1.The comparison of three different nucleic acid isolation methods in hepatitis B virus DNA detection with real-time quantitative PCR

Objective To investigate the influence of different HBV DNA extraction methods with Micro-Nucleic-Releaser (MNR), polysaccharide deposition method and boiling method in real-time quantitative and provide reference data for clinical lab regarding the PCR kit selection. Methods Three different HBV DNA isolation methods including MNR, polysaccharide deposition method and boiling method were used to study the extraction efficiency and anti-interference ability when HBV DNA virus in lipemia, haemolysis or jaundice serum samples was detected with real-time PCR. The traditional HBV DNA isolation method of Hydroxybenzene-Chloroform was used as control method. Results The quantitative results and amplification efficiency differed among different extraction methods. Complete hemolytic sample brought biggest influence, modest hemolytic sample give minimal influence, whereas the lipemia and jaundice did not bring significant influence to detection. The MNR give the best amplification efficiency and quantitativereproducibility. The extraction efficiency of boiling method was lowest. The fluorescent efficiency of real-timePCR from Hydroxybenzene-Chloroform methods was better than other methods, but it suffered from losingHBV DNA. Conclusions Real-time PCR system of HBV DNA isolation by MNR can provide an accurate,highly sensitive, and rapid method to quantify Hepatitis B virus. It is suitable to use in clinical laboratory.
2.Experimental study of a polypropylene mesh for moulding and supporting a breast during a breast reduction procedure.

OBJECTIVETo study the polypropylene mesh acted for the moulding and support.
METHODSFour mini-pigs were used in this experimental research. A polypropylene mesh was implanted under the skin on one side of each pig. An expander was thereafter placed in the deep layer to imitate the action of gravity to the skin and mesh. The specimens were collected in two different times for the biomechanics and histology examinations.
RESULTSThe biomechanical data were shown lower and the histological properties were found changeable in the expanded skin without the mesh support, compared with the normal skin. However, the changes did not occur in the expanded skin with the mesh support. Furthermore, the tensile strength and elastic modulus of the polypropylene mesh were significant less than the human skin.
CONCLUSIONThe Polypropylene mesh could prevent the extended skin effectively and has moulding and support effects.

3.Visuospatial impairment in patients with Parkinson's disease and its related factors

Objective To study the incidence of visuospatial impairment in patients with Parkinson's disease(PD)and its related factors.Methods Visuospatial impairment was assessed in 107 PD patients from 6 hospitals in Guangzhou during the period from April to June 2007 using cube copying test.The motor,neuropsyehiatric,and cognitive symptoms and the activities of daily life(ADL)of the patients were evaluated using Hoehn-Yahr scale,Unified Parkinson's Disease Rating Score-Ⅱ (UPDRS-Ⅱ),UPDRS-Ⅲ,UPDRS-Ⅳ,Hamilton Anxiety Scale(HAMA),Hamilton Rating Scale of Depression(HRSD),Fuld Object-Memory Evaluation(FONt),Rapid Verbal Retrieval Test(RVR),Wechsler Adult Intelligence Scale Block Design(WISC-BD),Wechsler Adult Intelligence Scale Digid Span(WAIS-DS),Mini-Mental State Examination(MMSE),and Neuropsychiatric Inventory(NPI).Spearman correlation analysis was performed to analyze the bivariate correlation between the visual-spatial ability and the related factors,and binary logistic regression analysis was used to identify the factors related to visual-spatial disorder.Results Visuospatial impairment was found in 59 of the 107 patients(55.14%).Logistic regression analysis identified the Hoehn-Yahr stage and duration after disease onset as the risk factors,while RVR and WISCoBD as the protective factors for visuospatial impairment.Conclusion The Hoehn-Yahr stage,duration from disease onset,speech fluency,and image recognition and construction ability are related to visuospatial impairment in the PD patients.The visuospatial ability of the PD patients can be improved by relieving their motor symptoms and trainings for speech and image recognition and construction abilities.
