1.IL-6 selectively activates STAT3 in U937 cells

Aim To investigate the signaling basis of the biological effects of IL-6 in U937 cells. Methods We determined the effect of IL-6 on the growth and differentiation of U937 cells and the effect of IL-6 on the activation of nuclear factors in U937 cells. Results IL-6 induced macrophage differentiation of U937 cells. Differentiated cells had strengthened ANAE (acid naphthyl acetate esterase)and NBT(nitroblue tetrazolium)-reducing activities and CD54 expression was upregulated. STAT3 could be remarkably activated by IL-6 in U937 cells and STAT3 activity was dependent on dose and time course of IL-6 treatment. However, NF-IL6,NF-κ B, AP-1 and other STAT members were not activated by IL-6. Conclusion IL-6 might induce terminal differentiation of U937 cells via JAK-STAT3 pathway.
2.Laparoscopic modified Swenson pull-through procedure for Hirschsprung's disease in children

Objective To introduce the experience of l ap aroscopic modified Swenson pull-through procedure for the treatment of Hirschspr ung's disease (HD). Methods A modified Swenson procedure was performed in 100 cases of HD, including 21 cases of short-segment phenotype, 72 cases of common phenotype, and 7 cases of long-segment phenotype. The patients w ere aged 8 months ~ 7 years, with a mean age of 1.5 years. A 0? or 30? laparos cope, with an inner diameter of 0.5 cm, was advanced from the umbilicular port, and operating instruments were introduced into the left and right lower quadaran t of the abdomen. The carbon dioxide was insufflated in the abdominal cavity at a pressure of 8~12 mmHg. Biopsies from the colonic seromuscular layer were made for pathological examinations. Laparoscopic dissection of blood vessels of the m esocolon was carried out closely near the colon, until the level 0.5~1 cm above the dentate line. The rectum and sigmoid was mobilized outside through the anus to complete rectal dissection. The proximal colon then was anastomosed to the re ctum by means of full-thickness suture and diagonal heart-shaped closure. Results All the cases were cured, without fatal cases. The operation time was 80~170 min (mean, 110 min), and the intraoperative blood loss was less than 10 ml. There were 5 cases of subcutaneous emphysema and 6 cases of urinary retention. On a follow-up in all the cases for 6~18 months, 5 cases of colitis, 2 cases of soiling, 1 case of anastomotic stricture, and 1 case of delayed anas tomotic leakage were observed. Conclusions Laparoscopic modifi ed Swenson pull-through procedure for Hirschsprung's disease is worthy of being recommended for its minimal invasion and simplicity of performance.

4.Isolation of a new natural compound from berry of Panax ginseng

Objective To study the constituents from the berry of Panax ginseng. Methods Compounds were separated and purified by column chromatographic methods. Their structures were elucidated by physicochemical properties and spectral analyses. Results Twelve compounds were isolated from water extracts of the berry of P. ginseng. They were identified as benzoic acid (Ⅰ), isoginsenoside-Rh3 (Ⅱ), ginsenoside-Rh2 (Ⅲ), ginsenoside-Rh1 (Ⅳ), ginsenoside-Rg1 (Ⅴ), ginsenoside-Re (Ⅵ), ginsenoside-Rd (Ⅶ), ginsenoside-Rc (Ⅷ), ginsenoside-Rb2 (Ⅸ), ginsenoside-Rb1 (Ⅹ), ?-sitosterol (Ⅺ), and 20-O-?-D-glucopyranosyl-20 (S)-protopanaxadiol named as compound K (ⅩⅡ). Conclusion The compound K is a new natural product.
5.Recovery of cardiac function in patients with frequent premature ventricular contraction after radiofrequcency catheter ablation by noninvasive hemodynamic detection technique

Objective To explore the change in cardiac function in patients with idiopathic frequent premature ventricular contraction(PVC) after radiofrequency catheter ablation(RFCA) and drug control by ultrasound Doppler and noninvasive hemodynamic detection technique.Methods Forty-eight patients with idiopathic frequent PVC were selected as the test group with a total of(16 391.03±10 873.01)beats per day.Another 55 healthy subjects were selected as the control group.Heart ultrasound Doppler and noninvasive hemodynamic detection technique were used in the two groups.Twenty-five patients were selected from the PVC group to receive RFCA,while noninvasive hemodynamic detection technique was adopted 3 days after operation.Results by ultrasound Doppler and noninvasive hemodynamic detection technique were compared between the two groups before and after operation.Results There was no significant difference in left ventricular ejection fraction(LVEF),left ventricular end-diastolic dimension(LVEDD),left ventricular end-systolic dimension (LVESD),left atrial diameter(LA)and the ratios of the peak mitral flow velocity in early diastole (E) to the peak atrial kick velocity (A) between the two groups.HI,SI,CI and C in PVC group were significantly lower than those in control group(P<0.01),but WA,O,WA/C and O/C were significantly higher than those in control group(P<0.01).HI,SI,CI and C obviously increased 3 days after operation(P<0.01),but WA,O,WA/C and O/C significantly decreased (P<0.01).Conclusion The cardiac functio of idiopathic PVC patients in the early stage may have been damaged,and RECA can reverse or delay the damaged cardiac function due to frequent PVC.
6.Analysis of clinical manifestations and prognosis of 68 cases of children with acute paraquat intoxication

Objective To study the clinical manifestations and influencing prognostic factors of children with acute paraquat in-toxication ,and to search for reasonable and effective treatments .Methods Retrospectively analyzed the clinical data of 68 children with acute paraquat intoxication from March 2005 to June 2012 .According to the amount of toxic dose and clinical symptoms ,all of them were divided into 3 groups ,mild type(22 cases) ,severe type(26 cases) and fulminant type(14 cases) ,retrospective analysis on clinical manifestation ,auxiliary examination and prognosis among the groups and following up the survival of these patients ,the risk factors which may affect the prognosis were explored .Results 66 cases were poisoned by taking orally and the other 2 cases were poisoned by the skin absorption .Gastrointestinal symptoms were the most common .Among 68 cases ,6 cases was lost the following up ,20 cases died(32 .26% mortality):all children from mild type survived ;8 patients from severe type and 12 patients from fulmi-nant type(30 .77% ,85 .71% ,mortality respectively ) .As the degree of the clinical classification deteriorated ,the mortality in-creased ,there was statistical signification between each other and so did the blood urea nitrogen and serum creatinine (P<0 .05) .By following up ,children who once passed the acute stage can recovery and the pulmonary fibrosis might be reversed by the chest CT follow-up .The logistic regression analysis indicated that children who had been diagnosed with acute kidney injury and multiple or-gan dysfunction syndrome showed the worse prognosis .Conclusion Acute kidney injury is a very important factor for the short term prognosis of paraquat intoxication ,while pulmonary fibrosis determine the long-term outcomes ,lung injury may be reversed in the course of the disease ,so we should take active treatments .
7.Significance and effects of emodin hypo-acid on regulating nitricoxide synthase of melanocyte in skin

BACKGROUND: Among multiple signals which affect the function of melanocyte, nitrogen monoxide (NO) has been thought as an important signal molecule. Emodin hypo-acid, effective component of rhubarb, can affect proliferation of melanocyte and regulate activity of tyrosinase in cells. OBJECTIVE: To study the effect of emodin hypo-acid on expression of nitricoxide synthase (NOS) of melanocyte in skin of guinea pig and definite the effective concentrations and mechanism of melanocyte in synthesizing melanin in living skin.DESIGN: Completely randomized grouping design and controlled study.SETTING: Department of Dermatology, Affiliated Hospital of Luzhou Medical College.MATERIALS: The experiment was completed at the Infection Laboratory of Affiliated Hospital of Luzhou Medical College and Laboratory of Histology and Embryology of Luzhou Medical College from January to June 2004. A total of 24 adult healthy male guinea pigs were randomly divided into 6 groups: control group, 2, 5, 10, 20 and 40 mg/L emodin hypo-acid groups with 4 in each group.METHODS: ① Emodin hypo-acid was diluted with dimethyl-sulfoxide into 2, 5, 10, 20 and 40 mg/L, then guinea pigs in emodin hypo-acid groups were injected subcutaneously with relevant dosages of emodin hypoacid which was provided by National Institute for the Control of Pharmaceutical and Biological Products into local skins of haunch and back, and the injected volume was 1 mL. Twenty-four hours later, 1 mL emodin hypo-acid of the same concentration was injected once again into one of the places which was injected before. Skin samples were selected after 48 hours and routine paraffin section was made. Skins which were not injected with any drugs were selected from control group and emodin hypoacid groups to regard as experimental control group. ② Expression of NOS was assayed with immunohistochemical method and melanocyte was identified under high-times optic microscope to observe stain and characteristic of cytoplasm. Five sections were randomly selected from each group. Every 20 cells on each section were measured with MLAS-1000 imaging analyzer and computer processing system was used to calculate average absorbency (A) of positive immune product in melanocyte. ③ Measurement data were compared with analysis of variance, and differences among groups were compared with SNK-q.MAIN OUTCOME MEASURES: Expression of NOS in melanocyte was assayed with immunohistochemical method and results were obtained with optic microscope and imaging analyzer.RESULTS: Average A value of positive immune product in melanocyte was lower in 2, 5, 10, 20 and 40 mg/L emodin hypo-acid groups than that in experimental control group (0.126±0.118, 0.103±0.082, 0.118±0.097,0.122±0.095, 0.112±0.078, 0.196±0.066, P < 0.05). There was no significant difference among emodin hypo-acid groups at various concentrations.CONCLUSION: ① Emodin hypo-acid can regulate expression of NOS in melanocyte through NO signal transduction pathway. ② Expression of NOS is not changed as the mass concentration of emodin hypo-acid varied from 2 to 40 mg/L.
8.Activation-status of IL-6-related nuclear factors in acute myeloid leukemia cells and their biological roles

Objective:To investigate the relation between the effects of IL 6 upon acute myeloid leukemia (AML) cells and its signal transduction mechanism Methods:Determined the effects of IL 6 on the growth of M1, R2, K562 and TF1 cells and the activation status of IL 6 induced nuclear factors (signal transducer and activator of transcription 3, STAT3 and nuclear factor IL6, NF IL6) by electrophoresis mobility shift assay (EMSA) Based upon these, blocked/reduced the activation of STAT3 or NF IL6 in M1 leukemia cells with STAT3/NF IL6 anti sense oligonucleotides (AS ODNs) and determined the oligonucleotides' effects upon IL 6 induced growth arrest of M1 cells Results:NF IL6 was constitutively activated in all these AML cells, while STAT3 was constitutively activated only in R2 and K562 cells Furthermore, STAT3 AS ODN reduced IL 6 induced growth arrest of M1 cells, while blocking/reducing of NF IL6 activation resulted in enhancing of IL 6 induced growth arrest Conclusion:JAK STATs pathway (especially STAT3) and Ras/MAPK pathway antagonize each other in proliferation regulation of AML cells
9.Effects of pancreatic kininogenase on myocardical fibrosis and serum nitric oxide level in spontaneously hypertensive rats

Objective To investigate the effects of pancreatic kininogenase on pressure,myocardical fibrosis and serum nitric oxide (NO) level in spontaneously hypertensive rats (SHR). Methods Twenty-four (fifteen weeks) male SHR were randomly divided into three groups:SHR group,pancreatic kininogenase group and captopril group(n=8 ),8 male Wistar kyoto rats with normal blood pressure were considered as control group. Pancreatic kininogenase was given by peritoneal injection (7.2 U?kg~ -1 ?d~ -1 ), captopril was given by intragastric administration(10 mg?kg~ -1 ?d~ -1 ), the rats in SHR group and rats with normol blood pressure in control group were treated with 0.9% NaCl(2 mL?kg~ -1 ?d~ -1 )administered through peritoneal injection. After four-week experiment, the pressure was measured in rats througth carotid artery,then the rats were sacrificed , and LVMI,CVF,PVCA and serum NO level were measured. Results In SHR group, SBP,LVMI,CVF and PVCA were higher, serum NO level was decreased obviously than those in control group (P0.05). Conclusion Pancreatic kininogenase can obviously reduce the blood pressure and reverse myocardial fibrosis,its mechanism may be concerned with the increasing of NO level in SHR.
10.A preliminary study of stent grafts seeded by CD34~+ stem cells in venous system

Objective To investigate the effect of CD34+ stem cells seeded stent graft in venous system. Method Twelve mongrel dogs were randomized into 2 groups. Stent grafts covered with PTFE or Dacron were implanted into infrarenal vena cava through a delivery system. In each group, 4 stent grafts were seeded by autogenous bone marrow CD34+ cells, and 2 treated by autogenous blood only as control. The grafts were explanted at 2 weeks. Light and electron microscopy were used to examine endothelialization of neointima. CD34 and Ⅷ factor stain were applied to identify endothelial cells. Results All stent grafts were patent except for one Dacron stent without seeding. Light, electron microscopy, CD34 and Ⅷ factor stain demonstrated that confluent endothelial cells appeared on the neointima of seeded grafts. No endothelial cells were detected in the non-seeding grafts. Conclusions Rapid endothelialization of stent grafts can be achieved through bone marrow CD34+ cells seeding in venous systems.
