1.Effect of basic fibroblast growth factor on healing of rabbit corneal wound caused by mustard gas

Objective: To study the effect of topical administration of basic fibroblast growth factor (bFGF) on healing of rabbit corneal wound caused by mustard gas. Methods: All the corneas of 8 New Zealand white rabbits were injured by liquid mustard at the concentration of 0.2%. One eye of each rabbit was treated with bFGF eye drops 6 times daily, the other eye with 0.25% chloromycetin eye drops as control. Computerized image analyzer was used to calculate the corneal epithelial healing rate. Results: The mean epithelial healing rate in bFGF-treated eyes was (1.276?0.152) mm 2/h, whereas in the control eyes it was (1.094?0.154) mm 2/h. Objective To investigate the relevance between protein expression and methylation of MG-MT and hMSH2 in glioma patimts.Methods Immunohistochemical and methylation specific PCR were adopted respectively to test on 275 cases of glioma patients for the protein expression and methylation situation of MGMT and hMSH2.Results The negative protein expression rate of MGMT and hMSH 2 in the tissue of brain golima were 47.2% and 62.5% respectively;the occurrence of methylation in gene promoter region were accordingly 41.8% and 22.4%.Statistical analysis revealed that MGMT promoter methylation in peripheral blood gene groups was related with the protein negative expression of tumor tissue (P<0.05),while there was no relationship between the protein expression of hMSH2 and its gene promoter methylation(P>0.05).Conclusion The meth-ylation of MGMT is a common molecular situation in the generation of brain glioma ,which may be connected with that of tumor.However,hMSH2 promoter methylation might not the main reason for inactivation of hMSH 2 pro-tein,there may be other important factors affecting its expression .
3.Therapeutic effects of combination of three drugs on healing of rabbit corneal wound caused by mustard gas

Objectives: To observe the therapeutic effects of collagen corneal shields (CCS) combined with fibronectin (FN) and epidermal growth factor (EGF) on healing of rabbit corneal wound caused by mustard gas, and to look for the new methods,new technology and new medicine to treat the injured cornea before eyelid edema observed in field hospital.Methods: After the left cornea was contaminated with mustard gas, 24 rabbits were equally divided into three groups(groups A, B, C) at random. Groups A,B,C were treated with PBS,CCS and CCS+FN+EGF,respectively. The corneas were stained with fluorescein and photographed 2, 8, 16, 24, 36,72 hours after contamination. The rate of corneal epithelial healing and percentage of damage were calculated. Results:The healing rates of groups A,B and C were (0.879±0.139)mm2/h, (1.223±0.166)mm2/h and (1.543±0.224)mm2/h,respectively and had statistically significant difference(P<0.01). The percentage of damage was 71.6%, 57.3% and 44.8%,respectively and also had statistically significant difference(P<0.01).Conclusions: The treatment with CCS can accelerate the corneal epithelial healing injured by mustard gas and the therapeutic effects will be better if CCS is combined with FN and EGF.
4.Genetic linkage analysis of localizing an autosomal dominant retinitis pigmentosa gene in a family

Objective:To localize the gene of autosomal dominant retinitis pigmentosa(ADRP) in a family. Methods: A large ADRP family was studied and 3 5 ml of venous blood from some family members was collected, and genomic DNA was extracted from the blood. Then two point linkage analysis between the known markers and the disease locus was performed. Results: Linkage analysis showed the maximum LOD score reached 2.732852 at marker D3S1292 (at recombination fraction ?=0.1). Conclusion: The gene responsible for ADRP is located in 3q21 eara.
5.Effect of δ-opioid receptor on bioactivity of human epidermal stem cells in vitro

Objective To observe the effect of δ-opioid receptor on proliferation and migration of human epidermal stem cells (hESCs) in vitro so as to offer treatment theory for skin injury.Methods hESCs from fresh foreskin tissues of normal young volunteers were isolated and cultured by enzyme digestion and differential adherence technique.Immunofluorescent staining was used to determine expression of integrin β1 and cytokeratin 19 (CK19) and flow cytometry was used for cell count.Second generation of cells were cultured for 5 consecutive days with keratinocyte serum-free medium (K-SFM) supplemented with 1 nmol/L (D-Ala2,K-Leu5)-enkephalin in Group A,with K-SFM supplemented with 1 nmol/L naltrindole and 1 nmol/L (D-Ala2,K-Leu5)-enkephalin in Group B,and with isolated K-SFM in Group C.Cellular division and proliferation were detected by MTT method.An in vitro 100 μm scratch-wound model was created on the confluent monolayer cells at 24 hours of incubation.Cells migrating from the wound margin were determined by inverted phase contrast microscope at 24,48,72,and 96 hours after wound formation,while wound closure rate was calculated at 72 hours.Results Primary cultured hESCs presented cobblestone-like shape after adherence growth,Immunofluorescence staining showed positive results for integrin β1 and CK19 and cell purity reached 95.6%.Moreover,MTT findings revealed proliferation of hESCs enhanced significantly in Group A,but lowered in Group B as compared to Group C (P < 0.05).hESCs migrated from the wound margin in all groups at 24 hours.However,more migrated cells were seen in Group A than in Group C and less in Group B than in Group C.Rate of wound closure was (89.5 ±0.7)% in Group A,(76.1 ±0.3)% in Group B,and (81.1 ±0.6)% in Group C at 72 hours,indicating significant differences among groups (P < 0.05).Conclusion Activation of δ-opioid receptor promotes the proliferation and migration of hESCs in vitro and may be implicated in wound healing.
6.The influence of heparin on the apoptosis and proliferation for K562 cells induced by vincristine

Objective To investigate the effect of heparin on the apoptosis and proliferation of human myeloid leukemia cell line K562 induced by vincristine.Methods K562 cells were pretreated by heparin for 1h,then cultured with 0.05 mg/L vincristine in 37℃ 5% CO2 for 24 h.Apoptosis of KS62 cells was evaluated by Hoechst 33342 staining,flow cytometer and DNA agarose gel electrophoresis after culture for 24 hours.The effect of heparin on KS62 cell proliferation and toxicitv was determined by Trypan blue staining and MTT assay.Results In the apeptosis induced group,the apeptosis rate was 40.10% dected by Hoechst 33342 fluorescence staining.The hepafin in different concentrations was found to be able to inhibit the apoptosis of K562 cells triggered by vincristine and the apoptosis rate was 32.47%,29.7%,25.5%,19.53% in the heparin groups of 25,50,100,200 U/ml,respectively.The apeptosis rate was significantly lower in the apeptosis induced group than in the heparin groups of 25,50,100,200 U/ml(P<0.05).The typical DNA ladder could be found in the apoptosis-induced group,and the DNA ladder gradually disappeared along with the increase of heparin(5~200 U/ml).The sub-G1 peak of K562 cells could be found in the induced group by FACS and the apoptosis rate was 21.61%.In the heparin groups of 25,50,100,200 U/ml,the sub-G1 Peak of K562 cells gradually dropped and the apoptosis rate was 13.64%,11.75%,8.59%,6.03%(P<0.05),respectively.After K562 cells were incubated with different hepafin concentrations(5~200 U/ml)for 24 hours,there was no difference compared with the normal control group in both the total live cell numbers and the cell proliferation rate measured by trypan blue staining and MTT assay(P>0.05).Conclusion The results suggested that heparin had no influence on KS62 cell toxicity and proliferation,but may inhibit the apoptosis of KS62 cells induced by vincristine.
7.Biomechanical properties of a magnesium alloy absorbable rib intramedullary nail

BACKGROUND: At present, the materials that are successful y applied in rib fracture internal fixation mainly include titanium, nitinol rib encircling bone bonding plate, imported poly-L-lactic acid absorbable rib nails and anatomical plate.
OBJECTIVE: To study the biomechanical characteristics of new magnesium al oy absorbable rib intramedul ary nail in the rib fracture fixation, and to compare with imported poly-L-lactic acid absorbable rib intramedul ary nail in order to provide scientific basis for clinical application.
METHODS: Thirty fresh adult fifth rib specimens were col ected, and the specimens were used to make models for the middle rib fractures. The specimens were fixed with AZ31B magnesium al oy absorbable rib intramedul ary nail (magnesium al oy group) and poly-L-lactic acid absorbable rib intramedul ary nail (poly-L-lactic acid group), and the normal rib specimen group was set as control. The biomechanical characteristics of the nails in each group were tested with stress analysis.
RESULTS AND CONCLUSION: Three-point bending strength measurement results showed that the bending strength in the magnesium alloy group was close to the normal value of the specimen (P > 0.05), and there was significant difference in the bending strength between magnesium alloy group and the poly-L-lactic acid group (P < 0.05). Torsional strength measurement showed that there was no significant difference in torsional strength between magnesium alloy group and the normal specimens, and the results showed that the magnesium alloy was better than poly-L-lactic acid in fixation (P < 0.05). Tensile tests showed that the tensile strength and anti-pulling force of the magnesium alloy fixation were better than those of poly-L-lactic acid fixation (P < 0.05). The results indicate that the magnesium alloy absorbable rib intramedullary nail is better than poly-L-lactic acid absorbable rib intramedullary nail in strength and tensile strength, which is the ideal fixation material for rib fixation.
8.Outcomes of traumatic flail chest treated by operative fixation versus conservative approach

Objective To compare the outcomes of traumatic flail chest with multiple injuries treated by operative fixation versus conservative approach. Methods The clinical data of 60 patients with traumatic flail chest with multiple injuries were retrospectively analysed, and the outcomes between operation group (treated by operative fixation, n=32)and non-operation group (treated by conservative approach, n=28) were compared. Results The mean time of hospital stay, ICU stay and mechanical ventilation was significantly shorter, and the prevalences of chest wall deformity, pulmonary infection, pulmonary atelectasis and respiratory failure were significantly lower in operation group than those in non-operation group (P<0.05). Six months after discharge, the pulmonary function parameters such as inspiratory capacity, forced vital capacity, forced expiratory volume in one second, peak expiratory flow, total lung capacity and maximal midexpiratory flow were significantly higher in operation group than those in non-operation group (P<0.05). Conclusion Traumatic flail chest with multiple injuries treated by operative fixation may lead to less flail chest associated complications. Operative fixation has short- and long-term benefits to flail chest.
9.Effect of Ginsenoside Rb _1 on apoptosis induced by hypoxia/hypoglycemia and reoxygenation in cultured rat hippocampal neurons

AIM: The aim of this work is to investigate the protective effects of Ginsenoside Rb 1(Rb 1) on apoptosis induced by hypoxia /hypoglycemia and reoxygenation in cultured rat hippocampal neurons. METHODS: Apoptosis were measured by flow cytometry; Morphological changes and neuronal necrosis were examined under microscope; The leakage of lactic dehydrogenase(LDH) and the product of nitric oxide(NO) were measured. RESULTS: hypoxia /hypoglycemia cultures for 5 h and reoxygenation induced neuronal apoptosis and necrosis,and significantly increased the leakage of LDH and the product of NO. The effects were enhanced with the extending time of reoxygenation. Rb 1 could significantly decrease the percentage of neuronal apoptosis and necrosis, and reduce the leakage of LDH and the product of NO. CONCLUSION: Rb 1 had an effect of anti-neuronal apoptosis. This effect might be related to the inhibition of the activity of NO synthase and NO production.

The effects of advanced glycation end products(AGE)on phosphorylation of ezrin/radixin/ moesin(ERM)protein in human umbilical vein endothelial ceils were detected by immunofluorescence cytochemistry and its mechanism was explored.AGE stimulated the phosphorylation of ERM protein in dose-and time-dependent manners(all P<0.05),which was involved in AGE receptor,Rho kinase,and p38 mitogen-activated protein kinase pathways.
