1.ANTI-APOPTOSIS EFFECT OF NADH ON NORMAL LIVER CELL LINES TREATED WITH CYANIDE

This study was to observe the anti apoptosis effect of NADH on a normal liver cell line inflicted with chemical hypoxia. L02 liver cells were treated with 3mmol/L KCN in the presence of different concentrations of NADH (0~600?g/ml) for 0.5, 2, 4 hours. The percentage of apoptotic cells was assayed with flow cytometry(FCM). Another three groups of experiment were done as following. Group Ⅰ was control. Groups Ⅱ and Ⅲ were treated with 3mmol/L KCN in the presence or absence of NADH (400?g/ml) for 2h. The expression of proteins of Bcl XL, Bcl 2 and Bax in the L02 cells were determined with FCM analysis. The results showed that NADH could obviously prevent L02 cells from apoptotic damage at the concentration of 400?g/ml. it could also significantly upregulate the expression of Bcl 2 and Bcl XL proteins and downregulate the expression of Bax proteins after cyanide challenge( P

To investigate the role of NADH in preventing apoptotic damage of human hepatic cells induced by chemotherapy drug cisplatin, the ultrastructural changes in hepatic cells were examined with transmission and scanning electron microscopy, propidium iodide (PI) staining was used to measure the apoptotic rate with flow cytometry, the expressions of p53 and bcl 2 gene were detected with RT PCR, the change in caspase 3 and caspase 8 of apoptotic molecules was examined with colorimetric assay. The results showed that, compared with the group of cisplatin, morphological apoptotic changes were not obvious, the apoptosis rate was significantly decreased, p53 mRNA expression was decreased, bcl 2 mRNA expression was increased, caspase 3 and caspase 8 activity levels were kept in a low level in the group of NADH /DDP. The study indicates that NADH can prevent apoptosis of human hepatic cells, and increase the possibility of using NADH to reduce side effects of chemotherapy.
3.EFFECTS OF REDUCED COENZYME Ⅰ ON REGULATION OF GENE EXPRESSION IN PC12 CELLS DAMAGED BY ROTENONE

To elucidate the mechanism of mitochondrial damage induced by rotenone and the possible biological function of NADH in repairing mitochondrial damage of PC12 cells, cytotoxicity test, immunocytofluorescence and flow cytometric analysis were used to investigate the changes of cell proliferation genes (c myc, c erbB 2), apoptosis inhibition genes bcl 2, p53 tumor suppressor protein, cell immediate early gene (c fos) and proliferating cell nuclear antigen (PCNA) in PC12 cells before and after exposure to rotenone. The results showed that rotenone could significantly inhibit the proliferation rate of PC12 cells and expression of c erbB 2, c myc, p53, and bcl 2 in PC12 cells, NADH could restore the proliferation activity of PC12 cell damaged by rotenone by gene regulation. It is suggested that rotenone could induce PC12 cells apoptosis not only by regulating mitochondria phosphorylation process, but also by down regulating the expression of oncogene proteins (C erbB 2, c myc), anti apoptotic gene protein (bcl 2), p53 tumor suppressor gene protein, and upregulating the expression of the immediate early gene c fos. Regulation of bcl 2, c myc, c erbB 2 and p53 might be involved in the repair of mitochondrial damage of PC12 cells.
4.THE MECHANISM UNDERLYING THE ANTAGONISTIC EFFECT OF REDUCED FORM COENZYME Ⅰ (NADH) AGAINST TOXIC EFFECT OF DOXORUBICIN ON MYOCARDIAL MITOCHONDRIA

To probe the effect of reduced form coenzyme Ⅰ(NADH) in antagonizing cardiac muscle toxicity induced by doxorubicin and its underlying mechanism. Primary culture of myocardial cells of SD rat and doxorubicin injury model were established. MTT assay, laser confocal microscopy, transmission electron microscopy and biological oxygen monitor were used to observe the morphology and function of mitochondria. The results showed that the killing rate was increased in the group of doxorubicin, and that in the group of NADH/doxorubicin was greatly decreased. Doxorubicin could induce ultrastructural damage of cardiomyocyte mitochondria, manifested as swelling, disintegration, disruption of cristae and fusion. Cardiac mitochondria were protected against injuries in the group treated with NADH/doxorubicin. There was a significant difference in the fluorescence intensity of mitochondria membrane potentional and ROS between the groups treated with doxorubicin and NADH/ doxorubicin. It is suggested that NADH can significantly antagonizing cardiac muscle toxicity induced by doxorubicin and can protect mitochondrial structure and function.
5.Effect of NADH on the damage of L02 cells undergoing X-ray irradiation

AIM To study effects of antioxidant NADH on damage of irradiated normal cell lines. METHODS L02 liver cells were cultured in RPMI 1640, exposed to X ray irradiation, and continued to culture in the presence or absence of NADH for 24 h. The cellular viability was determined by routine MTT method. Using fluorescence probe and confocal microscope, the level of cellular H 2O 2 was detected. Positive rate of bcl 2 and Bax protein expression in the L02 cells were analyzed by flow cytometry. RESULT NADH can not only antagonized growth inhibition of X ray irradiated L02 cells, decreased cellular H 2O 2 production, but also significantly increased positive rate of L02 cells expressed bcl 2 protein and decrease positive rate of L02 cells expressed Bax proteins ( P
6.Analysis on influence factors of medication compliance in elderly patients with hypertension

Objective To know the medication compliance in elderly patients with hypertension,to analyze the influence factors of medication compliance,and to propose forward intervention countermeasures.Methods The treatment of hypertension in elderly patients was investigated by questionnaire survey.Medication compliance and influence factors were analyzed.Results 166 cases with poor medication compliance,and the rate was 45.0%; 203 cases with good medication compliance,and the rate was 55.0%.It's showed significantly difference on degree of education between the observation group and control group (x2 =12.071,P=0.002).The proportion of peasant and worker was higher in the observation group than control group (78.9% vs.62.1%,x2 =12.817,P=0.002).The smoker (48.8%) and drinker (59.0%) in observation group more than smoker (27.1%) and drinker (31.0%) in control group(x2=18.481,P<0.001; x2=29.115,P< 0.001).The ratio of payout of-pocket for healthcare was higher in the observation group (21.1%) than control group (9.9%) (x2=9.084,P=0.003).The duration of hypertension was longer in the observation group (8.8±4.6) than control group (6.0±4.1) (t=1.806,P=0.036).The income per month was less in the observation group (1850±150) than control group (2150±200) (t=5.799,P< 0.001).There were regression relationship between medication compliance and degree of education,medical security and duration of hypertension.Conclusions According to the influence factors of medication compliance in elderly hypertensive patients,it's useful to improve medication compliance by enhancing health education and designing a personal treatment.

Objective To analyze and study the diagnosis of large granular lymphocyte leukemia(LGLL).Methods To report and discuss six cases weth LGLL we have found. Results 2 of T-LGLL and lof NK-LGLL had indolent process, mainly presenting with anemia and splenomegaly and good response to treatment, while; 1 of T-LGLL and 2 of NK-LGLL had aggressive process, their clinical characters are obviously general symptom, hepatomegaly,splenomegaly,these disease develop quickly and have bad prognosis. The immunophenotype of indolent LGLL is distinct from aggressive cases. Conclusion As a group of heterogeneous disease,its diagnosis should be based on clinical manifestation and immunophenotype and differentiated it carefully.

Objective To investigate the clinical and laboratory diagnosis of adult hemophagocytic syn-drome (HPS) . Methods Clinical data of 24 patients with HPS from 2000 to 2008 were retrospectively analyzed. Results Of the 24 HPS cases, 12 had a malignant associated hemophagocytic syndrome (MHAS), and 10 were fi-nally diagnosed by bone marrow immunohistochemist ;Of 12 cases in non-MAHS group,4 were with virus associated hemophagocytic syndrome (VAHS), and 4 were of the other infections, whereas 4 patients diagnosed of immune associated HS (MAS). There were significant difference in onset age, mortality, serum lactate dehydrogenase (LDH) and serum ferritin(FER) and neutrophilic NAP between non-MAHS group and MAHS group(P <0.01 ,P<0.05). In all cases bone marrow biopsy showed significant differences in cytological and pathological features between MAHS group and non-MAHS group. Conclusion Etiology,immunology,and bone marrow cell biopsy and pathology as well contribute to the diagnosis and typing of HPS and will give a guide to the therapy.

Paeoniae Radix Alba is the dried root of Paeonia lactiflora, which was first recorded in the Shennong's Classic of Materia Medica and listed as the top grade. It is a common blood-tonifying herb, and its chemical components are mainly monoterpenes and their glycosides, triterpenes, flavonoids and so on. Modern research has demonstrated that Paeoniae Radix Alba has the activities of anti-inflammation, pain easing, liver protection, and anti-oxidation, and thus it is widely used in clinical practice and has broad development prospects. In this paper, the research progress on the chemical composition, pharmacological effects, and quality control of Paeoniae Radix Alba were summarized. On this basis, the Q-markers of Paeoniae Radix Alba were predicted from the aspects of mass transfer and traceability, chemical composition specificity, and availability and measurability of chemical components, which will provide a scientific basis for the quality evaluation of Paeoniae Radix Alba.

10.Study on critical quality attributes of Qingjin Huatan Decoction based on serum pharmacochemistry.

Qingjin Huatan Decoction is a classic prescription with the effects of clearing heat, moistening lung, resolving phlegm, and relieving cough. In order to explore the critical quality attributes of Qingjin Huatan Decoction, we identified the blood components of Qingjin Huatan Decoction by ultra-performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-Q-TOF-MS) under the following conditions, chromatographic column: Acquity UPLC BEH C_(18) column(2.1 mm×100 mm, 1.7 μm); mobile phase: 0.1% formic acid acetonitrile(A)-0.1% formic acid in water(B); gradient elution; flow rate: 0.2 mL·min~(-1); column temperature: 30 ℃; injection volume: 5 μL. The electrospray ionization(ESI) source was used to collect data in both positive and negative ion modes under the following conditions, capillary voltage: 3 kV for the positive ion mode and 2 kV for the negative ion mode; ion source temperature: 110 ℃; cone voltage: 30 V; cone gas flow rate: 50 L·h~(-1); nitrogen degassing temperature: 350 ℃; degassing volume flow rate: 800 L·h~(-1); scanning range: m/z 50-2 000. In this experiment, a total of 66 related components of Qingjin Huatan Decoction were identified, including 22 prototype components and 44 metabolites. The results of this study preliminarily revealed the pharmacodynamic material basis of Qingjin Huatan Decoction in vivo, which has provided an experimental basis for the determination of quality markers of Qingjin Huatan Decoction and the development of new drugs.
