1.Analysis of the mutations of rhodopsin gene in autosomal dominant retinitis pigmentosa family

Objective To detect and analyse the mutations in rhodopsin gene of members in a family affected by autosomal dominant retinitis pigmentosa (ADRP). Methods Using the polymerase chain reaction (PCR), we amplified exon 1-5 of rhodopsin gene in patients with ADRP,and analyzed it with direct sequence measuement. Results The Gly-182-Asp mutation in the rhodopsin gene was detected in most of affected members of this ADRP family, but no mutation was detected in two affected members and the control ones. Conclusion We cannot regard the Gly-182-Asp mutation in the rhodopsin gene as the pathagenic factor of the ADRP family. It is likely there is a new gene next to the rhodopsin gene.
2.Thrombolytic drug infusion via carotid artery treating experimental central retinal artery occlusion

Objective To investigate the therapeutic effects of thrombolytic drug infusion via carotid artery on experimental central retinal artery occlusion (CRAO), and observe the changes of fibrinolytic activity in the systemic circulation. Methods To dissolve the thrombi in 15 cats (30 eyes) with CRAO established by laser irradiating a branch of central retinal artery after intravenous injection of photochemical drugs, urokinase (UK) was directly infused via carotid artery in 5 cats (10 eyes) in group A or intravenously injected in 5 cats (10 eyes) in group B, and isotonic saline solution was intravenously injected in 5 cats (10 eyes) in group C respectively. The patency of the artery was evaluated by fundus fluorescein angiography. Moreover, the changes of fibrinolitic activity in the blood were observed by blood biochemical examination. Results Four hours after UK infusion, the complete repatency proportion was 80% (5 cats 8 eys) in group A, and 50% (4 cats 5 eyes) in group B. There was significant difference between the two groups. 6.microRNA and its expression in lymphoma

MicroRNAs (miRNA), an abundant class of highly conserved small non-coding RNAs, suppress gene expression by binding to the 3' UTR of target mRNAs, which presents an entirely new approach to post-transcriptional regulation of gene expression. MiRNAs have been shown to play key roles in the regulation of diverse biological processes, such as development, inflammation, apoptosis, and tumorigenesis. Several studies have shown the dysregulated miRNAs in various lymphomas including the overexpression of miR-106a and miR-17-92 in T cell lymphoma;upregulated miR-155, miR-221 and miR-21 in diffuse large B-cell iymphoma and follicular cell lymphoma, and these miRNAs associated with subtypes of diffuse large B-cell lymphoma. Overexpression of the miR-17 cluster showed reduced levels of apoptosis, suggesting that the main effect of these miRNAs was to suppress cell death. Although miRNAs can act as both tumour suppressor and oncogenic molecules, the mechanistic explanation for this dysregulation is unclear but maybe linked to the defects in the miRNA biosynthetic machinery. As more evidence accumulates, it is likely that miRNAs will emerge as a new class of epigenetic regulator.
7.Effect of basic fibroblast growth factor on healing of rabbit corneal wound caused by mustard gas

Objective: To study the effect of topical administration of basic fibroblast growth factor (bFGF) on healing of rabbit corneal wound caused by mustard gas. Methods: All the corneas of 8 New Zealand white rabbits were injured by liquid mustard at the concentration of 0.2%. One eye of each rabbit was treated with bFGF eye drops 6 times daily, the other eye with 0.25% chloromycetin eye drops as control. Computerized image analyzer was used to calculate the corneal epithelial healing rate. Results: The mean epithelial healing rate in bFGF-treated eyes was (1.276?0.152) mm 2/h, whereas in the control eyes it was (1.094?0.154) mm 2/h.
