1.Application of the emergency command system in designated SARS hospitals

As soon as SARS hit Beijing, the Health Bureau of Beijing set Beijing Chest Hospital as a designated SARS hospital and initiated an emergency command system there under the direct guidance of the Bureau. Besides, 1 800 medical workers from 8 other hospitals were transferred on a temporary basis to the Chest Hospital to strengthen its workforce. In addition, experts teams for treatment, isolation and prevention, nursing, and a cooperation team for treatment with traditional Chinese medicine were formulated. Altogether 453 confirmed or suspect SARS cases were admitted. By June 24, 22 cases were referred to other hospitals, 20 suspect cases were excluded, and 26 cases died while all the rest were cured and discharged. The fatality rate was 5.79% . As practice shows, the initiation of the emergency command system pkyed a key role in the fight against SARS. It is suggested that in future the country establish and perfect an emergency public health system and that each third-tier hospital and above set up an infectious diseases department or solitary ward area.
2.Effects of different tension stress on the shape and F-actin of human peridontal ligament fibroblast in vitro

Objective To investigate the shape and F-actin of human periodontal ligament fibroblasts(HPDLF) under different tension stress in vitro so as to learn the stress-biological effects of HPDLF. Methods HPDLF were cultivated for 6 passages and observed morphologically and identified by immunocytochemistry to be positive anti-vimentin and negative anti-keratin. HPDLF were divided into four groups: control (without tension stress), static tension stress (5 kPa), dynamic tension stress group 1 (5 to 0 to 5 kPa at the frequency of 3/min) and dynamic tension stress group 2 (5 to 2.5 to 5 kPa at the frequency of 3/min). The cells of each group were observed at the different time points of 16, 24, 32 h. The projection areas and shapes of cells as well as the structure of F-actin were examined by laser scanning confocal microscope and immunity fluorescence technique. The relationship among tension stress, time, shape and the structure of F-actin of HPDLF was analysed. Results In the dynamic tension stress group 1 and 2, the shape and the arrangement of F-actin of some HPDLF underwent regular changes at 16, 24 h, but the changes appeared to be obvious at 32 h. In the static tension stress group, it was found the increase of interspace of HPDLF, but no obvious changes in the structure of F-actin. Conclusion Different tension stress patterns had different effects on the shape and F-actin of HPDLF. Especially in the dynamic tension stress group 2, it showed direct ratio between the cell projection areas, the average fluorescence density and different loading time.
3.Protection of Ulinastatin on gut barrier in septic rats

Objective To investigate the protective role of Ulinastatin UTI on gut barrier of septic rats. Methods Twenty-two SD rats were divided into three groups: sham laparatomy(S), cecal ligation and punture(CLP), and CLP plus UTI. Septic rat model was estabilished through CLP method. Fluorescence spectrometry was used to measure FITC-dextran concentration from bowel lumen to portal vein. Ultrastructure of intestinal mucosa was observed under transmission eletron microscope (TEM). Results Twenty-one hrs after CLP septic symptoms in CLP plus UTI group were milder than those in CLP group. 4.Methylation status of IGF -Ⅱgene P4 promoter in hepatocellular carcinoma

Objective To investigate the alternation of insulin -like growth factor -Ⅱ( IGF-Ⅱ) gene promoter P4 methylation status in hepatocellular carcinoma ( HCC) and explore its relationship with expression of P4 mRNA levels.Methods Liver specimens of 43 patients with HCC and normal liver specimens of 9 control patients were collected in operation .Tissue DNA and total RNA were extracted from these specimens .IGF-Ⅱ P4 methylation status and P4 mRNA expression levels were detected .Results (1)The incidence of IGF -ⅡP4 methyl-ation in HCC group was significantly lower than that in normal liver specimens (16.28%vs 88.89%,χ2 =19.12,P<0.01).(2)The expression level of IGF -ⅡP4 mRNA in HCC group was significantly higher than that that in normal liver specimens[(0.96 ±0.74) vs (0.25 ±0.19),t=5.48,P<0.01].(3)In HCC group,the IGF-Ⅱ P4 mRNA expression level with hypomethylation gene was significantly higher than that without hypomethylation gene [(1.18 ± 0.76) vs (0.32 ±0.27),t=5.28,P<0.01].Conclusion The hypomethylation alternation of IGF -Ⅱ P4 gene promoter which is accomplished by up -regulate P4 mRNA expression has a close relationship with HCC .
5.An in vitro natural degeneration model of chondrocytes derived from endplate of intervertebral discs of rats

OBJECTIVE: To set up a natural degeneration model of chondrocytes derived from endplate of intervertebral discs of rats in order to offer an appropriate carrier for the study on mechanism of intervertebral disc degeneration. METHODS: The method of enzyme digestion combined with natural subculture was used to set up the in vitro natural degeneration model of chondrocytes derived from the endplate of intervertebral disc of rats. The morphological appearances and microstructures of the chondrocytes of different generations were observed. The expression of collagen II in chondrocytes was detected by immunocytochemical method. RESULTS: The chondrocytes derived from the endplate of intervertebral disc expressed collagen II. After 13 days of culture, the chondrocytes of generation III showed that the ability of cell division descended, the nucleoli became unclear, the cells deformed obviously, fusiform shape with weak optical activity appeared, and the intercellular space was enlarged. There were vacuoles and lipid droplets in cytoplasm. The synthesis of collagen II, as well as the cell proliferation rate, descended notably. All results showed the natural degeneration process of the chondrocytes. CONCLUSION: The in vitro natural degeneration model of chondrocytes derived from endplate of intervertebral discs of rats was successfully established. This can offer the cytological basis for study on the mechanism of intervertebral disc degeneration.
6.Application of laparoscopic colectomy in mild colorectal perforation

Objective To explore the feasibility of removal of affected segments via laparoscopic surgery in patients with mild colorectal perforation. Methods The clinical data on sixty-one patients with colorectal perfo-ration who had been treated in our hospital between June 2006 and June 2016 were retrospectively analyzed. The patients were divided into an open surgery group(group OS,n=38)and a laparoscopic surgery group(group LS, n = 23). All the procedure-related data were collected and analyzed. P < 0.05 was considered statistically signifi-cant in this study. Results The volume of blood loss was smaller in group LS than in group OS[(100 ± 165)mL vs(200 ± 205)mL,P=0.032],hospital stay was shorter in group LS than in group OS[(7 ± 5.21)d vs.(10 ± 9.2)d ,P = 0.009],and rate of wound infection was lower in group LS than in group OS(4/23 vs. 13/38,P =0.039),while surgical duration was longer(213.7 ± 65.7 min vs. 173.5 ± 47.2 min,P = 0.047). There were no significant differences between the two groups in intraoperative blood transfusion[(0.8 ± 1.1)U vs.(0.4 ± 0.7)U , P=0.270],postoperative ICU stay[(2 ± 0.9)d vs.(2 ± 0.7)d,P=0.009],rate of postoperative complications 25/38 vs. 14/23),and hospitalization costs[(31 758 ± 29 283)yuan vs.(36 243 ± 27 584)yuan,P = 0.673]. Conclusions Laparoscopic colectomy is safe and feasible in the treatment of mild colorectal perforation.

Objective To evaluate the appearance of CT in patients with severe acute respiratory syndrome (SARS) in the recovery phase, and to study the correlation of CT findings with pulmonary function.Methods From June to August in 2003, 100 patient with confirmed SARS accepted examination in our hospital. Among them, 91 patients (39 men, 52 women, mean age 36.4 years, age range 19- 66 years) received CT examination and pulmonary function test on the same day. The interval between SARS onset and the examination ranged from 52 to 125 days (mean 87.4 days). CT appearances of pulmonary parenchymal abnormalities including distribution and extent of involvement were quantitatively analyzed, and four levels on CT scan including the aortic arch, the tracheal carina, the pulmonary venous confluence, and the dome of right diaphragm were selected to score the lesions. The correlation of CT scores with the results of pulmonary function tests was studied.Results Of the 91 cases, 47 patients had normal CT appearance in the recovery phase, whereas the other 44 patients still had parenchymal abnormalities, including residual ground-glass opacification and reticular shadow. Objective:
Schizophrenia (SCZ) is one of the most common and severe mental disorders. Modified electroconvulsive therapy (MECT) is the most effective therapy for all kinds of SCZ, and the underlying molecular mechanism remains unclear. This study is aim to detect the molecule mechanism by constructing the transcriptome dataset from SCZ patients treated with MECT and health controls (HCs).
Methods:
Transcriptome sequencing was performed on blood samples of 8 SCZ (BECT: before MECT; AECT: after MECT) and 8 HCs, weighted gene co-expression network analysis (WGCNA) was used to cluster the different expression genes, enrichment and protein-protein interaction (PPI) enrichment analysis were used to detect the related pathways.
Results:
Three gene modules (black, blue and turquoise) were significantly associated with MECT, enrichment analysis found that the long-term potentiation pathway was associated with MECT. PPI enrichment p-value of black, blue, turquoise module are 0.00127, <1×10-16 and 1.09×10-13, respectively. At the same time, EP300 is a key node in the PPI for genes in black module, which got from the transcriptome sequencing data.
Conclusion
It is suggested that the long-term potentiation pathways were associated with biological mechanism of MECT.

10.Implications of Approval of Cassiae Semen-based Health Food Products for Development and Supervision of Substances with both Edible and Medicinal Values

Cassiae Semen is a substance with both edible and medicinal uses approved by the National Health Administration, and it is widely used in health food products in China. In this study, an information database of Cassiae Semen-based health food products was constructed, and the common patterns of functional claims, major raw materials, and dosage forms were analyzed. The Apriori algorithm and network method were used to analyze the association rules of high-frequency ingredients with the three major functional claims: Maintaining blood lipid (cholesterol/triglyceride) level, controlling fat, and relaxing bowel. Secondly, based on the positioning and regulatory requirements of health food and the recent research progress in Cassiae Semen, this paper analyzed the problems in technology review and product development from safety, function claim, and quality controllability. Finally, suggestions were put forward to promote the inheritance and carry forward China's traditional healthcare culture and the achievement of the Healthy China Initiative. These suggestions included classifying and optimizing the access management of health food, innovating the traditional healthcare function claims, and strengthening the quality system construction and information disclosure of post-marketing evaluation.
