1. Alleviation of curcumin analog WZ35 on type 1 diabetes-induced renal injury via inhibiting inflammatory response

Objective: To study the alleviation of curcumin analog WZ35 on type 1 diabetic nephropathy (DN). Methods: Mice were treated with a single ip injection of streptozocin (STZ) to induce type 1 diabetes, while the control animals received the same volume of citrate buffer. The curcumin and its analog WZ35 (20 mg/kg) were ig administration for 9 weeks after the diabete obtained. The body weight and blood glucose were monitored every 7 d. Biochemistry analyzer was used to analyze the creatinine (Cr) and urea nitrogen (BUN) levels in serum. The histopathology of kidney tissue was detected by HE staining. RT-qPCR assay was used to evaluate the gene levels of inflammatory cytokines and chemokines. CD68 staining was used to evaluate the macrophages infiltration in kidney tissue. Results: Compared with negative control, the mice in diabetic group showed the reduced body weight, increased blood glucose, high Cr and BUN levels in serum, renal pathological damage, and increased inflammatory gene, and macrophages infiltration in kidney. While the administration with curcumin and its analog WZ35 could obviously attenuate the DN through inhibiting Cr and BUN in serum while had no effect on the body weight and blood glucose. Compared with curcumin, WZ35 could inhibit the gene expression of inflammatory cytokines and chemokines as well as infiltration of macrophages. Conclusion: The curcumin analog could attenuate DN through inhibiting inflammatory response.
2.Clinical analysis of conversion from gynecological laparoscopic surgery to laparotomy

Objective To discuss the causes and clinical significance of conversion to laparotomy during laparoscopic surgery.Method Three thousand two hundred and three cases who underwent laparoscopic surgery during the past five years were analyzed retrospectively.Results Three thousand one hundred and eighty cases underwent laparoscopic surgery,in which 23 cases were converted to laparotomy due to factors including severe pelvic adhesions(6 cases),complex disease with difficulties in laparoscopy (7 cases),massive haemorrhage(3 cases),bladder trauma(1 case),gastric trauma(1 case),intestinal trauma(1 case),as well as unexpected malignant genitalia neoplasm(4 cases).Conclusion The major causes for the conversion to laparotomy during gynecology laparoscopic surgery are severe pelvic adhesion and complex disease with difficulties in laparoscopy.Careful evaluation before surgery and conversion to laparotomy at the right moment can decrease the complication.
3.Optimum harvest study of Gentiana crassicaulis in Ludian.

The paper is aimed to study the difference in yield and quality at different harvest time and determine the optimum harvest of planting Gentiana in Ludian traditional harvest period. The authors analyzed the variation in fresh weight, dry weight, dry discount rate, length, diameter, volume and the content of gentiopicroside, loganin acid, alcohol-soluble extract and total ash and made a comprehensive appraisal of yield, appearance quality and intrinsic quality by gray relational distance ideal Comprehensive Evaluation method. The results showed that there is a big difference in yield and quality both 2-year-old and 3-year-old Gentiana harvested in traditional harvest period and the comprehensive evaluation more better when harvested more later. It can be seen, Gentiana harvested the later had a better yield and quality in Ludian traditional harvest period. The harvest of Gentiana can be appropriate delayed depending on the particular circumstances of production.

4.The Innate Immune Response in House Dust Mite-Induced Allergic Inflammation.

Hypersensitivity to house dust mite (HDM; Dermatophagoides sp.) allergens is one of the most common allergic responses, affecting up to 85% of asthmatics. Sensitization to indoor allergens is the strongest independent risk factor associated with asthma. Additionally, >50% of children and adolescents with asthma are sensitized to HDM. Although allergen-specific CD4+ Th2 cells orchestrate the HDM allergic response through induction of IgE directed toward mite allergens, activation of innate immunity also plays a critical role in HDM-induced allergic inflammation. This review highlights the HDM components that lead to activation of the innate immune response. Activation may due to HDM proteases. Proteases may be recognized by protease-activation receptors (PARs), Toll-like receptors (TLRs), or C-type lectin receptors (CTRs), or act as a molecular mimic for PAMP activation signaling pathways. Understanding the role of mite allergen-induced innate immunity will facilitate the development of therapeutic strategies that exploit innate immunity receptors and associated signaling pathways for the treatment of allergic asthma.

5.Expression,Purification and Characterization of [Gly14]-Humanin,a Novel Neuroprotective Peptide

Humanin(HN,its analogue [Gly14]-Humanin,HNG)was originally identified as an endogenous peptide that protects neuronal cells from apoptosis induced by various types of Alzheimer's disease-related insults.But the relative low content of this peptide in its natural sources limits its further characterization.An expression vector pET32a/HNG was corstructed and transformed it into E.coli BL21 trxB(DE3).HNG was expressed as a fusion protein in the soluble fraction and was purified by nickel affinity chromatography.Subsequently,the purified fusion protein was cleaved by enterokinase and was further purified by reverse-phase HPLC.A 23 mg recombinant HNG(rHNG)from 1 L bacterial culture was purified.The molecular weight of rHNG determined by ESI-MS was 2876.5 Da which was the expected size for correctly processed peptide.The N-terminal amino acid sequence of rHNG determined by Edman degradation method is identical to the theoretical sequence.Neuroprotective bioassay studies of rHNG exhibited its potential neuroprotective effect comparable to that of the natural HNG peptide.

8.Analysis of curative effect of chondroitin sulfate and glucnsamine sulfate on Kaschin-Beck disease

Objective To research the curative effect of chondroitin sulfate and glucosamine on Kashin-Beck disease(KBD). Methods According to Diagnosis for Kashin-Beck disease,80 patients of adult KBD were detected from Guanghui village Shangzhi city Heilongjiang province in July,2007,and they divided into treatment group and control group according to their condition,age and sex,40 person in each group. Treatment group was given chondroitin sulfate and glucosamine sulfate,and control group was given placebo(equivalent amount of starch). Bilateral knees X-ray films were shot before and after treatment (8th month),scale division magnifying glass was used to measure the width of joint space on X-ray films. Results The width of joint space respectively was (4.30±2.14) and (4.10±2.07)mm in control group before and after treatment,and treatment group respectively was (4.17±2.15),(4.16±2.11)mm. Medicine had no obviously role on joint space (F = 0.50,P > 0.05),Time and both of time and medicine had obvious role on joint space(F= 67.66,46.74,all P< 0.05). Joint space of treat group was thinner than control group(P < 0.05) before treatment,but thicker after treatment(P < 0.05). To compare with the width of before treatment,joint space width of control group became obviously narrow(P < 0.05). Conclusions Experimental group taking chondroitin sulfate and glucosamine sulfate alleviated knee joint space narrowing process of adults KBD patients compared with control group. It plays a protection role in articular cartilage and provides evidences for choosing drug and evaluating effect in the treatment of adults KBD.

10.Effects of ACE2/Apelin signaling on vascular autophagy regulation and hypertension pathogenesis

Autophagy provides protective effects on the maintenance of normal vascular structure and function. However, an overactivation of autophagy contributes to the pathogenesis of hypertension and other vascular diseases. The imbalance of vascular autophagy is involved in the occurrence and development of hypertensive vascular remodeling and injury, which is closely related to abnormal expression or activity of ACE2/Apelin signaling. This review will focus on the latest advancement and regulatory roles of ACE2/Apelin signaling in vascular autophagy and the pathogenesis of hypertension.
