2.MESH diagrams of Chinese in Beijing and its preliminary application in practice.

OBJECTIVETo establish the normal MESH diagrams of Chinese in Beijing, and to build a computerized MESH analysis system for orthodontic practice.
METHODSTwenty-eight subjects with normal occlusion were selected in Beijing and their lateral cephalograms were taken at the age of thirteen and eighteen, respectively. Individual MESH diagrams were then established for each subject mainly according to Moorrees' method from the cephalograms orientated in estimated natural head position. Male and female normal MESH diagrams were created. A computerized MESH analysis system was also developed.
RESULTSThe normal MESH diagrams of Chinese in Beijing, thirteen and eighteen years old respective, were established. The computerized MESH analysis system was constructed and used in orthodontic patients.
CONCLUSIONSMESH analysis is a proportional analysis method. It can show the results directly, succinctly and holistically. It is also a favorable complement and amendment to the commonly used angle and linear X-ray analysis methods.

Methods: A total of 42 Chang-Bai piglets were divided into 7 groups: control group, 1, 2, 4, 8, 12, and 24 hours wound group. 5.The influencing factors of caregivers in patient with nasopharyngeal carcinoma undergoing radiotherapy

Objective To investigate the level and influencing factors of care burden in caregivers of patients with nasopharyngeal carcinoma undergoing radiotherapy. Methods Totally 117 caregivers of patients with nasopharyngeal carcinoma undergoing radiotherapy and 117 patients were investigated with general information questionnaire, Zarit caregiver Burden Interiew (ZBI), Self-Perceived Burden Scale (SPBS), Karnofsky Performance Scale (KPS). Results The caregiver burden score of nasopharyngeal carcinoma undergoing radiotherapy was 28.44±18.16,in light level.The main influencing factors included the caregiver′s occupational status, physical health level, the patients of self-perceived burden level. Conclusion Nursing staff should pay attention to the self-perceived of caregivers and patients, and to provide caregivers professional and individualized care guidance and psychological counseling, so as to reduce the burden of care and improve the quality of care for patients.
6.The effects of TENS on metatarsus plantarflexion and inversion in stroke patients

Objective To evaluate the effects of TENS on metatarsus plantar flexion and inversion in stroke patients,and to explore its mechanism.Methods Thirty-two stroke patients with gastrocnemius spasticity were randomly divided into a control group (n=16) and a TENS group (n=16).All patients were treated with foot sup- ports,neurodevelopmental and manipulation therapies.In addition,the TENS group received TENS on the anterior tibialis,peroneus longus and brevis muscles.All patients were assessed in terms of their Chinese stroke scale(CSS) and H reflex scores before and after therapy.Results Compared with those in the control group,the H reflex scores in the TENS group were obviously decreased,while H reflex latency was prolonged and H/M was reduced. Gait in the TENS group was evidently improved.Conclusion TENS is an effective therapy to decrease gastrocnemi- us spasticity and to improve the gait of stroke patients.
7.Anticoagulant Ability and Heparinization of Decellularized Biomaterial Scaffolds.

In order to enhance the anticoagulant properties of decellularized biological materials as scaffolds for tissue engineering research via heparinized process, the decellularized porcine liver scaffolds were respectively immobilized with heparin through layer-by-layer self-assembly technique (LBL), multi-point attachment (MPA) or end-point attachment (EPA). The effects of heparinization and anticoagulant ability were tested. The results showed that the three different scaffolds had different contents of heparin. All the three kinds of heparinized scaffolds gained better performance of anticoagulant than that of the control scaffold. The thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (APTT) of EPA scaffold group were longest in all the groups, and all the three times exceeded the measurement limit of the instrument. In addition, EPA scaffolds group showed the shortest prepared time, the slowest speed for heparin release and the longest recalcification time among all the groups. The decellularized biological materials for tissue engineering acquire the best effect of anticoagulant ability in vitro via EPA heparinized technique.

8.Effect of continuous blood purification for management of multiple organ failure with acute renal failure.

OBJECTIVETo evaluate the effect of continuous blood purification (CBP) for treatment of multiple organ failure (MOF) with acute renal failure (ARF).
METHODSForty-seven patients with MOF underwent CBP for an average of 3.1-/+0.5 days averagely. Continuous veno-venous hemofiltration was performed at daytime, and the substitute fluids were infused with pre-dilution at the rate of 2000-4000 ml/h. The general conditions, clinical symptoms, and serum biochemical indexes of the patients were observed and MODS score was calculated.
RESULTSAfter CBP, the MODS score of the patients decreased significantly from 9.1-/+3.5 to 5.4-/+2.7 (P<0.01) and serum creatinine decreased from 451.3-/+134.5 to 223.7-/+100.2 micromol/L (P<0.05). Twenty-nine patients survived with the survival rate of 61.7%.
CONCLUSIONCBP is effective for treatment of MOF and may help reduce the mortality rate of MOF complicated by ARF.

9.Construction of shRNA eukaryotic expression vectors of pkm2 gene and their effect on drug resistant cell line of acute promyelocytic leukemia.

This study was aimed to construct the shRNA eukaryotic expression vectors of M2-pyruvate kinase gene (pkm2) and to investigate the effects of pkm2 gene interference on the drug resistance of acute promyelocytic leukemia (APL) cells in vitro. Three specific shRNAs of pkm2 gene were designed and cloned into PBSU6 vector containing a U6 promotor. The constructed plasmids were identified and proved by the restriction sequence analysis. Then the effect of pkm2-shRNA on the protein expression of endogenous PKM2 was detected in NB4R2 cells, a drug resistant cell line of APL by Western blot. The alteration of NB4R2 cell differentiation with the interference of pkm2 gene was also validated by nitroblue tetrazolium (NBT) reduction test. The results showed that three specific shRNA eukaryotic expression vectors targeting pkm2 were successfully constructed. The efficiency of pkm2 gene silence was proved at protein level. The interference of pkm2 gene could significantly enhance the cell differentiation in the drug resistant NB4R2 cell line. It is concluded that the DNA vector containing pkm2 targeting shRNA remarkably promotes the differentiation of NB4R2 cells, showing the prospects of developing the gene target drug.

10.The relationship of telomere and telomerase activity with outcome of aplastic anemia after immunosuppressive therapy.

OBJECTIVETo observe the changes of telomere length and telomerase activity in patients with aplastic anemia (AA), and relationship with immunosuppressive therapy (IST) efficacy, to explore the pathogenesis of AA and the role of telomere length in evaluating immunosuppressive therapy efficacy.
METHODS71 cases of AA patients between September 2010 and March 2013 were enrolled into this study. 3 ml peripheral blood specimens from this cohort of patients were collected to test the telomere length in peripheral blood mononuclear cell (PBMNC) with flow-FISH and detect telomerase activity with TRAP-PCR-ELISA method.
RESULTSTelomere length and age showed negative correlation (b=-0.387, P=0.001) in normal control, NSAA and SAA + VSAA groups, telomere length became shorter with the growth of age, and normal control group telomere length decreased along with the age growth slightly greater than the other two groups (NSAA, SAA+VSAA). Besides the effect of age on telomere length, no significant difference was observed between NSAA and SAA+VSAA groups (P=0.573), and NSAA, SAA+VSAA (30.957 ± 4.502,29.510 ± 5.911)groups were significantly shorter than normal control group (51.086±10.844) (P<0.01). Telomere length in NR group (25.357±4.848)was significantly lower than normal control group (51.086 ± 10.844) (P=0.005), telomere length in CR(32.808 ± 4.685)/PR groups (30.334±4.464) compared with normal control group had no significant difference (P=0.517, P=0.254). Telomere length below 29.21% obviously decreased outcomes of IST. Telomerase activity had significant difference (χ²=20.385, P<0.01). The telomerase activity had no significant difference in terms of age and gender in three groups, multiple comparison found that telomerase activities in SAA + VSAA (0.324±0.178) (P<0.01), and NSAA (0.234±0.175) groups (P=0.002) were significantly higher than normal control group (0.107±0.083).
CONCLUSIONTelomere length of PBMNC in AA patients was significantly shortened than normal control group with telomerase activity increased, and telomere shorted more apparently in NR group, these patients should adjust the treatment as early as possible. Telomeres could predict the curative effect of IST.
