1.Effect of Speed of Perioperative Fluid Infusion on Rate of Platelet Aggregation during Percutaneous Coronary Intervention

Objective To investigate the effect of different speed of perioperative fluid infusion on rate of platelet aggregation during percutaneous coronary intervention. Methods 60 patients accepted percutaneous coronary intervention (PCI) were divided into 2 groups. The control group (n=30) accepted fluid infusion in speed of 1.5~2 ml/kg·h, while the trial group (n=30) with the speed of 6~9 ml//kg·h after coronary arteriongraphy. Their blood pressure, heart beat, rate of platelet aggregation and osmotic pressure of plasma were recorded. Results It was not significantly different in rate of platelet aggregation before and 4 h after operation in the control group (P>0.05), and it was significantly lower 4 h and 1 d after operation in the trial group (P<0.05). Conclusion Fast fluid infusion perioperatively can lower the rate of platelet aggregation, which may improve the success of PCI and reduce the acute thrombosis
2.Purification and Identification of human Myocardial Troponin T

Objective To obtain high pure myocardiac Troponin T from myocardiac tissue. Methods After the myocardiac tissues were homogenized and purified with high salt solution, the Sephacryl S-300HR column chromatography was employed to separate the cTnT from cardiac proteins, and identified them with SDS-PAGE and Western blot. Results The crude cTn was purified by column chromatography, and there appeared three peaks. The second peak was single component, which could react with mouse anti-human Troponin T antibody, and its molecular weight was about 34kD. Conclusions In this experiment, high pure cTnT was obtained, which could pave a way for further research.
3.The clinical study on the treatment of peptic ulcer with blood clot and drug intervention in the department of internal medicine

Objective To study the clinical effect of drug intervention on the treatment of peptic ulcer with blood clots in the Department of internal medicine.Methods From 62 cases of peptic ulcer adherent blood clot were randomly divided into study group and control group according to the random number table,31 cases in each group,in each group.The control group was treated with esomeprazole infusion and subsequent oral treatment.The study group was given endoscopic hemostasis and subsequent oral esomeprazole treatment.Compare the two groups of curative effect,treatment profile and treatment before and after the study of the changes in the situation.Results The total effective rate of the study group was 90.32%,which was significantly better than that of the control group(P<0.05),which was significantly better than that of the control group 70.97%.Research group of rebleeding rate and transfer rate of surgery was significantly lower than that of control group(P<0.05),the study group,the time of hemostasis,the time of hospitalization significantly faster than that of the control group(P<0.05),study group medical expenses are significantly less than the control group(P<0.05).The two groups before treatment Blatchford score,Rockall score,SF-36 score no significant difference,after treatment in the two groups of the three scores were compared with those before treatment significantly optimized(P<0.05)study group the score optimization was significantly better than the control group(P<0.05).Conclusion Peptic ulcer adhesion blood clot give endoscopic therapy can greatly enhance the efficacy,reduce bleeding and transfer the risk of surgery,more effectively improve the acute upper digestive tract bleeding symptoms and signs,improve the life quality of the patients.
4.Screening and characterization of human scFv antibodies against nasopharyngeal carcinoma

Objective To screen the anti-nasopharyngeal carcinoma scFv from a human anti-nasopharyngeal carcinoma single-chain phage antibody library, and identify its characteristics. Methods The single-chain phage antibody library was subjected to three rounds of positive and negative cell panning and enrichment, and then it was selected by ELISA. The binding specificity of phage antibodies with naso-pharyngeal carcinoma cells was confirmed by immunohistochemistry. Results After panning, enrichment and testing by ELISA, 3 phage an-tibody clones reacting with CNE2 more strongly than HUVEC and NP69 were picked out from 4212 clones. One clone, HNSAO33, was fur-ther analyzed after DNA sequencing. The results of immunohistochemistry with cultured cells were similar to those of ELISA. HNSAO33 spe-cifically reacted to nasopharyngeal carcinoma cells in most human nasopharyngeal carcinoma tissue sections except a few human normal naso-pharyngeal tissue sections. The distinction of positive rates was of a great statistical significance. Conclusion ELISA and immunohisto-chemistry results confirmed HNSAO33 specifically bind with nasopharyngeal carcinoma cells. The seFv fragment against nasopharyngeal carci-noma may be further developed and applied in clinical diagnosis and therapy of nasopharyngeal carcinoma.
5.Expression of scFv SA3 against hepatoma fused with enhanced green fluorecsent protein and its targeted ability in vivo

Objective To express and purify the human scFv antibody,SA3,against the hepatoma fused to enhanced green fluorecsent protein,and to observe the targeted capacity of fusion protein EGFP-SA3 in vivo.Methods SA3 and EGFP genes were cloned into plasmid pET-25b( + )to construct the recombinant plasmid EGFP-SA3/pET-25b ( + ),followed by DNA sequencing.Then it was transformed into E.coli BL21 ( DE3 ) and induced for fusion expression of EGFP-SA3with IPTG.The expressed fusion protein EGFP-SA3 was purified and detected with SDS-PAGE.HepG2 cells were incubated with the fusion protein EGFP-SA3 in vitro,and the binding bioactivity was observed under the fluorecsent microscope.Further more,we injected the EGFP-SA3 by caudal vein into nude mice planted by hepatoma and observed the whole body fluorescence image of EGFP.Results SA3 and EGFP genes were successfully cloned into pET-25b( + ),which was confirmed by restriction enzyme Nco I-Xho I or Nco I-Eco RI.A band migrated at the position 750 bp,same to EGFP gene,emerged when recombinant plasmid was digested by restriction enzyme Nco I-Eco RI.Similarly,a band,about 1 500 bp,emerged when digested by Nco I-Xho I.The open-reading frame was confirmed by DNA sequencing.Fusion protein EGFP-SA3 was expressed as inclusion body.After purification and refolding,the result of immunofluorecsence detection verified that EGFP-SA3could specifically bind to HepG2 cells and maximum tumor penetration was at 24 h after the injection.Conclusion The purified fusion protein EGFP-SA3 has strong binding capacity to HepG2cells,indicating the scFv SA3 has a potential value as a targeting molecule for diagnosis and targeted therapy for liver cancer.
6.Early diagnosis value of procalcitonin in severe brain damage combined with pulmonary infection

Objective To evaluate the early diagnosis value of procalcitonin (PCT) in severe brain damage combined with pul‐monary infection .Methods The brain injury patients in the hospital from January to October 2014 were enrolled in the study and divided into infectious group whose infection had occurred within 5 days after admitting to hospital and non‐infectious group who had not suffered from infection .The blood samples of the patients were collected within 2 h and 3 days after admitting to hospital and detected for PCT concentration .The Early diagnosis value of PCT in brain damage combined with pulmonary infection was e‐valuated and compared with white blood cells (WBC) ,neutrophile granulocyte(N)and hypersensitive C‐reactive protein(hs‐CRP) . Results The incidence of pulmonary infection within 5 days of severe brain injury was 22 .9% (41/179) .There were statistically differences of PCT ,WBC ,N and hs‐CRP between infectious group and non‐infectious group(P< 0 .05) .The areas under curve (AUC) of PCT ,WBC ,N and hs‐CRP were 0 .83 ,0 .80 ,0 .78 and 0 .82 respectively .The combination of PCT+WBC+ hs‐CRP had the highest diagnostic value since its AUC was 0 .87 .PCT had a satisfied diagnostic veracity since it had good sensitivity ,specificity and positive predictive value in the diagnosis of brain damage combined with pulmonary infection .Conclusion PCT could be an ear‐ly diagnosis indicator in severe brain damage combined with pulmonary infection ,and the diagnostic veracity is higher when com‐bined with WBC and hs‐CRP .An antimicrobial treatment is recommended when PCT concentration of brain damage patient rises , especially when combined with WBC and hs‐CRP concentration elevating .
7.Cluster analysis of changes in physique among Japanese children and adolescents during 1900-2015

Objective:
To analyze changes in physique change among children and adolescents in Japan from 1900 to 2015, and to provide the scientific basis for national constitution development strategy in China.
Methods:
Data on height and weight of Japanese children and adolescents aged 6-17 years provided by the Ministry of Foreign Affairs. Cluster analysis was performed to analyze the data.
Results:
The 5 time periods of highest physique growth rate of Japanese boy and girls were 1948-1950, 1950-1955, 1955-1960, 1960-1965, 1965-1970, and the 5 time periods of lowest growth rate were 1939-1948, 2005-2010, 2000-2005, 1900-1905, 2010-2015, 1995-2000. Physique growth of Japanese children and adolescents showed a relatively constant increasing trend before World War II and the late 20th century, and a rapid increasing trend during 20 years after World War II and a decreasing trend during and short period after World War II. In the cluster analysis, the lower half of male students fell into a single time period (1939-1948) showing the highest rate of decline.
Conclusion
Physical changes of Japanese children and adolescents are closely related to the socio-economic environment, and rapid increasing trend is observed during 20 years after World War II. Based on experiences from Japan, rapid socio-economic growth in China provides an important opportunity to promote physical development of children and adolescents. Cautions should be paid to the risk of overweight and obesity accompanied by economic growth.
8.Effect of gamma-aminobutyric acid on malignant phenotypes of hepatocellular carcinoma cell line HepG-2.

OBJECTIVE:
To determine the effect of gamma-aminobutyric acid(GABA) on proliferation and malignant phenotypes of hepatocellular carcinoma cell line HepG-2.
METHODS:
HepG-2 cells were cultured by routine method, and then treated with different concentrations of GABA. The proliferation of HepG-2 cells was measured through MTT, doubling time and cell cycles by flow cytometry. The malignant phenotypes were investigated by soft agar colony formation assay.
RESULTS:
Compared with the control group, GABA efficiently stimulated the proliferation of HepG-2 cells in a dose-dependent manner and affected the distribution of cell cycles of HepG-2 cells. The doubling time of the control group and the GABA-treated group were 39.0, 30.6, 30.0, 27.3, 26.6, and 38.2 h, respectively. The colony formation rates were 3.2%, 4.2%, 5.4%, 6.6%,6.5%, and 3.5%, respectively. Tumorigenicity testing showed that the average weights of tumor was 1.382 g, and 0.285 g for the 2 groups. The difference between the control group and the GABA-treated groups was significant (P<0.01).
CONCLUSION
GABA can enhance the proliferation and malignant phenotypes of HepG-2 cells.

9.Nutritional status and energy metabolism in amyotrophic lateral sclerosis patients

Amyotrophic lateral sclerosis (ALS) is the most common motor neuron disease characterized by progressive muscle wasting and weakness, bulbar paralysis and pyramidal sign. Due to rapid deterioration in several years, patients always die from respiratory failure. Until now, no effective treatment has been discovered. Nutrition and metabolism are fundamental in all human existence, which may also correlate with the development and progression of ALS. Early assessment of nutritional and metabolic status and individualized dietary intervention may prolong life-span of patients with ALS. Therefore, the article reviewed the nutritional status and energy metabolism in ALS patients, and sought proper dietary management patterns.
10.The role of myeloid-derived suppressor cells in chronic osteomyelitis

Inhibitory cells derived from bone marrow are a kind of inhibitory cells derived from bone marrow.These cells are not only related to tumor growth,but also participate in the inflammatory immune process.Therefore,we established a rat model of chronic osteomyelitis,and used gemcitabine to inhibit the cell growth ratio of MDSCs.We detected the ratio of MDSCs in bone marrow and spleen of rats by flow cytometry and immunofluorescence,detected the changes of inflammatory factors in peripheral blood by ELISA,and analyzed the inflammatory factors(TNF-α,PCT,IL-4,IL-10,IL-11)in peripheral blood of normal rats,osteomyelitis rats and rats after gemcitabine inhibition.The results showed that the proportion of MDSCs cells in bone marrow and spleen of osteomyelitis model rats was increased,but it was significantly decreased in gemcitabine group(P<0.05).Levels of inflammatory factors(TNF-α,PCT,IL-4,IL-10,IL-17,IFN-γ,TGF-β)were positively correlated with the change of MDSCs cell proportion(P<0.05).From the results,it can be inferred that the change of the proportion of MDSCs cells in rat osteomyelitis is positively related to the inflammatory factors,and gemcitabine can reduce inflammatory factors by inhibiting MDSCs.
