. 6.Clinical analysis on hyponatremia in acute craniocerebral injury

Objecfive To explore pathological mechanism and treatment of central hyponatrem-ia. Methods Synchronous assay was made to detect changes of atrial natriuretic peptide(ANP),brain natriuretic peptide(BNP),endogenous digitalis-like substance(EDLS),antideuretic hormone (ADH),Na+ concentrations in blood and urine as well as osmotic pressure of plasma and urine in 68 pa-tients with traumatic brain injury(TBI). Results Of all,there were 27 patients with hyponatremia,mostly in patients with severe or critical TBI.There found syndrome of inappropriate secretion of antidi-uretic hormone(SIADH)in 7 patients and cerebral salt wasting syndrome(CSWS)in 20. Conclu-sions The central hyponatremia in patients with TBI may be related to the increased secretion of EDLS and ADH.The decrease of ANP and BNP in blood has no direct effect on Na+ concentration in blood.In-travenous injection of extrinsic thyrotropin releasing hormone(TRH)may inhibit dilutional hyponatremia resulted from increased secretion of ADH in TBI patients.

8.Experimental study of water and electrolyte changes following seawater immersion of animals with intestinal rupture

Objective To evaluate the changes in water, electrolyte and acid-base balance after seawater immersion in cases of open abdominal trauma associated with intestinal rupture, and to obtain a theoretical basis for the early treatment of open abdominal injury with intestinal rupture in naval combat. Methods A canine model of open abdominal trauma with intestinal rupture was established in 26 healthy adult dogs, and they were divided randomly into three groups. All animals were subjected to abdominal wall incision and intestinal rupture. Seawater immersion group(n=10) was immersed into artificial seawater after trauma; normal saline solution group(n=6) was immersed into normal saline solution after trauma; control group (n=10) had no immersion. The 3 groups were observed for changes in water, electrolyte and acid-base balance, and the results were analysed and compared. Results Signficant disturbance of water, electrolyte and acid-base imbalance were observed in the seawater immersion groups, but no significant changes of these parameters were seen in the control group and normal saline group. Conclusion Seawater immersion is the main factor leading to the disturbance of body metabolism after open abdominal trauma with intestinal rupture.
9.Esthetic effect of resin infiltration for the treatment of post-orthodontic white spot lesions

Objective:To evaluate the effects of resin infiltration in the treatment of post-orthodontic white spot lesions (WSLs)and compare it with microabrasion.Methods:1 0 subjects with 66 teeth with noncavitated and unrestored WSLs after multibracket treatment were recruited.A simple randomized,split-mouth and controlled design was used to allocate WSLs to the resin infiltration and micro-abrasion groups(n =33).The area of the WSLs were measured photographically by using image analysis software before treatment (T0),1 week(T2)and 3 months(T3)after treatment.Data were analyzed with multifactorial analysis of the variances with repeated measures at an a level of 5% and a power of 80%.Results:The area of the white spot lesions(R)decreased significantly in both groups(P <0.05).At T3 the success rate of resin infiltration group(88.68%)was significantly higher than that of microabrasion group (64.67%)(P <0.05).Conclusion:Resin infiltration is more effective than microabrasion in the treatment of post-orthodontic WSLs.
10.Translucency,opalescence and fluorescence properties of six light-cured resin composites

Objective:To evaluate the translucency,opalescence and fluorescence reflection of 6 light-cured resin composites. Methods:Disks of 6 resin composites including Filtek Supreme XT-YT(XT),Filtek Supreme XT-A3B(3M),Clearfil Majesty-A3 (MJ),Gradia Direct-A3(GC),Venus-A3(VE)and Charisma Diamond-A3(CA)were prepared in the diameter of 9 mm with the thickness 2.0 mm.PR-650 spectral scanning colorimeter was used to measure and caculate translucency parameters(TP),opal-escence parameter(OP)and fluorescence reflection(FL)and to draw the fluorescence reflection curves.Results:Except XT and MJ,TP value of other resin composites were in the range of natural tooth.Only OP value of XT was in the range of natural tooth en-amel.FL value of XT was close to that of natural tooth dentin.Fluorescent peak wavelength of GC,MJ,VE and CA were in the range of natural tooth,but fluorescent peak height was higher than that of natual tooth.TP was positively correlated with OP(r =0.85,P <0.05).There was no statistic correlation between TP and FL,between OP and FL.Conclusion:The 6 resin composites can simulate optical property of natual tooth,but the opalescence of resin composites is defective.
