1.Risk assessment of acute kidney injury after intracoronary stent implantation

Objective To investigate the risk factors of acute kidney injury(AKI) after intracoronary stent implantation in order to provide the basis for clinical prophylaxis and treatment.Methods Retrospectively analyzed 626 consecutive patients who underwent isolated intracoronary stent implantation in our institution from January 2007 to July 2011.Multivariate logistic regression model was constructed to identify the risk factors for the development of AKI defined as a serum creatinine (SCr) 130 to 199 μ mol/L or estimated creatinine clearance(Ccr) 30 to 60 ml/min per 1.73 m2.Results Ninety-three patients of 626 (14.9%) underwent isolated intracoronary stent implantation developed AKI.The results of the multivariate forward stepwise logistic regression analysis found that risk factors for the development of AKI following isolated intra-coronary stent implantation was associated with age (OR =1.570,95% CI 1.308-1.885),ejection fraction (EF) ≤ 30%(OR =11.526,95% CI 2.452-54.177),hypotension during perioperative and postoperation (OR =11.074,95% CI 2.439-50.282),operation duration(OR =1.032,95% CI 1.012-1.051),sex (OR =0.010,95% CI 0.001-0.086),NYHA class Ⅲ & Ⅳ (OR =0.209,95% CI 0.059-0.737),peripheral vascular disease (OR =0.528,95% CI 0.286-0.973),chronic obstructive pulmonary diseases (OR =0.546,95% CI 0.304-0.982),preoperation Cr (OR=1.418,95%CI 1.216-1.654) (and all P<0.05).Conclusion AKI is the common complications after intracoronary stent implantation,especially age,EF ≤ 30%,hypotension during perioperative and postoperation,operation duration are independent risk factors.
2.EXPERIMENTAL PATHOLOGICAL STUDY OF MYOCARDIAL DAMAGE INDUCED BY CATECHOLAMINE

The experimental myocardial damages were induced in Wistar rats by noradrenaline infusion (20ug/kg body wt/minute)for a period of 90 minutes daily for three consecutive days. The animals weresacrificed few minutes (acute phase), 7 days(subacute phase),and 30~90 days (chronic phase)afterinfusion. Specimens of myocardium were examined by histopathological, immunohistochemical and ultrastructural methods. Multiple focal necrosis, deficient of myoglobin in myocytes, myofibrillar fragmentation and dissolution were observed in the acute phase. The mdrphofogical feature simulated themyocardial infarctinn without coronary occlusion. Focal myocardial atrophy, scar f0rmation of myocardium, proliferation of fibroblasts and infiltraiton of the neo-colleagenous fibrils in necrotic myocytes were observed in the subacute and chronic phase. This feacture simulated the early cardiomyopathy.
3.Electroacupuncture of 2 Hz induces long-term depression of synaptic transmission in the spinal dorsal horn in rats with neuropathic pain

Objective: To observe the effect of 2 Hz electroacupuncture (EA) on long term depression (LTD) of synaptic transmission in the spinal dorsal horn in rats with neuropathic pain, so as to explore the central mechanisms of the antinociceptive effects of 2 Hz electroacupuncture on neuropathic pain. Methods: The neuropathic pain models were produced by tight ligation of the L5/L6 spinal nerves in Sprague Dawley rats. The C fiber evoked field potentials in the spinal dorsal horn were recorded with extracellular recording techniques. The parameters of the electroacupuncture were as follows: frequency of 2 Hz, wavelength of 0.6 ms, intensity of 1,2,3 mA lasting 10 min for each intensity, stimulation time of 30 min. The positive stimulating electrode was placed in acupoint “sanyinjiao” and the negative electrode in “zusanli”. 4.Effect of Itk knock down in Coxsackievirus-induced myocarditis model

Objective To study the effect of Itk down regulation on PBMC cell proliferation and inflammatory cytokines production in a Coxsackievirus-induced myocarditis model.Methods BALB/c mice were injected via caudal vein with plasmid Itk-shRNA then infected with CVB3.The change of Itk protein expression,cell proliferation,cytokines production and mice survival rate of mice were observed in the fourth days after infected.Results Itk mRNA in groups of mice transfected with Itk-shRNA was reduced about 40% in spleen cells,compared with that in control groups or shRNAnon groups (P<0.05).PBMC proliferation and serum cytokines were significantly inhibited by transfected with Itk-shRNA.Conclusion Knocking down Itk expression can inhibit mice inflammatory reaction.
5.Establishment of subseries cell lines from tongue cancer single cell and detection of cancer stem cell markers.

OBJECTIVETo establish subseries cell lines from single, cancer cell of Tca8113M1 cell line and detect the cancer stem cell markers in the different subseries cell lines.
METHODSThe subseries cell lines from single cancer cell of Tca8113M1 cell line were established by limiting dilution assay in vitro. The characteristic of tumorigenicity and CD44, CD184, extracellular soluble antigen (ESA) of the cancer stem cell markers were detected by xenotransplantation and flow cytometry respectively.
RESULTSTotal 192 single cells of Tca8113M1 cell line were cultured and were deposited as one cell per well. There were 12 subpopulations origin from 192 single cells spheroid cultures. The ratio was 6.25% (12/192). In the different subpopulations, the tumorigenicity and expression of CD44 and ESA were at high levels, but the expression of CD184 was in different level. There were three kinds morphology of colonies derived from single cancer cells, holoclone, meroclone and paraclone. Cell line could be derived from carcinoma cell holoclones by cell culture. Meroclone and paraclone did not exist in cell culture in vitro.
CONCLUSIONTongue cancer stem cell may exist in Tca8113M1 cell line, cell line can be established and holoclone is the origin of cell line. This is a novel approach to the identification and enrichment for cancer stem cell.

6.Mediating role of neutrophil-lymphocyte ratio in association between polycyclic aromatic hydrocarbons and mild cognitive impairment

Background As a group of environmental pollutants, polycyclic aromatic hydrocarbons (PAHs) are neurotoxic and may cause mild cognitive impairment (MCI) by inducing inflammation. Whether neutrophil-lymphocyte ratio (NLR), an inflammatory indicator, plays a mediating role in the relationship between PAHs exposure and MCI is unclear yet. Objective To investigate a potential mediating role of NLR in the association between exposure to PAHs and MCI in coke oven plant workers. Methods Eleven urine hydroxylated PAHs (OH-PAHs) of 530 coke oven plant workers were determined by high performance liquid chromatography tandem mass spectrometry. NLR was derived from participants' routine blood examination results using a fully automated haematology analyser. The associations between urinary OH-PAHs and MCI were analyzed by binary logistic regression, the associations between urinary OH-PAHs and NLR were analyzed by multiple linear regression, and the role of NLR in the relationship between urinary OH-PAHs and MCI was evaluated by mediating effect analysis. Results After controlling for confounding factors and other OH-PAHs, the results of binary logistic regression showed that for every e-fold (e is the base of the natural logarithm) increase in the concentration of 2-hydroxynaphthalene (2-OHNap) and 1-hydroxyphenanthrene (1-OHPhe), the OR (95%CI) values of reporting MCI positive were 1.21 (1.02, 1.43) and 1.25 (1.04, 1.51) respectively. For each unit increase of NLR, the OR (95%CI) of reporting MCI positive was 1.56 (1.12, 2.18). The results of multiple linear regression showed that each unit increase in natural log-transformed levels of 1-OHPhe was associated with 0.05 (95%CI: 0.01, 0.10) increase of NLR. The results of mediating effect analysis showed that the association between urinary 1-OHPhe and MCI was partially mediated by peripheral blood NLR, with a mediation ratio of 9.8%. Conclusion Exposure to PAHs in coke oven plant workers may increase the risk of reporting MCI positive partially through increased NLR in peripheral blood.
