2.Correlation of Color Vision to Anxiety and Depression in Patients with Parkinson's Disease

Objective:To investigate the correlation between the change of color vision and anxiety or depression in patients with Parkinson's disease (PD). Methods:From November, 2018 to November, 2019, 63 PD patients from the PD database of Department of Neurology, Dalian Friendship Hospital were selected, and they were tested with Farnsworth-Munsell 100 Hue Test (FMT), Hamilton Anxiety Scale (HAMA), Hamilton Depression Scale (HAMD), Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS) and Hospital Anxiety and Depression Scale (HADS). Their age, gender, course of disease, education level, Hoehn-Yahr (H-Y) stage, Unified Parkinson's Disease Rating Scale part III (UPDRSIII), levodopa dose, and scores of Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA) were collected as related factors, and anlyzed with correlation analysis and multivariate regression. 3. Wiskot-Aldrich syndrome-like lower expressed in colon cancer tissues and inhibited the stemness of human colon cancer

Objective To explore the expression of Wiskot ̄Aldrich syndrome-like(WASL)in colon cancer tissues and its function on colon cancer stemness. Methods The UALCAN database was used to analyze the expression of WASL in colon cancer tissues and its correlation among the gender, lymphnode stage and metastasis in colon cancer patients. Immunohistofluorescence and immunocyte fluorescence were used to verify the date from UALCAN. Western blotting was used to detect the expression and transfection efficiency of WASL in human normal colorectal mucosal cells (FHC cells) and colon cancer cells HCT-116, SW-480. Sphere formation assay and colony formation were used to detect the effect of WASL on the stemness of colon cancer. Hypoxia assay, serum deprivation, drug resistance assay were used to detect the stemness of colon cancer cells. Results WASL was lower expressed in colon cancer tissues (P<0. 05). WASL was lower expressed in colon cancer tissues than peri-cancer tissues. WASL was obviously lower expressed in HCT-116 and SW-480 than FHC cells. WASL overexpression stable cells were constructed suscessfully in HCT-116 and SW-480.Real-time PCR showed WASL obviously down-regulated the expression of cancer stemcells (CSCs) related markers [octamer binding transcription factor 4 (OCT4), sex determining region Y-box 2 (SOX2), Lin28, krüppel-like factor (KLF)]. Sphere formation assay and colony formation assay showed WASL could inhibited the sphere formation and colony formation. Hypoxia assay and serum deprivation showed WASL inhibited the resistance of cancer cells to hypoxia and serum deprivation. Drug resistance assay showed WASL inhibit the resistance of cancer cells to 5-fluorouracil(5-FU). Conclusion WASL is lower expressed in colon cancer tissues and cells, and is related with poor prognosis of colon cancer. Overexpression of WASL can inhibit the stemness of colon cancer cells.
4.Ginsenoside Rg3 regulates CaM gene expression through PI3K/AKT signal system to promote apoptosis of gastric cancer BGC-823 cell

Objective: To investigate Ginsenoside Rg3 interfering the expression of CaM through PI3K/AKT signaling pathway to affect the biological activity of gastric cancer BGC-823 cells. Methods: After the culture and passage of gastric cancer BGC-823 cells, Western blotting was used to detect the expression of p-AKT and CaM protein in gastric cancer BGC-823 cells treated with IGF-1 and/ or Rg3; The effect of IGF-1 and/or Rg3 on the proliferation of BGC-823 cells was detected by MTT assay; The effect of IGF-1 and/or Rg3 on the invasion of BGC-823 cells was detected by Transwell assay; Effect of IGF-1 and/or Rg3 on apoptosis of BGC-823 cells was detected by Flow Cytometry. Results: Western blotting results showed that the expression of p-AKT and CaM protein increased in BGC-823 cells with the prolongation of IGF-1 treatment (all P<0.05); Compared with the blank control group, Rg3 significantly inhibited the proliferation of BGC-823 cells, while IGF-1 and IGF-1+Rg3 significantly promoted the cell proliferation (all P<0.05); Compared with the blank control group, Rg3 significantly reduced the invasion of BGC-823 cells, while IGF-1 and IGF-1+Rg3 significantly promoted the invasion of BGC-823 cells (all P<0.05);Flow cytometry showed that compared with the blank control group, Rg3 significantly promoted the apoptosis of BGC-823 cells, while IGF-1 and IGF-1+Rg3 significantly inhibited the apoptosis of BGC-823 cells (all P< 0.05). Conclusion: Ginsenoside Rg3 inhibits the expression of CaM by blocking PI3K/AKT signaling pathway, thereby promoting the apoptosis of gastric cancer BGC-823 cells.
5.Effect of Fingomod on the Inflammatory Response of Cerebral Ischemia-reperfusion Injury in Rats

Objective To investigate the effect of Fingomod(FTY720)on the cerebral ischemia-reperfusion injury in rats. Methods SPF rats(250 g~300 g)of 8 weeks old were divided into sham operation group,blank control group,group of Clopidogrel and group of FTY720.Rat model of ischemic stroke was made by tMCAO. The behavioral disorder scores were scored in different groups after different drug treatments. And observing the area of infarctus tissue by the TTC method and the nerve cells changed by HE method in different groups. The levels of inflammatory factors IL-1β and IL-6 in brain tissue of rats were changed by method of ELISA. The level of p-p38MAPK and NF-κB in brain tissue of rats were determined by Western-blot method. Results(1)FTY720 can alleviate the behavior disorder of rats(P<0.01);(2)FTY720 can reduce the area of infarctus tissue(P<0.01)and the injury of neurology cells in rats;(3)FTY720 can reduce the level of p-p38MAPK and NF-κB(P<0.01)in the ischemic brain tissue of rats;4,FTY720 can be reduced The level of IL-1β(P<0.01)and IL-6(P<0.01)in the ischemic brain tissue. Conclusion Fingomod(FTY720)has a protective effect on the cerebral ischemia-reperfusion rat model by decreasing the level of inflammatory response.
6. Mediating effect of township leaders' perceived organizational support on occupational stress and job satisfaction

OBJECTIVE: To investigate the job satisfying degree of township leaders and to explore the mediating effect of perceived organizational support on occupational stress and job satisfaction.METHODS: A total of 1 545 township leaders from Huludao City,Fengcheng City and Fushun City in Liaoning Province were selected as study subjects by cluster random sampling method.The questionnaire of Effort-Reword Imbalance,the Minnesota Satisfaction Questionnaire and the Perceived Organizational Support were used to investigate their occupational stress,job satisfaction and perceived organizational support.RESULTS: The total score of occupational stress in township leaders was(66.1 ± 6.8),and job satisfaction was(71.1 ± 13.1).The score of perceived organizational support was(42.2 ± 9.9).Occupational stress and perceived organizational support had direct prediction role on job satisfaction(P<0.01).The mediation effect of perceived organizational support between occupational stress and job satisfaction was 0.190,accounting for 48.3% of the total effect.CONCLUSION: Organizational support plays a part of mediating role on occupational stress and job satisfaction.
7.Protective mechanism of hydrogen on oxidative stress-induced retinal aging

AIM: To investigate the protective mechanism of hydrogen on retinal senescence induced by oxidative stress.
METHODS: Mice were randomly divided into three groups: control group, model group and treatment group. Animal models of retinal oxidative stress injury were established by injecting sodium iodate solution into mice caudal vein. Harvesting the mice retina, Western-blot was used to detect the level of proteins related to DNA damage response such as ATM, NF-κB, cyclin D1 and HMGB1 that associated with DNA repair.
RESULTS: SA-β-gal staining showed that the blue-green deposits in treatment group were reduced than that in model group. The expression of DNA damage reactive protein in treatment group ATM, cyclin D1, NF-κB(0.10±0.009, 0.32±0.01, 0.19±0.002)were significantly lower than those in the model groups(0.77±0.08, 0.70±0.02, 0.36±0.01), and the differences were statistically significant(all P<0.01). At the same time, the expression of DNA repair protein HMGB1 in treatment group(0.927±0.06)were notably higher than that in model group(0.383±0.07)and the difference was statistically significant(P<0.01).
CONCLUSION: H2 can attenuate senescence by inhibiting oxidative-stress induced DNA damage.
8. Construction of evaluation index system for emergency rescue ability of nursing staffs

OBJECTIVE: To construct an evaluation index system for the emergency rescue ability of nursing staffs.METHODS: An index system was established through literature analysis and Delphi method to conduct two rounds of investigations on 21 experts engaged in nursing work. The analytic hierarchy process was used to determine index weight at all levels. RESULTS: The positive coefficients of expert consultations of the two rounds of Delphi method were 95. 24% and100. 00% respectively. The authority levels of the experts were 0. 898 and 0. 908,and the coordination coefficient of expert opinion was 0. 255-0. 467( P < 0. 01). The emergency rescue ability evaluation index system for nursing staffs was successfully established,which included 4 first-level indicators,10 second-level indicators and 54 third-level indicators.CONCLUSION: The emergency rescue ability evaluation index system for nursing staffs is scientific and practical. It provides a strong basis for formulating the training plans for nursing staffs to participate in emergency rescue work.
9.Screening of host RNA binding proteins interacting with Nelson Bay reovirus σ NS by immunoprecipitation combined with mass spectrometry

Objective :
To screen the RNA binding proteins interacting with NBV nonstructural protein σNS in host
cells and to analyze their bioinformatics functions.
Methods :
In this study , the eukaryotic expression vector pEF⁃HA⁃MB⁃S3 of NBV σ NS was constructed and transfected into HEK293T cells after verification. After RNase A treatment , the obtained protein lysate was enriched by immunoprecipitation to enrich σNS binding proteins , identified and analyzed by LC⁃MS/MS mass spectrometry , and the properties and specific functions of proteins were discovered with the help of related Bioinformatics tools.
Results :
In this study , 32 candidate RNA binding proteins interacting with NBV σNS proteins were successfully screened , and the results of bioanalysis showed that these proteins were mainly located in cytoplasm and nucleus , and were mainly involved in biological processes such as cell metabolism , biological regulation , virus translation and transcription.
Conclusion
This study preliminarily analyzes the function of RNA binding proteins interacting with NBV σNS , which lays a foundation for further study on the mechanism of σNS protein in NBV life cycle.
10. Chemical constituents from Vitex negundo var. heterophylla

Objective: To study the chemical constituents in the twigs of Vitex negundo var. heterophylla. Methods: A variety of silica gel column chromatography, Sephadex LH-20 gel column chromatography, and HPLC methods were used for the separation and purification of chemical composition. Their structures were identified on the basis of physicochemical property and spectral data. Results: Eight compounds were obtained and identified as apigenin-6-C-β-D-glucopyranosyl 8-C-α-L-arabinopyranoside (1), isoorientin (2), luteolin-6-C-α-L-arabinopyranosyl-8-C-β-D-glucopyranoside (3), luteolin-6,8-di-C-α-L-arabinopyranoside (4), apigenin-6-α-L- arabinopyranosyl-8-C-β-D-glucopyranoside (5), luteolin-6-C-α-L-arabinopyranosyl 8-C-β-L-arabinopyranoside (6), luteolin-6-C-β-L- arabinopyranosyl-8-C-β-D-glucopyranoside (7), and luteolin-7-O-β-D-glucopyranoside (8). Conclusion: Compounds 1 and 3-7 are first isolated from the plants of Vitex Linn.
