Objective:To investigate the effects of granulocyte-macrophage colony-stimulating factor on expressions of CC chemokine receptor 2(CCR2) mRNA and protein in cultured human THP-1 cell lines.Methods:THP-1 cell was stimulated by GM-CSF of different concentrations and time intervals.The mRNA expression of CCR2 was tested by RT-PCR,The protein expression of CCR2 was analyzed by Western blotting.Results:RT-PCR and Western blotting showed that THP-1 cell in control group expressed a low-level of CCR2 mRNA and protein.After stimulated by GM-CSF,the expression of CCR2 mRNA and protein increased in a concentration-and time-dependent manner.Conclusion:GM-CSF could stimulate THP-1 cell to express CCR2 mRNA and protein,in an concentration-and time-dependent manner.

6.A Comparative Study of Dietary Intake among Urban Japanese and Chinese Aged 50$\sim$79

This study examined the difference in dietary intake between middle−aged and elderly Japanese and Chinese. Volunteers aged 50∼79 living in two cities in both Japan and China were recruited in local community service centers and were asked to complete a 3−day diet recording. The final results were based on 356 subjects (166 Chinese and 190 Japanese). In men, the Japanese subjects significantly consumed more energy, with a large proportion of the energy coming from carbohydrates, dietary fiber, vitamin A, vitamin B2, vitamin C and they consumed less iron, and a lower proportion of energy from protein and fat than in the Chinese subjects. In women, the Japanese subjects had significantly higher intakes of energy, the proportion of energy from carbohydrates, dietary fiber, calcium, vitamin A, vitamin B1, vitamin B2, vitamin C and they consumed less iron, with the proportion of energy from protein and from fat in comparison with the Chinese women. Fat intake provided more than 29% of the total dietary energy in Chinese subjects. Daily intake of food items was significantly greater in the Japanese participants, both men and women (18.54±2.85, 20.11±2.40, respectively), than in the Chinese subjects (14.11±3.62, 15.51±4.15, respectively) (p<0.01 by Mann Whitney U−test). The present study suggests that the middle−aged and elderly Chinese subjects should increase their variety of foods consumed while decreasing their consumption of high−fat foods. For the Japanese subjects, the higher intake of total calories among the woman should also be noted.

7.Protection of δ-opioid receptor stimulation against injured myocardial cells by hydrogen peroxide

AIM To study protective effect of [D-Ala~2, D-Leu~5]-enkephalin (DADLE) against hydrogen peroxide (H_2O_2) induced myocardial cell injury and its possible mechanisms. METHODS Myocardial cells were isolated from neonatal rats and cultured for 48 h. Then the cells were randomly assigned into normal control, H_2O_2(200 μmol·L~(-1)), H_2O_2+DADLE(1 μmol·L~(-1)), H_2O_2+DADLE +naltrindole(10 μmol·L~(-1)) and H_2O_2+DADLE +U0126(10 nmol·L~(-1)) groups and cultured for another 48 h.[~3H]TdR incorporation assay and flow cytometry were used to measure the cell proliferation and apoptosis rate. The lactate dehydrogenase (LDH) activities in culture supernatant measured by using LDH activity kit. The superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in cells were measured with xanthine oxidase method and color reaction of thiobarbituric acid, respectively. The expressions of extracellular signal-regulated kinase (ERK) and phosphorylated-ERK (p-ERK) were observed with Western blot. RESULTS ① Compared with normal control group, the incorporation of [~3H]TdR in myocardial cells of H_2O_2 group was significantly lower, apoptosis rate was higher, LDH activity and MDA content in cells were higher, while SOD activity in cells was lower. In addition, the ratio of IA_( p-ERK) /IA_( ERK) was decreased. ② Compared with H_2O_2 group, the incorporation of [3H]TdR in H_2O_2+DADLE group was significantly higher, apoptosis rate was lower, LDH activity and MDA content in cells decreased, while SOD activity increased significantly. The ratio of IA_( p-ERK) /IA_( ERK) was increased. ③ δ-Opioid receptor antagonist naltrindole and ERK antagonist U0126 inhibited this effect of DADLE on the above index changes induced by H_2O_2. CONCLUSION The δ-opioid receptor has protective effect against H_2O_2-induced myocardial cell injury, and its possible mechanism may be related to its promotion of antioxide capacity and ERK phosphorylation.
8.Use of inflammatory markers of induced sputum for assessing the severity of late onset asthma and differential diagnosis

Objective To investigate the relationship between the eosinophil (Eos) percentage and levels of eosinophil cationic protein (ECP) in induced sputum and the severity of late onset asthma(LOA) and differential diagnosis. Methods Fifty-four patients with LOA were selected. The percentage of eosinophil and levels of ECP were measured by Wrigbt′s stain and immuno-CAP system respectively. The lung function was also evaluated. The same indexes were measured in 25 elderly patients with chronic obstructive pulmonary disease (COPD) and 10 healthy elderly subjects as control. A vector for a mutant Chinese Helicobacter pylori (Hp) strains knock out pathogenicity island (PAI) was constructed, which was the basis to establish a Chinese Hp mutant deleting pathogenicity island. Genetic engineering techniques such as polymerase chain reaction, plasmid extraction, agarose gel electrophoresis, restriction analysis, ligation, preparation of competence cell and transformation were used to make two cloning fragments containing two end regions of PAI and a selectable chloramphenicol resistance marker between them, and then engineering the recombined fragments into pBluescript plasmid. The result showed that restriction analysis demonstrated that the engineering mutant vector had been recombined. It suggested that we constructed an engineering mutant vector which targeted to delete the PAI in Chinese Hp strains. It will be useful for addressing the role of PAI in the pathogenesis of Hp infection.
10.Clinical significance of detecting serum sCD_(44V6) in non-small cell lung carcinoma

Objective To explore the clinical significance of detecting serum sCD44V6 in non?- small cell lung carcinoma. Methods Levels of serum sCD44V6 in 42 patients with lung carcinoma and in 30 healthy persons were measured by ELISA. Results There was obvious difference of serum sCD44V6 levels between patients with lung carcinoma and healthy persons. There was no difference between patients with squamous cell carcinoma and with adenocarcinoma. sCD44V6 have the higher positive rate and specificity. Conclusions sCD44V6 would be a good way to non?- small cell lung carcinoma, and could be useful to the diagnosis of early?- stage non?- small cell lung carcinoma.
