Objective To investigate the effect of 12-lipoxygenase(12-LOX) inhibitor inducing the apoptosis of human hepatocellular carcinoma cell line HepG2 on the expression of survivin gene.Methods HepG2 cells were cultivated in RPMI-1640 medium.12-LOX mRNA was analyzed by reverse transcription polymerase chain reaction(RT-PCR).The effect of baicalein on the proliferation of the cells was detected by thiazolyl blue tetrazoliumbromide(MTT) method.The subcellular structures were observed under electron microscope.DNA ladder pattern on agarose gel electrophores was used to evaluate the apoptotic index of hepG2 cells.The expression of survivin mRNA was analyzed by RT-PCR.Results 12-LOX mRNA was expressed in human HepG2 cells.At concentrations from 20 to 80 ?mol?L-1,baicalein inhibited the proliferation of HepG2 cells in a concentration-and time-dependent manner from 24 to 72 h.There were significant differences between any other groups(P
2.CT quantitative diagnosis in fatty liver: a clinical study

Objective To establish the CT criteria of quantitative diagnosis for liver steatosis by means of studying the CT features of fatty liver cases proven histologically. Methods Twenty-eight cases of fatty liver were underwent non-enhanced CT scan, and the attenuation of liver parenchyma was measured. To differentiate the degree of fatty liver, the mean CT value and the relative density of hepatic vessels were observed. The quantitative diagnosis was made according to the CT number threshold and the criteria of relative density of hepatic vessels, respectively. Results Among the 28 cases, there were 17 cases of mild steatosis with mean CT number of 46 HU (32-65 HU), 7 cases of middle degree fatty liver with mean CT number of 28 HU (15-38 HU), and 4 cases of sever fatty liver with mean CT number of 0.2 HU (-7-11 HU). For the relative density of hepatic vessels, 16 of the 17 cases of mild fatty liver had a appearance of hepatic vessels immersion and 1 mild case had reverse hepatic vessels display, 6 of 7 middle degree cases had reverse hepatic vessels display with 1 case having the appearance of hepatic vessels immersion, and all the 4 case of sever steatosis had the appearance of reverse hepatic vessels display with sharp contrast between vessels and the liver parenchyma. 3.ERG11 mutations associated with azole resistance in Candida albicans isolates from vulvovaginal candidosis patients

Objective:To investigate the azole susceptibility of Candida albicans (C. albicans) from vulvovaginal candidosis patients and to analyze the relationship between ERG11 gene mutations in these isolates and azole resistance.
Methods:Three hundred and two clinical isolates of Candida species were collected. Azole susceptibility was tested in vitro in microdilution studies. The ERG11 genes of 17 isolates of C. albicans (2 susceptibles, 5 dose-dependent resistants and 10 resistants) were amplified and sequenced.
Results:Of the 302 isolates collected, 70.2%were C. albicans, of which 8.5%, 3.8%and 4.2%were resistant to fluconazole, itraconazole and voriconazole, respectively. In total, 27 missense mutations were detected in ERG11 genes from resistant/susceptible dose-dependent isolates. Among them, Y132H, A114S, and Y257H substitutions were most prevalent and were known to cause fluconazole resistance. G464S and F72S also has been proved to cause fluconazole resistance. Two novel substitutions (T285A, S457P) in hotspot regions were identified.
Conclusions:Twenty seven mutations in the ERG11 gene were identified in azole-resistant C. albicans isolates, which indicated a possible relation with the increase in resistance to azole drugs and the recurrence of vulvovaginal candidosis. The relationship of two novel substitutions (T285A, S457P) with fluconazole resistance needs to be further verified by site-directed mutagenesis.
4.Effects of Clostridium Butyricum Combined with Antibiotics on Postoperative Inflammatory Factors and Gastrointestinal Function of Patients with Acute Appendicitis after Laparoscopic Surgery

OBJECTIVE:To investigate the effects of clostridium butyricum combined with antibiotics on inflammatory factors and gastrointestinal function of patients with acute appendicitis after laparoscopic surgery. METHODS:A total of 102 patients with acute appendicitis undergoing laparoscopic surgery in our hospital during Jan. 2014-Jan. 2015 were divided into observation group and control group according to odd-even admission number,with 51 cases in each group. Control group was given Cefuroxime sodi-um for injection,Metronidazole injection and Cefixime dispersible tablets for anti-infective therapy after surgery. Observation group was additionally given Clostridium butyricum capsule 1260 mg,tid,for consecutive 3 d,on the basis of control group. The levels of serum inflammatory factors(TNF-α,IL-6),gastrin(GAS)and motilin(MTL)were compared between 2 groups before surgery and 3 d after surgery. The time of gastrointestinal function recovery(the time of bowel sounds,the time of first flatus,the time of postoperative first defecation)and the occurrence of compliance were recorded in 2 groups. RESULTS:Before surgery,there was no statistical significance in the levels of serum inflammatory and GAS,MTL factors between 2 groups (P>0.05). 3 d after sur-gery,the levels of TNF-α,IL-6 and CRP in 2 groups were decreased significantly,while the levels of GAS and MTL were in-creased significantly;above indexes of observation group were significantly better than those of control group,with statistical sig-nificance(P<0.05). The time of bowel sounds,the time of first flatus and the time of postoperative first defecation in observation group were significantly shorter than control group,with statistical significance(P<0.05). The incidence of complications in obser-vation group was 3.92%,which was significantly lower than 15.69%of control group(P<0.05). CONCLUSIONS:Clostridium bu-tyricum combined with antibiotics can effectively relieve inflammatory state of patients with acute appendicitis after laparoscopic sur-gery,promote gastrointestinal function recovery,and reduce the occurrence of complications.
